| 1 | Cranial irradiation impairs intrinsic excitability and synaptic plasticity of hippocampal CA1 pyramidal neurons with implications for cognitive function显示文摘Radiation therapy is a standard treatment for head and neck tumors.However,patients often exhibit cognitive impairments following radiation therapy.Previous studies have revealed that hippocampal dysfunction,specifically abnormal hippocampal neurogenesis or neuroinflammation,plays a key role in radiation-induced cognitive impairment.However,the long-term effects of radiation with respect to the electrophysiological adaptation of hippocampal neurons remain poorly characterized.We found that mice exhibited cognitive impairment 3 months after undergoing 10 minutes of cranial irradiation at a dose rate of 3 Gy/min.Furthermore,we observed a remarkable reduction in spike firing and excitatory synaptic input,as well as greatly enhanced inhibitory inputs,in hippocampal CA1 pyramidal neurons.Corresponding to the electrophysiological adaptation,we found reduced expression of synaptic plasticity marker VGLUT1 and increased expression of VGAT.Furthermore,in irradiated mice,long-term potentiation in the hippocampus was weakened and GluR1 expression was inhibited.These findings suggest that radiation can impair intrinsic excitability and synaptic plasticity in hippocampal CA1 pyramidal neurons. | Min-Yi Wu Wen-Jun Zou Pei Yu Yuhua Yang Shao-Jian Li Qiang Liu Jiatian Xie Si-Qi Chen Wei-Jye Lin Yamei Tang | 2022 | Neural Regeneration Research2022,17,10: | 5 |
| 2 | Characterization and potential diagnostic application of monoclonal antibodies specific to rabies virus显示文摘Objective:Rabies is invariably a fatal encephalomyelitis that is considered to be a serious public health problem.It is necessary to develop standard rabies virus diagnostic tools,especially for diagnosing the strains prevalent in China.Methods:Monoclonal antibodies(MAbs)specific to rabies virus were produced and characterized by enzyme linked immunosorbent assay(ELISA),isotyping,affinity assay,immunofluorescence assay(IFA),and immunocytochemistry.The MAb,whose affinity was higher for antigen,was used to establish an antigen captureELISA(AC-ELISA)detection system and test the efficiency by using clinical samples.Results:The heavy chain subclasses of two MAbs were all determined to be IgG2a.The 3C7 MAb showed stronger reactivity with rabies virus protein than the 2C5 MAb in an ELISA analysis,whereas the 3C7 MAb showed the highest affinity for antigen.IFA and immunocytochemistry results also indicated that the two MAbs could recognize rabies virus protein in its native form in cell samples.Data obtained using clinical samples showed that rabies virus could be detected by AC-ELISA detection system using the 3C7 MAb.Conclusion:It was potentially useful for the further development of highly sensitive,easily handled,and relatively rapid detection kits/tools for rabies surveillance in those areas where rabies is endemic,especially in China. | Xinjian Liu Xiaomin Feng Qi Tang Zhongcan Wang Zhenning Qiu Yuhua Li Changjun Wang Zhenqing Feng Jin Zhu Xiaohong Guan | 2010 | The Journal of Biomedical Research2010,24,5: | 5 |

| 6 | Design of communication relay mission for supporting lunar-farside soft landing显示文摘Chang'E-IV will be the first soft-landing and rover mission on the lunar farside. The relay satellite, which is located near the Earth-Moon L2 point for relay communication, is the key to the landing mission. Based on an analysis of the characteristics of the task and the technical difficulties associated with the relay satellite system, the overall design scheme of the relay communication mission is proposed in terms of trajectory design and communication system design among other aspects. First, according to the complex dynamic environment, a mission orbit that serves as an uninterrupted communication link is presented. A short-duration and low-energy transfer trajectory with lunar flyby is discussed. Orbital correction and a low-cost control strategy for orbit maintenance in the Earth-Moon L2 point region are provided. Second,considering the existing technical constraints, the requirement of relay communication in different stages and the design schemes of frequency division and redundant relay communication system are introduced. Finally,based on the trajectory design index and the performance of the communication system, the overall design scheme of the relay communication mission is proposed. | 13 | lncR-GAS5 upregulates the splicing factor SRSF10 to impair endothelial autophagy,leading to atherogenesis显示文摘Long noncoding RNAs(lncRNAs)play a critical role in the regulation of atherosclerosis.Here,we investigated the role of the lncRNA growth arrest-specific 5(lncR-GAS5)in atherogenesis.We found that the enforced expression of lncR-GAS5 contributed to the development of atherosclerosis,which presented as increased plaque size and reduced collagen content.Moreover,impaired autophagy was observed,as shown by a decreased LC3II/LC3I protein ratio and an elevated P62 level in lncR-GAS5-overexpressing human aortic endothelial cells.By contrast,lncR-GAS5 knockdown promoted autophagy.Moreover,serine/arginine-rich splicing factor 10(SRSF10)knockdown increased the LC3II/LC3I ratio and decreased the P62 level,thus enhancing the formation of autophagic vacuoles,autolysosomes,and autophagosomes.Mechanistically,lncR-GAS5 regulated the downstream splicing factor SRSF10 to impair autophagy in the endothelium,which was reversed by the knockdown of SRSF10.Further results revealed that overexpression of the lncR-GAS5-targeted gene miR-193-5p promoted autophagy and autophagic vacuole accumulation by repressing its direct target gene,SRSF10.Notably,miR-193-5p overexpression decreased plaque size and increased collagen content.Altogether,these findings demonstrate that lncR-GAS5 partially contributes to atherogenesis and plaque instability by impairing endothelial autophagy.In conclusion,lncR-GAS5 overexpression arrested endothelial autophagy through the miR-193-5p/SRSF10 signaling pathway.Thus,miR-193-5p/SRSF10 may serve as a novel treatment target for atherosclerosis. | Yuhua Fan Yue Zhang Hongrui Zhao Wenfeng Liu Wanqing Xu Lintong Jiang Ranchen Xu Yue Zheng Xueqing Tang Xiaohan Li Limin Zhao Xin Liu Yang Hong Yuan Lin Hui Chen Yong Zhang | 2023 | Frontiers of Medicine2023,17,2: | 1 |

| 17 | Effect of orbital shadow at an Earth-Moon Lagrange point on relay communication mission显示文摘The shadow effect is an important constraint to be considered during the implementation of exploration missions. In this paper, for the Earth-Moon Lagrange point L2 relay communication mission, shadow effect issues on a periodic orbit about L2 are investigated. A systematic analysis based on the time domain and phase space is performed including the distribution, duration, and frequency of shadows. First, the LindstedtPoincare and second-order differential correction methods are used in conjunction with the DE421 planetary ephemeris to achieve a mission trajectory family in a high-precision ephemeris model. Next, on the basis of a conical shadow model, the influence of different orbital phases and amplitudes on the shadow is analyzed. The distribution of the shadow is investigated as well. Finally, the configuration of the shadow and its characteristics are studied. | 20 | Mapping of de novo mutations in primary biliary cholangitis to a disease-specific co-expression network underlying homeostasis and metabolism显示文摘Primary biliary cholangitis(PBC) is an autoimmune disease involving dysregulation of a broad array of homeostatic and metabolic processes. Although considerable single-nucleotide polymorphisms have been unveiled, a large fraction of risk factors remains enigmatic. Candidate genes with rare mutations that tend to confer more deleterious effects need to be identified. To help pinpoint cellular and developmental mechanisms beyond common noncoding variants, we integrate whole exome sequencing with integrative network analysis to investigate genes harboring de novo mutations. Prominent convergence has been revealed on a network of disease-specific co-expression comprised of 55 genes associated with homeostasis and metabolism. The transcription factor gene MEF2 D and the DNA repair gene PARP2 are highlighted as hub genes and identified to be up-and down-regulated, respectively, in peripheral blood data set. Enrichment analysis demonstrates that altered expression of MEF2 D and PARP2 may trigger a series of molecular and cellular processes with pivotal roles in PBC pathophysiology.
