| 2 | Chinese expert consensus statement on the diagnosis and treatment of fulminant myocarditis显示文摘Fulminant myocarditis is primarily caused by infection with any number of a variety of viruses. It arises quickly, progresses rapidly, and may lead to severe heart failure or circulatory failure presenting as rapid-onset hypotension and cardiogenic shock,with mortality rates as high as 50%–70%. Most importantly, there are no treatment options, guidelines or an expert consensus statement. Here, we provide the first expert consensus, the Chinese Society of Cardiology Expert Consensus Statement on the Diagnosis and Treatment of Fulminant Myocarditis, based on data from our recent clinical trial(NCT03268642). In this statement, we describe the clinical features and diagnostic criteria of fulminant myocarditis, and importantly, for the first time,we describe a new treatment regimen termed life support-based comprehensive treatment regimen. The core content of this treatment regimen includes(i) mechanical life support(applications of mechanical respirators and circulatory support systems,including intraaortic balloon pump and extracorporeal membrane oxygenation),(ii) immunological modulation by using sufficient doses of glucocorticoid, immunoglobulin and(iii) antiviral reagents using neuraminidase inhibitor. | 4 | Satellite-Based Estimation of Daily Average Net Radiation under Clear-Sky Conditions显示文摘Daily average net radiation(DANR) is an important variable for estimating evapotranspiration from satellite data at regional scales, and is used for atmospheric and hydrologic modeling, as well as ecosystem management. A scheme is proposed to estimate the DANR over large heterogeneous areas under clear-sky conditions using only remotely sensed data.The method was designed to overcome the dependence of DANR estimates on ground data, and to map spatially consistent and reasonably distributed DANR, by using various land and atmospheric data products retrieved from MODIS(Moderate Resolution Imaging Spectroradiometer) data. An improved sinusoidal model was used to retrieve the diurnal variations of downward shortwave radiation using a single instantaneous value from satellites. The downward shortwave component of DANR was directly obtained from this instantaneous value, and the upward shortwave component was estimated using satellite-derived albedo products. Four observations of air temperature from MOD07 L2 and MYD07 L2 data products were used to derive the downward longwave component of DANR, while the upward longwave component was estimated using the land surface temperature(LST) and the surface emissivity from MOD11 L2. Compared to in situ observations at the cropland and grassland sites located in Tongyu, northern China, the root mean square error(RMSE) of DANR estimated for both sites under clear-sky conditions was 37 W m-2and 40 W m-2, respectively. The errors in estimation of DANR were comparable to those from previous satellite-based methods. | 5 | A targeted sequencing approach to find novel pathogenic genes associated with sporadic aortic dissection显示文摘Aortic dissection(AD) is a heterogeneous genetic disease of the aorta with high mortality and poor prognosis. However, only few genetic causes of AD have been explored till date. After conducting a broad literature review focused on identifying potential pathogenic pathways, we designed a panel containing 152 AD-associated genes to conduct massively parallel targeted nextgeneration sequencing of 702 sporadic aortic dissection patients and 163 matched healthy controls. After validation by Sanger sequencing, we identified 21 definitely pathogenic and 635 likely pathogenic variants in 61.25%(430/702) of patients. In these patients, 34.88%(150/430) harbored more than one variant that was either definitely or likely to be pathogenic. Among the candidate genes, we identified 546 likely pathogenic variants in 47.72%(335/702) of patients. Importantly, we identified 94 lossof-function(LOF) variants in 45 genes in AD patients, but only five LOF variants in the controls(P=1.34×10^(-4)). With a burden test, we highlighted RNF213 as an important new gene for AD pathogenesis. We also performed transcriptome sequencing of human aorta tissues to evaluate the expression levels of these newly identified genes. Our study has compiled a comprehensive genetic map of sporadic AD in the Han Chinese population. | 6 | Variants of genes encoding collagens and matrix metalloproteinase system increased the risk of aortic dissection显示文摘Aortic dissection(AD) is a devastating,heterogeneous condition of aorta.The homeostasis between collagens and matrix metalloproteases(MMPs)/tissue inhibitors of MMPs(TIMPs) system in the extracellular matrix plays an important role for structure and functions of aorta.However,our knowledge on association between variants of genes in this system and pathogenesis of AD is very limited.We analyzed all yet known coding human genes of collagens(45 genes),MMPs/TIMPs(27genes) in 702 sporadic AD patients and in 163 matched healthy controls,by using massively targeted next-generation and Sanger sequencing.To define the pathogenesis of potential disease-causing candidate genes,we performed transcriptome sequencing and pedigree co-segregation analysis in some genes and generated Col5a2 knockout rats.We identified 257 pathogenic or likely pathogenic variants which involved 88.89%(64/72) genes in collagens-MMPs/TIMPs system and accounted for 31.05%(218/702) sporadic AD patients.In them,84.86%patients(185/218) carried one variant,12.84%two variants and 2.30%more than two variants.Importantly,we identified 52 novel probably pathogenic loss-of-function(LOF) variants(20 nonsense,16 frameshift,14 splice sites,one stop-loss,one initiation codon) in 11.06%(50/452) AD patients,which were absent in 163controls(P=2.5×10^(-5)).Transcriptome sequencing revealed that identified variants induced dyshomeostasis in expression of collagens-TIMPs/MMPs systems.The Col5α2^(-/-) rats manifested growth retardation and aortic dysplasia.Our study provides a first comprehensive map of genetic alterations in collagens-MMPs/TIMPs system in sporadic AD patients and suggests that variants of these genes contribute largely to AD pathogenesis. | Zongzhe Li Chengming Zhou Lun Tan Peng Chen Yanyan Cao Chenze Li Xianqing Li Jiangtao Yan Hesong Zeng Dao-Wu Wang Dao-Wen Wang | 2017 | Science China(Life Sciences)2017,60,1: | 4 |

| 8 | Hierarchical responses of soil organic and inorganic carbon dynamics to soil acidification in a dryland agroecosystem,China显示文摘Soil acidification is a major global issue of sustainable development for ecosystems. The increasing soil acidity induced by excessive nitrogen(N) fertilization in farmlands has profoundly impacted the soil carbon dynamics. However, the way in which changes in soil p H regulating the soil carbon dynamics in a deep soil profile is still not well elucidated. In this study, through a 12-year field N fertilization experiment with three N fertilizer treatments(0, 120, and 240 kg N/(hm^2·a)) in a dryland agroecosystem of China, we explored the soil p H changes over a soil profile up to a depth of 200 cm and determined the responses of soil organic carbon(SOC) and soil inorganic carbon(SIC) to the changed soil p H. Using a generalized additive model, we identified the soil depth intervals with the most powerful statistical relationships between changes in soil p H and soil carbon dynamics. Hierarchical responses of SOC and SIC dynamics to soil acidification were found. The results indicate that the changes in soil p H explained the SOC dynamics well by using a non-linear relationship at the soil depth of 0–80 cm(P=0.006), whereas the changes in soil p H were significantly linearly correlated with SIC dynamics at the 100–180 cm soil depth(P=0.015). After a long-term N fertilization in the experimental field, the soil p H value decreased in all three N fertilizer treatments. Furthermore, the declines in soil p H in the deep soil layer(100–200 cm) were significantly greater(P=0.035) than those in the upper soil layer(0–80 cm). These results indicate that soil acidification in the upper soil layer can transfer excess protons to the deep soil layer, and subsequently, the structural heterogeneous responses of SOC and SIC to soil acidification were identified because of different buffer capacities for the SOC and SIC. | 10 | Modelling the formation and dissolution behavior of alumina agglomerate in the cryolite显示文摘The presence of alumina agglomerates seriously affects the current efficiency of the aluminum electrolysis process.Clarify the dynamic dissolution process of agglomerates is essential to improve the current efficiency of aluminum electrolysis.A mathematical model is proposed to describe the different phenomena from the formation until complete dissolution of agglomerates.Considering permeation and solidification processes of cryolite,a semi-analytical mathematical model is developed to formulate the formation,melting and dissolution processes of agglomerates,and the time duration for each stage is deduced.Porosity and heat mass transfer of agglomerates are explored based on the packing theory and mechanism of heat mass transfer in wet porous media.Dimensionless approach is applied to investigate the main factors affecting the dissolution stages and porosity of agglomerates.The results show that the superheat has a great influence on the formation and melting stage,the diameter of agglomerates can reach 14.93 mm for 200 particles agglomerated.The density decreases with the increase of agglomerated particle number,which varies in the range of 2.27-2.28 g/cm^(3).The average dissolution rate of agglomerates is about 1.83×10^(-5)-2.95×10^(-5) kg/s within the range of alumina concentration in this study. | Jiaqi Wang Mao Li Benjun Cheng Hesong Li | 2024 | Particuology2024,86,3: | 0 |
| 11 | Development of a Prognostic Index for Kidney Cancer Based on Hypoxia Landscape显示文摘Objective:Kidney cancer is a group of cancers occurred in the kidney.Hypoxia is a condition characterized by insuff-cient oxygen supply in the body or specifie organs.It has been proven to play an essential role in the pathogenesis and development of various cancer.However,the roles and mechanisms of hypoxia in kidney cancer have not been investigated clearly.In this study,we comprehensively analyzed the roles of hypoxia in kidney cancer.Methods:The RNA-Seq data of kidney cancer were downloaded from TCGA dataset.The survival of patients with diffentially expressed hypoxia-related genes was analyzed with the survival package.The prognostic value of hypoxia-related genes was evaluated with univariate Cox regression analysis.The correlation of hypoxia-related genes to immune ells infiltration and gene mutation in kidney cancer was assessed.We identifed 7 hypoxia-related genes of kidney cancer,with which we developed a hypoxia-related genes-based prognostic index using multivariate Cox regression analysis to establish the prog-nostic model.Results:The high-risk group showed lower survival compared with the low-risk group.HRGPI was an independent pre-dictor of kidney cancer,and it was associated with malignant stages.Hypoxia-related genes were correlated to activation of inflammatory pathways,infiltration of inflammatory ellls,and expression of inhibitory immune checkpoints,indicat ing that hypoxia was related to im-mune response in kidney cancer.Lastly,we found that hypoxia was related to missense mutation in kidney cancer.Conclusion:Hy-poxia is a risk factor of kidney cancer.It can be regarded as the prognostic indicator and therapeutic target for the treatment of kidney cancer in the future. | Wang Hesong Feng Yanyan Wang Ting Feng Yuyin Jiang Haixu Wang Tingjian Huang Guangrui | 2021 | 肿瘤预防与治疗2021,34,11: | 0 |
| 12 | Discrimination of TCM constitutions by biochemical and routine urine indexes显示文摘Objective:To investigate whether the specific traditional Chinese medicine(TCM)constitution of individuals can be defined by certain biological indexes instead of answering the questionnaire,and to explore the possibility of discriminating nine TCM constitutions from each other simultaneously using biological indexes.Methods:Blood and urine samples from 152 individuals with nine TCM constitutions were collected,and the related biological indexes were analyzed combining ANOVA,multiple comparison,discriminant analysis,and support vector machine.Results:We found that 4 out of 24 blood routine indexes,7 out of 10 urine routine indexes,and 12 out of 32 biochemical indexes showed differences among the constitutions.High-sensitivity C-reactive protein,apolipoprotein A1,and alkaline phosphatase were potential candidates for screening out individuals with unbalanced constitutions.Combining uric acid,high-density lipoprotein,apolipoprotein A1,creatine kinase,total protein,aspartate aminotransferase,total bile acid,dehydrogenase,sodium,and calcium levels had the potential to directly distinguish the nine TCM constitutions from each other.Among these indexes,the highest ratio of discriminant analysis between two constitutions was 95.5%,while the lowest was 66.1%.Conclusion:Our results suggest that some biochemical and urine indexes are related to various TCM constitutions,and thus they have the potential to be used for TCM constitution classification. | Xiaoling Liu Pengfei Zhao Jianhua Zhen Shen Zhang Hesong Wang Yuxiu Sun Wei Wang Tingjian Wang Kaiwen Hu Guangrui Huang | 2022 | Journal of Traditional Chinese Medical Sciences2022,9,2: | 0 |

| 14 | In utero delivery of mRNA to the heart,diaphragm and muscle with lipid nanoparticles显示文摘Nanoparticle-based drug delivery systems have the potential to revolutionize medicine,but their low vascular permeability and rapid clearance by phagocytic cells have limited their medical impact.Nanoparticles delivered at the in utero stage can overcome these key limitations due to the high rate of angiogenesis and cell division in fetal tissue and the under-developed immune system.However,very little is known about nanoparticle drug delivery at the fetal stage of development.In this report,using Ai9 CRE reporter mice,we demonstrate that lipid nanoparticle(LNP)mRNA complexes can deliver mRNA in utero,and can access and transfect major organs,such as the heart,the liver,kidneys,lungs and the gastrointestinal tract with remarkable efficiency and low toxicity.In addition,at 4 weeks after birth,we demonstrate that 50.99±5.05%,36.62±3.42%and 23.7±3.21%of myofiber in the diaphragm,heart and skeletal muscle,respectively,were transfected.Finally,we show here that Cas9 mRNA and sgRNA complexed to LNPs were able to edit the fetal organs in utero.These experiments demonstrate the possibility of non-viral delivery of mRNA to organs outside of the liver in utero,which provides a promising strategy for treating a wide variety of devastating diseases before birth. | Kewa Gao Jie Li Hengyue Song Hesong Han Yongheng Wang Boyan Yin Diana L.Farmer Niren Murthy Aijun Wang | 2023 | Bioactive Materials2023,,7: | 0 |
