1Aberrant DNA methylation in 5′ regions of DNA methyltransferase genes in aborted bovine clones显示文摘High rate of abortion and developmental abnormalities is thought to be closely associated with inefficient epigenetic reprogramming of the transplanted nuclei during bovine cloning. It is known that one of the important mechanisms for epigenetic reprogramming is DNA methylation. DNA methylation is established and maintained by DNA methyltransferases (DNMTs), therefore, it is postulated that the inefficient epigenetic reprogramming of transplanted nuclei may be due to abnormal expression of DNMTs. Since DNA methylation can strongly inhibit gene expression, aberrant DNA methylation of DNMT genes may disturb gene expression. But presently, it is not clear whether the methylation abnormality of DNMT genes is related to developmental failure of somatic cell nuclear transfer embryos. In our study, we analyzed methylation patterns of the 5′ regions of four DNMT genes including Dnmt3a, Dnmt3b, Dnmt1 and Dnmt2 in four aborted bovine clones. Using bisulfite sequencing method, we found that 3 out of 4 aborted bovine clones (AF1, AF2 and AF3) showed either hypermethylation or hypomethylation in the 5′ regions of Dnmt3a and Dnmt3b, indicating that Dnmt3a and Dnmt3b genes are not properly reprogrammed. However, the individual AF4 exhibited similar methylation level and pattern to age-matched in vitro fertilized (IVF) fetuses. Besides, we found that the 5′ regions of Dnmt1 and Dnmt2 were nearly completely unmethylated in all normal adults, IVF fetuses, sperm and aborted clones. 4Evaluation of triage strategies for high-risk human papillomavirus-positive women in cervical cancer screening:A multicenter randomized controlled trial in different resource settings in China显示文摘Objective:We aimed to evaluate the effectiveness of different triage strategies for high-risk human papillomavirus(hrHPV)-positive women in primary healthcare settings in China.Methods:This study was undertaken in 11 rural and 9 urban sites.Women aged 35-64 years old were enrolled.HrHPV-positive women were randomly allocated to liquid-based cytology(LBC),visual inspection with acetic acid and Lugol’s iodine(VIA/VILI)(rural only)triage,or directly referred to colposcopy(direct COLP).At 24 months,hrHPV testing,LBC and VIA/VILI were conducted for combined screening.Results:In rural sites,1,949 hrHPV-positive women were analyzed.A total of 852,218 and 480 women were randomly assigned to direct COLP,LBC and VIA/VILI.At baseline,colposcopy referral rates of LBC or VIA/VILI triage could be reduced by 70%-80%.LBC(n=3 and n=7)or VIA/VILI(n=8 and n=26)could significantly decrease the number of colposcopies needed to detect one cervical intraepithelial neoplasia(CIN)2 or worse and CIN3+compared with direct COLP(n=14 and n=23).For the 24-month cumulative detection rate of CIN2+,VIA/VILI triage was 0.50-fold compared with LBC triage and 0.46-fold with the direct COLP.When stratified by age,baseline LBC triage+performed best(P<0.001),peaking among women aged 35-44 years(Ptrend=0.002).In urban sites,1,728 women were hrHPV genotyping test positive.A total of 408,571 and 568women were randomly assigned to direct COLP for HPV16/18+,direct COLP for other hrHPV subtypes+,and LBC triage for other hrHPV subtypes+.LBC(n=12 and n=31)significantly decreased the number of colposcopies needed to detect one CIN2+and CIN3+compared with direct COLP(n=14 and n=44).HPV16/18+increased the 24-month cumulative detection rate of CIN2+(17.89%,P<0.001).Conclusions:LBC triage for hrHPV-positive women in rural settings and direct COLP for HPV16/18+women and LBC triage for other hrHPV subtype+women in urban settings might be feasible strategies.Le Dang Linghua Kong Yuqian Zhao Yi Dai Li Ma Lihui Wei Shulan Zhang Jihong Liu Mingrong Xi Long Chen Xianzhi Duan Qing Xiao Guzhalinuer Abulizi Guonan Zhang Ying Hong Qi Zhou Xing Xie Li Li Mayinuer Niyazi Zhifen Zhang Jiyu Tuo Yiling Ding Youlin Qiao Jinghe Lang 2022Chinese Journal of Cancer Research2022,34,5:2

6A 12-bit 250-MS/s Charge-Domain Pipelined Analog-to-Digital Converter with Feed-Forward Common-Mode Charge Control显示文摘A feed-forward Common-Mode(CM) charge control circuit for a high-speed Charge-Domain(CD)pipelined Analog-to-Digital Converter(ADC) is presented herein.This study aims at solving the problem whereby the precision of CD pipelined ADCs is restricted by the variation in input CM charge,which can compensate for CM charge errors caused by a variation in CM charge input in real time.Based on the feed-forward CM charge control circuit,a 12-bit 250-MS/s CD pipelined ADC is designed and realized using a 1 P6 M 0.18-μm CMOS process.The ADC achieved a Spurious Free Dynamic Range(SFDR) of 78.1 d B and a Signal-to-Noise-and-Distortion Ratio(SNDR) of 64.6 d B for a 20.1-MHz input:a SFDR of 74.9 d B and SNDR of 62.0 d B were achieved for a 239.9-MHz input at full sampling rate.The variation in signal-to-noise ratio was less than 3 d B over a 0–1.2 V input CM voltage range.The power consumption of the prototype ADC is only 85 m W at 1.8 V supply,and it occupies an active die area of 2.24 mm^2.Zongguang Yu Xiaobo Su Zhenhai Chen Jiaxuan Zou Jinghe Wei Hong Zhang Yan Xue 2018Tsinghua Science and Technology2018,23,1:2

8Single-molecule imaging reveals the stoichiometry change of epidermal growth factor receptor during transactivation by β_2-adrenergic receptor显示文摘Stimulation of G protein-coupled receptors(GPCRs) can lead to the transactivation of the epidermal growth factor receptors(EGFR). The cross-communication between the two signaling pathways regulates several important physiological or pathological processes. However, the molecule mechanism underlying EGFR transactivation remains poorly understood. Here, we aim to study the GPCR-mediated EGFR transactivation process using the single-molecule fluorescence imaging and tracking approach.We found that although EGFR existed as monomers at the plasma membrane of resting cells, they became dimers and thus diffused slower following the activation of β2-adrenergic receptor(β2-AR) by isoproterenol(ISO). We further proved thatβ2-AR-mediated changes of EGFR in stoichiometry and dynamics were mediated by Src kinase. 12IL-13/IL-13RA2 signaling promotes colorectal cancer stem cell tumorigenesis by inducing ubiquitinated degradation of p53显示文摘Cancer stem cells(CSCs)are considered tumor-initiating cells and the main drivers of disease progression.Targeting these rare cancer cells,however,remains challenging with respect to therapeutic benefit.Here,we report the up-regulation of IL-13RA2 expression in colorectal cancer(CRC)tissues and spheroid cells.The expression of IL-13RA2 was positively correlated with canonical stemness markers in CRC.We further demonstrated that the level of IL-13 was up-regulated in the serum of CRC patients.Biologically,recombinant IL-13(rIL13)stimulation promoted the sphere formation,proliferation,and migration of CRC cells in vitro and enhanced tumorigenesis in vivo.This phenotype could be reversed by knocking down IL-13RA2.Mechanistically,IL-13 activated autophagy by inducing LC3I/LC3II transformation in CRC-CSCs,which was crucial for the biological functions of IL-13.We further demonstrated that IL-13RA2 acted as a modular link of the E3 ligase UBE3C and the substrate p53 protein,enhancing the interaction of UBE3C and p53,thereby inducing the K48-linked ubiquitination of p53.In conclusion,the IL-13/IL-13RA2 signaling cascade promotes CRC-CSC self-renewal and tumorigenesis by inducing p53 ubiquitination,adding an important layer to the connection between IL-13 and p53,which can be translated into novel targeted therapies.Baoyu He Jing Liang Qianqian Qin Yuqin Zhang Shuo Shi Jinghe Cao Zhixin Zhang Qingli Bie Rou Zhao Li Wei Baogui Zhang Bin Zhang 2024Genes & Diseases2024,11,1:0
13Design of Low-Power Modern Radar SoC Based on ASIX显示文摘With the rapid development in spaceflights and aeroplanes,the demand for low-power and miniaturization techniques has become insistent in modern radar systems. A new framework for low-power modern radar System on a Chip(SoC) based on ASIX core is presented. Pivotal modules and low-power design flows are described in detail. The dynamic clock-distribution mechanism of the power management module and the influence of the chip power are both stressed. This design adopts the SMIC 0.18-?m 1P6M Salicide CMOS process,the area is 7.825mm 7.820 mm,there are approximately 2 million gates and the frequency is 100 MHz. The results show that the modern radar SoC passes the test on modern radar application system and meets the design requirements. The chip incurs power savings of 42.79% during the fore-end phase and 12.77% during the back-end phase. 14Transcriptomic characteristics and impaired immune function of patients who retest positive for SARS-CoV-2 RNA显示文摘Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection,has become a global public health crisis.Some patients who have recovered from COVID-19 subsequently test positive again for SARS-CoV-2 RNA after discharge from hospital.How such retest-positive(RTP)patients become infected again is not known.In this study,30 RTP patients,20 convalescent patients,and 20 healthy controls were enrolled for the analysis of immunological characteristics of their peripheral blood mononuclear cells.We found that absolute numbers of CD4^(+)T cells,CD8^(+)T cells,and natural killer cells were not substantially decreased in RTP patients,but the expression of activation markers on these cells was significantly reduced.The percentage of granzyme B-producing T cells was also lower in RTP patients than in convalescent patients.Through transcriptome sequencing,we demonstrated that high expression of inhibitor of differentiation 1(ID1)and low expression of interferon-induced transmembrane protein 10(IFITM10)were associated with insufficient activation of immune cells and the occurrence of RTP.These findings provide insight into the impaired immune function associated with COVID-19 and the pathogenesis of RTP,which may contribute to a better understanding of the mechanisms underlying RTP.Dongyao Wang Dong Wang Min Huang Xiaohu Zheng Yiqing Shen Binqing Fu Hong Zhao Xianxiang Chen Peng Peng Qi Zhu Yonggang Zhou Jinghe Zhang Zhigang Tian Wuxiang Guan Guiqiang Wang Haiming Wei 2021Journal of Molecular Cell Biology2021,13,10:0
15A 4 Gbps current-mode transmitter for 12-bit 250 MSPS ADC显示文摘A 4 Gbps transmitter for a 12-bit 250 MSPS pipelined ADCs is presented.A low power current mode(CM) output driver with reverse scaling technique is proposed.A high speed,low power combined serializer is implemented to convert 12 bit parallel data into a serial data stream.The whole transmitter is used in a 12-bit250 MSPS pipelined ADC for the digital output buffer and fabricated in 180 nm 1.8 V 1P5M CMOS technology.Test results show that the transmitter provides an eye height greater than 800 mV for data rates of both 2 Gbps and 4 Gbps,the 12-bit 250 MSPS ADC achieves the SNR of 69.92 dBFS and SFDR of 81.17 dB with 20.1 MHz input at full sampling speed.The ADC with the 4 Gbps transmitter consumes the power consumption of 395 mW,where the power consumption of transmitter is 75 mW.The ADC occupies an area of 2.5×3.2 mm^2,where the active area of the transmitter block is 0.5×1.2 mm^2.Zhenhai Chen Zongguang Yu Jinghe Wei Dejin Zhou Xiaobo Su Jiaxuan Zou 2017Journal of Semiconductors2017,38,8:0
16Improved ECG signal compression method for E-healthcare显示文摘For achieving a higher compression ratio(CR)in compression sensing,the time-sparse bio-signals,such as electrocardiograph(ECG),are generally directly filtered via a dynamic or fixed threshold,however,inevitably leading to the loss of critical diagnostic bio-information.We propose a compression scheme to reduce the transmitting loss.Instead of the directly utilizing the original ECG data,the residuals between original and synthetic ECG signals are applied as the input signal.We employ the dynamic model to guarantee the consistency between the synthetic ECG signals waves(P,Q,R,S,and T)and the originals.The feasibility of the proposed method is tested through operating on the recorded ECG signals from a healthy human.During the process of building simulation platform,the sparsity,percentage root mean difference(PRD)versus sampling frequency,and signals reconstruction algorithm are fully taken into account.Before compression,we set the threshold to filter the residual waves,in which utilizing the residuals as input data by setting the thresold as 0.01 mV and 0.08 mV resulted the amount reduction of the transmitting data by 18%and 81.2%,respectively.And the simulation results show that CR can reach 2.75 when the PRD value is less than 9%.Gao Miao Yu Zhiguo Li Qingqing Wei Jinghe Gu Xiaofeng 2019The Journal of China Universities of Posts and Telecommunications2019,26,4:0
17High Precision Mix-Signal Capacitor Mismatch Error Calibration Method for Charge-Domain Pipelined ADC显示文摘A mix-signal high precision capacitor mismatch error calibration method for charge domain pipelined ADCs is proposed. The calibration method calibrates the capacitors one by one based on binary search.Charge errors caused by the capacitor mismatch in and between pipelined sub-stage circuits can be compensated by the proposed calibration method.
