| 1 | Episodic rifting of continental marginal basins and tectonic events since 10 Ma in the South China Sea显示文摘Integrated study on the tectonic, basin filling and thermal evolution of the Tertiary basins on the northern margin of the South China Sea has indicated that the rift continental margin on the northern South China Sea is not a passive margin and that the western margin is characteristic of the transformextension. Episodic rifting and the thermal events since 10 Ma in the area have been documented by the integrated analysis of dynamic process. | 3 | A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. | 6 | Anlotinib as third- or further-line therapy for short-term relapsed small-cell lung cancer: subgroup analysis of a randomized phase 2 study (ALTER1202)显示文摘Patients with small-cell lung cancer (SCLC) relapse within months after completing previous therapies. This study aimed to investigate the efficacy and safety of anlotinib as third- or further-line therapy in patients with short-term relapsed SCLC from ALTER1202. Patients with short-term relapsed SCLC (disease progression within 3 months after completing ≥ two lines of chemotherapy) in the anlotinib (n = 67) and placebo (n = 34) groups were analyzed. The primary endpoint was progression-free survival (PFS). The secondary endpoints included overall survival, objective response rate (ORR), disease control rate, and safety. Anlotinib significantly improved median PFS/OS (4.0 vs. 0.7 months, P < 0.0001)/(7.3 vs. 4.4 months, P = 0.006) compared with placebo. The ORR was 4.5%/2.9% in the anlotinib/placebo group (P = 1.000). The DCR in the anlotinib group was higher than that in the placebo group (73.1% vs. 11.8%, P < 0.001). The most common adverse events (AEs) were hypertension (38.8%), loss of appetite (28.4%), and fatigue (22.4%) in the anlotinib group and gamma-glutamyl transpeptidase elevation (20.6%) in the placebo group. No grade 5 AEs occurred. For patients with short-term relapsed SCLC, third- or further-line anlotinib treatment was associated with improved survival benefit. | 9 | LIN28 coordinately promotes nucleolar/ribosomal functions and represses the 2C-like transcriptional program in pluripotent stem cells显示文摘LIN28 is an RNA binding protein with important roles in early embryo development,stem cell differentiation/re-programming,tumorigenesis and metabolism.Previous studies have focused mainly on its role in the cytosol where it interacts with Let-7 microRNA precursors or mRNAs,and few have addressed LIN28's role within the nucleus.Here,we show that LIN28 displays dynamic temporal and spatial expression during murine embryo development.Maternal LIN28 expression drops upon exit from the 2-cell stage,and zygotic LIN28 protein is induced at the forming nucleolus during 4-cell to blas-tocyst stage development,to become dominantly expressed in the cytosol after implantation.In cultured pluripotent stem cells(PSCs),loss of LIN28 led to nucleolar stress and activation of a 2-cell/4-cell-like transcriptional program characterized by the expression of endogenous retrovirus genes.Mechanistically,LIN28 binds to small nucleolar RNAs and rRNA to maintain nucleolar integrity,and its loss leads to nucleolar phase separation defects,ribosomal stress and activation of P53 which in turn binds to and activates 2C transcription factor Dux.LIN28 also resides in a complex containing the nucleolar factor Nucleolin(NCL)and the transcrip-tional repressor TRIM28,and LIN28 loss leads to reduced occupancy of the NCL/TRIM28 complex on the Dux and rDNA loci,and thus de-repressed Dux and reduced rRNA expression.Lin28 knockout cells with nucleolar stress are more likely to assume a slowly cycling,translationally inert and anabolically inactive state,which is a part of previously unappreciated 2C-like transcriptional program.These findings elucidate novel roles for nucleolar LIN28 in PSCs,and a new mechanism linking 2C program and nucleolar functions in PSCs and early embryo development. | Zhen Sun Hua Yu Jing Zhao Tianyu Tan Hongru Pan Yuqing Zhu Lang Chen Cheng Zhang Li Zhang Anhua Lei Yuyan Xu Xianju Bi Xin Huang Bo Gao Longfei Wang Cristina Correia Ming Chen Qiming Sun Yu Feng Li Shen Hao Wu Jianlong Wang Xiaohua Shen George Q.Daley Hu Li Jin Zhang | 2022 | Protein & Cell2022,13,7: | 4 |
| 10 | Geology, Geochronology, and Hf Isotopic Composition of the Pha Lek Fe Deposit, Northern Laos: Implications for Early Permian Subduction-Related Skarn Fe Mineralization in the Truong Son Belt显示文摘The Truong Son metallogenic belt in central Laos and Vietnam is an important Fe-Cu-Sn-Au polymetallic ore district. The Pha Lek Fe deposit is closely related to Late Carboniferous– Early Permian I-type granitic magmatism, and contains >50 Mt @ 45% to 50% of Fe ore. Ore minerals occur mainly as magnetite and hematite in the skarn alteration zone between a granitic pluton and metamorphosed Middle–Upper Devonian carbonates. The granitic pluton comprises granodiorite and granite, with zircon U-Pb dating indicating synchronous emplacement at 288.2±1.3 and 284.9±1.2 Ma, respectively. Zircons from these granitoids have ε_(Hf)(t) values of 2.9–11.2 and relatively young TDM2 ages(<1.0 Ga), indicating an origin by partial melting of depleted mafic crust or magma mixing. Previous studies have shown that these granitoids have high Y, Yb, and K_2O contents, and low Sr and Na_2O contents, which are interpreted as the melting of mafic continental crust. Pyrite of the main mineralization stage yields an ^(187)Re/^(188)Os-^(187)Os/^(188)Os isochron age of 287±17 Ma, indicating that mineralization is associated with Pha Lek granitic magmatism. A Late Carboniferous–Early Permian subduction-related skarn-type Fe mineralization model is proposed for the Pha Lek deposit. | 12 | Stable isotope geochemical characteristics of dissolved inorganic carbon in the Jiulong River Estuary, Fujian Province, China显示文摘The isotopic composition of dissolved inorganic carbon(DIC) in estuarine environments has been studied for its significant role in determining the isotopic composition of inorganic/organic matter and its applications to the study of various natural processes. | 13 | Oxidation-strengthened disulfide-bridged prodrug nanoplatforms with cascade facilitated drug release for synergetic photochemotherapy显示文摘One of the major barriers in utilizing prodrug nanocarriers for cancer therapy is the slow release of parent drug in tumors.Tumor cells generally display the higher oxidative level than normal cells,and also displayed the heterogeneity in terms of redox homeostasis level.We previously found that the disulfide bond-linkage demonstrates surprising oxidationsensitivity to form the hydrophilic sulfoxide and sulphone groups.Herein,we develop oxidation-strengthened prodrug nanosystem loaded with pyropheophorbide a(PPa)to achieve light-activatable cascade drug release and enhance therapeutic efficacy.The disulfide bond-driven prodrug nanosystems not only respond to the redox-heterogeneity in tumor,but also respond to the exogenous oxidant(singlet oxygen)elicited by photosensitizers.Once the prodrug nanoparticles(NPs)are activated under irradiation,they would undergo an oxidative self-strengthened process,resulting in a facilitated drug cascade release.The IC50 value of the PPa@PTX-S-S NPs without irradiation was 2-fold higher than those of NPs plus irradiation.In vivo,the PPa@PTX prodrug NPs display prolonged systemic circulation and increased accumulation in tumor site.The PPa@PTXS-S NPs showed much higher efficiency than free PTX or the PPa@PTX-C-C NPs to suppress the growth of 4 T1 tumors.Therefore,this novel oxidation-strengthened disulfide-bridged prodrug-nanosystem has a great potential in the enhanced efficacy of cancer synergetic photochemotherapy. | Bin Yang Lin Wei Yuequan Wang Na Li Bin Ji Kaiyuan Wang Xuanbo Zhang Shenwu Zhang Shuang Zhou Xiaohui Yao Hang Song Yusheng Wu Haotian Zhang Qiming Kan Tao Jin Jin Sun | 2020 | Asian Journal of Pharmaceutical Sciences2020,15,5: | 2 |

| 16 | Estimation of chlorophyll content in Brassica napus based on unmanned aerial vehicle images显示文摘The chlorophyll content has a direct effect on photosynthesis of crops.In order to explore a quick and convenient method for estimating the chlorophyll content of Brassica napus and facilitate efficient crop monitoring,we measured the actual value of chlorophyll with a SPAD-502 chlorophyll detector,and collected aerial images of B.napus with an unmanned aerial vehicle(UAV)carrying a RGB camera in this study.The total number of 270samples collected images were divided into regions according to the planting conditions of different B.napus varieties in the field.Then,according to the empirical formula,there were 36 colors’characteristic parameters calculated and combined.To estimate the chlorophyll content of rape,189 samples were included in the modeling set,while the other 81 samples were enrolled in the validation set for testing the accuracy of this model.After the combination of R(red),G(green)and B(blue)color channels,the results showed that the color characteristics B/(R+G),b,B/G,(G-B)/(G+B),g-b were highly connected with the measured value of chlorophyll SPAD,and the correlation coefficient between the combination based on B/(R+G)and SPAD value was 0.747.With R2=0.805,RMSE=3.343,and RE=6.84%,the regression model created using random forest had superior outcomes,according to the model comparison.This study offers a new method for quickly estimating the amount of chlorophyll in rapeseed and a workable reference for crop monitoring using the UAV platform. | Yayi Huang Qiming Ma Xiaoming Wu Hao Li Kun Xu Gaoxiang Ji Fang Qian Lixia Li Qian Huang Ying Long Xiaojun Zhang Biyun Chen Changhua Liu | 2022 | Oil Crop Science2022,7,3: | 1 |
