2Segmentation of Hematoxylin-Eosin stained breast cancer histopathological images based on pixel-wise SVM classifier显示文摘Hematoxylin-Eosin(HE) staining is the routine diagnostic method for breast cancer(BC), and large amounts of HE stained histopathological images are available for analysis. It is emergent to develop computational methods to efficiently and objectively analyze these images, with the aim of providing potentially better diagnostic and prognostic information for BC. This work focuses on analyzing our in-house HE stained histopathological images of breast cancer tissues. Since tumor nests(TNs) and stroma morphological characteristics can reflect the biological behaviors of breast invasive ductal carcinoma(IDC), accurate segmentation of TNs and the stroma is the first step towards the subsequent quantitative analysis. We first propose a method based on the pixel-wise support vector machine(SVM) classifier for segmenting TNs and the stroma, then extract four morphological characters related to the TNs from the images and investigate their relationships with the patients' 8-year disease free survival(8-DFS). The evaluation result shows that the classification based segmentation method is able to distinguish between TNs and stroma with 87.1% accuracy and 80.2% precision,suggesting that the proposed method is promising in segmenting HE stained IDC histopathological images. 3Stress-induced CsPbBr3 nanocrystallization on glass surface:Unexpected mechanoluminescence and applications显示文摘In this work,we discovered an unexpected mechanoluminescence (ML) phenomena occurring when transforming amorphous into crystalline,due to the stress-induced precipitation of CsPbBr3 perovskite nanocrystals on glass surface.It is revealed that,unlike the conventional thermal-induced phase transformation mechanism,the breakage of bonding of glass network provides the energy for nucleation and growth,and the shear stress avoids the long-range migration of structural units for crystallization.Such unique ML phenomenon enables the visualization of dynamical force that is inaccessible by common strategy,and so,opens up some novel applications,such as the pressure-sensitive 'glassy pencil' to learn people's writing habits,and the pb^2+-detection with good sensitivity and selectivity.These findings not only demonstrate an effective route for the preparation of perovskite materials in a green,time-saving,low cost,and scalable way,enrich the knowledge of glass crystallization mechanism,but also exploit a useful avenue to quantitatively visualize the dynamical force.Xiaoqiang Xiang Hang Lin Renfu Li Yao Cheng Qingming Huang Ju Xu Congyong Wang Xueyuan Chen Yuansheng Wang 2019Nano Research2019,12,5:8
4Q-switching of waveguide lasers based on graphene/WS_2 van der Waals heterostructure显示文摘We report on the operation of passively Q-switched waveguide lasers at 1 μm wavelength based on a graphene∕WS_2 heterostructure as a saturable absorber(SA). The gain medium is a crystalline Nd:YVO_4 cladding waveguide produced by femtosecond laser writing. The nanosecond waveguide laser operation at 1064 nm has been realized with the maximum average output power of 275 m W and slope efficiency of 37%. 5Oxygenation,inflammatory response and lung injury during one lung ventilation in rabbits using inspired oxygen fraction of 0.6 vs.1.0显示文摘Maintaining adequate oxygenation during one-lung ventilation(OLV) requires high inspired oxygen fraction(FiO_2).However,high FiO_2 also causes inflammatory response and lung injury.Therefore,it remains a great interest to clinicians and scientists to optimize the care of patients undergoing OLV.The aim of this study was to determine and compare oxygenation,inflammatory response and lung injury during OLV in rabbits using FiO_2 of 0.6 vs.1.0.After 30 minutes of two-lung ventilation(TLV) as baseline,30 rabbits were randomly assigned to three groups receiving mechanical ventilation for 3 hours:the sham group,receiving TLV with 0.6 FiO_2;the 1.0 FiO_2 group,receiving OLV with 1.0 FiO_2;the 0.6 FiO_2 group,receiving OLV with 0.6 FiO_2.Pulse oximetry was continuously monitored and arterial blood gas analysis was intermittently conducted.Histopathologic study of lung tissues was performed and inflammatory cytokines and the mRNA and protein of nuclear factor kappa B(NF-κB) p65 were determined.Three of the 10 rabbits in the 0.6 FiO_2 group suffered hypoxemia,defined by pulse oximetric saturation(SpO_2) less than 90%.Partial pressure of oxygen(PaO_2),acute lung injury(ALI) score,myeloperoxidase(MPO),tumor necrosis factor-a(TNF-α),interleukin-6(IL-6),mRNA and protein of NF-kB p65 were lower in the 0.6 FiO_2group than in the 1.0 FiO_2 group.In conclusion,during OLV,if FiO_2 of 0.6 can be tolerated,lung injury associated with high FiO_2 can be minimized.Further study is needed to validate this finding in human subjects.Zeping Xu Lianbing Gu Qingming Bian Pengyi Li Lijun Wang Jingyuan Zhang Yanning Qian 2017The Journal of Biomedical Research2017,31,1:7
6RLS and LMS blind adaptive multi-user detection method and comparison in acoustic communication显示文摘RLS and LMS blind adaptive multi-user detection algorithm and multi-user detector was proposed to solve the problem of multi-user signal detection problem encountered in underwater acoustic communication networks.In simulation analysis,RLS and the LMS blind adaptive multi-user detector were designed and tested for synchronous and asynchronous multi-user communication process.The results of SIR comparison and MMSE comparison show that,both of the two methods can realize blind adaptive detection when any user change in multi-user communication,during this process,the training communication sequences are not needed.The RLS algorithm has about 5 dB higher in SIR compared with LMS algorithm,and the convergence velocity of RLS algorithm is also higher than LMS algorithm when the communication users change.RLS algorithm has better ability in multi-user detection than that of LMS algorithm,and it has great attraction and guiding significance for solving the problem of multiple access interference(MAI) in multi-user communication.WANG Zhongqiu WANG Hongru MENG Qingming 2015Instrumentation2015,2,2:7
7Phase I study of CAR-T cells with PD-1 and TCR disruption in mesothelin-positive solid tumors显示文摘Programmed cell death protein-1(PD-1)-mediated immunosuppression has been proposed to contribute to the limited clinical efficacy of chimeric antigen receptor T(CAR-T)cells in solid tumors.We generated PD-1 and T cell receptor(TCR)deficient mesothelin-specific CAR-T(MPTK-CAR-T)cells using CRISPR-Cas9 technology and evaluated them in a dose-escalation study.A total of 15 patients received one or more infusions of MPTK-CAR-T cells without prior lymphodepletion.No dose-limiting toxicity or unexpected adverse events were observed in any of the 15 patients.The best overall response was stable disease(2/15 patients).Circulating MPTK-CAR-T cells peaked at days 7–14 and became undetectable beyond 1 month.TCR-positive CAR-T cells rather than TCR-negative CAR-T cells were predominantly detected in effusion or peripheral blood from three patients after infusion.We further confirmed the reduced persistence of TCR-deficient CAR-T cells in animal models.Our results establish the preliminary feasibility and safety of CRISPR-engineered CAR-T cells with PD-1 disruption and suggest that the natural TCR plays an important role in the persistence of CAR-T cells when treating solid tumors.Zhenguang Wang Na Li Kaichao Feng Meixia Chen Yan Zhang Yang Liu Qingming Yang Jing Nie Na Tang Xingying Zhang Chen Cheng Lianjun Shen Jiaping He Xun Ye Wei Cao Haoyi Wang Weidong Han 2021Cellular & Molecular Immunology2021,18,9:7
8An analytical biomarker for treatment of patients with recurrent B-ALL after remission induced by infusion of anti-CD19 chimeric antigen receptor T(CAR-T) cells显示文摘Anti-CD19 chimeric antigen receptor-modified T(CAR-T-19) cells have emerged as a powerful targeted immunotherapy for B-cell lineage acute lymphoblastic leukemia with a remarkable clinical response in recent trials. Nonetheless, few data are available on the subsequent clinical monitoring and treatment of the patients, especially those with disease recurrence after CAR-T-19 cell infusion. Here, we analyzed three patients who survived after our phase I clinical trial and who were studied by means of biomarkers reflecting persistence of CAR-T-19 cells in vivo and predictive factors directing further treatment. One patient achieved 9-week sustained complete remission and subsequently received an allogeneic hematopoietic stem cell transplant. Another patient who showed relapse after 20 weeks without detectable leukemia in the cerebrospinal fluid after CAR-T-19 cell treatment was able to achieve a morphological remission under the influence of stand-alone low-dose chemotherapeutic agents. The third patient gradually developed extensive extramedullary involvement in tissues with scarce immune-cell infiltration during a long period of hematopoietic remission after CAR-T-19 cell therapy. Long-term and discontinuous increases in serum cytokines(mainly interleukin 6 and C-reactive protein) were identified in two patients(Nos. 1 and 6) even though only a low copy number of CAR molecules could be detected in their peripheral blood. This finding was suggestive of persistent functional activity of CAR-T-19 cells. 9Attribution analysis based on Budyko hypothesis for land evapotranspiration change in the Loess Plateau, China显示文摘Land evapotranspiration(ET) is an important process connecting soil, vegetation and the atmosphere, especially in regions that experience shortage in precipitation.Since 1999, the implementation of a large-scale vegetation restoration project has significantly improved the ecological environment of the Loess Plateau in China.However, the quantitative assessment of the contribution of vegetation restoration projects to long-term ET is still in its infancy.In this study, we investigated changes in land ET and associated driving factors from 1982 to 2014 in the Loess Plateau using Budyko-based partial differential methods.Overall, annual ET slightly increased by 0.28 mm/a and there were no large fluctuations after project implementation.An attribution analysis showed that precipitation was the driving factor of inter-annual variability of land ET throughout the study period;the average impacts of precipitation, potential evapotranspiration, and vegetation restoration on ET change were 61.5%, 11.5% and 26.9%, respectively.These results provide an improved understanding of the relationship between vegetation condition change and climate variation on terrestrial ET in the study area and can support future decision-making regarding water resource availability.HE Guohua ZHAO Yong WANG Jianhua GAO Xuerui HE Fan LI Haihong ZHAI Jiaqi WANG Qingming ZHU Yongnan 2019Journal of Arid Land2019,11,6:5

12Urban-rural disparities in mild cognitive impairment and its functional subtypes among community- dwelling older residents in central China显示文摘background Substantial variations in the prevalence of mild cognitive impairment(MCI)and its subtypes have been reported,although mostly in geographically defined developed countries and regions.Less is known about MCI and its subtypes in rural areas of less developed central China.Aims The study aimed to compare the prevalence of MCI and its subtypes in residents aged 65 years or older in urban and rural areas of Hubei Province,China.Methods Participants aged 65 years or older were recruited between 2018 and 2019.Inperson structured interviews and clinical and neuropsychological assessments were performed at city health community centres and township hospitals.results Among 2644 participants without dementia,735 had MCI,resulting in a prevalence of 27.8%for total MCI,20.9%for amnestic MCI(aMCI)and 6.9%for non-amnestic MCI(naMCI).The prevalence of MCI in urban and rural areas was 20.2%and 44.1%,respectively.After adjusting for demographic factors,the prevalence of total MCI,aMCI and naMCI differed significantly between rural and urban areas(adjusted odds ratio(OR)2.10,1.44 and 3.76,respectively).Subgroup analysis revealed an association between rural socioeconomic and lifestyle disadvantage and MCI and its subtypes.Conclusions Our findings suggest that the prevalence of MCI among urban residents in central China is consistent with that in other metropolis areas,such as Shanghai,but the prevalence in rural areas is twice that in urban areas.Prospective studies and dementia prevention in China should focus on rural areas.Dan Liu Lin Li Lina An Guirong Cheng Cong Chen Mingjun Zou Bo Zhang Xuguang Gan Lang Xu Yangming Ou Qingming Wu Ru Wang Yan Zeng 2021General Psychiatry2021,34,5:3

14Biomimetic polyetheretherketone microcarriers with specific surface topography and self-secreted extracellular matrix for large-scale cell expansion显示文摘Reusable microcarriers with appropriate surface topography,mechanical properties,as well as biological modification through decellularization facilitating repeated cell culture are crucial for tissue engineering applications.Herein,we report the preparation of topological polyetheretherketone(PEEK)microcarriers via gas-driven and solvent exchange method followed by hydrothermal treatment at high temperature and pressure.After hydrothermal treated for 8 h,the resulting topological PEEK microcarriers exhibit walnut-like surface topography and good sphericity as well as uniform size distribution of 350.24619.44 mm.And the average width between ravine-patterned surface of PEEK microcarriers is 7806290 nm.After repeated steam sterilization by autoclaving for three times,topological PEEK microcarriers show nearly identical results compared with previous ones indicating strong tolerance to high temperature and pressure.This is a unique advantage for large-scale cell expansion and clinical applications.Moreover,PEEK microcarriers with special topography possess higher protein adsorption efficiency.In addition,the reutilization and biofunctionalization with repeated decellularization of topological PEEK microcarriers show highly beneficial for cell adhesion and proliferation.Therefore,our study is of great importance for new generation microcarriers with micro-and nano-scale surface feature for a broad application prospect in tissue engineering.Qingming Ji ZongliangWang Zixue Jiao Yu Wang Zhenxu Wu Peng Wang Yuhang Zhu Shuo Sun Yi Liu Peibiao Zhang 2020Regenerative Biomaterials2020,7,1:3

16The G-protein α subunit GhGPA positively regulates Gossypium hirsutum resistance to Verticillium dahliae via induction of SA and JA signaling pathways and ROS accumulation显示文摘Verticillium wilt, a devastating disease in cotton caused by Verticillium dahliae, reduces cotton quality and yield. Heterotrimeric GTP-binding proteins, consisting of Ga, Gb, and Gc subunits, transducers of receptor signaling, function in a wide range of biological events. However, the function of Ga proteins in the regulation of defense responses in plants is largely unexplored, except for a few reports on model species. In the present study, a cotton G-protein a-subunit-encoding gene(GhGPA) was isolated from Verticillium wilt-resistant Gossypium hirsutum(upland cotton) cv. ND601. GhGPA transcription was up-regulated under V. dahliae stress, with higher expression in tolerant than in susceptible cotton cultivars.Subcellular localization revealed GhGPA to be located in the plasma membrane. GhGPA shows high(85.0%) identity with Arabidopsis AT2 G26300(AtGPA1), and AtGPA1 gpa1-4 mutants displayed susceptibility to V. dahliae. Ectopic expression of GhGPA successfully restored the resistance of Arabidopsis gpa1-4 mutants to Verticillium wilt and made them more resistant than the wild type. Overexpression of GhGPA in Arabidopsis markedly increased the resistance and resulted in dramatic up-regulation of pathogenesis-related(PR) genes and increased in H2 O2 accumulation and salicylic acid(SA) and jasmonic acid(JA) contents. However, suppressing GhGPA expression via virus-induced gene silencing(VIGS)increased susceptibility to Verticillium wilt, down-regulated the expression of PR and marker genes in SA and JA signaling pathways, and reduced H2 O2 content. The contents of SA and JA in Arabidopsis gpa1-4 and VIGS cotton were lower than those in the wild type and empty-vector control. However,GhGPA-overexpressing Arabidopsis contained more SA and JA than the wild type when inoculated with V. dahliae. Thus, GhGPA plays a vital role in Verticillium wilt resistance by inducing SA and JA signaling pathways and regulating the production of reactive oxygen species. 17Human hepatopoietin augmenter of liver regeneration──A hepatotrophic factor for liver regeneration, and its potential antihepatitis effect in vivo显示文摘The effect of human hapatopoietin i.e. augmenter of liver regeneration (hALR) was determined on hepatocyte DNA synthesis, and on CCl 4-induced hepatitis in animal in vitro and in vivo. It is found that ALR could directly stimulate DNA synthesis of hepatocytes in primary culture in a dose-dependent manner. In 30% hepatectomied rats, significant DNA synthesis occurred in control rats ; even so, exogenous hALR gene encoding protein increased DNA synthesis of regeneration liver by 2.3 fold compared with control rats. A lower dose of CCl 4 was administrated in rats and the effect of ALR on DNA synthesis of regenerating liver 48 h after CCl 4 administration was analyzed. Few Budr-labeled hepatocytes were visible in control rats. However, exogenous hALR markedly increased the number of labeled cells in a dose-dependent manner. Statistical analysis showed that 10 or 40 μg·kg -1 ALR protein stimulated DNA synthesis by 1.7 and 4.8 fold respectively. In addition to enhancing cell growth, adiministration of hALR achieved a significant improvement in reversing the lethality of rat hepatic failure when compared with that of control group; the elevation of cytosolic enzymes was dramatically suppressed by exogenous hALR in CCl 4-treated mice in vivo and in vitro; histologically, hepatocytes around the central vein were necrotic, and the degree of hepatocyte necrosis in control mice was more prominent than that in the mice given 40 μg·kg-1 hALR 48 h after CCl 4 administration. We also noted that hALR had a strong antihepatitis effect in vitro which was determined with primary cultured rat hepatocytes. These findings suggest that hALR protects the integrity of hepatocytes against severe hepatitis, and indicate that ALR may be an important regulator of liver regeneration and play a major role in liver injury repair. We demonstrated that bone regeneration in the rat skull defect model was significantly accelerated with low-intensity NIR stimulation. In vitro studies showed that NIR stimulation could promote the osteoblast differentiation in bone mesenchymal stem cells(BMSCs) and MC3T3-E1 cells, which was associated with increased ubiquitination of the core circadian clock protein Cryptochrome 1(CRY1) in the nucleus. We found that the reduction of CRY1 induced by NIR light activated the bone morphogenetic protein(BMP) signaling pathways, promoting SMAD1/5/9 phosphorylation and increasing the expression levels of Runx2 and Osterix. NIR light treatment may act through sodium voltage-gated channel Scn4a, which may be a potential responder of NIR light to accelerate bone regeneration. 19Impact of large-scale vegetation restoration project on summer land surface temperature on the Loess Plateau, China显示文摘A large-scale afforestation project has been carried out since 1999 in the Loess Plateau of China. However, vegetation-induced changes in land surface temperature (LST) through the changing land surface energy balance have not been well documented. Using satellite measurements, this study quantified the contribution of vegetation restoration to the changes in summer LST and analyzed the effects of different vegetation restoration patterns on LST during both daytime and nighttime. The results show that the average daytime LST decreased by 4.3°C in the vegetation restoration area while the average nighttime LST increased by 1.4°C. The contributions of the vegetation restoration project to the changes in daytime LST and nighttime LST are 58% and 60%, respectively, which are far greater than the impact of climate change. The vegetation restoration pattern of cropland (CR) converting into artificial forest (AF) has a cooling effect during daytime and a warming effect at nighttime, while the conversion of CR to grassland has an opposite effect compared with the conversion of CR to AF. 20Blood flow activation in rat somatosensory cortex under sciatic nerve stimulation revealed by laser speckle imaging显示文摘In many functional neuroimaging research the change of local cerebral blood flow (CBF) induced by sensory stimulation is regarded as an indicator of the change in cortical neuronal activity although a precise and full spatio-temporal description of local CBF response coupled to neural activity has still not been laid out. Using the laser speckle imaging technique a relatively large exposed area in somatosensory cortex of rat was imaged for the observation of the variations of CBF during sciatic nerve stimulation. The results showed that cerebral blood flow activation was spatially localized and discretely distributed in the targeted microvasculature. Individual arteries, veins and capillaries in different diameters were activated with the time going.
