1Effects of flavonoids from Allium mongolicum Regel on growth performance and growth-related hormones in meat sheep显示文摘This study was conducted to investigate the effects of different doses of flavonoids from Allium mongolicum Regel on the production performance and neuroendocrine hormones in meat sheep and to determine the optimum dosage of Allium mongolicum Regel flavonoids to add to the basal diet of dry lotfeeding meat sheep. Sixty meat sheep(initial body weight = 39.9 ± 3.2 kg; 6-month-old) were randomly assigned to 4 groups(15 sheep per group). The sheep in the control group were fed a basal diet, and the 3 experimental groups were fed the basal diet supplemented with flavonoids at 11.22 and 33 mg/kg. Blood samples were collected via the jugular vein at d 0.15, 30, 45, and 60 to determine the neuroendocrine hormone levels. The fasting weight of the sheep was measured during the experimental period, and feed offered and refusals were recorded daily. The basal diet supplemented with flavonoids from 11 to 33 mg/kg significantly increased the daily weight gain and average daily feed intake(P<0.05) and significantly decreased the feed conversion ratio(P<0.05), but there were no differences among the supplementation groups(P>0.05). Starting on d 30, the growth hormone(GH) and insulin-like growth factor-1(IGF-1)levels in the sera of the sheep in the supplementation groups increased significantly(P<0.05), and the increases occurred in a time-dependent manner. Compared with control group, after d 30, the serum corticosterone(CORT) levels were reduced in the sheep that consumed the basal diet supplemented with22 mg/kg flavonoids(P<0.05), but among the other experimental groups, there was a non-significant effect(P>0.05). The serum adrenocorticotropic hormone(ACTH) levels were increased by the supplementation of flavonoids, but compared with the control group, the effect was not significant. The basal diet supplemented with flavonoids at levels from 11 to 33 mg/kg had a significant effect on the production performance and neuroendocrine hormone levels of meat sheep, and the effect occurred in a time-dependent manner. 2Exosome-based small RNA delivery:Progress and prospects显示文摘RNA interfering(RNAi), mediated by small interfering RNAs and microRNAs, is currently one of the most promising tools of gene therapy. Small RNAs are capable of inducing specific post-transcriptional gene silencing, providing a potentially effective platform for the treatment of a wide array of diseases. However, similar to other nucleic acid-based drugs,the major hurdle of RNAi therapy is lack of efficient and non-immunogenic delivery vehicles. Currently, viruses, synthetic polymers, and lipid-based carriers are among the most widely studied vehicles for small RNA delivery. However, many drawbacks are reported to be associated with these delivery vehicles. There is a pressing need to replace them with more efficient and better-tolerated approaches. Exosomes secreted from the endocytic compartment of live cells, are a subtype of endogenous extracellular vesicles that transfer genetic and biochemical information among different cells, thus playing an important role in cellcell communication. Recently, accumulating attention has been focused on harnessing exosomes as nanaocarriers for small RNAs delivery. Due to their natural role in shuttling endogenous nucleic acid in our body, exosomes may exhibit higher delivery efficiency, lower immunogenicity, and better compatibility than existing foreign RNA carriers. Importantly,exosomes own intrinsic homing capacity that can guide small RNAs across natural membranous barriers. Moreover, such a capacity can be further improved by adding appropriate targeting moieties. 3Formalin-induced astrocyte glutamate-glutamine cycle involved in rat spinal cord central sensitization显示文摘BACKGROUND:Central sensitization,a state of increased excitability of nociceptive neurons in the spinal dorsal horn following peripheral tissue injury and/or inflammation,is an important mechanism underlying hyperalgesia and neuropathic pain.Participation of the glutamate-glutamine cycle in central sensitization of the spinal cord remains poorly understood. OBJECTIVE:To determine whether the astrocyte-neuronal glutamate-glutamine cycle is involved in formalin-induced central sensitization in the spinal cord. DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment was performed at the Institute of Orthopedics,Second Hospital,Lanzhou University,China from September 2007 to August 2008. MATERIALS:Methionine sulfoximine(MSO,0.1 mmol/L),glutamine(0.25 mmol/L),and formalin were used for this study. METHODS:A total of 43 male,Sprague Dawley rats,aged 4 months,were randomly assigned to a sham operation group(n = 6) and a model group(n = 37).Rats in the model group received intrathecal infusion in the spinal cord.7 days later,37 model rats were randomly divided into PBS, MSO,glutamine,MSO + glutamine and formalin subcutaneous injection alone groups.The PBS, MSO,glutamine,MSO + glutamine groups were respectively intrathecally injected with PBS,MSO, glutamine,MSO + glutamine(50μL each),and then infused with 10μL of saline.Rats from the sham operation group were not subjected to intrathecal infusion in the spinal cord.At 15 minutes after intrathecal injection,a rat model of formalin-induced inflammatory pain was established by subcutaneous injection of 5%formalin(50μL) in the left hindpaw. MAIN OUTCOME MEASURES:Changes in spontaneous nociceptive behavior(licking/biting or flinching) were observed following formalin injection into the rat hindpaw. RESULTS:Compared with the PBS group,duration of licking/biting was significantly shortened,and flinching frequency was significantly diminished in the MSO group(P<0.05).Compared with the MSO group,duration of licking/biting was significantly prolonged,and flinching frequency was significantly increased in the MSO + glutamine group(P<0.05).There was no significant difference in inflammatory pain behaviors among the sham operation,PBS,glutamine,MSO + glutamine,and formalin subcutaneous injection alone groups(P>0.05). 4Dual-modulation difference stimulated emission depletion microscopy to suppress the background signal显示文摘Stimulated emission depletion(STED)nanoscopy is one of the most well-developed nanoscopy techniques that can provide subdiffraction spatial resolution imaging.Here,we introduce dual-modulation difference STED microscopy(dmdSTED)to suppress the background noise in traditional STED imaging.By applying respective time-domain modulations to the two continuous-wave lasers,signals are distributed discretely in the frequency spectrum and thus are obtained through lock-in demodulation of the corresponding frequencies.The background signals can be selectively eliminated from the effective signal without compromise of temporal resolution.We used nanoparticle,fixed cell,and perovskite coating experiments,as well as theoretical demonstration,to confirm the effectiveness of this method.We highlight dmdSTED as an idea and approach with simple implementation for improving the imaging quality,which substantially enlarges the versatility of STED nanoscopy.Wensheng Wang Chuankang Li Zhengyi Zhan Zhimin Zhang Yubing Han Cuifang Kuang Xu Liu 2022Advanced Photonics2022,4,4:1

6Intracellular distribution and internalization pathways of guanidinylated bioresponsive poly(amido amine)s in gene delivery显示文摘Guanidinylated bioresponsive poly(amido amine)s polymers, CAR-CBA and CHL-CBA, weresynthesized by Michael-type addition reaction between guanidine hydrochloride(CAR) orchlorhexidine(CHL) and N,N-cystaminebisacrylamide(CBA). Previous studies have shownthat both polymers had high transfection efficiencies as gene delivery carriers. In this study,we investigated the nucleolus localization abilities and cellular internalization pathways ofthese two polymers in gene delivery. Each polymer condensed plasmid DNA(p DNA) andformed nanoparticle complexes, and then their transfection studies were performed inMCF-7 cells. Both complexes were found enriched in nucleolus after cellular transfection,and their transfection efficiencies were significantly improved when transfection was per-formed on MCF-7 cells arrested at M phase. The transfection efficiency of CAR-CBA-pDNAwas inhibited by chlorpromazine, and cell endosomes were disrupted after being exposedto CAR-CBA-pDNA. In regards to CHL-CBA-pDNA, its transfection efficiency was not affected by three types of endocytosis inhibitors used in the study, and CHL-CBA-pDNA showed no effect on endosomes. Cellular lactate dehydrogenase release and membrane morphology were changed after cells were transfected by the two complexes. The results indicated that both CAR-CBA and CHL-CBA polymers demonstrated good nucleolus localization abilities. It was beneficial for transfection when cells were arrested at M phase. 9Design, Synthesis, Biological Activity and Molecular Docking Study of Coumarin Derivatives Bearing 2-MethyIbiphenyl Moiety显示文摘A hybrid pharmacophore approach was used to design and synthesize a series of coumarin derivatives bearing 2-methylbiphenyl moiety, which were evaluated for their in vitro anticancer activities against four cancer cell lines(MCF-75 A549, H460 and HT29) and PD-1/PD-L1 inhibitory activities. Moreover, several compounds with excellent anticancer activities were selected to evaluate the cytotoxicities against one normal cell line(HEK-293). The most promising compound llo showed the best anticancer activities against the four tested cancer cell lines with the IC50 values of 6.45, 8.65, 6,57 and 8.13 gmol/L, respectively, and displayed weak cytotoxicity on the normal cell(HEK-293). Furthermore, screening of PD-1 /PD-L1 inhibitory activity revealed that compound llo could effectively inhibit the binding of PD-1/PD-L1, and the binding interactions of compound llo with PD-L1 protein were explored by molecular docking. 10A Survey Report on Sugarcane Production in Guangxi Zhuang Autonomous Region in 2013显示文摘In order to fully understand sugarcane production in Guangxi in 2013,the sample survey combined with data collection and field survey was carried out in sugarcane production areas in Nanning,Hechi,Liuzhou,Chongzuo,Baise and Laibin from May 28 to June 36 in2013. It was found that the sugarcane growing area in Guangxi in 2013 declined compared with that in 2012; the rate of emergence remained the same as previous year,and as for the plant number of plant crop and ratoon crop per hectare,there was a decline of 4000 and 2000 seedlings respectively compared with the figure in 2012; borer damage rate declined on the whole while the incidence of sugarcane smut increased.Due to the growth in the cost of planting,the growers were less enthusiastic for management. 11Preparation of highly stable diclofenac potassium pellet with microcrystalline cellulose by extrusionespheronization显示文摘The main purpose of this study is to prepare highly stable diclofenac potassium(DP)pellet with microcrystalline cellulose(MCC)by extrusionespheronization.Using MCC,DP pellets were prepared and the stability was investigated.Related compounds of DP pellets were analyzed by High Pressure Liquid Chromatography(HPLC).After stability test of 60℃/75%RH for 10 d,the values of two main related compounds were 0.94%and 2.17%,respectively.Compatibility tests show that instability of DP was mainly caused by MCC.To improve the stability of DP in presence of MCC,different kinds of stabilizers were investigated.Upon addition of 1.5%(w/w)sodium hydroxide,the primary related compound of pellets was reduced to be 0.159%after stability test of 60℃/75%RH for 50 d.This study demonstrated that MCC induced decomposition of DP upon exposure to moisture could be prohibited by addition of sodium hydroxide.The mechanisms were discussed and residual hydroxyl free radicals in excipients were responsible for decomposition of DP.Finally,this formulation of DP is highly stable with sustained-release behavior.Yueqi Wang Hao Huang Chungang Zhang Yilin Tang Jinzhuo Li Xing Tang Cuifang Cai 2013Asian Journal of Pharmaceutical Sciences2013,8,6:0
12Observation on the Effect of Spontaneous Ventilation Single-Port Thoracoscopy and Conventional Thoracoscopy on T Lymphocyte Subsets and Inflammatory Factors in Patients with Non-Small Cell Lung Cancer显示文摘Objective:To explore the effects of two types of surgical approaches,spontaneous ventilation single-port thoracoscopy and conventional thoracoscopy,on T lymphocyte subsets and inflammatory factors in patients with non-small cell lung cancer.Methods:Fifty-eight patients with non-small cell lung cancer who underwent surgery in the Affiliated Hospital of Hebei University from January 2020 to December 2021 were selected as the study subjects.All of them underwent standard lobectomy and lymph node dissection.Among them,32 patients who underwent spontaneous ventilation single-port thoracoscopy were included in the study group,while 26 patients who underwent conventional thoracoscopy were included in the control group.The perioperative related indices,lung function,immune factor levels,and inflammatory factor levels were compared between the two groups.Results:In the study group,the perioperative incision length,intraoperative blood loss,operation time,and number of lymph node dissections were 4.28±2.01,77.89±12.02,87.21±16.11,and 20.69±4.45,respectively.In the control group,the perioperative incision length,intraoperative blood loss,operation time,and number of lymph node dissections were 6.32±2.45,84.98±16.98,95.88±14.89,and 21.45±4.36,respectively.There was no statistical difference between the study group and the control group in the number of lymph node dissections and intraoperative blood loss;the lung function in the study group was significantly better than that in the control group;the levels of T cell subsets CD4+,CD8+,CD4+/CD8+,CD16+CD56+of the study group were 46.36±5.87,30.98±4.12,1.19±0.23,and 17.41±6.25,respectively,while those in the control group were 35.78±4.12,34.14±3.87,1.04±0.24,and 12.45±5.56,respectively;the levels of immunoglobulin IgG,IgM,and IgA of the study group were 10.45±2.14,1.21±0.24,and 1.26±0.25,respectively,while those of the control group were 8.78±1.78,1.06±0.12,and 1.06±0.26,respectively,with statistical differences;the levels of CRP,IL-6,IL-8,and TNF-αof the control group were 21.87±4.26,98.01±9.58,111.03±9.96,and 123.05±9.77,respectively,while those of the study group were 19.47±3.89,89.12±8.96,104.32±9.12,and 112.98±9.16,respectively,in which the comparison was statistically significant.Conclusion:In the surgical treatment of non-small cell lung cancer,spontaneous ventilation single-port thoracoscopic surgery has several advantages,which include less trauma and bleeding.It reduces the impact of surgical trauma on the immune function of the body,improves the postoperative lung function and inflammatory stress of the body,as well as accelerates the recovery of patients.It is an alternative to open lung lobectomy for the treatment of lung cancer.Hefei Li Cuifang Liu Yanan Wang Shaoyong Dong Haijiang Liao Ce Li Xuguang Zhang Mo Deng 2022Proceedings of Anticancer Research2022,6,5:0
13Hot-melt sub-and outercoating combined with enteric aqueous coating to improve the stability of aspirin tablets显示文摘Aspirin is apt to hydrolyze. In order to improve its stability, a new method has been developed involving the application of hot-melt sub-and outercoating combined with enteric aqueous coating. The main aim was to investigate the influence of these factors on the stability of ASA and understand how they work. Satisfactory storage stability were obtained when the aspirin tablet core coated with Eudragit L30D55 film was combined with glycerin monostearate(GMS) as an outercoat. Hygroscopicity testing indicated that the moisture penetrating into the tablet may result in a significant change in the physical properties of the coating film observed by scanning electron microscopy. Investigation of the compatibility between the drug and film excipients shows that the talc and methacrylic acid had a significant catalytic effect on ASA. A hypothesis was proposed that the hydrolysis of ASA enteric coated tablets(ASA-ECT) was mostly concentrated in the internal film and the interfaces between the film and tablet core. In conclusion, hot-melt coating technology is an alternative to subcoating or outercoating. 14End-to-end computational design for an EUV solar corona multispectral imager with stray light suppression显示文摘An extreme ultraviolet solar corona multispectral imager can allow direct observation of high temperature coronal plasma,which is related to solar flares,coronal mass ejections and other significant coronal activities.This manuscript proposes a novel end-to-end computational design method for an extreme ultraviolet(EUV)solar corona multispectral imager operating at wavelengths near 100 nm,including a stray light suppression design and computational image recovery.To suppress the strong stray light from the solar disk,an outer opto-mechanical structure is designed to protect the imaging component of the system.Considering the low reflectivity(less than 70%)and strong-scattering(roughness)of existing extreme ultraviolet optical elements,the imaging component comprises only a primary mirror and a curved grating.A Lyot aperture is used to further suppress any residual stray light.Finally,a deep learning computational imaging method is used to correct the individual multi-wavelength images from the original recorded multi-slit data.In results and data,this can achieve a far-field angular resolution below 7',and spectral resolution below 0.05 nm.The field of view is±3 R☉along the multi-slit moving direction,where R☉represents the radius of the solar disk.The ratio of the corona's stray light intensity to the solar center's irradiation intensity is less than 10-6 at the circle of 1.3 R☉.Jinming Gao Yue Sun Yinxu Bian Jilong Peng Qian Yu Cuifang Kuang Xiangzhao Wang Xu Liu Xiangqun Cui 2024Astronomical Techniques and Instruments2024,1,1:0
15Synthesis of lithium and aluminum complexes supported by [OC(Bu^t)CHP(Ph_2)=NBu^t]^-ligand and catalysis of [R_2Al{OC(Bu^t)-CHP(Ph_2)=NBu^t}] (R=Me,Et) and [Me_2Al{1-{OC(Ph)CH}-3-R^1-5-MeC_3HN_2}] (R^1=Me,Bu^t) in the ring-opening polymerization of ε-capr显示文摘A series of lithium and aluminum complexes bearing [OC(But)CHP(Ph2)=NBut]-ligand were synthesized and characterized. Reaction of ButC(O)CH2Br with Ph2PNHBut afforded [Ph2P(NHBut)CH2C(O)But]+Br-(1). Treatment of 1 with excess of NaH in THF generated ligand precursor Ph2P(CH2C(O)But)=NBut (2). Reaction of 2 with AlR3 (R = Me,Et) gave N,O-chelate aluminum complexes [R2Al{OC(But)CHP(Ph2)=NBut}] (3,R = Me; 4,R = Et). Lithiation of 2 with an equiv. of LiBun formed lithium complex [Li{OC(But)CHP(Ph2)=NBut}] (5). Reaction of the lithium complex with an equiv. of AlCl3 yielded [Cl2Al{OC(But) CHP(Ph2)=NBut}] (6). Complex 6 was also obtained by reaction of 3 with an equiv. of AlCl3. Compounds 2-6 were characterized by NMR spectroscopy,elemental analysis and single crystal X-ray diffraction techniques (for 2,3 and 6).
