| 1 | Improvement of oxytetracycline production mediated via cooperation of resistance genes in Streptomyces rimosus显示文摘Increasing the self-resistance levels of Streptomyces is an effective strategy to improve the production of antibiotics.To increase the oxytetracycline(OTC) production in Streptomyces rimosus,we investigated the cooperative effect of three co-overexpressing OTC resistance genes:one gene encodes a ribosomal protection protein(otrA) and the other two express efflux proteins(otrB and otrC).Results indicated that combinational overexpression of otrA,otrB,and otrC(MKABC) exerted a synergetic effect.OTC production increased by 179%in the recombinant strain compared with that of the wild-type strain M4018.The resistance level to OTC was increased by approximately two-fold relative to the parental strain,thereby indicating that applying the cooperative effect of self-resistance genes is useful to improve OTC production.Furthermore,the previously identified cluster-situated activator OtcR was overexpressed in MKABC in constructing the recombinant strain MKRABC;such strain can produce OTC of approximately7.49 g L^((-1)),which represents an increase of 19%in comparison with that of the OtcR-overexpressing strain alone.Our work showed that the cooperative overexpression of self-resistance genes is a promising strategy to enhance the antibiotics production in Streptomyces. | Shouliang Yin Xuefeng Wang Mingxin Shi Fang Yuan Huizhuan Wang Xiaole Jia Fang Yuan Jinliang Sun Tiejun Liu Keqian Yang Yuxiu Zhang Keqiang Fan Zilong Li | 2017 | Science China(Life Sciences)2017,60,9: | 10 |
| 2 | The localization of type 2 diabetes susceptibility gene loci in northern Chinese Han families显示文摘We conducted a genome-wide scan, in which 358 well distributed fluorescent dye-labeled microsatellite marker sets were applied in 32 Chinese Han type 2 diabetes families from Northern China to search for the susceptibility gene loci. The data collected from screening all the chromosomes of genome were genotyped by using genescan and genotyping software, then, parametric and non-parametric multipoint test, and affected sib-pair analysis as well, were used to analyze the data. We identified some susceptibility gene loci residing in chromosomes 1,12,18,20, respectively, or precisely, located around D1S214, D1S207, D1S218, D1S235, D12S336, D18S61 and D20S118. | 4 | Effects of Sini San used alone and in combination with fluoxetine on central and peripheral 5-HT levels in a rat model of depression显示文摘OBJECTIVE: To investigate the effects of Sini San and fluoxetine on the levels of central and peripheral 5-HT in a rat model of depression, and provide new insight into the treatment of depression with integrated Chinese-Western Medicine.METHODS: A rat model of depression was established by chronic mild stress(CMS). Model rats received either Sini San, fluoxetine, a combination of the two drugs, or no drug treatment. Healthy naive rats were used as controls. Open field and sucrose preference tests were used to assess depression-like behavior. ELISA and immunohistochemistry were used to determine central and peripheral levels of 5-HT.RESULTS: In the group with no drug treatment,central 5-HT expression decreased while peripheral5-HT concentrations increased as CMS continued.Four weeks after CMS, Sini San alone was less effective in reducing depression-like behavior than fluoxetine alone or in combination with Sini San,but combined use was more effective than fluoxetine alone. Eight weeks after CMS, Sini San alone or in combination with fluoxetine was more effective in reducing depression-like behavior than fluoxetine alone. Furthermore Sini San and fluoxetine used alone or in combination notably increased central 5-HT expression and decreased peripheral 5-HT levels in the rat model.CONCLUSION:The results of the present study indicate that there is a synergistic action between the two medicines in the treatment of depression. Sini San exhibited a relatively long lag before its effects were observed; however, by eight weeks the Traditional Chinese Medicine appeared at least as effective as fluoxetine. | 7 | High-throughput RNA sequencing reveals the anti-inflammatory mechanism of baicalin on Propionibacterium acnes-induced acne in rabbits显示文摘Objective:Propionibacterium acnes(P.acnes)plays an important role in the development of acne,an inflammatory skin disease with a high-incidence.In this study,we used high-throughput RNA sequencing(RNA-seq)to reveal the anti-inflammatory mechanism of baicalin on P.acnes-induced acne in rabbits.Methods:The Kligman method was used to induce acne in the ears of New Zealand rabbits.The effect of baicalin on the acne model was evaluated by the number of acne lesions and hematoxylin and eosin(H&E)staining of acne tissues.Enzyme-linked immunoabsorbent assay was used to measure the protein expression levels of tumor necrosis factor a(TNFA),interleukin-1 b(IL1B),IL6,and IL8 in the serum of rabbits.RNA-seq was performed to investigate the mechanism of anti-inflammatory activities of baicalin on acne.Immunohistochemical analysis and Western blot were used to validate the expression levels of related proteins in acne tissues.Results:Baicalin treatment significantly reduced the number of acne lesions and lesions of the ear as well as levels of serum inflammatory cytokines.RNA-seq data showed that baicalin treatment globally suppressed inflammation,especially the TNF signaling pathway and Staphylococcus aureus infection pathway,in the rabbit acne model.Conclusion: Baicalin effectively ameliorates P. acnes-induced acne in rabbits by suppressingthe inflammatory response in rabbits. | Xingzhe Yang Guangrui Huang Leiming You Honghao Sheng Yuxiu Sun Yuyin Feng Qingyi Lu Anlong Xu | 2019 | Journal of Traditional Chinese Medical Sciences2019,6,3: | 2 |

| 15 | Metal confined in metal-organic framework-based mixed matrix membranes for efficient butadiene recognition separation显示文摘The efficient separation of butadiene(1,3-C_(4)H_(6))from C_(4)hydrocarbons is a critical step in petrochemical processes.However,the traditional cryogenic distillation suffers from energy-intensity and serious environmental stress,necessitating the development of alternative technologies for efficient 1,3-C_(4)H_(6)separation.Herein,a 1,3-C_(4)H_(6)recognition mixed matrix membrane is reported via incorporating metal copper encapsulated a metal-organic framework(CuBTC@Cu)into elastic poly(dimethylsiloxane)(PDMS).The resulting CuBTC@Cu/PDMS membrane can efficient separate 1,3-C_(4)H_(6)from various C_(4)hydrocarbons including 1,3-C_(4)H_(6)/n-C_(4)H_(8),1,3-C_(4)H_(6)/iso-C_(4)H_(8),1,3-C_(4)H_(6)/n-C_(4)H_(10)and 1,3-C_(4)H_(6)/iso-C_(4)H_(10),yielding superior selectivity of 5.11,6.35,4.78,and 10.30,respectively,with 1,3-C_(4)H_(6)permeability of 53240 Barrer.Notably,the appropriateπ-complexation interaction between butadiene molecules and CuBTC@Cu as well as suitable transmission channel size enable the membrane only permeable to 1,3-C_(4)H_(6)and block the permeation of other C_(4)hydrocarbons,showing a unique 1,3-C_(4)H_(6)recognition behavior in membrane separation.The concept of affinity-relying separation combining molecular sieving would open a new direction for designing gas membranes for efficient light hydrocarbon separations. | Xixi Cen Yuxiu Sun Caijiao Yu Zhihua Qiao Chongli Zhong | 2023 | Aggregate2023,4,5: | 0 |
| 16 | Predicting of Covalent Organic Frameworks for Membrane-based Isobutene/1,3-Butadiene Separation:Combining Molecular Simulation and Machine Learning显示文摘Efficient separation of C4 olefins is of critical importance and a challenging task in petrochemical industry. Covalent organic frameworks(COFs) could be used as promising candidates for membrane-based isobutene/1,3-butadiene(i-C4H8/C4H6) separation. Owing to large amounts of COFs appearing, however, the rapid prediction of optimal COFs is imperative before experimental efforts. In this work, we combine molecular simulation and machine learning to study COF membranes for efficient isolation of i-C4H8 over C4H6. Using molecular simulation, four potential COF membranes, which possess both high membrane performance score (MPS) value and moderate membrane selectivity were screened out and the mechanism of membrane separation further revealed is an adsorption dominated process. Further, random forest(RF) model with high prediction accuracy(R2>0.84) was obtained and used for elucidating key factors in controlling the membrane selectivity and i-C4H8 permeability. Ultimately, the optimal COF features were obtained through structure-performance relationship study. | 18 | Discrimination of TCM constitutions by biochemical and routine urine indexes显示文摘Objective:To investigate whether the specific traditional Chinese medicine(TCM)constitution of individuals can be defined by certain biological indexes instead of answering the questionnaire,and to explore the possibility of discriminating nine TCM constitutions from each other simultaneously using biological indexes.Methods:Blood and urine samples from 152 individuals with nine TCM constitutions were collected,and the related biological indexes were analyzed combining ANOVA,multiple comparison,discriminant analysis,and support vector machine.Results:We found that 4 out of 24 blood routine indexes,7 out of 10 urine routine indexes,and 12 out of 32 biochemical indexes showed differences among the constitutions.High-sensitivity C-reactive protein,apolipoprotein A1,and alkaline phosphatase were potential candidates for screening out individuals with unbalanced constitutions.Combining uric acid,high-density lipoprotein,apolipoprotein A1,creatine kinase,total protein,aspartate aminotransferase,total bile acid,dehydrogenase,sodium,and calcium levels had the potential to directly distinguish the nine TCM constitutions from each other.Among these indexes,the highest ratio of discriminant analysis between two constitutions was 95.5%,while the lowest was 66.1%.Conclusion:Our results suggest that some biochemical and urine indexes are related to various TCM constitutions,and thus they have the potential to be used for TCM constitution classification. | Xiaoling Liu Pengfei Zhao Jianhua Zhen Shen Zhang Hesong Wang Yuxiu Sun Wei Wang Tingjian Wang Kaiwen Hu Guangrui Huang | 2022 | Journal of Traditional Chinese Medical Sciences2022,9,2: | 0 |
