1Synergistic effect of fluorination on both donor and acceptor materials for high performance non-fullerene polymer solar cells with 13.5% efficiency显示文摘A high performance polymer solar cells(PSCs) based on polymer donor PM6 containing fluorinated thienyl benzodithiophene unit and n-type organic semiconductor acceptor IT-4 F containing fluorinated end-groups were developed. In addition to complementary absorption spectra(300–830 nm) with IT-4 F, the PM6 also has a deep HOMO(the highest occupied molecular) level(-5.50 e V), which will lower the open-circuit voltage(V_(oc)) sacrifice and reduce the E_(loss) of the IT-4 F-based PSCs. Moreover, the strong crystallinity of PM6 is beneficial to form favorable blend morphology and hence to suppress recombination. As a result, in comparison with the PSCs based on a non-fluorinated D/A pair of PBDB-T:ITIC with a medium PCE of 11.2%, the PM6:IT-4 Fbased PSCs yielded an impressive PCE of 13.5% due to the synergistic effect of fluorination on both donor and acceptor, which is among the highest values recorded in the literatures for PSCs to date. Furthermore, a PCE of 12.2% was remained with the active layer thickness of up to 285 nm and a high PCE of 11.4% was also obtained with a large device area of 1 cm^2. In addition, the devices also showed good storage, thermal and illumination stabilities with respect to the efficiency. 2Regulatory Effect of E2,IL-6 and IL-8 on the Growth of Epithelial Ovarian Cancer Cells显示文摘To determine the regulatory effects of estrogen and cytokine IL-6 and IL-8 on the growth of epithelial ovarian cancer (OVCA), we first examined the status of estrogen receptors (ERα and ERβ), IL-6 receptor (IL-6Rα and gp130), and IL-8 receptor (IL-8RA and IL-8RB) on five epithelial OVCA cell lines by semiquantitative RT-PCR and Western blot analysis. Results showed that the expressions of these receptors were variable on the five cells.Those OVCA cells expressing the receptors were selected to study related molecular mechanism. MTT assay was performed to observe the effects of 17β-estradiol (E2), IL-6 and IL-8 on cell proliferation. We discovered that E2 markedly promoted the proliferation of CAOV-3 and OVCAR-3 cell in a time- and dose-dependent manner.Tamoxifen (Txf), an ER inhibitor, completely blocked the proliferation of the E2-induced cells, and IL-6- or/and IL-8-neutralizing antibody only showed partially blocking activity. IL-6 and IL-8 were able to significantly stimulate CAOV-3 and OVCAR-3 cell proliferation in a time- and dose-dependent manner, which had a potential synergistic effect on CAOV-3 cells but not on OVCAR-3 cells. The cell proliferation induced by these two cytokines was abolished completely by their specific neutralizing antibodies, partially by Txf, but not by unrelated goat IgG.Taken together, our results suggested that estrogen, IL-6 and IL-8 could modulate OVCA growth by forming a reciprocal cascade with amplifying effect. 3Influence of Different Al Contents on Microstructure, Tensile and Wear Properties of Zn-based Alloy显示文摘The influence of different Al contents on the microstructure, hardness, tensile and wear behavior of Zn-based alloys was investigated in the present study. The test results show that alloy with 27% Al content shows the higher ultimate strength and elongation percent at room and elevated temperatures. It also exhibits the higher wear resistance at all loads in view of the higher tightness of Zn-27Al alloy. Increasing Al content from 33% to 48% results in improvement of ultimate strength and elongation percent at room and elevated temperatures. However, the ultimate strength and elongation percent of the ZA48 alloy are still lower than that of Zn-27Al alloys. 4Characterization of OP9 as authentic mesenchymal stem cell line显示文摘Mesenchymal stem cells (MSCs) are multipotent stem cells capable of differentiating into various cell types,including osteocytes,chondrocytes,adipocytes,myocytes,and tenocytes.However,the difficulty or failure in expanding the mouse MSCs in vitro greatly hampered important research in animal models.The OP9,a stromal cell line from mouse bone marrow,has hematopoietic supportive capacity.Here,we report that the OP9 has the immunophenotype (CD45-,CD11b-,FLK-1-,CD31-,CD34-,CD44+,CD29+,Sca-1+,CD86-,and MHCII-) identical to canonical mouse MSCs.The expression of CD140a+,CD140b+,α-SMA+ and Calponin+ suggested the perivascular origin of OP9.Functionally,the OP9 had strong clonogenic ability and could be induced into osteocytes,chondrocytes and adipocytes.The lymphocyte transformation test (LTT) and mixed leukocyte reaction (MLR) showed that the OP9 could suppress T lymphocyte proliferation stimulated by nonspecific mitogens (PHA) or allogeneic lymphocytes (BALB/c T cells).Finally,the migration of OP9 could be efficiently induced by bFGF,IGF-1,IL-3,PDGF-BB,TGF-β1 and TGF-β3.In conclusion,the OP9 were bona fide MSCs,and such homogenous cell line will be helpful to delineate biological features of MSCs at the stem cell level.Jiao Gao Xin-Long Yan Ren Li Yi Liu Wenyan He Shengkun Sun Yu Zhang Bing Liu Jiaxiang Xiong Ning Mao 1 2010Journal of Genetics and Genomics2010,37,7:3
5High-performance all-polymer solar cells enabled by a novel low bandgap non-fully conjugated polymer acceptor显示文摘The non-fully conjugated polymer as a new class of acceptor materials has shown some advantages over its small molecular counterpart when used in photoactive layers for all-polymer solar cells(all-PSCs),despite a low power conversion efficiency(PCE)caused by its narrow absorption spectra.Herein,a novel non-fully conjugated polymer acceptor PFY-2TS with a low bandgap of~1.40 eV was developed,via polymerizing a largeπ-fused small molecule acceptor(SMA)building block(namely YBO)with a non-conjugated thioalkyl linkage.Compared with its precursor YBO,PFY-2TS retains a similar low bandgap but a higher LUMO level.Moreover,compared with the structural analog of YBO-based fully conjugated polymer acceptor PFY-DTC,PFY-2TS shows a similar absorption spectrum and electron mobility,but significantly different molecular crystallinity and aggregation properties,which results in optimal blend morphology with a polymer donor PBDB-T and physical processes of the device in all-PSCs.As a result,PFY-2TS-based all-PSCs achieved a PCE of 12.31%with a small energy loss of 0.56 eV enabled by the reduced non-radiative energy loss(0.24 eV),which is better than that of 11.08%for the PFY-DTC-based ones.Our work clearly demonstrated that non-fully conjugated polymers as a new class of acceptor materials are very promising for the development of high-performance all-PSCs.Qunping Fan Ruijie Ma Tao Liu Jianwei Yu Yiqun Xiao Wenyan Su Guilong Cai Yuxiang Li Wenhong Peng Tao Guo Zhenghui Luo Huiliang Sun Lintao Hou Weiguo Zhu Xinhui Lu Feng Gao Ellen Moons Donghong Yu He Yan Ergang Wang 2021Science China Chemistry2021,64,8:3

7Bacterial and archaeal community structures in the Arctic deepsea sediment显示文摘Microbial community structures in the Arctic deep-sea sedimentary ecosystem are determined by organic matter input, energy availability, and other environmental factors. However, global warming and earlier ice-cover melting are affecting the microbial diversity. To characterize the Arctic deep-sea sediment microbial diversity and its relationship with environmental factors, we applied Roche 454 sequencing of 16 S r DNA amplicons from Arctic deep-sea sediment sample. Both bacterial and archaeal communities' richness, compositions and structures as well as taxonomic and phylogenetic affiliations of identified clades were characterized. Phylotypes relating to sulfur reduction and chemoorganotrophic lifestyle are major groups in the bacterial groups; while the archaeal community is dominated by phylotypes most closely related to the ammonia-oxidizing Thaumarchaeota(96.66%) and methanogenic Euryarchaeota(3.21%). 8Specnuezhenide suppresses diabetes-induced bone loss by inhibiting RANKL-induced osteoclastogenesis显示文摘Diabetes osteoporosis is a chronic complication of diabetes mellitus(DM)and is associated with osteoclast formation and enhanced bone resorption.Specnuezhenide(SPN)is an active compound with anti-inflammatory and immunomodulatory properties.However,the roles of SPN in diabetic osteoporosis remain unknown.In this study,primary bone marrow macrophages(BMMs)were pretreated with SPN and were stimulated with receptor activator of nuclear factor kappa B ligand(RANKL;50 ng/mL)to induce osteoclastogenesis.The number of osteoclasts was detected by tartrate-resistant acid phosphatase(TRAP)staining.The protein levels of cellular oncogene fos/nuclear factor of activated T cells c1(c-Fos/NFATc1),nuclear factor kappa-B(NF-κB),and mitogen-activated protein kinases(MAPKs)were evaluated by western blot analysis.NF-κB luciferase assays were used to examine the role of SPN in NF-κB activation.The DM model group received a high-glucose,high-fat diet and was then intraperitoneally injected with streptozotocin(STZ).Micro-CT scanning,serum biochemical analysis,histological analysis were used to assess bone loss.We found that SPN suppressed RANKL-induced osteoclast formation and that SPN inhibited the expression of osteoclast-related genes and c-Fos/NFATc1.SPN inhibited RANKL-induced activation of NF-κB and MAPKs.In vivo experiments revealed that SPN suppressed diabetes-induced bone loss and the number of osteoclasts.Furthermore,SPN decreased the levels of bone turnover markers and increased the levels of runt-related transcription factor 2(RUNX2),osteoprotegerin(OPG),calcium(Ca)and phosphorus(P).SPN also regulated diabetes-related markers.This study suggests that SPN suppresses diabetes-induced bone loss by inhibiting RANKL-induced osteoclastogenesis,and provides an experimental basis for the treatment of diabetic osteoporosis.Xiaoshuang Ye Juanjuan Jiang Juan Yang Wenyan Yan Luyue Jiang Yan Chen 2022Acta Biochimica et Biophysica Sinica2022,54,8:2
9Unidirectional and stage-dependent roles of Notch1 in Wnt-responsive Lgr5+ cells during mouse inner ear development显示文摘Wnt and Notch signaling play crucial roles in the determination of the prosensory domain and in the differentiation of hair cells(HCs)and supporting cells during mouse inner ear development;however,the relationship between the two signaling pathways in the mouse cochlea remains largely unknown.Here,we investigated the interactions between Notch and Wnt signaling on the basis of the bidirectional regulation of Notch1 specifically in Wnt-responsive Lgr5+progenitors during different cochlear development stages.We found that the downregulation of Notch1 in Lgr5+cells from embryonic day(E)14.5 to E18.5 can drive the quiescent Lgr5+cells to re-enter the cell cycle and differentiate into extra HCs,whereas the upregulation of Notch1 expression did not affect the proliferation or differentiation of otic progenitor cells.No effect was observed on the upregulation or downregulation of Notch1 in Lgr5+cells from E10.5 to E14.5.We concluded that the roles of Notch1 in Wnt-responsive Lgr5+cells are unidirectional and stage dependent and Notch1 serves as a negative regulator for Lgr5+progenitor activation during cochlear differentiation.Our findings improved the understanding of the interactions between Notch and Wnt signaling in cochlear development.Hui Jiang Shan Zeng Wenli Ni Yan Chen Wenyan Li 2019Frontiers of Medicine2019,13,6:1

11Pertussis toxin–induced inflammatory response exacerbates intracerebral haemorrhage and ischaemic stroke in mice显示文摘Background Stroke is a devastating disease,including intracerebral haemorrhage(ICH)and ischaemic stroke.Emerging evidences indicate that systemic inflammatory cascades after stroke contribute to brain damage.However,the direct effects and features of systemic inflammation on brain injury,especially comparing between ischaemic and haemorrhagic stroke,are still obscure.Methods Pertussis toxin(PT)was used to build a pro-inflammatory milieu after ICH and ischaemic stroke in mouse model.The neurodeficits,stroke lesion,immune response and blood–brain barrier(BBB)destruction were assessed.Results In ICH mouse model,PT-induced systemic inflammation exacerbated neurological deficits,and enlarged haemorrhage lesion and perihaematomal oedema.We also found promoted leucocyte infiltration and inflammatory cytokine release into the brain after PT treatment.Moreover,the integrity of the BBB was further disrupted after receiving PT.Furthermore,we demonstrated that PT enhanced brain inflammation and aggravated stroke severity in middle cerebral artery occlusion mouse model.Conclusions Our results suggest that PT increases inflammatory response that exacerbates brain injury after ICH or ischaemic stroke in mouse model.Ming Zou Yan Feng Yuwhen Xiu Yan Li Ying Zhang Junwan Fan Haowen Li Jingli Cao Wenyan He Wei-Na Jin 2022Stroke & Vascular Neurology2022,7,1:1
12Chemometric identification of canonical metabolites linking critical process parameters to monoclonal antibody production during bioprocess development显示文摘A deeper understanding of the biological events occurring when bioprocess parameters changed will be of great value in improving the monoclonal antibodies (mAbs) production. Design of experiment (DoE) was applied to investigate the effect of process parameters (pH, temperature shift and dissolve oxygen (DO)) on protein titer. The key metabolites connecting the critical process parameters (CPPs) with monoclonal antibody production were identified by different chemometrics tools. Finally, the biological events of marker metabolites relating with titer improvement were concluded. pH and temperature shift were identified as CPPs that affect the target protein titer. A series of metabolites influenced by the altered CPPs and correlated with protein titer were screened by principal component analysis (PCA) and Pearson' correlation test. The marker metabolites and their pathways linking CPPs to target protein titer in different culture phases were summarized. 14Programming of Regulatory T Cells In Situ for Nerve Regeneration and Long-Term Patency of Vascular Grafts显示文摘Rapid integration into the host tissue is critical for long-term patency after small diameter tissue engineering vascular grafts(sdTEVGs)transplantation.Neural recognition may be required for host integration and functionalization of the graft.However,immune rejection and inflammation hinder nerve regeneration of sdTEVGs.Here,a CRISPR/dCas9-nanocarrier was used for targeted programming of regulatory T cells(Treg cells)in situ to promote nerve regeneration of sdTEVGs by preventing excessive inflammation.Treg cells and(C-C chemokine receptor)CCR2+macrophage recruitment occurred after transplantation.The nanodelivery system upregulated ten eleven translocation(TET2)in Treg cells in vitro.Reprogrammed Treg cells upregulated anti-inflammatory cytokines and decreased the proportion of CCR2+macrophages.IL-6 concentrations decreased to the levels required for nerve regeneration.Implantation of CRISPR/dCas9 nanodelivery system-modified sdTEVGs in rats resulted in Treg cell editing,control of excessive inflammation,and promoted nerve regeneration.After 3 months,nerve regeneration was similar to that observed in normal blood vessels;good immune homeostasis,consistency of hemodynamics,and matrix regeneration were observed.Neural recognition promotes further integration of the graft into the host,with unobstructed blood vessels without intimal hyperplasia.Our findings provide new insights into vascular implant functionalization by the host.Yanhong Wang Fangchao Xue Yanzhao Li Lin Lin Yeqin Wang Shanlan Zhao Xingli Zhao Yong Liu Ju Tan Gang Li Haoran Xiao Juan Yan Hao Tian Min Liu Qiao Zhang Zhaojing Ba Lang He Wenyan Zhao Chuhong Zhu Wen Zeng 2022Research2022,,4:1

18A new discovered ABCA1 gene polymorphisms and the association of ABCA1 SNPs with coronary artery disease and plasma lipids in Chinese population显示文摘Objective: Single nucleotide polymorphisms (SNP) of ATP-binding cassette transporter A1 (ABCA1) gene are related to plasma lipid and susceptibility to coronary artery disease (CAD). Our first goal was to screen all 50 coding regions of ABCA1 to find new SNPs. Our second goal was to investigate the frequency distribution of R1587K and M883I polymorphisms of ABCA1 gene, which are the variant occurred most frequently, in Chinese people and to evaluate their association with the CAD phenotype and plasma lipids. Methods: Single-strand conformation polymorphism (SSCP) and DNA sequence were used for confirming new SNP of ABCA1, and restriction fragment length polymorphism (RFLP) were applied for confirming genotypes of R1587K and M883I in 112 CAD cases and 108 healthy people. Results: We discovered a new ABCA1 SNP in Chinese population, which converse 233 amino acids from Methionine to Valine (M233V). This new ABCA1 SNP located in exon7, and might potentially modulate the biological function of lipid metabolism. For R1587K and M883I SNPs, the K allele and I allele frequency was 28.9%and 31.1%, respectively. The K allele at R1587K conferred lower mean values of HDL-C in a dose-dependent manner in both CAD patients and healthy people. However, 883I allele was not associated with plasma lipid level. Neither 1587KK nor 883II associated with increased risk of CAD.
