1Tree ring based streamflow reconstruction for the Upper Yellow River over the past 1234 years显示文摘We established a Juniperus przewalski tree ring width chronology, based on tree ring cores collected from the A’nyêmaqên Mountains. Statistical analysis showed that the chronology was highly correlated with instrumental streamflow records from previous August to current July from the Tangnaihai Station in the upper reaches of the Yellow River, with a correlation coefficient of 0.656. Streamflow for the upper reaches of the Yellow River was reconstructed for the past 1234 years. Low flow periods for the 11-year averaged streamflow reconstruction were definite as lower than mean plus 1 standard deviation, and high flow periods were higher than mean minus 1 standard deviation. Over the past 1234 years, high flows occurred 18 times, and low flows occurred 12 times. The main low flow periods were identified as AD 1140–1156, AD 1295–1309, AD 1473–1500, and AD 1820–1847, and the main high flow periods were identified as AD 846–873, and AD 1375–1400. Extremely low streamflow over the reconstruction period was seen during the late 15th century, coinciding with a widespread drought phenomenon, which took place in the northeastern Tibetan Plateau over the same period. Reconstructed streamflow shows significant low-frequency variability, which is in line with drought variability of neighboring regions, as inferred from tree rings and other proxies. 2Isolation and biological analysis of tumor stem cells from pancreatic adenocarcinoma显示文摘AIM: To explore the method of isolation and biological analysis of tumor stem cells from pancreatic adenocarcinoma cell line PANC-1. METHODS: The PANC-1 cells were cultured in Dulbecco modified eagle medium F12 (1:1 volume) (DMEM-F12) supplemented with 20% fetal bovine serum (FBS). Subpopulation cells with properties of tumor stem cells were isolated from pancreatic adenocarcinoma cell line PANC-1 according to the cell surface markers CD44 and CD24 by flow cytometry. The proliferative capability of these cells in vitro were estimated by 3-[4,5-dimehyl-2-thiazolyl]-2,5-diphenyl-2H-tetrazolium bromide (MTT) method. And the tumor growth of different subpopulation cells which were injected into the hypodermisof right and left armpit of nude mice was studied,and expression of CD44 and CD24 of the CD44+CD24+ cell-formed nodules and PANC-1 cells were detected by avidin-biotin-peroxidase complex (ABC) immunohistochemical staining. RESULTS: The 5.1%-17.5% of sorted PANC-1 cells expressed the cell surface marker CD44,57.8% -70.1% expressed CD24,only 2.1%-3.5% of cells were CD44+ CD24+. Compared with CD44-CD24-cells,CD44+CD24+ cells had a lower growth rate in vitro. Implantation of 104 CD44-CD24-cells in nude mice showed no evidenttumor growth at wk 12. In contrast,large tumors were found in nude mice implanted with 103 CD44+CD24+ cells at wk 4 (2/8),a 20-fold increase in tumorigenic potential (P < 0.05 or P < 0.01). There was no obvious histological difference between the cells of the CD44+CD24+ cell-formed nodules and PANC-1 cells. CONCLUSION: CD44 and CD24 may be used as the cell surface markers for isolation of pancreatic cancer stem cells from pancreatic adenocarcinoma cell line PANC-1. Subpopulation cells CD44+CD24+ have properties of tumor stem cells. 3A 457-year reconstruction of precipitation in the southeastern Qinghai-Tibet Plateau,China using tree-ring records显示文摘This paper presents a 457-year reconstruction of precipitation in the southeastern Qinghai-Tibet Plateau using tree-ring records.Tree-ring samples were collected from the Hengduan Mountains in the southeastern part of the Qinghai-Tibet Plateau,China.A nearly 500-year chronology was developed using tree-ring width records.Correlation analysis shows moisture is the main factor limiting tree growth in this region.Ring-widths were significantly positively correlated with the Palmer Drought Severity Index(PDSI) and precipitation in many months.The highest correlation coefficient was found between the annual growth of trees and precipitation from the previous September to the current June(0.738).Based on this relationship,we reconstructed the precipitation history from 1509 to 2006.The reconstruction explains 54.4%(Radj2=53.5%,N=49,F=56.12) of the actual precipitation variation during the calibration period(1958-2006).During the reliable period of the reconstruction(1549-2006),some low-frequency climate signals are included,indicating this region has been getting wetter in the last 20 years.The reconstruction documents six apparently dry and five pluvial periods and the 17th century dry period lasted longer than any other.When compared with other recent studies,this study and these earlier reconstructions show a similar trend in the variation of drought and pluvial.Further spatial correlation analysis confirms that the reconstructed precipitation adequately represents the rainfall history of the entire Hengduan Mountain area.The Multi-taper method,a type of spectral analysis,reveals that precipitation in this area had significant(P<0.01) spectral peaks at 3-5 a,60 a and 79-85 a.GOU XiaoHua YANG Tao GAO LinLin DENG Yang YANG MeiXue CHEN FaHu 2013Chinese Science Bulletin2013,58,10:16
4Persistence of side population cells with high drug efflux capacity in pancreatic cancer显示文摘AIM: To investigate the persistence of side population (SP) cells in pancreatic cancer and their role and mechanism in the drug resistance. METHODS: The presentation of side population cells in pancreatic cancer cell line PANC-1 and its proportion change when cultured with Gemcitabine, was detected by Hoechst 33342 staining and FACS analysis. The expression of ABCB1 and ABCG2 was detected by real- time PCR in either SP cells or non-SP cells. RESULTS: SP cells do exist in PANC-1, with a median of 3.3% and a range of 2.1-8.7%. After cultured with Gemcitabine for 3 d, the proportion of SP cells increased significantly (3.8% ± 1.9%, 10.7% ± 3.7%, t = 4.616, P = 0.001 < 0.05). ABCB1 and ABCG2 expressed at higher concentrations in SP as compared with non-SP cells (ABCB1: 1.15 ± 0.72, 5.82 ± 1.16, t = 10.839, P = 0.000 < 0.05; ABCG2: 1.16 ± 0.75, 5.48 ± 0.94, t = 11.305, P = 0.000 < 0.05), which may contribute to the efflux of fluorescent staining and drug resistance. 7Pancreas duodenal homeobox-1 expression and significance in pancreatic cancer显示文摘AIM: To study the correlations of Pancreas duodenal homeobox-1 with pancreatic cancer characteristics, including pathological grading, TNM grading, tumor metastasis and tumor cell proliferation. METHODS: Reverse transcriptase-polymerase chain reaction (RT-PCR) was used to detect PDX-1 mRNA expression in pancreatic cancer tissue and normal pancreatic tissue. The expression of PDX-1 protein was measured by Western blot and immunohistochemistry. Immunohistochemistry was also used to detect proliferative cell nuclear antigen (PCNA). Correlations of PDX-1 with pancreatic cancer characteristics, including pathological grading, TNM grading, tumor metastasis and tumor cell proliferation, were analyzed by using χ2 test. RESULTS: Immunohistochemistry showed that 41.1% of pancreatic cancers were positive for PDX-1 expression, but normal pancreatic tissue except islets showed no staining for PDX-1. In consistent with the result of imunohistochemistry, Western blot showed that 37.5% of pancreatic cancers were positive for PDX-1. RT-PCR showed that PDX-1 expression was significantly higher in pancreatic cancer tissues than normal pancreatic tissues (2-3.56 ± 0.35 vs 2-8.76 ± 0.14, P < 0.01). Lymph node metastasis (P < 0.01), TNM grading (P < 0.05), pathological grading (P < 0.05) and tumor cell proliferation (P < 0.01) were significantly correlated with PDX-1 expression levels. CONCLUSION: PDX-1 is re-expressed in pancreatic cancer, and PDX-1-positive pancreatic cancer cells show more malignant potential compared to PDX-1-negative cells. 8Generation of electricity from CO_2 mineralization: Principle and realization显示文摘Current CO2 reduction and utilization technologies suffer from high energy consuming. Thus, an energy favourable route is in urgent demanding. CO2 mineralization is theoretically an energy releasing process for CO2 reduction and utilization, but an approach to recovery this energy has so far remained elusive. For the first time, here we proposed the principle of harvesting electrical energy directly from CO2 mineralization, and realized an energy output strategy for CO2 utilization and reduction via a CO2-mineralization fuel cell(CMFC) system. In this system CO2 and industrial alkaline wastes were used as feedstock, and industrial valuable Na HCO3 was produced concomitantly during the electricity generation. The highest power density of this system reached 5.5 W/m2, higher than many microbial fuel cells. The maximum open circuit voltage reached 0.452 V. Moreover, this system was demonstrated viable to low concentration CO2(10%) and other carbonation process. 9Clinical effect of manual reduction of humeroradial joint in the treatment of typeⅠ-Ⅲfresh Monteggia fracture in children显示文摘Purpose:To evaluate the efficacy of closed reduction on the humeroradial joint in the treatment of Bado typeⅠ,ⅡandⅢfresh Monteggia fractures in children and investigate the effect of clinical factors,including Bado classification,age and time of treatment on the success rate of closed reduction.Methods:We retrospectively studied the data of children<10 years old with fresh Monteggia fractures(injury within two weeks)treated by manual reduction with plaster immobilization from January 2014 to April 2019.All patients were followed up in the outpatient department every two weeks for 4-6 weeks until plaster removal and then 3,6 and 12 months.Online or telephone interview was provided for some inconvenient patients after 6 months.Mackay criteria were used to evaluate the clinical effect.Radiographic data were collected and reviewed to assess the reduction of the humeroradial joint.Function of the elbow joint and forearm was evaluated and risk factors related to the failure of reduction were assessed.The successful manual reduction was analyzed from three aspects,respectively Bado fracture type(Ⅰ,Ⅱ,Ⅲ),patient age(<3 year,3-6 years,>6 years)and time interval from injury to treatment(group A,<1 day;group B,1-3 days;group C,>3 days).Results:Altogether 88 patients were employed in this study,including 58 males(65.9%)and 30 females(34.1%)aged from 1 to 10 years.There were 29 cases(33.0%)of Bado typeⅠMonteggia fractures,16(18.2%)typeⅡand 43(48.7%)typeⅢ.Successful manual reduction was achieved in 79 children(89.8%)at the last follow-up.The failed 9 patients received open surgery.Mackay criteria showed 100%goodexcellent rate for all the patients.The success rate of manual reduction was 89.7%,87.5%and 90.7%in Bado typeⅠ,ⅡandⅢcases,respectively,revealing no significant differences among different Bado types(x2=0.131,p=0.937).Successful closed reduction was achieved in 13 toddlers(13/13,100%),38 preschool children(28/42,90.5%)and 28 school-age children(28/33,84.8%),suggesting no significant difference either(x2=2.375,p=0.305).However time interval from injury to treatment showed that patients treated within 3 days had a much higher rate of successful manual reduction:67 cases(67/71,94.4%)in group A,10 cases(10/11,90.9%)in group B,and 2 cases(2/6,33.3%)in group C(x2=22.464,p<0.001).Fisher's test further showed significant differences between groups A and C(p=0.001)and groups B and C(p=0.028).Conclusion:Closed reduction is a safe and effective method for treating fresh Monteggia fractures in children.The reduction should be conducted as soon as possible once the diagnosis has been made.Yin-Qiang Cao Jia-Zhong Deng Yuan Zhang Xiao-Wei Yuan Tao Liu Jun Li Xiang Li Pan Gou Ming Li Xing Liu 2020Chinese Journal of Traumatology2020,23,4:5

11In vitro pancreas duodenal homeobox-1 enhances the differentiation of pancreatic ductal epithelial cells into insulin-producing cells显示文摘AIM: To observe whether pancreatic and duodenal homeobox factor-1 enhances the differentiation of pancreatic ductal epithelial cells into insulin-producing cells in vitro. METHODS: Rat pancreatic tissue was submitted to digestion by collegenase, ductal epithelial cells were separated by density gradient centrifugation and then cultured in RPMI1640 medium with 10% fetal bovine serum. After 3-5 passages, the cells were incubated in a six-well plate for 24 h before transfection of recombination plasmid XlHbox8VP16. Lightcycler quantitative real-time RT-PCR was used to detect the expression of PDX-1 and insulin mRNA in pancreatic epithelial cells. The expression of PDX-1 and insulin protein was analyzed by Western blotting. Insulin secretion was detected by radioimmunoassay. Insulin- producing cells were detected by dithizone-staining. RESULTS: XlHbox8 mRNA was expressed in pancreatic ductal epithelial cells. PDX-1 and insulin mRNA as well as PDX-1 and insulin protein were signifi cantly increased in the transfected group. The production and insulin secretion of insulin-producing cells differentiated from pancreatic ductal epithelial cells were higher than those of the untransfected cells in vitro with a significant difference (1.32 ± 0.43 vs 3.48 ± 0.81, P < 0.01 at 5.6 mmol/L; 4.86 ± 1.15 vs 10.25 ± 1.32, P < 0.01 at 16.7 mmol/L). CONCLUSION: PDX-1 can differentiate rat pancreaticductal epithelial cells into insulin-producing cells in vitro. 12Age and Body Size of the Shangcheng Stout Salamander Pachyhynobius shangchengensis(Caudata:Hynobiidae)from Southeastern China显示文摘Age and body size a re critical for understanding life history evolution and ecology.In this study,the age and body size of the Shangcheng stout salamander,Pachyhynobius shangchengensis,from a population in Anhui Province,China,were studied by skeletochronology.The mean age was 8.8±0.2(mean±SD)years in females and 9.6±0.2 in males and ranged 5-13 years for both sexes.The mean age was significantly different between sexes.The mean body size and mass were(100.21±0.91)mm and(31.76±0.73)g in females,and(105.31±1.23)mm and(37.14±1.12)g in males,respectively.Males were significantly larger and heavier than females,indicating sexual size dimorphism.There was a significant positive correlation among body size,body mass,and age,suggesting that the oldest individuals are larger and heavier.The growth rate in males was significantly higher than in females.The present study provides preliminary data on life-history traits which can be helpful for future studies of this species and other hynobiid salamanders.Jianli XIONG Jianping GOU Yong HUANG Baowei ZHANG Hongtao REN Tao PAN 2020Asian Herpetological Research2020,11,3:2

17Multi-objective and multi-scheme research on water and sediment regulation potential of reservoirs in the upper Yellow River显示文摘The problems of the deteriorating relation between water and sediment,and the escalating conflict between water supply and demand in the upper Yellow River(YR),need to be addressed.Reservoirs of Longyangxia,Liujiaxia,and Heishanxia(Long-Liu-Hei)in the upper YR are taken as the research object,and a multi-objective,water-sediment optimal operation model for cascade reservoirs Long-Liu-Hei has been developed.This operation model considers the comprehensive requirements of water supply,power generation,flood control,and especially,water and sediment regulation.The improved Non-dominated Sorting Genetic Algorithms(NSGA-II)method is used to demonstrate the water and sediment regulation potential of three scenario schemes under long-term conditions,and the adjustable water allocation of four schemes under typical year conditions are calculated.It is recommended to use the mode of generating hydropower depending on the available water amount as the best operating mode under the conditions of a long time series and for typical years.This recommended operation mode has verified the rationality and accuracy of the reservoir operations model.At the same time,compared with the calculation results for water and sediment regulation after the construction of the Heishanxia,the current research indicates:(1)Under the recommended model,the potential for water and sediment regulation under the long-term series and typical years is once every four years.(2)Inter-basin water transfer greatly improves the cascade power generation.When the adjustable water volume is converted into the controlled volume for water and sediment regulation,the intensity frequency of water and sediment regulation of cascade reservoirs are improved.(3)With the intensification of the contradiction between the supply of and demand for water resources in the upper YR in the future,the amount of transferable water exhibits a decreasing trend,and the regulation of water and sediment in the upper YR is facing huge challenges.(4)Through joint operation with the Longyangxia and Liujiaxia reservoirs(Long-Liu),Heishanxia will play an important role in the regulation and control of power generation,water supply,flood control,and ice prevention in the upper YR.The current research results can provide a theoretical basis and technical support for water and sediment regulation in the YR basin.Tao Bai Jia Yu Wenting Jin Jiaquan Wan Shaojie Gou Xu Ma Panpan Ma 2023International Journal of Sediment Research2023,38,2:1
