1Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 2A review on tribology of polymer composite coatings显示文摘Self-lubricating polymer composite coatings,with tailorable tribological and mechanical properties,have been widely employed on mechanical parts to reduce friction and wear,which saves energy and improves the overall performance for applications such as aerospace satellite parts,shafts,gears,and bushings.The addition of functional fillers can overcome the limitations of single-polymer coatings and extend the service life of the coatings by providing a combination of low friction,high wear resistance,high load bearing,high temperature resistance,and high adhesion.This paper compares the heat resistance,and the tribological and mechanical properties of common polymer matrices,as well as the categories of functional fillers that improve the coating performance.Applicable scopes,process parameters,advantages,and limitations of the preparation methods of polymer coatings are discussed in detail.The tribological properties of the composite coatings with different matrices and fillers are compared,and the lubrication mechanisms are analyzed.Fillers reduce friction by promoting the formation of transfer films or liquid shear films.Improvement of the mechanical properties of the composite coatings with fillers of different morphologies is described in terms of strengthening and toughening mechanisms,including a stress transfer mechanism,shear yielding,crack bridging,and interfacial debonding.The test and enhancement methods for the adhesion properties between the coating and substrate are discussed.The coating adhesion can be enhanced through mechanical treatment,chemical treatment,and energy treatment of the substrate.Finally,we propose the design strategies for high-performance polymer composite coating systems adapted to specific operating conditions,and the limitations of current polymer composite coating research are identified.Yilong REN Lin ZHANG Guoxin XIE Zhanbo LI Hao CHEN Hanjun GONG Wenhu XU Dan GUO Jianbin LUO 2021Friction2021,9,3:13

4Whole genome sequencing of 10K patients with acute ischaemic stroke or transient ischaemic attack: design, methods and baseline patient characteristics显示文摘Background and purpose Stroke is the second leading cause of death worldwide and the leading cause of mortality and long-term disability in China,but its underlying risk genes and pathways are far from being comprehensively understood.We here describe the design and methods of whole genome sequencing(WGS)for 10914 patients with acute ischaemic stroke or transient ischaemic attack from the Third China National Stroke Registry(CNSR-III).Methods Baseline clinical characteristics of the included patients in this study were reported.DNA was extracted from white blood cells of participants.Libraries are constructed using qualified DNA,and WGS is conducted on BGISEQ-500 platform.The average depth is intended to be greater than 30×for each subject.Afterwards,Sentieon software is applied to process the sequencing data under the Genome Analysis Toolkit best practice guidance to call genotypes of single nucleotide variants(SNVs)and insertion-deletions.For each included subject,21 fingerprint SNVs are genotyped by MassARRAY assays to verify that DNA sample and sequencing data originate from the same individual.The copy number variations and structural variations are also called for each patient.All of the genetic variants are annotated and predicted by bioinformatics software or by reviewing public databases.Results The average age of the included 10914 patients was 62.2±11.3 years,and 31.4%patients were women.Most of the baseline clinical characteristics of the 10914 and the excluded patients were balanced.Conclusions The WGS data together with abundant clinical and imaging data of CNSR-III could provide opportunity to elucidate the molecular mechanisms and discover novel therapeutic targets for stroke.Si Cheng Zhe Xu Yang Liu Jinxi Lin Yong Jiang Yilong Wang Xia Meng Anxin Wang Xinying Huang Zhimin Wang Guohua Chen Songdi Wu Zhengchang Jia Yongming Chen Xuerong Qiu Jun Wu Binbin Song Weizhong Ji Zhongping An Wenjun Xue Lili Zhao Yu Geng Hongyan Li Hao Li Yongjun Wang 2021Stroke & Vascular Neurology2021,6,2:8
5Low serum albumin levels predict poor outcome in patients with acute ischaemic stroke or transient ischaemic attack显示文摘Background To examine the relationship of serum albumin with poor functional outcome and mortality in patients with acute ischaemic stroke(AIS)or transient ischaemic attack(TIA),and perform a meta-analysis to summarise the association.Methods We analysed data from the Third China National Stroke Registry(CNSR-Ⅲ).Patients were divided into four groups based on serum albumin levels at admission.The outcomes included poor functional outcome(modified Rankin Scale(mRS)score of 3 to 6)and mortality at 3 months and 1 year.Multiple logistic regression models and Cox regression models were used to evaluate the association,respectively.We used a fixed-effect model to calculate the risk ratio for poor functional outcome and a random-effect model for mortality in the meta-analysis.Results A total of 13618 patients were enrolled.During the 3-month follow-up period,compared with 40 to 44.9 g/L group,patients in<35 g/L group had an increased risk of poor functional outcome and mortality(adjusted OR 1.37(95%CI 1.12 to 1.67);adjusted HR 2.13(95%CI 1.41 to 3.23)).The relationship in per 10 g/L decreased serum albumin with prognosis was consistently inversed(adjusted OR 1.17(95%CI 1.01 to 1.35);adjusted HR 1.86(95%CI 1.30 to 2.64)).Also,low serum albumin levels were independently correlated with clinical outcomes at 1 year.In the meta-analysis,the OR for poor functional outcome pooled 3 studies per 1 g/L decrease was 1.03(95%CI 1.02 to 1.05),and the HR for mortality pooled 5 studies was 1.07(95%CI 1.03 to 1.11).Conclusions Low serum albumin levels predict poor functional outcome and mortality in patients with AIS or TIA.Hongyu Zhou Anxin Wang Xia Meng Jinxi Lin Yong Jiang Jing Jing Yingting Zuo Yilong Wang Xingquan Zhao Hao Li Yongjun Wang 2021Stroke & Vascular Neurology2021,6,3:7
6Awareness intervention for Beijing neurologists regarding secondary prevention of cerebral infarction/transient ischemia Cross-sectional investigation显示文摘BACKGROUND: Stroke prevention guidelines should be made available to neurologists for clinical application to aid in uniformity, timing, preciseness, and acceptance of disease. OBJECTIVE: To investigate the awareness of neurologists in some Beijing hospitals of intervention in secondary prevention of cerebral infarction/transient ischemia. DESIGN: Cross-sectional study. SETTING: Beijing Tiantan Hospital, Beijing Anzhen Hospital, General Hospital of Beijing Military Area, Command of Chinese PLA, Beijing Chuiyangliu Hospital, Beijing 6th Hospital, Beijing Hepingli Hospital, and Beijing Daxing District Hospital. PARTICIPANTS: A total of 28 (associate) chief physicians, 58 attending physicians, and 54 resident physicians who engaged in clinical treatment of cerebrovascular diseases were selected from 8 hospitals in Beijing from March to April 2007. All physicians provided informed consent. METHODS: Self-made closed questionnaires were provided for data collection, consisting of 16 questions that were single choice or multiple choice. Specifically, questions 1-7 focused on awareness of blood pressure regulation in different patients and first choice of decompression drug; questions 8-12 focused on awareness of lipid regulation; and questions 13-16 focused on awareness of anti-blood platelet drugs applied in secondary prevention. The scores ranged from 0-100 points, and each question was worth 6.25 points. The scores positively correlated with the awareness rate. To test leveling real-time, the survey lasted for a maximum of 20 minutes. One questionnaire was independently finished by each subject in the survey. MAIN OUTCOME MEASURES: Awareness intervention among neurologists during secondary prevention of cerebral infarction/transient ischemia and questionnaire scores. RESULTS: 140 subjects were included in the final analysis. ① The awareness rate among neurologists for intervention during secondary prevention of cerebral infarction/transient ischemia ranged from 0.7-57.9%, the scores ranged between 0-56 points, and the mean score was 26 points. ② Scores of resident physicians were 0-56 points with a mean score of 26 points; attending physicians scored 6-50 points, with a mean score of 26 points; and chief physicians scored 6-50 points, with a mean score of 23 points. There were no significant differences among the various physicians (F = 0.771, t = 0.465, P > 0.05). CONCLUSION: Awareness among neurologists of intervention during secondary prevention of cerebral infarction/transient ischemia is not ideal. 8Revealing the critical effect of solid electrolyte interphase on the deposition and detriment of Co(Ⅱ) ions to graphite anode显示文摘'Dissolution,migration,and deposition'of transition metal ions (TMIs) result in capacity degradation of lithium-ion batteries (LIBs).Understanding such detrimental mechanism of TMIs is critical to the development of LIBs with long cycle life.In most previous works,TMIs were directly introduced into the electrolyte to investigate such a detrimental mechanism.In these cases,the TMIs are deposited directly on the fresh anode surface.However,in the practical battery system,the TMIs are deposited on the anode covered with solid electrolyte interphase (SEI) film.Whether the pre-presence of SEI film on anode surface influences the deposition and detriment of TMIs is unclear.In this work,the deposition of Co element on graphite anode with and without SEI film were systematically studied.The results clearly show that,in comparison with that of fresh graphite (SEI-free),the presence of SEI film aggravates the deposition of Co ions due to the Li^(+)–Co^(2+) ion exchange between the SEI and Co^(2+)-containing electrolyte without the driving of the electric field,leading to faster capacity fading of graphite anode.Therefore,how to regulate electrolytes and film-forming additives to design the components of SEI and prevent its exchange with TMIs,is a crucial way to inhibit the deposition and detriment of TMIs on graphite anode.Qiming Xie Jiawei Chen Lidan Xing Xianggui Zhou Zekai Ma Binhong Wu Yilong Lin Hebing Zhou Weishan Li 2022Journal of Energy Chemistry2022,31,6:2
9Analysis of Internal Moisture Change of Camphor Seeds during Drying by Low-Field NMR显示文摘In this study,low-field nuclear magnetic resonance(LF-NMR)was used to collect the transverse relaxation signals of camphor seeds that had been dried at different temperatures and for different durations.The transverse relaxation time of the hydrogen nucleus was obtained by SIRT(Simultaneous Iterative Reconstruction Technique)inversion,and the correlation between the moisture content on dry basis and the amplitude of NMR signal of camphor seeds sampled at different drying stages was analyzed.The results showed that water existed in three main forms strongly bound water,weakly bound water and free water in camphor seeds.During the drying process,the peak position of strongly bound water remained unchanged,but the peak positions of weakly bound water and free water fluctuated.Drying rate increased with drying temperature increasing.In addition,there was a very significant linear relationship between the total NMR signal amplitude and moisture content on dry basis,and the correlation coefficient reached 0.984 4.Yan XUAN Yilong XIANG Jing YANG Buhong GAO Ying TANG Feng LIN 2019Agricultural Biotechnology2019,8,4:2

11PolyvasculaR Evaluation for Cognitive Impairment and vaScular Events (PRECISE)-a population-based prospective cohort study: rationale, design and baseline participant characteristics显示文摘Background and purpose Cardiovascular diseases and dementia are two major diseases in the elderly.Atherosclerosis is associated with future vascular events and cognitive impairment.The PolyvasculaR Evaluation for Cognitive Impairment and vaScular Events(PRECISE)study is a population-based prospective cohort study with comprehensive evaluation of multiterritorial artery stenosis and plaque using advanced vascular imaging techniques and prospective collection of vascular events and cognitive assessments.Methods Between May 2017 and September 2019,the PRECISE study enrolled 3067 community-dwelling adults with ages between 50 and 75 years cluster sampled from six villages and four communities of Lishui city in China.Data are collected in face-to face interviews at baseline,2-year and 4-year follow-up visits.Brain MRI including high-resolution sequences for intracranial and carotidal arteries and CT angiography for thoracoabdominal arteries were performed at baseline and will be rescanned after 4 years.Cardiovascular/cerebrovascular events and cognitive assessment will be prospectively collected after the enrollment.Blood and urine samples were collected and biomarkers were tested at baseline.Results A total of 3067 subjects were enrolled,among which 53.5%were female with an average age of 61.2±6.7 years.Among them,2.8%,8.1%,43.1%and 21.6%had a history of stroke,coronary heart diseases,hypertension and diabetes mellitus,respectively.Conclusions The PRECISE study is a population-based prospective cohort study with comprehensive evaluation of atherosclerotic stenosis and plaque using advanced vascular imaging techniques.Data from this cohort provide us an opportunity to precisely evaluate polyvascular atherosclerosis and its association with future vascular events and cognitive impairment.Yuesong Pan Jing Jing Xueli Cai Yilong Wang Suying Wang Xia Meng Chunlai Zeng Jiong Shi Jiansong Ji Jinxi Lin Lingchun Lyu Zhe Zhang Lerong Mei Shiping Li Shan Li Wanlin Zhu Hao Li Tiemin Wei Yongjun Wang 2021Stroke & Vascular Neurology2021,6,1:1
12An experimental investigation on decoupling performance for a lateral axis micromachined gyroscope with varying environmental parameters显示文摘This paper carries out an experiment study of decoupling performance for a novel lateral axis micromachined gyroscope with varying environmental parameters. The non-ideal mathematical model for the coupling mechanism of the gyroscope is established through the gyro dynamic response matrix. The coupling components varying with ambient pressure and temperature induced by stiffness coupling,damping coupling and electrostatic force coupling are semi-analytically discussed. The overall coupling ratio is evaluated via experiments in the custom-built installation. The testing results show that the decoupling performance of the gyroscope is sensitive to the environmental parameters and all the non-ideal errors are determined as a function of ambient pressure and temperature. The coupling error varies from 0.05% to 0.25% within the pressure range of 100 Pa-100 kPa. The characteristics of coupling with temperature are measured from 20°C to 100°C with a variation from 0.35% to 0.41%. The results also indicate that within the range of measured ambient pressure and temperature,the minimum coupling ratio occurs at 100 Pa and room temperature. The overall performance of the gyroscope is tested under the pressure of about 2000 Pa and room temperature to achieve a relatively low coupling ratio. The scale factor is measured to be 7.8 mV(°) -1 s-1 with nonlinearity about 0.45% in the full-scale range of 600(°) s-1. 14A large-sized basal ankylopollexian from East Asia, shedding light on early biogeographic history of Iguanodontia显示文摘A presumably mostly quadrupedal ankylopollexian iguanodontian, Bayannurosaurus perfectus gen. et sp.nov., is reported here, and is represented by an excellently well-preserved skeleton from the Lower Cretaceous Bayingebi Formation of Inner Mongolia, China. The diagnosis of the taxon includes several autapomorphies, notably a dorsally directed, strap-like posterodorsal process of the jugal and a horizontally oriented preacetabular process of the ilium. The nearly complete caudal series retains eight posterior-most caudals with procoelous, trapezoidal centra in dorsal view, and the last three caudals are fully fused. The discovery of B. perfectus opens a critical new window on the early evolution and intercontinental dispersal of Iguanodontia. The skeleton displays a transitional morphology between nonhadrosauriform ankylopollexians and Hadrosauriformes. A phylogenetic analysis indicates that Bayannurosaurus is positioned higher on the tree than Hypselospinus, but below Ouranosaurus just outside of Hadrosauriformes. 15肺癌患者KDR基因酪氨酸激酶结构域的突变和多态性研究(英文)显示文摘Objective: Although the kinase insert domain-containing receptor (KDR) gene play an very important role in the metastasis of cancer and is also as one of the molecular targets used in cancer therapy, mutation in the tyrosine kinase (TK) domain of the KDR gene has not been reported. Here we detected the mutations and polymorphisms in the TK domain of KDR gene in human lung cancer patients and to give the basic evidence and clue for cancer prevention and target therapy. Methods: The entire sequence of exons 21, 22, 23 and 27 (which contain the coding sequence of tyrosine phosphorylation) in the TK domain of KDR gene in the patients with lung cancer and control healthy individuals were assayed by PCR and DNA sequencing. We also analyzed one non-coding single nucleotide polymorphisms (SNPs) in the KDR gene. Results: No mutations were found in exon 22, 23 and 27. One heterozygous mutation of c.+2837 in exon 21 was found at a frequency of 2.08% (2/96) in the patients with lung cancer and none were detected in the healthy control individuals. The mutation was from a G to a A resulting in substitution of arginine with histidine residue. 16Evaluation of temporal compositing algorithms for annual land cover classification using Landsat time series data显示文摘In this paper, four widely used temporal compositing algorithms, i.e.median, maximum NDVI, medoid, and weighted scoring-basedalgorithms, were evaluated for annual land cover classification usingmonthly Landsat time series data. Four study areas located in California,Texas, Kansas, and Minnesota, USA were selected for image compositingand land cover classification. Results indicated that images compositedusing weighted scoring-based algorithms have the best spatial fidelitycompared to other three algorithms. In addition, the weighted scoringbasedalgorithms have superior classification accuracy, followed bymedian, maximum NDVI, and medoid in descending order. However, themedian algorithm has a significant advantage in computational efficiencywhich was ~70 times that of weighted scoring-based algorithms, andwith overall classification accuracy just slightly lower (~0.13% onaverage) than weighted scoring-based algorithms. Therefore, werecommended the weighted scoring-based compositing algorithms forsmall area land cover mapping, and median compositing algorithm forthe land cover mapping of large area considering the balance betweencomputational complexity and classification accuracy. 17Naringin,neohesperidin and their corresponding dihydrochalcones as bioactive substances:a symphony of bitter–sweet显示文摘Bitter is generally undesirable,although it is an important part of flavor.Bitter substances exhibit diverse health-promoting activities,which is in line with the famous Chinese saying‘a good medicine tastes bitter’.Naringin(NAG)and neohesperidin(NHP),two important flavanones that give bitterness to citrus fruits,show various pharmacological activities.Interestingly,their hydrogenation products,i.e.naringin dihydrochalcone(NDC)and neohesperidin dihydrochalcone(NHDC),undergo a dramatic taste shift from bitter to intensely sweet,which can be 300 and 1000 times sweeter than sucrose,respectively.Such sweeteners not only provide a sweet taste without the burden of increased calorie intake and glycemia,but also may exert multiple bioactivities.This review summarizes common dietary bitter and sweet compounds with sensory scores.Taste conversions induced by structural changes from bitter NAG and NHP to sweet NDC and NHDC are particularly discussed.In addition,the taste-sensing mechanisms,pharmacological characteristics,dietary distribution,synthesis,and food industry applications of these bitter–sweet interchangeable compounds are outlined.In conclusion,the bitter NAG and NHP are promising therapeutic candidates for management of diverse etiologically complex diseases while their corresponding dihydrochalcones NDC and NHDC are promising sweeteners,which might be a blessing for those who need to control sugar intake.Ruoqi Wang Wenwen You Heng Lin Yunlin Cao Changjie Xu Kunsong Chen Yilong Liu Xian Li 2023Food Quality and Safety2023,7,4:0
18Dt1 inhibits SWEET-mediated sucrose transport to regulate photoperiod-dependent seed weight in soybean显示文摘Soybean is a photoperiod-sensitive short-day crop whose reproductive period and yield are markedly affected by day-length changes.Seed weight is one of the key traits determining the soybean yield;how-ever,the prominent genes that control the final seed weight of soybean and the mechanisms underlying the photoperiod's effect on this trait remain poorly understood.In this study,we identify SwW19 as a major locus controlling soybean seed weight by QTL mapping and determine Dt1,an orthologous gene of Arabidopsis TFL1 that is known to govern the soybean growth habit,as the causal gene of the SW19 locus.We showed that Dt1 is highly expressed in developing seeds and regulates photoperiod-dependent seed weight in soybean.Further analyses revealed that the Dt1 protein physically interacts with the sucrose transporter GmSWEET10a to negatively regulate the import of sucrose from seed coat to the embryo,thus modulating seed weight under long days.However,Dt1 does not function in seed development under short days due to its very low expression.Importantly,we discovered a novel natural allelic variant of Dt1(H4 haplotype)that decouples its pleiotropic effects on seed size and growth habit;i.e.,this variant remains functional in seed development but fails to regulate the stem growth habit of soybean.Collectively,our findings provide new insights into how soybean seed development responds to photoperiod at different latitudes,offering an ideal genetic component for improving soybean's yield by manipulating its seed weightandgrowth habit.Xiaoming Li Zhonghui Chen Haiyang Li Lin Yue Cuirong Tan Hongjie Liu Yilong Hu Yuhua Yang Xiani Yao Lingping Kong Xiang Huangi Bin Yu Chunyu Zhang Yuefeng Guan Baohui Liu Fanjiang Kong Xingliang Hou 2024Molecular Plant2024,17,3:0
