1Newborn screening with targeted sequencing:a multicenter investigation and a pilot clinical study in China显示文摘Different newborn screening(NBS) programs have been practiced in many countries since the 1960 s. It is of considerable interest whether next-generation sequencing is applicable in NBS. We have developed a panel of 465 causative genes for 596 early-onset, relatively high incidence, and potentially actionable severe inherited diseases in our Newborn Screening with Targeted Sequencing(NESTS) program to screen 11,484 babies in 8 Women and Children’s hospitals nationwide in China retrospectively. The positive rate from preliminary screening of NESTS was 7.85%(902/11,484). With 45.89%(414/902) follow-up of preliminary positive cases, the overall clinically confirmative diagnosis rate of monogenic disorders was 12.07%(50/414), estimating an average of 0.95%(7.85% × 12.07%) clinical diagnosis rate, suggesting that monogenic disorders account for a considerable proportion of birth defects. The disease/gene spectrum varied in different regions of China. NESTS was implemented in a hospital by screening 3923 newborns to evaluate its clinical application. The turn-around time of a primary report, including the sequencing period of < 7 days, was within 11 days by our automatic interpretation pipeline. 2Characteristics of distribution of Mycobacterium tuberculosis lineages in China显示文摘The genotyping methods of Mycobacterium tuberculosis would dramatically improve our understanding of the molecular epidemiology of tuberculosis. 3,929 isolates, from a National Survey of Drug-Resistant Tuberculosis in 2007 in China, were successfully genotyped by large sequence polymorphisms and 15 loci variable number tandem repeats. We found that 2,905(2,905/3,929, 73.9%) cases belonged to Lineage 2, dominated in the east and central regions, 975 cases(975/3,929, 24.8%) were Lineage 4, highly prevailed in the west regions, and 36 and 13 cases were Lineage 3 and Lineage 1, respectively. We also explored the associations between lineages(Lineage 2 vs. Lineage 4) and clinical characteristics by logistic regression. For Lineage 2, the risk factors were Han-ethnicity population and fever. However, for Lineage 4, they were occupation(farmer), and degree of education(non-literate). 14The heterogeneous immune landscape between lung adenocarcinoma and squamous carcinoma revealed by single-cell RNA sequencing显示文摘A thorough interrogation of the immune landscape is crucial for immunotherapy strategy selection and prediction of clinical responses in non-small-cell lung cancer(NSCLC)patients.Single-cell RNA sequencing(scRNA-seq)techniques have prompted the opportunity to dissect the distinct immune signatures between lung adenocarcinoma(LUAD)and lung squamous cell carcinoma(LUSC),the two major subtypes of NSCLC.Here,we performed scRNA-seq on 72,475 immune cells from 40 samples of tumor and matched adjacent normal tissues spanning 19 NSCLC patients,and drew a systematic immune cell transcriptome atlas.Joint analyses of the distinct cellular compositions,differentilly expressed genes(DEGs),cell-cell interactions,pseudotime trajectory,transcriptomic factors and prognostic factors based on The Cancer Genome Atlas(TCGA),revealed the central roles of cytotoxic and effector T and NK cells and the distinct functional macrophages(Mo)subtypes in the immune microenvironment heterogeneity between LUAD and LUSC.The dominant subtype of Mo was FABP4-Mp in LUAD and SPP1-Mo in LUSC.Importantly,we identifieda novel lymphocyte-related Mo cluster,which we named SELENOP-Mo,and further established its antitumor role in both types,especially in LUAD.Our comprehensive depiction of the immune heterogeneity and definition of Mp clusters could help design personalized treatment for lung cancer patients in clinical practice.Chengdi Wang Qiuxiao Yu Tingting Song Zhoufeng Wang Lujia Song Ying Yang Jun Shao Jingwei Li Yinyun Ni Ningning Chao Li Zhang Weimin Li 2022Signal Transduction and Targeted Therapy2022,7,9:1

17The top cited articles on glioma stem cells in Web of Science显示文摘BACKGROUND: Glioma is the most common intracranial tumor and has a poor patient prognosis. The presence of brain tumor stem cells was gradually being understood and recognized, which might be beneficial for the treatment of glioma. OBJECTIVE: To use bibliometric indexes to track study focuses on glioma stem cell, and to investigate the relationships among geographic origin, impact factors, and highly cited articles indexed in Web of Science. METHODS: A list of citation classics for glioma stem cells was generated by searching the database of Web of Science-Expanded using the terms 'glioma stem cell' or 'glioma, stem cell'' or 'brain tumor stem cell'. The top 63 cited research articles which were cited more than 100 times were retrieved by reading the abstract or full text if needed. Each eligible article was reviewed for basic information on subject categories, country of origin, journals, authors, and source of journals. Inclusive criteria: (1) articles in the field of glioma stem cells which was cited more than 100 times; (2) fundamental research on humans or animals, clinical trials and case reports; (3) research article; (4) year of publication: 1899-2012; and (5) citation database: Science Citation Index-Expanded. Exclusive criteria: (1) articles needing to be manually searched or accessed only by telephone; (2) unpublished articles; and (3) reviews, conference proceedings, as well as corrected papers. RESULTS: Of 2 040 articles published, the 63 top-cited articles were published between 1992 and 2010. The number of citations ranged from 100 to 1 754, with a mean of 280 citations per article. These citation classics came from nineteen countries, of which 46 articles came from the United States. Duke University and University of California, San Francisco led the list of classics with seven papers each. The 63 top-cited articles were published in 28 journals, predominantly Cancer Research and Cancer Cell, followed by Cell Stem Cell and Nature. CONCLUSION: Our bibliometric analysis provides a historical perspective on the progress of glioma stem cell research. 20Efficacy of early prone or lateral positioning in patients with severe COVID-19:a single-center prospective cohort显示文摘Background:Position intervention has been shown to improve oxygenation,but its role in non-invasively ventilated patients with severe COVID-19 has not been assessed.The objective of this study was to investigate the efficacy of early position intervention on non-invasively ventilated patients with severe COVID-19.Methods:This was a single-center,prospective observational study in consecutive patients with severe COVID-19 managed in a provisional ICU at Renmin Hospital of Wuhan University from 31 January to 15 February 2020.Patients with chest CT showing exudation or consolidation in bilateral peripheral and posterior parts of the lungs were included.Early position intervention(prone or lateral)was commenced for>4 hours daily for 10 days in these patients,while others received standard care.Results:The baseline parameters were comparable between the position intervention group(n=17)and the standard care group(n=35).Position intervention was well-tolerated and increased cumulative adjusted mean difference of SpO_(2)/FiO_(2)(409,95%CI 86 to 733)and ROX index(26,95%CI 9 to 43)with decreased Borg scale(−9,95%CI−15 to−3)during the first 7 days.It also facilitated absorption of lung lesions and reduced the proportion of patients with high National Early Warning Score 2(≥7)on days 7 and 14,with a trend toward faster clinical improvement.Virus shedding and length of hospital stay were comparable between the two groups.Conclusions:This study provides the first evidence for improved oxygenation and lung lesion absorption using early position intervention in non-invasively ventilated patients with severe COVID-19,and warrants further randomized trials.Zhong Ni Kaige Wang Ting Wang Yuenan Ni Wei Huang Ping Zhu Tao Fan Ye Wang Bo Wang Jun Deng Zhicheng Qian Jiasheng Liu Wenhao Cai Shanling Xu Yu Du Gang Wang Zongan Liang Weimin Li Jianfei Luo Fengming Luo Dan Liu 2020Precision Clinical Medicine2020,3,4:0
