Fusion mechanism of 2019-nCoV and fusion inhibitors targeting HR1 domain in spike protein显示文摘Very recently,a novel coronavirus,2019-nCoV,emerged in Wuhan,China and then quickly spread worldwide, resulting in>17,388 confirmed cases and 361 deaths as of 3 February 2020,thus calling for the development of safe and effective therapeutics and prophylatics.1,2 Similar to severe acute respiratory syndrome(SARS)-CoV,2019-nCoV belongs to lineage B betacoronavirus, and it has the ability to utilize human angiotensin-converting enzyme 2(ACE2)as a receptor to infect human cells. | Shuai Xia Yun Zhu Meiqin Liu Qiaoshuai Lan Wei Xu Yanling Wu Tianlei Ying Shuwen Liu Zhengli Shi Shibo Jiang Lu Lu | 2020 | Cellular & Molecular Immunology2020,17,7: | 27
RBD-Fc-based COVID-19 vaccine candidate induces highly potent SARS-CoV-2 neutralizing antibody response显示文摘The pandemic of coronavirus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has posed serious threats to global health and economy, thus calling for the development of safe and effective vaccines. The receptorbinding domain(RBD)in the spike protein of SARS-CoV-2 is responsible for its binding to angiotensin-converting enzyme 2(ACE2)receptor.It contains multiple dominant neutralizing epitopes and serves as an important antigen for the development of COVID-19 vaccines. Here,we showed that immunization of mice with a candidate subunit vaccine consisting of SARS-CoV-2 RBD and Fc fragment of human IgG,as an immunopotentiator, elicited high titer of RBD-specific antibodies with robust neutralizing activity against both pseudotyped and live SARS-CoV-2 infections. The mouse antisera could also effectively neutralize infection by pseudotyped SARS-CoV-2 with several natural mutations in RBD and the IgG extracted from the mouse antisera could also show neutralization against pseudotyped SARS-CoV and SARS-related coronavirus(SARSr-CoV).Vaccination of human ACE2 transgenic mice with RBD-Fc could effectively protect mice from the SARS-CoV-2 challenge. These results suggest that SARS-CoV-2 RBD-Fc has good potential to be further developed as an effective and broad-spectrum vaccine to prevent infection of the current SARS-CoV-2 and its mutants,as well as future emerging SARSr-CoVs and re-emerging SARS-CoV. | Zezhong Liu Wei Xu Shuai Xia Chenjian Gu Xinling Wang Qian Wang Jie Zhou Yanling Wu Xia Cai Di Qu Tianlei Ying Youhua Xie Lu Lu Zhenghong Yuan Shibo Jiang | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 7
Antibody Cocktail Exhibits Broad Neutralization Activity Against SARS-CoV-2 and SARS-CoV-2 Variants显示文摘Severe acute respiratory syndrome coronavirus 2(SARS-Co V-2)has precipitated multiple variants resistant to therapeutic antibodies. In this study,12 high-affinity antibodies were generated from convalescent donors in early outbreaks using immune antibody phage display libraries.Of them, two RBD-binding antibodies(F61 and H121)showed high-affinity neutralization against SARS-Co V-2,whereas three S2-target antibodies failed to neutralize SARS-Co V-2.Following structure analysis,F61 identified a linear epitope located in residues G446–S494,which overlapped with angiotensinconverting enzyme 2(ACE2)binding sites, while H121 recognized a conformational epitope located on the side face of RBD,outside from ACE2 binding domain.Hence the cocktail of the two antibodies achieved better performance of neutralization to SARS-Co V-2.Importantly,these two antibodies also showed efficient neutralizing activities to the variants including B.1.1.7 and B.1.351,and reacted with mutations of N501 Y,E484 K,and L452 R,indicated that it may also neutralize the recent India endemic strain B.1.617.The unchanged binding activity of F61 and H121 to RBD with multiple mutations revealed a broad neutralizing activity against variants, which mitigated the risk of viral escape. Our findings revealed the therapeutic basis of cocktail antibodies against constantly emerging SARS-Co V-2 variants and provided promising candidate antibodies to clinical treatment of COVID-19 patients infected with broad SARS-Co V-2 variants. | Yuanyuan Qu Xueyan Zhang Meiyu Wang Lina Sun Yongzhong Jiang Cheng Li Wei Wu Zhen Chen Qiangling Yin Xiaolin Jiang Yang Liu Chuan Li Jiandong Li Tianlei Ying Dexin Li Faxian Zhan Youchun Wang Wuxiang Guan Shiwen Wang Mifang Liang | 2021 | Virologica Sinica2021,36,5: | 3

Evaluation of spatial epitope computational tools based on experimentally-confirmed dataset for protein antigens显示文摘Antibody molecules interact with antigen proteins through the epitope area, where the epitope residues are found to be discontinuous or spatial or conformational rather than linear on the protein surface. There are various computational algorithms to predict the spatial epitopes, and each of them have an outstanding performance based on their individual testing dataset. In this work,an independent dataset was created through collection of the epitope residual sites which have been confirmed by experiments. Based on this dataset,6 popular web-servers developed for B-cell structural epitope prediction, including SEPPA,CEP,DiscoTope,ElliPro,PEPOP and BEpro, were evaluated and compared according to sensitivity, the positive predictive value, the successful pick-up rate and the area under the curve of the receiver operator characteristic(AUC).The results showed that the general performance of spatial epitope prediction tools did obtain substantial advancement, and SEPPA gave the best performance among the 6 tools. However, the current prediction accuracy was still far from satisfaction. Moreover, our comparison elucidated that the performance of the web-servers was significantly affected by their training datasets and the algorithms adopted. In this sense, the results of our research may improve the design of B-cell epitope prediction tools and provide additional clues when the users utilize these tools in their related research. | XU XiaoLian SUN Jing LIU Qi WANG XiaoJing XU TianLei ZHU RuiXin WU Di CAO ZhiWei | 2010 | Chinese Science Bulletin2010,55,20: | 2

Analysis and Design of the Control Strategy of Three-roll Rewinder显示文摘By dividing the rewinding process of a three-roll rewinder into three zones, mathematical equations were formulated and diagrams were drawn to determine the structure factors of the rewinder, such as the diameters and spatial distribution of the rewinding rolls. The qualitative and quantitative analysis results show that the roll gap of the first two rewinding rolls and the angle between the line that connects the center points of the first two rewinding rolls and the vertical line play an important role in determining the ideal log diameter, which is related to the production capacity of the rewinder. Additionally, the ideal roll gap has a significant effect on the spatial distribution of the three rewinding rolls, the rewinding dead zone, and the rewinding quality of the log, which should be taken into full consideration when designing and improving rewinders to enhance production efficiency and satisfy ultra-high quality requirements. | TianLei Liu | 2017 | Paper And Biomaterials2017,2,2: | 1
Molecular cytogenetic analyses of two new wheat-rye 6RL translocation lines with resistance to wheat powdery mildew显示文摘Rye(Secale cereale)is a valuable gene donor for wheat improvement, especially for its resistance to diseases.Developing rye-derived resistance sources is important for wheat breeding. In the present study, two wheat-rye derivatives, designated JS016 and JS110,were produced by crossing common wheat cultivar Yangmai 23 with Pakistani rye accession W2A.Using sequential genomic in situ hybridization(GISH)and multicolor fluorescence in situ hybridization(mc-FISH),JS016 and JS110 were identified as a T6BS.6RL translocation line and a T6BS.6BL6RL translocation line, respectively.Ten newly 6RL chromosome arm-specific markers were developed and used to confirm the 6RL translocation. The wheat 55K single-nucleotide polymorphism(SNP)array further verified the molecular cytogenetic identification results above and clarified their breakpoints at 430.9 and 523.0 Mb of chromosome 6B in JS016 and JS110,respectively.Resistance spectrum and allelism test demonstrated that JS016 and JS110 possessed novel powdery mildew resistance gene(s)that was derived from the 6RL translocation but differed from Pm20.Moreover,JS016 and JS110 had better agronomic traits than the previously reported 6RL translocation line carrying Pm20.To efficiently transfer and detect the 6RL translocation from JS016 and JS110,one 6RL-specific Kompetitive allele specific PCR(KASP)marker was developed and validated in high throughput marker-assisted selection(MAS). | Shanying Zhu Haonan Du Fuyu Su Jin Wang Qingfeng Meng Tianlei Liu Rui Guo Zhaozhao Chen Huanhuan Li Wenxuan Liu Pengtao Ma Huagang He | 2023 | The Crop Journal2023,11,2: | 1

Semisynthesis of homogeneous spike RBD glycoforms from SARS-CoV-2 for profiling the correlations between glycan composition and function显示文摘Vaccines have been the primary remedy in the global fight against coronavirus disease 2019(COVID-19).The receptor-binding domain(RBD)of the spike protein,a critical viral immunogen,is affected by the heterogeneity of its glycan structures and relatively low immunogenicity. Here,we describe a scalable synthetic platform that enables the precise synthesis of homogeneously glycosylated RBD,facilitating the elucidation of carbohydrate structure–function relationships.Five homogeneously glycosylated RBDs bearing biantennary glycans were prepared, three of which were conjugated to T-helper epitope(Tpep)from tetanus toxoid to improve their weak immune response.Relative to natural HEK293-derived RBD,synthetic RBDs with biantennary N-glycan elicited a higher level of neutralising antibodies against SARS-CoV-2 in mice.Furthermore,RBDs containing T_(pep)elicited significant immune responses in transgenic mice expressing human angiotensin-converting enzyme 2.Our collective data suggest that trimming the N-glycans and T_(pep)conjugation of RBD could potentially serve as an effective strategy for developing subunit vaccines providing efficient protection. | Farong Ye Cheng Li Feng-Liang Liu Xinliang Liu Peng Xu Rong-Hua Luo Wenping Song Yong-Tang Zheng Tianlei Ying Biao Yu Ping Wang | 2024 | National Science Review2024,11,2: | 0
Multiple evaluations of atmospheric behavior between Criegee intermediates and HCHO: Gas-phase and air-water interface reaction显示文摘Given the high abundance of water in the atmosphere, the reaction of Criegee intermediates(CIs)with(H_(2)O)_(2) is considered to be the predominant removal pathway for CIs.However, recent experimental findings reported that the reactions of CIs with organic acids and carbonyls are faster than expected. At the same time, the interface behavior between CIs and carbonyls has not been reported so far. Here, the gas-phase and air-water interface behavior between Criegee intermediates and HCHO were explored by adopting high-level quantum chemical calculations and Born-Oppenheimer molecular dynamics(BOMD)simulations.Quantum chemical calculations evidence that the gas-phase reactions of CIs+HCHO are submerged energy or low energy barriers processes. The rate ratios speculate that the HCHO could be not only a significant tropospheric scavenger of CIs,but also an inhibitor in the oxidizing ability of CIs on SO_(x) in dry and highly polluted areas with abundant HCHO concentration. The reactions of CH_(2)OO with HCHO at the droplet’s surface follow a loop structure mechanism to produce i)SOZ(■),ii)BHMP(HOCH_(2)OOCH_(2)OH),and iii)HMHP(HOCH_(2)OOH).Considering the harsh reaction conditions between CIs and HCHO at the interface(i.e.,the two molecules must be sufficiently close to each other),the hydration of CIs is still their main atmospheric loss pathway. These results could help us get a better interpretation of the underlying CIs-aldehydes chemical processes in the global polluted urban atmospheres. | Tianlei Zhang Mingjie Wen Chao Ding Yongqi Zhang Xiaohui Ma Zhuqing Wang Makroni Lily Junhai Liu Rui Wang | 2023 | Journal of Environmental Sciences2023,,5: | 0
Orthologous genesPm12andPm21from twowild relatives of wheat show evolutionary conservation but divergent powdery mildew resistance显示文摘Wheat powdery mildew, caused by Blumeria graminis f.sp.tritici(Bgt),is a devastating disease that threatens wheat production worldwide.Pm12,which originated from Aegilops speltoides,a wild relative of wheat, confers strong resistance to powdery mildew and therefore has potential use in wheat breeding.Using susceptible mutants induced by gamma irradiation,we physically mapped and isolated Pm12 and showed it to be orthologous to Pm21 from Dasypyrum villosum, also a wild relative of wheat. The resistance function of Pm12 was validated via ethyl methanesulfonatemutagenesis,virus-induced gene silencing, and stable genetic transformation.Evolutionary analysis indicates that the Pm12/Pm21 loci in wheat species are relatively conserved but dynamic. Here,we demonstrated that the two orthologous genes,Pm12 and Pm21,possess differential resistance against the same set of Bgt isolates.Overexpression of the coiledcoil domains of both PM12 and PM21 induces cell death in Nicotiana benthamiana leaves. However, their full-length forms display different cell death-inducing activities caused by their distinct intramolecular interactions.Cloning of Pm12 will facilitate its application in wheat breeding programs. This study also gives new insight into two orthologous resistance genes,Pm12 and Pm21,which show different race specificities and intramolecular interaction patterns. | Shanying Zhu Cheng Liu Shuangjun Gong Zhaozhao Chen Rong Chen Tianlei Liu Renkang Liu Haonan Du Rui Guo Genying Li Miaomiao Li Renchun Fan Zhiyong Liu Qian-Hua Shen Anli Gao Pengtao Ma Huagang He | 2023 | Plant Communications2023,4,2: | 0
Effects of Nitrogen-Regulating Gene AreA on Growth,Pathogenicity, and Fumonisin Synthesis of Fusarium proliferatum显示文摘Rice spikelet rot disease not only threatens rice yields but also poses risks to humans and animals due to the production of the category 2B carcinogen fumonisins by the pathogen Fusarium proliferatum.Nitrogen(N)metabolism is known to have a significant influence on fungal growth and the synthesis of secondary metabolites.AreA is a global N regulatory gene belonging to the GATA transcription factor family. In this study,we observed that theΔAreA mutant exhibited a notable reduction in growth rate and conidium production.Pathogenicity experiments revealed thatΔAreA had almost lost its ability to infect rice spikelets. | SUN Lei CHEN Xu ZHOU Qianya ZHANG Tianlei YU Qian LIU Lianmeng HUANG Shiwen WANG Ling | 2024 | Rice science2024,31,1: | 0

Uncertainty-aware complementary label queries for active learning显示文摘Many active learning methods assume that a learner can simply ask for the full annotations of some training data from annotators. These methods mainly try to cut the annotation costs by minimizing the number of annotation actions.Unfortunately, annotating instances exactly in many realworld classification tasks is still expensive. To reduce the cost of a single annotation action,we try to tackle a novel active learning setting, named active learning with complementary labels(ALCL).ALCL learners ask only yes/no questions in some classes. After receiving answers from annotators,ALCL learners obtain a few supervised instances and more training instances with complementary labels, which specify only one of the classes to which the pattern does not belong. There are two challenging issues in ALCL:one is how to sample instances to be queried, and the other is how to learn from these complementary labels and ordinary accurate labels. For the first issue,we propose an uncertainty-based sampling strategy under this novel setup. | Shengyuan LIU Ke CHEN Tianlei HU Yunqing MAO | 2023 | Frontiers of Information Technology & Electronic Engineering2023,24,10: | 0
Discovery and druggability evaluation of pyrrolamide-type GyrB/ParE inhibitor against drug-resistant bacterial infection显示文摘The bacterial ATP-competitive GyrB/ParE subunits of type II topoisomerase are important anti-bacterial targets to treat super drug-resistant bacterial infections.Herein we discovered novel pyrrolamide-type GyrB/ParE inhibitors based on the structural modifications of the candidate AzD5099 that was withdrawn from the clinical trials due to safety liabilities such as mitochondrial toxicity. The hydroxyisopropyl pyridazine compound 28 had a significant inhibitory effect on Gyrase(GyrB,IC_(50)=49 nmol/L)and a modest inhibitory effect on TopoⅣ(ParE,IC_(50)=1.513μmol/L)of Staphylococcus aureus. It also had significant antibacterial activities on susceptible and resistant Gram-positive bacteria with a minimum inhibitory concentration(MIC)of less than 0.03μg/mL,which showed a time-dependent bactericidal effect and low frequencies of spontaneous resistance against S.aureus.Compound 28 had better protective effects than the positive control drugs such as DS-2969(5)and AZD5099(6)in mouse models of sepsis induced by methicillin-resistant Staphylococcus aureus(MRSA)infection. It also showed better bactericidal activities than clinically used vancomycin in the mouse thigh MRSA infection models. Moreover, compound 28 has much lower mitochondrial toxicity than AZD5099(6)as well as excellent therapeutic indexes and pharmacokinetic properties. At present, compound 28 has been evaluated as a pre-clinical drug candidate for the treatment of drug-resistant Gram-positive bacterial infection. On the other hand, compound 28 also has good inhibitory activities against stubborn Gram-negative bacteria such as Escherichia coli(MIC=1μg/mL),which is comparable with the most potent pyrrolamide-type GyrB/ParE inhibitors reported recently. In addition, the structure-activity relationships of the compounds were also studied. | Xintong Zhao Jing Feng Jie Zhang Zunsheng Han Yuhua Hu Hui-Hui Shao Tianlei Li Jie Xia Kangfan Lei Weiping Wang Fangfang Lai Yuan Lin Bo Liu Kun Zhang Chi Zhang Qingyun Yang Xinyu Luo Hanyilan Zhang Chuang Li Wenxuan Zhang Song Wu | 2023 | Acta Pharmaceutica Sinica B2023,13,12: | 0

Development of amulti-channel readout electronics verification system for MWDC detectors显示文摘Background The Multi-Wire Drift Chamber(MWDC)is among the significant detectors for CSR External Target Experiment(CEE).We require the MWDC detector’s front-end readout circuits to have a high integration, high count rate, and big dynamic range in order to measure the tracks of fore-angle products.Purpose In this paper,MWDC readout electronics verification system is shown, which is based on the front-end amplifier chip and Switched Capacitor Array(SCA)chip independently established by the project team. Results In the dynamic input range of 14 fC–950 fC,the readout electronics’energy measurement linearity is greater than 1%.The test results with MWDC illustrate that with a 55Fe source an energy resolution of 22.4%was acquired, and with cosmic rays the residual error is less than 400um. | Jiapeng Xu Canwen Liu Tianlei Pu Wei Liu Junwei Yan Zhi Qin Feng Li Zhi Deng Yi Qian Jie Kong | 2022 | Radiation Detection Technology and Methods2022,6,3: | 0
Occurrence Regularity of Major Damage Generations of Nilaparvata lugens(Stl) in Single Cropping Medium Rice Region in Sandu County of Guizhou Province显示文摘In order to clarify occurrence regularity of major damage generations of Nilaparvata lugens( Stl) in Sandu Shui Autonomous County( Sandu County for short),Guizhou Province,a karst single cropping medium rice region, the population dynamics of N. lugens were systematically studied using the methods of light trap and systematic field survey from 2009 to 2011. The results showed that the number of immigration generations caught by light trap and the actual population of N. lugens in fields were different among three years. The daily maximum trapping numbers of N. lugens amounted to 6 688 in 2009,which were 15. 3 and 2. 5 times of that in 2010 and 2011,respectively. The peak numbers in prediction nursery were 10 777,4 630 and 1 615 heads per hundred hills in 2009,2010 and 2011,respectively. The forth and fifth generations of N. lugens were the major damage generations in single cropping medium rice fields, and the initial immigration day was significantly related with primary-peak day. The occurrence degree of adults and nymphs of N. lugens varied with years and transplanting periods, and the peak days of N. lugens nymphs were from late July to early August in 2010 and 2011. The occurrence peaks of adults and nymphs in late transplanting paddy field were later than that in the early one. Meanwhile, the coexisted various growth periods of rice, due to time differences of transplanting, were conducive to inhabitation and reproduction of N. lugens. The population dynamics of spiders and Cyrtohinus lividipennis well synchronied with that of N. lugens, but predatory capacity of spiders and C. lividipennis was not enough to control N. lugens. | Liu Tianlei Jin Daochao Yang Hong Ai Zhengxian Zhou Zhaoxia | 2014 | Plant Diseases and Pests2014,5,2: | 0
