2Development of cell-based pseudovirus entry assay to identify potential viral entry inhibitors and neutralizing antibodies against SARS-CoV-2显示文摘Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)is the causative virus of the coronavirus disease 2019(COVID-19)pandemic.To establish a safe and convenient assay system for studying entry inhibitors and neutralizing antibodies against SARS-CoV-2,we constructed a codon-optimized,full-length C-terminal mutant spike(S)gene of SARS-CoV-2.We generated a luciferase(Luc)-expressing pseudovirus containing the wild-type or mutant S protein of SARS-CoV-2 in the envelope-defective HIV-1 backbone.The key parameters for this pseudovirus-based assay,including the S mutants and virus incubation time,were optimized.This pseudovirus contains a Luc reporter gene that enabled us to easily quantify virus entry into angiotensin-converting enzyme 2(ACE2)-expressing 293T cells.Cathepsin(Cat)B/L inhibitor E64d could significantly block SARS-CoV-2 pseudovirus infection in 293T-ACE2 cells.Furthermore,the SARS-CoV-2 spike pseudotyped virus could be neutralized by sera from convalescent COVID-19 patients or recombinant ACE2 with the fused Fc region of human IgG1.Thus,we developed a pseudovirus-based assay for SARS-CoV-2,which will be valuable for evaluating viral entry inhibitors and neutralizing antibodies against this highly pathogenic virus.Jie Hu Qingzhu Gao Changlong He Ailong Huang Ni Tang Kai Wang 2020Genes & Diseases2020,7,4:15

4A Genomic Variation Map Provides Insights into the Genetic Basis of Spring Chinese Cabbage (Brassica rapa ssp.pekinensis)Selection显示文摘Chinese cabbage is the most consumed leafy crop in East Asian countries.However,premature bolting induced by continuous low temperatures severely decreases the yield and quality of the Chinese cabbage, and therefore restricts its planting season and geographic distribution.In the past 40years,spring Chinese cabbage with strong winterness has been selected to meet the market demand.Here,we report a genome variation map of Chinese cabbage generated from the resequencing data of 194 geographically diverse accessions of three ecotypes.In-depth analyses of the selection sweeps and genome-wide patterns revealed that spring Chinese cabbage was selected from a specific population of autumn Chinese cabbage around the area of Shandong peninsula in northern China.We identified 23 genomic loci that underwent intensive selection,and further demonstrated by gene expression and haplotype analyses that the incorporation of elite alleles of VERNALISATION INSENTIVE 3.1(BrVIN3.1)and FLOWER LOCUS C 1(BrFLC1)is a determinant genetic source of variation during selection.Moreover,we showed that the quantitative response of BrVIN3.1 to cold due to the sequence variations in the cis elements of the BrVlN3.1 promoter significantly contributes to bolting-time variation in Chinese cabbage.Collectively, our study provides valuable insights into the genetic basis of spring Chinese cabbage selection and will facilitate the breeding of bolting-resistant Varieties by molecular-marker-assisted selection,transgenic or gene editingapproaches.Tongbing Su Weihong Wang Peirong Li Bin Zhang Pan Li Xiaoyun Xin Honghe Sun Yangjun Yu Deshuang Zhang Xiuyun Zhao Changlong Wen Gang Zhou Yuntong Wang Hongkun Zheng Shuancang Yu Fenglan Zhang 2018Molecular Plant2018,11,11:6
5Analysis of the Global Swell Distributions Using ECMWF Re-Analyses Wind Wave Data显示文摘The existence of three well-defined tongue-shaped zones of swell dominance,termed as 'swell pools',in the Pacific,the Atlantic and the Indian Oceans,was reported by Chen et al.(2002)using satellite data.In this paper,the ECMWF Re-analyses wind wave data,including wind speed,significant wave height,averaged wave period and direction,are applied to verify the existence of these swell pools.The swell indices calculated from wave height,wave age and correlation coefficient are used to identify swell events.The wave age swell index can be more appropriately related to physical processes compared to the other two swell indices.Based on the ECMWF data the swell pools in the Pacific and the Atlantic Oceans are confirmed,but the expected swell pool in the Indian Ocean is not pronounced.The seasonal variations of global and hemispherical swell indices are investigated,and the argument that swells in the pools seemed to originate mostly from the winter hemisphere is supported by the seasonal variation of the averaged wave direction.The northward bending of the swell pools in the Pacific and the Atlantic Oceans in summer is not revealed by the ECMWF data.The swell pool in the Indian Ocean and the summer northward bending of the swell pools in the Pacific and the Atlan-tic Oceans need to be further verified by other datasets.ZHANG Jie WANG Weili GUAN Changlong 2011Journal of Ocean University of China2011,10,4:6
6Combined genomic, transcriptomic, and metabolomic an alyses provide in sights into chayote (Sechium edule) evolution and fruit development显示文摘Chayote(Sechium edule)is an agricultural crop in the Cucurbitaceae family that is rich in bioactive components.To enhance genetic research on chayote,we used Nanopore third-generation sequencing combined with Hi-C data to assemble a draft chayote genome.A chromosome-level assembly anchored on 14 chromosomes(N50 contig and scaffold sizes of 8.40 and 46.56 Mb,respectively)estimated the genome size as 606.42 Mb,which is large for the Cucurbitaceae,with 65.94%(401.08 Mb)ofthe genome comprising repetitive sequences;28,237 protein-coding genes were predicted.Comparative genome analysis indicated that chayote and snake gourd diverged from sponge gourd and that a whole-genome duplication(WGD)event occurred in chayote at 25±4 Mya.Transcriptional and metabolic analysis revealed genes involved in fruit texture,pigment,fl avor,fl avonoids,antioxidants,and plant hormones during chayote fruit development.The analysis of the genome,transcriptome,and metabolome provides insights into chayote evolution and lays the groundwork for future research on fruit and tuber development and genetic improvements in chayote.Anzhen Fu Qing Wang Jianlou Mu Lili Ma Changlong Wen Xiaoyan Zhao Lipu Gao Jian Li Kai Shi Yunxiang Wang Xuechuan Zhang Xuewen Zhang Fengling Wang Donald Grierson Jinhua Zuo 2021Horticulture Research2021,8,1:6
7Pain threshold, anxiety and other factors affect intensity of postoperative pain in gastric cancer patients: A prospective cohort study显示文摘Objective: This prospective cohort study explored factors related to postoperative pain in gastric cancer patients.Methods: A total of 236 patients who underwent gastrectomy were enrolled. All patients enrolled in the study completed the Hospital Anxiety and Depression Scale(HADS) questionnaire and Life Orientation Test-Revised(LOT-R) questionnaire on the day before surgery. Heat pain threshold(HPT), cold pain threshold(CPT) and pressure pain threshold(PPT) were measured for all patients one day prior to surgery and demographic details were collected. All patients were connected to a patient-controlled intravenous analgesia(PCIA) pump at the end of the surgery. The occurrence of postoperative pain was used as a dependent variable, and multivariate logistic regression analyses were conducted to screen for factors affecting postoperative pain.Results: In total, 83 patients(35.2%) had postoperative pain. Body mass index(BMI) ≥28 kg/m^(2) [odds ratio(OR): 2.67;95% confidence interval(95% CI): 1.07-6.67], total gastrectomy(OR: 2.64;95% CI: 1.42-4.91),preoperative anxiety score ≥8(OR: 2.37;95% CI: 1.12-5.02), heat pain threshold ≤4.9 s(OR: 2.14;95% CI:1.06-4.32), pressure pain threshold ≤4 g(OR: 2.05;95% CI: 1.05-4.03), and female gender(OR: 1.99;95% CI:1.04-3.83) were risk factors for postoperative pain.Conclusions: Obesity, wide range of gastrectomy, high preoperative anxiety, low HPT and PPT, and female gender are associated with increased risk for postoperative pain.Hongyu Tan Jin Wei Shuo Li Ling Yu Hongwei Sun Ke Ji Yinkui Wang Changlong Li 2021Chinese Journal of Cancer Research2021,33,3:5

9Exploration and functionalization of M1-macrophage extracellular vesicles for effective accumulation in glioblastoma and strong synergistic therapeutic effects显示文摘Glioblastoma multiforme(GBM)is a highly aggressive brain tumor with an extremely low survival rate.New and effective approaches for treatment are therefore urgently needed.Here,we successfully developed M1-like macrophage-derived extracellular vesicles(M1EVs)that overcome multiple challenges via guidance from two macrophage-related observations in clinical specimens from GBM patients:enrichment of M2 macrophages in GBM;and origination of a majority of infiltrating macrophage from peripheral blood.To maximize the synergistic effect,we further functionalized the membranes of M1EVs with two hydrophobic agents(the chemical excitation source CPPO(C)and the photosensitizer Ce6(C))and loaded the hydrophilic hypoxiaactivated prodrug AQ4N(A)into the inner core of the M1EVs.After intravenous injection,the inherent nature of M1-derived extracellular vesicles CCA-M1EVs allowed for blood-brain barrier penetration,and modulated the immunosuppressive tumor microenvironment via M2-to-M1 polarization,which increased hydrogen peroxide(H_(2)O_(2))levels.Furthermore,the reaction between H_(2)O_(2) and CPPO produced chemical energy,which could be used for Ce6 activation to generate large amounts of reactive oxygen species to achieve chemiexcited photodynamic therapy(CDT).As this reaction consumed oxygen,the aggravation of tumor hypoxia also led to the conversion of non-toxic AQ4N into toxic AQ4 for chemotherapy.Therefore,CCA-M1EVs achieved synergistic immunomodulation,CDT,and hypoxia-activated chemotherapy in GBM to exert a potent therapeutic effect.Finally,we demonstrated the excellent effect of CCA-M1EVs against GBM in cell-derived xenograft and patient-derived xenograft models,underscoring the strong potential of our highly flexible M1EVs system to support multi-modal therapies for difficult-to-treat GBM.Xiaojun Wang Hui Ding Zongyang Li Yaonan Peng Hui Tan Changlong Wang Guodong Huang Weiping Li Guanghui Ma Wei Wei 2022Signal Transduction and Targeted Therapy2022,7,4:3

11The genome and transcriptome analysis of snake gourd provide insights into its evolution and fruit development and ripening显示文摘Snake gourd(Trichosanthes anguina L.),which belongs to the Cucurbitaceae family,is a popular ornamental and food crop species with medicinal value and is grown in many parts of the world.Although progress has been made in its genetic improvement,the organization,composition,and evolution of the snake gourd genome remain largely unknown.Here,we report a high-quality genome assembly for snake gourd,comprising 202 contigs,with a total size of 919.8 Mb and an N50 size of 20.1 Mb.These findings indicate that snake gourd has one of the largest genomes of Cucurbitaceae species sequenced to date.The snake gourd genome assembly harbors 22,874 protein-coding genes and 80.0%of the genome consists of repetitive sequences.Phylogenetic analysis reveals that snake gourd is closely related to sponge gourd but diverged from their common ancestor~33–47 million years ago.The genome sequence reported here serves as a valuable resource for snake gourd genetic research and comparative genomic studies in Cucurbitaceae and other plant species.In addition,fruit transcriptome analysis reveals the candidate genes related to quality traits during snake gourd fruit development and provides a basis for future research on snake gourd fruit development and ripening at the transcript level.Lili Ma Qing Wang Jianlou Mu Anzhen Fu Changlong Wen Xiaoyan Zhao Lipu Gao Jian Li Kai Shi Yunxiang Wang Xuewen Zhang Xuechuan Zhang Zhangjun Fei Donald Grierson Jinhua Zuo 2020Horticulture Research2020,7,1:2
12Molecularly tagged genes and quantitative trait loci in cucumber with recommendations for QTL nomenclature显示文摘Cucumber,Cucumis sativus L.(2n=2x=14),is an important vegetable crop worldwide.It was the first specialty crop with a publicly available draft genome.Its relatively small,diploid genome,short life cycle,and selfcompatible mating system offers advantages for genetic studies.In recent years,significant progress has been made in molecular mapping,and identification of genes and QTL responsible for key phenotypic traits,but a systematic review of the work is lacking.Here,we conducted an extensive literature review on mutants,genes and QTL that have been molecularly mapped or characterized in cucumber.We documented 81 simply inherited trait genes or major-effect QTL that have been cloned or fine mapped.For each gene,detailed information was compiled including chromosome locations,allelic variants and associated polymorphisms,predicted functions,and diagnostic markers that could be used for marker-assisted selection in cucumber breeding.We also documented 322 QTL for 42 quantitative traits,including 109 for disease resistances against seven pathogens.By alignment of these QTL on the latest version of cucumber draft genomes,consensus QTL across multiple studies were inferred,which provided insights into heritable correlations among different traits.Through collaborative efforts among public and private cucumber researchers,we identified 130 quantitative traits and developed a set of recommendations for QTL nomenclature in cucumber.This is the first attempt to systematically summarize,analyze and inventory cucumber mutants,cloned or mapped genes and QTL,which should be a useful resource for the cucurbit research community.Yuhui Wang Kailiang Bo Xingfang Gu Junsong Pan Yuhong Li Jinfeng Chen Changlong Wen Zhonghai Ren Huazhong Ren Xuehao Chen Rebecca Grumet Yiqun Weng 2020Horticulture Research2020,7,1:2

18Influences of Reservoir Heterogeneity and Anisotropy on CO_2 Sequestration and Heat Extraction for CO_2-Based Enhanced Geothermal System显示文摘Enhanced geothermal systems(EGS) have a great potential to extract geothermal energy and have attracted much interest. In this paper, based on a 3D thermal-hydrologic model considering CO_2 sequestration, the influences of reservoir heterogeneity and anisotropy on CO_2 sequestration and heat extraction in CO_2-based EGS are investigated. Different heterogeneous reservoirs and homogeneous reservoir are compared, and different ratios among reservoir permeability components are compared. The results show that greater reservoir heterogeneity enhances CO_2 sequestration and restrains heat extraction. Higher ratio between horizontal(x and y directions) and vertical permeability components enhances CO_2 sequestration and heat extraction, and vertical permeability component has a little effect. With the increasing ratio between x-directional(perpendicular to the line of the injection well and the production well) and y-directional(perpendicular to x direction) reservoir permeability components(i.e. kx:ky), both CO_2 sequestration amount and steady-state heat extraction rate first increase and then decrease, and thermal breakthrough time increases, showing that there exists an optimum kx:ky, which is about 1:1.
