| 1 | Two soybean bHLH factors regulate response to iron deficiency显示文摘Iron is an indispensable micronutrient for plant growth and development. Limited bioavailability of Fe in the soil leads to iron deficiency chlorosis in plants and yield loss. In this study, two soybean basic helix-loophelix transcription factors, Gmb HLH57 and Gmb HLH300,were identified in response to Fe-deficiency. Both transcription factors are expressed in roots and nodules,and are induced by Fe deficiency; these patterns were confirmed in transgenic hairy roots expressing constructs of the endogenous promoters fused to a GUS reporter gene. Bimolecular fluorescence complementation, yeast two-hybrid and coimmunoprecipitation(co-IP) assays indicated a physical interaction between Gmb HLH57 and Gmb HLH300. Studies on transgenic soybeans Reseoverexpressing Gmb HLH57 and Gmb HLH300 revealed that overexpression of each transcription factor, alone,results in no change of the responses to Fe deficiency,whereas overexpression of both transcription factors upregulated the downstream Fe uptake genes and increased the Fe content in these transgenic plants.Compared to wild type, these double overexpression transgenic plants were more tolerant to Fe deficiency.Taken together, our findings establish that Gmb HLH57 and Gmb HLH300 are important transcription factors involved in Fe homeostasis in soybean. | Lin Li Wenwen Gao Qi Peng Bin Zhou Qihui Kong Yinghui Ying Huixia Shou | 2018 | Journal of Integrative Plant Biology2018,60,7: | 10 |
| 2 | Development and utilization of a new chemically-induced soybean library with a high mutation density显示文摘Mutagenized populations have provided important materials for introducing variation and identifying gene function in plants. In this study, an ethyl methanesulfonate(EMS)-induced soybean(Glycine max) population,consisting of 21,600 independent M_2 lines, was developed.Over 1,000 M_(4(5))families, with diverse abnormal phenotypes for seed composition, seed shape, plant morphology and maturity that are stably expressed across different environments and generations were identified. Phenotypic analysis of the population led to the identification of a yellow pigmentation mutant, gyl, that displayed significantly decreased chlorophyll(Chl) content and abnormal chloroplast development. Sequence analysis showed that gyl is allelic to Minn Gold, where a different single nucleotide polymorphism variation in the Mg-chelatase subunit gene(ChlI1a) results in golden yellow leaves. A cleaved amplified polymorphic sequence marker was developed and may be applied to marker-assisted selection for the golden yellow phenotype in soybean breeding. | 5 | Increased prediction value of biomarker combinations for the conversion of mild cognitive impairment to Alzheimer's dementia显示文摘Background:Progression of mild cognitive impairment(MCI)to Alzheimer's disease(AD)dementia can be predicted by clinical features and a combination of biomarkers may increase the predictive power.In the present study,we investigated whether the combination of olfactory function and plasma neuronal-derived exosome(NDE)Aβ1-42 can best predict progression to AD dementia.Methods:Eighty-seven MCI patients were enrolled and received cognitive assessment at 2-year and 3-year follow-up to reevaluate cognition.In the meanwhile,80 healthy controls and 88 AD dementia patients were enrolled at baseline as well to evaluate the diagnose value in cross-section.Olfactory function was evaluated with the sniffin sticks(SS-16)and Aβ142 levels in NDES were determined by ELISA.Logistic regression was performed to evaluate the risk factors for cognitive decline in MCI at 2-year and 3-year revisits.Results:In the cross cohort,lower SS-16 scores and higher AB142 levels in NDES were found in MCI and AD dementia compared to healthy controls.For the longitudinal set,8 MCI individuals developed AD dementia within 2 years,and 16 MCI individuals developed AD dementia within 3 years.The two-parameter combination of SS-16 scores and Aβ1-42 level in NDEs showed better prediction in the conversion of MCI to AD dementia at 2-year and 3-year revisits.Moreover,after a 3-year follow-up,SS-16 scores also significantly predicted the conversion to AD dementia,where lower scores were associated with a 10-fold increased risk of developing AD dementia(p=0.006).Similarly,higher AB1-42 levels in NDES in patients with MCI increased the risk of developing AD dementia by 8.5-fold(p=0.002).Conclusion:A combination of two biomarkers NDE AB1-42 and SS-16 predicted the conversion of MCI to AD dementia more accurately.These findings have critical implications for understanding the pathophysiology of AD dementia and for developing preventative treatments for cognitive decline. | Aonan Zhao Yuanyuan Li Yi Yan Yinghui Qiu Binyin Li Wei Xu Ying Wang Jun Liu Yulei Deng | 2020 | Translational Neurodegeneration2020,9,3: | 2 |

| 12 | Phosphate-dependent regulation of vacuolar trafficking of OsSPX-MFSs is critical for maintaining intracellular phosphate homeostasis in rice显示文摘Vacuolar storage of inorganic phosphate(Pi)is essential for Pi homeostasis in plants.The SPX-MFS family proteins have been demonstrated to be vacuolar Pi transporters in many plant species.Transcriptional regulation of the predominant transporter among rice SPX-MFSs,OsSPX-MFS3,was only moderately suppressed by Pi starvation.Thus,post-transcriptional mechanisms were hypothesized to regulate the activity of OsSPX-MFS3.In this study,we found that the tonoplast localization of OsSPX-MFSs is inhibited under Pi-depleted conditions,resulting in their retention in the pre-vacuolar compartments(PVCs).A yeast two-hybrid screen identified that two SNARE proteins,OsSYP21 and OsSYP22,interact with the MFS domain of OsSPX-MFS3.Further genetic and cytological analyses indicate that OsSYP21 and OsSYP22 facilitate trafficking of OsSPX-MFS3 from PVCs to the tonoplast.Although a homozygous frameshift mutation in OsSYP22 appeared to be lethal,tonoplast localization of OsSPX-MFS3 was significantly inhibited in transgenic plants expressing a negative-dominant form of OsSYP22(OsSYP22-ND),resulting in reduced vacuolar Pi concentrations in OsSYP22-ND plants.Under Pi-depleted conditions,the interaction between OsSYP22 and OsSPX-MFS3 was disrupted,and this process depended on the presence of the SPX domain.Deleting the SPX domains of OsSPX-MFSs resulted in their tonoplast localization under both Pi-depleted and Pi-replete conditions.Complementation of the osspx-mfs1/2/3 triple mutants with the MFS domain or the SPX domain of OsSPX-MFS3 confirmed that the MFS and SPX domains are responsive to Pi transport activity and Pi-dependent regulation,respectively.These data indicated that the SPX domains of OsSPX-MFSs sense cellular Pi(InsP)levels and,under Pi-depleted conditions,inhibit the interaction between OsSPX-MFSs and OsSYP21/22 and subsequent trafficking of OsSPX-MFSs from PVCs to the tonoplast. | Runze Guo Qi Zhang Kun Qian Yinghui Ying Wenying Liao Lening Gan Chuanzao Mao Yong Wang James Whelan Huixia Shou | 2023 | Molecular Plant2023,16,8: | 0 |
| 13 | Immediate and long-term outcomes after treat-all among people living with HIV in China:an interrupted time series analysis显示文摘Background In 2003,China implemented free antiretroviral therapy(ART)for people living with HIV(PLHIV),establishing an eligibility threshold of CD4<200 cells/μl.Subsequently,the entry criteria were revised in 2012(eligibility threshold:CD4≤350 cells/μl),2014(CD4≤500 cells/μl),and 2016(treat-all).However,the impact of treat-all policy on HIV care and treatment indicators in China is unknown.We aimed to elucidate the immediate and long-term impact of the implementation of treat-all policy in China.Methods Anonymized programmatic data on ART initiation and collection in PLHIV who newly started ART were retrieved between 1 January 2015 and 31 December 2019,from two provincial and municipal Centers for Disease Control and Prevention and ten major infectious disease hospitals specialized in HIV care in China.We used Poisson and quasi-Poisson segmented regression models to estimate the immediate and long-term impact of treat-all on three key indicators:monthly proportion of 30-day ART initiation,mean CD4 counts(cells/μl)at ART initiation,and mean estimated time from infection to diagnosis(year).We built separate models according to gender,age,route of transmission and region.Results Monthly data on ART initiation and collection were available for 75,516 individuals[gender:83.8%males;age:median 39 years,interquartile range(IQR):28-53;region:18.5%Northern China,10.9%Northeastern China,17.5%Southern China,49.2%Southwestern China].In the first month of treat-all,compared with the contemporaneous counterfactual,there was a significant increase in proportion of 30-day ART initiation[+12.6%,incidence rate ratio(IRR)=1.126,95%CI:1.033-1.229;P=0.007]and mean estimated time from infection to diagnosis(+7.0%,IRR=1.070,95%CI:1.021-1.120;P=0.004),while there was no significant change in mean CD4 at ART initiation(IRR=0.990,95%CI:0.956-1.026;P=0.585).By December 2019,the three outcomes were not significantly different from expected levels.In the stratified analysis,compared with the contemporaneous counterfactual,mean CD4 at ART initiation showed significant increases in Northern China(+3.3%,IRR=1.033,95%CI:1.001-1.065;P=0.041)and Northeastern China(+8.0%,IRR=1.080,95%CI:1.003-1.164;P=0.042)in the first month of treat-all;mean estimated time from infection to diagnosis showed significant increases in male(+5.6%,IRR=1.056,95%CI:1.010-1.104;P=0.016),female(+14.8%,IRR=1.148,95%CI:1.062-1.240;P<0.001),aged 26-35(+5.3%,IRR=1.053,95%CI:1.001-1.109;P=0.048)and>50(+7.8%,IRR=1.078,95%CI:1.000-1.161;P=0.046),heterosexual transmission(+12.4%,IRR=1.124,95%CI:1.042-1.213;P=0.002)and Southwestern China(+12.9%,IRR=1.129,95%CI:1.055-1.208;P<0.001)in the first month of treat-all.Conclusions The implementation of treat-all policy in China was associated with a positive effect on HIV care and treatment outcomes.To advance the work of rapid ART,efforts should be made to streamline the testing and ART initiation process,provide comprehensive support services,and address the issue of uneven distribution of medical resources. | Xinsheng Wu Guohui Wu Ping Ma Rugang Wang Linghua Li Yinghui Sun Junjie Xu Yuwei Li Tong Zhang Quanmin Li Yuecheng Yang Lijing Wang Xiaoli Xin Ying Qiao Bingxue Fang Zhen Lu Xinyi Zhou Yuanyi Chen Qi Liu Gengfeng Fu Hongxia Wei Xiaojie Huang Bin Su Hui Wang Huachun Zou | 2023 | Infectious Diseases of Poverty2023,12,4: | 0 |
