Microstructure and mechanical properties of Mg-5Li-1Al sheets prepared by accumulative roll bonding显示文摘Ultrafine-grain and high-strength Mg-5Li-1Al sheets were prepared by accumulative roll bonding(ARB)process. Evolution of microstructure and mechanical properties of ARB-processed Mg-5Li-1Al sheets was investigated. Results show that, during ARB process, the evolution of deformation mechanism of t Mg-5Li-1Al alloy is as follows: twinning deformation, shear deformation, forming macro shear zone, and finally dynamic recrystallization(DRX). The grain refining mechanism changes from twin DRX to rotation DRX. With the increase in ARB cycles, strength of the Mg-5Li-1Al sheets is enhanced, whilst elongation varies slightly. With the increase in rolling cycles, anisotropy of mechanical properties decreases. It is conclusive that strain hardening and grain refinement dominate the strengthening mechanism of Mg-5Li-1Al alloy. | Legan Hou Tianzi Wang Ruizhi Wu Jinghuai Zhang Milin Zhang Anping Dong Baode Sun Sergey Betsofen Boris Krit | 2018 | Journal of Materials Science & Technology2018,34,2: | 13
The long rather than the short allele of 5-HTTLPR predisposes Han Chinese to anxiety and reduced connectivity between prefrontal cortex and amygdala显示文摘The short allele of the serotonin-transporter gene is associated with higher risk for anxiety and depression in Caucasians, but this association is still unclear in Asians. Here, we addressed this issue using behavioral and multi-modal MRI approaches in a large group of healthy Han Chinese participants (n = 233). In contrast to findings in Caucasians, we found that long-allele (L) carriers had higher anxiety scores. In another group (n = 64) experiencing significant levels of depression or anxiety, the L-allele frequency was also significantly higher. In healthy participants, L-carriers had reduced functional and anatomical connectivity between the amygdala and prefrontal cortex (PFC), which was correlated with anxiety or depression scores. Our findings demonstrated that in Chinese Han participants, in contrast to Caucasians, the L-allele confers vulnerability to anxiety or depression and weakens top-down emotional control between the PFC and amygdala. Therefore, ethnic background should be taken into account in gene-related studies and their potential clinical applications. | Haixia Long Bing Liu Bing Hou Chao Wang Jin Li Wen Qin Dawei Wang Yuan Zhou Keith M. | 3 | Separate Neural Networks for Gains and Losses in Intertemporal Choice显示文摘An important and unresolved question is how human brain regions process information and interact with each other in intertemporal choice related to gains and losses. Using psychophysiological interaction and dynamic causal modeling analyses, we investigated the functional interactions between regions involved in the decisionmaking process while participants performed temporal discounting tasks in both the gains and losses domains. We found two distinct intrinsic valuation systems underlying temporal discounting in the gains and losses domains:gains were specifically evaluated in the medial regions, including the medial prefrontal and orbitofrontal cortices, and losses were evaluated in the lateral dorsolateral prefrontal cortex. In addition, immediate reward or punishment was found to modulate the functional interactions between the dorsolateral prefrontal cortex and distinct regions in both the gains and losses domains: in the gains domain, the mesolimbic regions; in the losses domain, the medial prefrontal cortex, anterior cingulate cortex, and insula. These findings suggest that intertemporal choice of gains and losses might involve distinct valuation systems, and more importantly, separate neural interactions may implement the intertemporal choices of gains and losses. These findings may provide a new biological perspective for understanding the neural mechanisms underlying intertemporal choice of gains and losses. | Yang-Yang Zhang Lijuan Xu Zhu-Yuan Liang Kun Wang Bing Hou Yuan Zhou Shu Li Tianzi Jiang | 2018 | Neuroscience Bulletin2018,34,5: | 6
Complement C7 is a novel risk gene for Alzheimer's disease in Han Chinese显示文摘Alzheimer's disease is the most common neurodegenerative disease, and has a high level of genetic heritability and population heterogeneity. In this study,we performed the whole-exome sequencing of Han Chinese patients with familial and/or early-onset Alzheimer's disease, followed by independent validation, imaging analysis and function characterization. We identified an exome-wide significant rare missense variant rs3792646(p.K420Q)in the C7 gene in the discovery stage(P=1.09×10^(-6),odds ratio=7.853)and confirmed the association in different cohorts and a combined sample(1615 cases and 2832 controls,P_(combined)=2.99×10^(-7),odds ratio=1.930).The risk allele was associated with decreased hippocampal volume and poorer working memory performance in early adulthood, thus resulting in an earlier age of disease onset.Overexpression of the mutant p.K420Q disturbed cell viability, immune activation and β-amyloid processing.Electrophysiological analyses showed that the mutant p.K420Q impairs the inhibitory effect of wild type C7 on the excitatory synaptic transmission in pyramidal neurons. These findings suggested that C7 is a novel risk gene for Alzheimer’s disease in Han Chinese. | Deng-Feng Zhang Yu Fan Min Xu Guihong Wang Dong Wang Jin Li Li-Li Kong Hejiang Zhou Rongcan Luo Rui Bi Yong Wu Guo-Dong Li Ming Li Xiong-Jian Luo Hong-Yan Jiang Liwen Tan Chunjiu Zhong Yiru Fang Chen Zhang Nengyin Sheng Tianzi Jiang Yong-Gang Yao | 2019 | National Science Review2019,6,2: | 6
An App knock-in rat model for Alzheimer’s disease exhibiting Aβand tau pathologies, neuronal death and cognitive impairments显示文摘A major obstacle in Alzheimer’s disease(AD)research is the lack of predictive and translatable animal models that reflect disease progression and drug efficacy.Transgenic mice overexpressing amyloid precursor protein(App)gene manifest non-physiological and ectopic expression of APP and its fragments in the brain, which is not observed in AD patients. The App knock-in mice circumvented some of these problems, but they do not exhibit tau pathology and neuronal death. We have generated a rat model, with three familiar App mutations and humanized Aβsequence knocked into the rat App gene.Without altering the levels of full-length APP and other APP fragments, this model exhibits pathologies and disease progression resembling those in human patients:deposit of Aβplaques in relevant brain regions, microglia activation and gliosis, progressive synaptic degeneration and AD-relevant cognitive deficits.Interestingly,we have observed tau pathology, neuronal apoptosis and necroptosis and brain atrophy, phenotypes rarely seen in other APP models. This App knock-in rat model may serve as a useful tool for AD research, identifying new drug targets and biomarkers, and testing therapeutics. | Keliang Pang Richeng Jiang Wei Zhang Zhengyi Yang Lin-Lin Li Makoto Shimozawa Simone Tambaro Johanna Mayer Baogui Zhang Man Li Jiesi Wang Hang Liu Ailing Yang Xi Chen Jiazheng Liu Bengt Winblad Hua Han Tianzi Jiang Weiwen Wang Per Nilsson Wei Guo Bai Lu | 2022 | Cell Research2022,32,2: | 6
ASAF: altered spontaneous activity fingerprinting in Alzheimer’s disease based on multisite fMRI显示文摘Several monocentric studies have noted alterations in spontaneous brain activity in Alzheimer's disease (AD), although there is no consensus on the altered amplitude of low-frequency fluctuations in AD patients. The main aim of the present study was to identify a reliable and reproducible abnormal brain activity pattern in AD. The amplitude of local brain activity (AM), which can provide fast mapping of spontaneous brain activity across the whole brain, was evaluated based on multisite rs-fMRI data for 688 subjects (215 normal controls (NCs), 221 amnestic mild cognitive impairment (aMCI) 252 AD). Two-sample t-tests were used to detect group differences between AD patients and NCs from the same site. Differences in the AM maps were statistically analyzed via the Stouffer's meta-analysis. Consistent regions of lower spontaneous brain activity in the default mode network and increased activity in the bilateral hippocampus/parahippocampus, thalamus, caudate nucleus, orbital part of the middle frontal gyrus and left fusiform were observed in the AD patients compared with those in NCs. Significant correlations (P?0.05, Bonferroni corrected) between the normalized amplitude index and Mini-Mental State Examination scores were found in the identified brain regions, which indicates that the altered brain activity was associated with cognitive decline in the patients. Multivariate analysis and leave-one-site-out cross-validation led to a 78.49% prediction accuracy for single-patient classification. The altered activity patterns of the identified brain regions were largely correlated with the FDG-PET results from another independent study. These results emphasized the impaired brain activity to provide a robust and reproducible imaging signature of AD. | Jiachen Li Dan Jin Ang Li Bing Liu Chengyuan Song Pan Wang Dawei Wang Kaibin Xu Hongwei Yang Hongxiang Yao Bo Zhou Alexandre Bejanin Gael Chetelat Tong Han Jie Lu Qing Wang Chunshui Yu Xinqing Zhang Yuying Zhou Xi Zhang Tianzi Jiang Yong Liu Ying Han | 2019 | Science Bulletin2019,64,14: | 3
Influence of rolling directions on microstructure, mechanical properties and anisotropy of Mg-5Li-1Al-0.5Y alloy显示文摘Mg-5Li-1Al-0.5Y alloy was rolled with different directions. The microstructure, mechanical properties and texture of the specimens were investigated with optical microscope, tensile tester and X-ray diffraction. The results show that changing rolling directions can refine the grain size of as-rolled alloys.Meanwhile, rolling directions have an obvious influence on the mechanical properties and texture of Mg-5Li-1Al-0.5Y alloy, thus affecting the anisotropy of the alloy. The sheet,of which the RD(rolling direction)and ND(normal direction)are both changed between two passes, possesses the smallest anisotropy. From the texture results, changing rolling directions reduces the maximum pole density, making the highest point distribution region excursion and the highest point distributes more scatteredly. | Tianzi Wang Tianlong Zhu Jianfeng Sun Ruizhi Wu Milin Zhang | 2015 | Journal of Magnesium and Alloys2015,3,4: | 2

A potential ethnic difference in the association between 5-HTTLPR polymorphisms and the brain default mode network显示文摘The serotonin-transporter-linked polymorphic region(5-HTTLPR)is associated with mood disorders. This association is thought to be due to amygdala hyperresponsiveness to negative emotional stimuli as a result of reduced frontal cortical control. In Caucasians, the short form is associated with this effect, but in Han Chinese we recently found that the long form is involved.Serotonin receptors have rich expression in default mode network(DMN)regions and the recent studies have found an association between the short form of the 5-HTTLPR and DMN functional connectivity(FC)in Caucasians. The present study has investigated whether there may also be anethnic difference in this influence of 5-HTTLPR on the DMN.We recruited 233 young Han Chinese subjects and calculated the resting-state default-network FC.Our study found that the L carriers had decreased FC in the bilateral medial prefrontal cortex, right parahippocampal gyrus, left middle temporal gyrus, and increased FC in left precuneus(Pcu)compared to SS.The PCC-Pcu FC in L carriers was significantly negatively correlated with the depression scores. Our findings, therefore, suggest that there is also a difference between Caucasian and Han Chinese subjects in the association between the different forms of the5-HTTLPR and DMN functional connectivity. | Haixia Long Bing Liu Bing Hou Chao Wang Keith M. | 15 | Effects of childhood trauma on aggressive behaviors andhippocampal function:the modulation of COMT haplotypes显示文摘Background:Aggression is a commonly hostile behavior linked to the hippocampal activity.Childhood trauma(CT)exposure has been associated with altered sensitization of the hypothalamic-pituitary-adrenal(HPA)axis and hippocampal volumewhich could increase violent aggressive behaviors.Additionally,Catechol-O-methyltransferase(COMT),the major dopamine metabolism enzyme,is impli-cated in stress responsivity, including aggression.Hence,CT exposure may affect aggression through the effect on the hippocampal function, which might also be modulated by the COMT variations. Objectives:This study examined whether both CT and haplotypes of COMT moderate hippocampal function and thus affect human aggressive behavior. Methods:We obtained bilateral hippocampal functional connectivity maps using resting state functional magnetic resonance imag-ing(MRI)data.COMT haplotype estimation was performed using Haploview 4.2 and PHASE 2.1.Then we constructed a moderated mediation model to study the effect of the CTQ×COMT on aggressive behavior. Results:Three major haplotypes were generated from thirteen single nucleotide polymorphisms(SNPs)within the COMT gene and formed three haplotypes corresponding to high, medium, and low enzymatic activity of COMT.The results showed interactive re-lationships between the Childhood Trauma Questionnaire(CTQ)and COMT with respect to the functional connectivity(FC)of the bilateral hippocampus(HIP)-orbital frontal cortex(OFC).Specifically,CT experience predicted lower negative HIP-OFC coupling in the APS and HPS haplotypes corresponding to the medium and high enzymatic activity of COMT,but greater FC in the LPS haplotypes corresponding to the low enzymatic activity. We also observed a conditional mediation effect of the right HIP-OFC coupling in the link between COMT and aggressive behavior that was moderated by CT experience. Conclusions:These results suggest that CT and COMT have a combined effect on aggressive behavior through hippocampal function. This mediation analysis sheds light on the influence of childhood experience on aggressive behavior in different genetic backgrounds. | Chao Wang Linfei Zhu Wenyu Zheng Hanyuzhu Peng Jiaojian Wang Yue Cui Bing Liu Tianzi Jiang | 2023 | Psychoradiology2023,3,1: | 0

Reproducible Abnormalities and Diagnostic Generalizability of White Matter in Alzheimer’s Disease显示文摘Alzheimer’s disease(AD)is associated with the impairment of white matter(WM)tracts. The current study aimed to verify the utility of WM as the neuroimaging marker of AD with multisite diffusion tensor imaging datasets[321 patients with AD,265 patients with mild cognitive impairment(MCI),279 normal controls(NC)],a unified pipeline, and independent site cross-validation.Automated fiber quantification was used to extract diffusion profiles along tracts.Random-effects meta-analyses showed a reproducible degeneration pattern in which fractional anisotropy significantly decreased in the AD and MCI groups compared with NC.Machine learning models using tract-based features showed good generalizability among independent site cross-validation.The diffusion metrics of the altered regions and the AD probability predicted by the models were highly correlated with cognitive ability in the AD and MCI groups. We highlighted the reproducibility and generalizability of the degeneration pattern of WM tracts in AD. | Yida Qu Pan Wang Hongxiang Yao Dawei Wang Chengyuan Song Hongwei Yang Zengqiang Zhang Pindong Chen Xiaopeng Kang Kai Du Lingzhong Fan Bo Zhou Tong Han Chunshui Yu Xi Zhang Nianming Zuo Tianzi Jiang Yuying Zhou Bing Liu Ying Han Jie Lu Yong Liu Multi-Center Alzheimer’s Disease Imaging(MCADI)Consortium | 2023 | Neuroscience Bulletin2023,39,10: | 0
Identification of novel loci influencing refractive error in East Asian populations using an extreme phenotype design显示文摘The global 'myopia boom' has raised significant international concerns. Despite a higher myopia prevalence in Asia, previous large-scale genome-wide association studies(GWASs) were mostly based on European descendants. Here, we report a GWAS of spherical equivalent(SE) in 1852 Chinese Han individuals with extreme SE from Guangzhou(631 <-6.00 D and 574 > 0.00 D) and Wenzhou(593 <-6.00 D and54 >-1.75 D), followed by a replication study in two independent cohorts with totaling 3538 East Asian individuals. The discovery GWAS and meta-analysis identify three novel loci, which show genome-wide significant associations with SE, including 1 q25.2 FAM163 A, 10 p11.22 NRP1/PRAD3, and 10 p11.21 ANKRD30 A/MTRNR2 L7, together explaining 3.34% of SE variance. 10 p11.21 is successfully replicated. The allele frequencies of all three loci show significant differences between major continental groups(P < 0.001). The SE reducing(more myopic) allele of rs10913877(1 q25.2 FAM163 A) demonstrates the highest frequency in East Asians and much lower frequencies in Europeans and Africans(EAS = 0.60,EUR = 0.20, and AFR = 0.18). The gene-based analysis additionally identifies three novel genes associated with SE, including EI24, LHX5, and ARPP19. These results provide new insights into myopia pathogenesis and indicate the role of genetic heterogeneity in myopia epidemiology among different ethnicities. | Xiaotong Han Tianzi Liu Xiaohu Ding Jialin Liu Xingyan Lin Decai Wang Moeen Riaz Paul N.Baird Zhi Xie Yuan Cheng Yi Li Yuki Mori Masahiro Miyake Hengtong Li Ching-Yu Cheng Changqing Zeng Kyoko Ohno-Matsui Xiangtian Zhou Fan Liu Mingguang He | 2022 | Journal of Genetics and Genomics2022,49,1: | 0
Promoting Metro Operation Safety by Exploring Metro Operation Accident Network显示文摘Understanding the causation of accidents is essential to promote metro operation safety. In terms of 243 reported metro operation accident cases in China, a directed weighted network was constructed based on complex network theory, where nodes and directed edges denotes factors and event chains respectively. To reveal the key causal factors, the topological characteristics of metro operation accident network(MOAN) were analyzed from both global and local views. The results show that facility-type factors are more closely related to the occurrence of the accidents from the perspectives of average path length and cascading effects. Accident types like train delay and train suspension are the great risk recipients. Key causal factors with large out-degree, out-strength, betweenness centrality and cluster coefficient, such as communication and signal failure, vehicle failure and piling into the train should be noticed. The research framework proposed in the paper is not only applicable to China’s metro operation system, but also appropriate for other transportation system safety studies. | Qi SUO Liyuan WANG Tianzi YAO Zihao WANG | 2021 | Journal of Systems Science and Information2021,9,4: | 0
Modified fiber qualities of the transgenic cotton expressing a silkworm fibroin gene显示文摘A silkworm gene for fibroin was introduced into the upland cotton WC line by Agrobacterium-mediated transformation. PCR detection for fibroin, nptII and gus genes, Kanamycin (Km)-resistance analysis and GUS-histochemical assay were conducted on 30 regenerated plants from 9 callus lines, and 17 positive plants were obtained by these 5 screening methods. By Km-resistance analysis and PCR for fibroin, 6 homozygous lines in T3 were obtained. Southern blot and Northern bolt demonstrated that the fibroin gene was inserted into the genome of these 6 lines, stably inherited and expressed. Compared to the control, the surface structure of mature fiber in the 6 lines was significantly distorted and an increased number of convolution was observed by scanning electron microscopy (SEM). Fiber quality traits analysis indicated that fiber elongation of the 6 homozygous lines was all increased and fiber strength of 3 lines was enhanced. These results indicated that fibroin expression influenced cotton fiber structure and quality, suggesting that fibroin has great potential for improving cotton fiber quality by genetic engineering. | LI FeiFei WU ShenJie Lǔ FenNi CHEN TianZi JU Ming WANG HaiHai JIANG YanJie ZHANG Jie GUO WangZhen ZHANG TianZhen | 2009 | Chinese Science Bulletin2009,54,7: | 0
