Patient-derived non-small cell lung cancer xenograft mirrors complex tumor heterogeneity显示文摘Objective:Patient-derived xenograft(PDX)models have shown great promise in preclinical and translational applications, but their consistency with primary tumors in phenotypic, genetic, and pharmacodynamic heterogeneity has not been well-studied.This study aimed to establish a PDX repository for non-small cell lung cancer(NSCLC)and to further elucidate whether it could preserve the heterogeneity within and between tumors in patients. Methods:A total of 75 surgically resected NSCLC specimens were implanted into immunodeficient NOD/SCID mice.Based on the successful establishment of the NSCLC PDX model,we compared the expressions of vimentin,Ki67,EGFR,and PD-L1 proteins between cancer tissues and PDX models using hematoxylin and eosin staining and immunohistochemical staining. In addition,we detected whole gene expression profiling between primary tumors and PDX generations. We also performed whole exome sequencing(WES)analysis in 17 first generation xenografts to further assess whether PDXs retained the patient heterogeneities. Finally, paclitaxel, cisplatin, doxorubicin, atezolizumab, afatininb, and AZD4547 were used to evaluate the responses of PDX models to the standard-of-care agents. Results:A large collection of serially transplantable PDX models for NSCLC were successfully developed. The histology and pathological immunohistochemistry of PDX xenografts were consistent with the patients’tumor samples.WES and RNA-seq further confirmed that PDX accurately replicated the molecular heterogeneities of primary tumors.Similar to clinical patients,PDX models responded differentially to the standard-of-care treatment, including chemo-,targeted-and immuno-therapeutics.Conclusions:Our established PDX models of NSCLC faithfully reproduced the molecular, histopathological, and therapeutic characteristics,as well as the corresponding tumor heterogeneities, which provides a clinically relevant platform for drug screening, biomarker discovery, and translational research. | Xuanming Chen Cheng Shen Zhe Wei Rui Zhang Yongsheng Wang Lili Jiang Ke Chen Shuang Qiu Yuanli Zhang Ting Zhang Bin Chen Yanjun Xu Qiyi Feng Jinxing Huang Zhihui Zhong Hongxia Li Guowei Che Kai Xiao | 2021 | Cancer Biology & Medicine2021,18,1: | 5
Plant multiscale networks:charting plant connectivity by multi-level analysis and imaging techniques显示文摘In multicellular and even single-celled organisms, individual components are interconnected at multiscale levels to produce enormously complex biological networks that help these systems maintain homeostasis for development and environmental adaptation.Systems biology studies initially adopted network analysis to explore how relationships between individual components give rise to complex biological processes.Network analysis has been applied to dissect the complex connectivity of mammalian brains across different scales in time and space in The Human Brain Project. In plant science, network analysis has similarly been applied to study the connectivity of plant components at the molecular, subcellular, cellular, organic, and organism levels.Analysis of these multiscale networks contributes to our understanding of how genotype determines phenotype. In this review,we summarized the theoretical framework of plant multiscale networks and introduced studies investigating plant networks by various experimental and computational modalities. We next discussed the currently available analytic methodologies and multi-level imaging techniques used to map multiscale networks in plants. Finally,we highlighted some of the technical challenges and key questions remaining to be addressed in this emerging field. | Xi Zhang Yi Man Xiaohong Zhuang Jinbo Shen Yi Zhang Yaning Cui Meng Yu Jingjing Xing Guangchao Wang Na Lian Zijian Hu Lingyu Ma Weiwei Shen Shunyao Yang Huimin Xu Jiahui Bian Yanping Jing Xiaojuan Li Ruili Li Tonglin Mao Yuling Jiao Sodmergen Haiyun Ren Jinxing Lin | 2021 | Science China(Life Sciences)2021,64,9: | 3
Stabilized cobalt-free lithium-rich cathode materials with an artificial lithium fluoride coating显示文摘Iron-substituted cobalt-free lithium-rich manganese-based materials, with advantages of high specific capacity, high safety, and low cost, have been considered as the potential cathodes for lithium ion batteries. However, challenges, such as poor cycle stability and fast voltage fade during cycling under high potential, hinder these materials from commercialization. Here,we developed a method to directly coat LiF on the particle surface of Li_(1.2)Ni_(0.15)Fe_(0.1)Mn_(0.55O2).A uniform and flat film was successfully formed with a thickness about 3 nm,which can effect-ively protect the cathode material from irreversible phase transition during the deintercalation of Li^(+).After surface coating with 0.5wt%LiF,the cycling stability of Li_(1.2)Ni_(0.15)Fe_(0.1)Mn_(0.55O2) cycled at high potential was significantly improved and the voltage fade was largely suppressed. | Wei Liu Jinxing Li Hanying Xu Jie Li Xinping Qiu | 2022 | International Journal of Minerals,Metallurgy and Materials2022,29,5: | 3
Recent progress in Chinese fusion research based on superconducting tokamak configuration显示文摘Fusion energy is a promising source of clean energy, which could solve energy shortages and environmental pollution.Research into controlled fusion energy has been ongoing for over half a century.China has created a clear roadmap for magnetic confinement fusion development, where superconducting tokamaks will be used in commercial fusion reactors. The Experimental Advanced Superconducting Tokamak(EAST)is the world’s first fully superconducting tokamak with upper and lower divertors, which aims at longpulse,steady-state,H-mode operation, and 101-s H-mode discharge had been achieved. In 2007,China joined the International Thermonuclear Experimental Reactor(ITER)and became one of its seven members.Thirteen procurement packages are undertaken by China, covering superconducting magnets, power supplies,plasma-facing components(PFCs),diagnostics, etc. To bridge the gap between the ITER and fusion demonstration power plants(DEMOs),China is planning to build the Chinese Fusion Engineering Testing Reactor(CFETR)to demonstrate related technologies and physics models. The engineering design of the CFETR was completed in 2020,and Comprehensive Research Facilities for Fusion Technology(CRAFT)are being constructed to explore the key technologies used in the CFETR. | Jinxing Zheng Jinggang Qin Kun Lu Min Xu Xuru Duan Guosheng Xu Jiansheng Hu Xianzu Gong Qing Zang Zhihong Liu Liang Wang Rui Ding Jiming Chen Pengyuan Li Lei Xue Lijun Cai Yuntao Song | 2022 | The Innovation2022,3,4: | 3
Transparent,stretchable,temperature-stable and self-healing ionogel-based triboelectric nanogenerator for biomechanical energy collection显示文摘A flexible and stable power supply is essential to the rapid development of wearable electronic devices. In this work,a transparent,flexible,temperature-stable and ionogel electrode-based self-healing triboelectric nanogenerator(IS-TENG)was developed. The ionogel with excellent stretchability(1,012%),high ionic conductivity(0.3 S·m^(−1))and high-temperature stability(temperature range of−77 to 250℃)was used as the electrode of the IS-TENG.The IS-TENG exhibited excellent transparency(92.1%)and stability. The output performance did not decrease when placed in a 60℃oven for 48 h.In addition, the IS-TENG behaved like a stable output in the range of−20 to 60℃.More importantly, the IS-TENG could also achieve self-healing of electrical performance at temperatures between−20 and 60℃and its output can be restored to its original state after healing. When the single-electrode IS-TENG with an area of 3 cm×3 cm was conducted under the working frequency of 1.5 Hz,the output values for open-circuit voltage,short-circuit current,short-circuit transferred charge, and maximum peak power density were 189 V,6.2μA,57 nC,and 2.17 W·m^(−2),respectively.The IS-TENG enables to harvest biomechanical energy, and drive electronic devices.Furthermore, the application of IS-TENGs as self-driven sensors for detecting human behavior was also demonstrated, showing good application prospects in the field of wearable power technology and self-driven sensing. | Weiqiang Liao Xiukun Liu Yuqi Li Xu Xu Jinxing Jiang Shaorong Lu Dequan Bao Zhen Wen Xuhui Sun | 2022 | Nano Research2022,15,3: | 2
Studies on inner wall structure of tracheids in Taxus chinensis with resin casting method显示文摘The resin casting method has been applied to studying the inner wall structure of tracheids in normal and compression wood, Taxus chinensis in gymnosperm. Not only the structures which are visible by the conventional method, but also the three-dimensional structures of pits pair in intertracheids unvisualized by the normal way have been observed. Moreover, it is found that there exist many differences in the angle, trend and occurrence of helical thickenings and distribution of warty layers on the inner walls of tracheids in normal and compression wood. | Jianhua Xu Yuxi Hu Jinxing Lin | 1999 | Chinese Science Bulletin1999,44,15: | 2
Influence of different illuminations with xenon or microwave sulfur lamp on jointing and tillering of wheat显示文摘The emission of microwave sulfur lamp is mainly composed of visible light. The sulfur lamp, producing little infrared radiation, has high efficiency, long duration of illumination and less energy consumption. In order to probe the agricultural application of the lamp, experiments were carried out with wheat (cv. Yangmai 158) to emphatically study illuminating effects of xenon or sulfur lamp on tillering, jointing and net photosynthetic rate during tillering and jointing periods. Results indicated that there is almost no difference in net photosynthetic rate of wheat leaves growing under different lamps. Xenon lamp significantly advanced the time of jointing, decreased number of tillers, number of total leaves per plant and leaf size, as well as inhibited root growth. In contrast with xenon lamp, sulfur lamp evidently delays the time of heading and grain maturation, increased number of heads per plant, head length, total number of grains per plant, head weight and total grain weight per plant, | CHEN Qilin LIU Yuequn YU Xinjian CHEN Jinxing WENG Jun XU Chunhe | 2002 | Chinese Science Bulletin2002,47,17: | 2

Study on Chemical Synthesis and Insecticidal Activity of Peptideκ-CTx-btg02显示文摘[Objectives]This study was conducted to investigate the chemical synthesis of peptideκ-CTx-btg02.[Methods]Linear peptideκ-CTx-btg02 was synthesized by solid phase peptide synthesis(SPPS).After oxidation and folding of the linear peptide, mass spectrometry identification and high performance liquid chromatography(HPLC)purification were performed. Then, the MTT method and insect injection method were applied to study its insecticidal activity.[Results]The peptideκ-CTx-btg02 was successfully synthesized by the SPPS method, and identified for the formation of disulfide bonds by mass spectrometry identification, and the purity after HPLC separation was greater than 95%.The MTT experiment showed that the peptideκ-CTx-btg02 could inhibit the growth of insect cells sf9,with a half effective dose of 0.13 nM.The insect injection experiment showed that the peptideκ-CTx-btg02 could effectively kill Tenebrio molitor, with a half lethal dose of 15.6 nM.The results of the electrophysiological experiment showed that 10μM peptideκ-CTx-btg02 had no blocking activity on murine acetylcholineα2β3 andα3β4.Therefore,the peptideκ-CTx-btg02 had a good inhibitory effect on the growth of insect cells, highly effective insecticidal activity and weak mammalian toxicity.[Conclusions]This study lays a foundation for the development of new, safe and efficient peptide biological insecticides. | Qin CHEN Jinxing FU Min XU Bingmiao GAO | 2021 | Agricultural Biotechnology2021,10,4: | 1

Regulation of artificial supramolecular transmembrane signal transduction by selenium-containing artificial enzyme receptors显示文摘Signal transduction across lipid bilayers is of profound importance in biological processes. In biological systems, natural enzymes mediate biochemical effects by binding to substrates and facilitating the conversion of external signals into physiological responses.Sequential transmission of biological signals from one enzyme to the next promotes signal transduction with feedforward and feedback mechanisms.Reconstructing these processes in an artificial system provides potential applications and offers a new way to understand fundamental biological processes in depth. However, the design of artificial signal transduction systems regulated by artificial enzyme receptors in a predictable and intelligent manner remains a challenge.Herein, benefiting from the polarity-regulated characteristics of Se-containing compounds with artificial glutathione peroxidase(GPx)activity,we constructed an artificial transmembrane signaling receptor with a Se-containing GPx-like recognition head group,a membrane-anchoring group, and a pre-enzyme end group. The artificial supramolecular signal transduction system containing such signal transduction receptors extends the range of signaling systems based on enzyme regulation, which provides a new way to study natural signal processes in cells and artificially regulated biological processes. | Shengda Liu Yunpeng Xing Tengfei Yan Yijia Li Ruizhen Tian Jinxing Hou Jiayun Xu Tingting Wang Junqiu Liu | 2023 | Nano Research2023,16,1: | 0
