1Molecular Epidemiological Analysis of Echovirus 19 Isolated From an Outbreak Associated With Hand, Foot, and Mouth Disease (HFMD) in Shandong Province of China显示文摘Objective To elucidate the genetic characterization and molecular epidemiological features of Echovirus 19(E19)isolates collected from an outbreak associated with hand,foot and mouth disease(HFMD)in Tai'an city of Shandong Province of China from July to September,2003.Methods Thirty seven Echovirus 19 isolates were isolated from stool specimens and throat swabs collected during the outbreak,then major capsid(VP1)genomic sequence was determined,and phylogenetic tree was done based on the VP1 sequences among these 37 and other E19 viruses deposited in the Genbank.Also a representative strain named CHN-SD03-TN12 was selected for sequencing of 5'-untranslated regions(5'-UTR).Results The identity rate was about 98.9%-100% among all these 37 E19 viruses.The genetic relationships between these 37 E19 isolates and other strains reported were also depicted.The identity rate was about 78.4%-78.9% compared with E19 reference strain Burke.The substitutions in the sequence of 5'-UTR resulted in changes in the conjectural properties of 5'-UTR of E19 viruses.Conclusion The genetic features of E19 viruses isolated during the outbreak in Shandong Province in 2003 may be associated with a genetic and antigenic drift that changes the virulence of the Shandong isolates,but the molecular changes in Shandong E19 viruses contributing to their phenotype remain to be further illuminated.However,the sequences described in this paper substantiate the changes taken place in capsid VP1 and 5'UTR regions.These substitutions may contribute to their tropism and virulence,and play a significant role in pathogenesis and clinical manifestations of the disease.ZHEN ZHU WEN-BO XU AI-QIANG XU HAI-YAN WANG YONG ZHANG LI-ZHI SONG Hui-LiYANG YANLI FENGJI 2007Biomedical and Environmental Sciences2007,20,4:22
2An investigation of surface deformation after fully mechanized,solid back fill mining显示文摘The surface deformation after fully mechanized back filling mining was analyzed.The surface deformation for different backfill materials was predicted by an equivalent mining height model and numerical simulations.The results suggest that:(1) As the elastic modulus,E,of the backfill material increases the surface subsidence decreases.The rate of subsidence decrease drops after E is larger than 5 GPa;(2) Fully mechanized back fill mining technology can effectively control surface deformation.The resulting surface deformation is within the specification grade I,which means surface maintenance is not needed.A site survey showed that the equivalent mining height model is capable of predicting and analyzing surface deformation and that the model is conservative enough for engineering safety.Finally,the significance of establishing a complete error correction system based on error analysis and correction is discussed.Li Jian Zhang Jixiong Huang Yanli Zhang Qiang Xu Junming 2012International Journal of Mining Science and Technology2012,22,4:23
3RNA-Seq analysis of yak ovary: improving yak gene structure information and mining reproduction-related genes显示文摘RNA-Seq,a high-throughput(HT)sequencing technique,has been used effectively in large-scale transcriptomic studies,and is particularly useful for improving gene structure information and mining of new genes.In this study,RNA-Seq HT technology was employed to analyze the transcriptome of yak ovary.After Illumina-Solexa deep sequencing,26826516 clean reads with a total of 4828772880 bp were obtained from the ovary library.Alignment analysis showed that 16992 yak genes mapped to the yak genome and 3734 of these genes were involved in alternative splicing.Gene structure refinement analysis showed that7340 genes that were annotated in the yak genome could be extended at the 5′or 3′ends based on the alignments been the transcripts and the genome sequence.Novel transcript prediction analysis identified 6321 new transcripts with lengths ranging from 180 to 14884 bp,and 2267 of them were predicted to code proteins.BLAST analysis of the new transcripts showed that1200?4933 mapped to the non-redundant(nr),nucleotide(nt)and/or SwissProt sequence databases.Comparative statistical analysis of the new mapped transcripts showed that the majority of them were similar to genes in Bos taurus(41.4%),Bos grunniens mutus(33.0%),Ovis aries(6.3%),Homo sapiens(2.8%),Mus musculus(1.6%)and other species.Functional analysis showed that these expressed genes were involved in various Gene Ontology(GO)categories and Kyoto Encyclopedia of Genes and Genomes pathways.GO analysis of the new transcripts found that the largest proportion of them was associated with reproduction.The results of this study will provide a basis for describing the normal transcriptome map of yak ovary and for future studies on yak breeding performance.Moreover,the results confirmed that RNA-Seq HT technology is highly advantageous in improving gene structure information and mining of new genes,as well as in providing valuable data to expand the yak genome information.LAN DaoLiang XIONG XianRong WEI YanLi XU Tong ZHONG JinCheng ZHI XiangDong WANG Yong LI Jian 2014Science China(Life Sciences)2014,57,9:23
4Plant-derived phosphocholine facilitates cellular uptake of anti-pulmonary fibrotic HJT-sRNA-m7显示文摘Pulmonary fibrosis, a progressive chronic disease with a high mortality rate, has limited treatment options. Currently, lung transplantation remains the only effective treatment. Here we report that a small RNA, HJT-sRNA-m7, from a Chinese herbal medicine Hong Jing Tian(HJT, RHODIOHAE CRENULATAE RADIX ET RHIZOMA, Rhodiola crenulata) can effectively reduce the expressions of fibrotic hallmark genes and proteins both in alveolar in vitro and in mouse lung tissues in vivo. We also discovered over one hundred oil-soluble chemicals from HJT decoctions, most of which are found in lipid extracts from other Chinese herbals decoctions, including Pu Gong Ying(PGY, TARAXACI HERBA, Taraxacum mongolicum), Chuan Xin Lian(CXL, changed to 'ANDROGRAPHIS HERBA, Andrographis paniculata'), and Jin Yin Hua(JYH, lonicera japonica or Honeysuckle). We identified the active component in these decoctions as two forms of phosphocholines, PC(18:0/18:2) and PC(16:0/18:2). These PCs potentially could form liposomes with small RNAs to enter human alveolar and gastric cells. 5The Genetic Architecture of Flowering Time and Photoperiod Sensitivity in Maize as Revealed by QTL Review and Meta Analysis显示文摘The control of flowering is not only important for reproduction,but also plays a key role in the processes of domestication and adaptation.To reveal the genetic architecture for flowering time and photoperiod sensitivity,a comprehensive evaluation of the relevant literature was performed and followed by meta analysis.A total of 25 synthetic consensus quantitative trait loci(QTL)and four hot-spot genomic regions were identified for photoperiod sensitivity including 11 genes related to photoperiod response or flower morphogenesis and development.Besides,a comparative analysis of the QTL for flowering time and photoperiod sensitivity highlighted the regions containing shared and unique QTL for the two traits.Candidate genes associated with maize flowering were identified through integrated analysis of the homologous genes for flowering time in plants and the consensus QTL regions for photoperiod sensitivity in maize(Zea mays L.).Our results suggest that the combination of literature review,meta-analysis and homologous blast is an efficient approach to identify new candidate genes and create a global view of the genetic architecture for maize photoperiodic flowering.Sequences of candidate genes can be used to develop molecular markers for various models of marker-assisted selection,such as marker-assisted recurrent selection and genomic selection that can contribute significantly to crop environmental adaptation.Jie Xu Yaxi Liu Jian Liu Moju Cao Jing Wang Hai Lan Yunbi Xu Yanli Lu Guangtang Pan Tingzhao Rong 2012Journal of Integrative Plant Biology2012,54,6:14
6Curative effect of acupuncture and moxibustion on insomnia: a randomized clinical trial显示文摘OBJECTIVE: To observe the effect of acupuncture and moxibustion on insomnia and explore its mechanism. METHODS: One hundred and twenty patients were randomly divided into an experiment group and a control group. Sixty patients in the experiment group were treated once a day with acupuncture at Baihui (GV 20), Sishencong (EX-HN 1), Shenmai (BL 62), and Zhaohai (KI 6) and with moxibustion at Baihui (GV 20) and Sishencong (EX-HN 1). Sixty patients in the control group were acupunctured once a day at Shenmen (HT 7), Neiguan (PC 6), and Sanyinjiao (SP 6).The Pittsburgh Sleep Quality Index (PSQI) was used to compare sleep improvement between the two groups. RESULTS: The total effective rate was 87.7% in the experiment group and 76.3% in the control group. The PSQI scores and the total score were lower after treatment than before treatment in bothgroups. However, the reduction in the experiment group was greater than that in the control group in sleeping quality, time to fall asleep, sleeping disorder, and daytime function (P<0.05). 7Clinical applications of MARSALA for preimplantation genetic diagnosis of spinal muscular atrophy显示文摘Conventional PCR methods combined with linkage analysis based on short tandem repeats(STRs) or Karyomapping with single nucleotide polymorphism(SNP) arrays, have been applied to preimplantation genetic diagnosis(PGD) for spinal muscular atrophy(SMA), an autosome recessive disorder. However, it has limitations in SMA diagnosis by Karyomapping, and these methods are unable to distinguish wildtype embryos with carriers effectively. Mutated allele revealed by sequencing with aneuploidy and linkage analyses(MARSALA) is a new method allowing embryo selection by a one-step next-generation sequencing(NGS) procedure, which has been applied in PGD for both autosome dominant and X-linked diseases in our group previously. In this study, we carried out PGD based on MARSALA for two carrier families with SMA affected children. As a result, one of the couples has given birth to a healthy baby free of mutations in SMA-causing gene. It is the first time that MARSALA was applied to PGD for SMA, and we can distinguish the embryos with heterozygous deletion(carriers) from the wild-type(normal) ones accurately through this NGS-based method. In addition, direct mutation detection allows us to identify the affected embryos(homozygous deletion), which can be regarded as probands for linkage analysis, in case that the affected family member is absent. 8Status and development of high-power laser facilities at the NLHPLP显示文摘In this paper, we review the status of the multifunctional experimental platform at the National Laboratory of High Power Laser and Physics(NLHPLP). The platform, including the SG-II laser facility, SG-II 9th beam, SG-II upgrade(SG-II UP) facility, and SG-II 5 PW facility, is operational and available for interested scientists studying inertial confinement fusion(ICF) and a broad range of high-energy-density physics. These facilities can provide important experimental capabilities by combining different pulse widths of nanosecond, picosecond, and femtosecond scales. In addition, the SG-II UP facility, consisting of a single petawatt system and an eight-beam nanosecond system, is introduced including several laser technologies that have been developed to ensure the performance of the facility. 9Simulation of CO_2 variations at Chinese background atmospheric monitoring stations between 2000 and 2009: Applying a CarbonTracker model显示文摘We carried out a downscaling treatment over China using the CarbonTracker numerical model,which was applied using double grid nesting technology(3°×2°over the whole globe,1°×1°over China),simulating and analyzing atmospheric CO2concentrations over 10 recent years(2000–2009).The simulation results agreed very well with observed data from four background atmospheric monitoring stations in China(The periods for which the simulation results and observed values be compared were January2000 to December 2009 for the WLG station and June 2006 to December 2009 for the SDZ,LFS,and LAN stations),giving correlation coefficients of>0.7.The high-resolution simulation data correlated slightly better than the low resolution simulation data with the observed data for three of the regions’atmospheric background stations.Further analysis of the annual,seasonal CO2concentration variations at the background stations showed that the CO2concentration increased each year over the study period,with an average annual increase of more than 5%,and annual increases of more than 7%at the Shangdianzi and Lin’an stations.Seasonal CO2variations were greater at the Longfengshan station than at the Shangdianzi or Lin’an stations.However,the CO2concentrations were higher at the Shangdianzi and Lin’an stations because they are greatly affected by human activities in the Jingjinji and Changjiang Delta economic zones.Spatial distribution in CO2concentrations and fluxes were higher in eastern than in western China.CHENG YanLi AN XingQin YUN FangHua ZHOU LingXi LIU LiXin FANG ShuangXi XU Lin 2013Chinese Science Bulletin2013,58,32:8
10Experimental study on the mechanical properties and consolidation mechanism of microbial grouted backfill显示文摘Backfill mining technology is the practice of returning waste materials underground for both disposal and geotechnical stability,however,a challenge with current technologies is that they commonly require cement-based binders which have a relatively high environmental impact.Finding alternatives to cement-based binders can improve environmental performance and this paper proposes microbial grouted backfill(MGB)as a potential solution.In this paper,the effects of the cementation solution concentration(CSC),volume ratio of bacterial solution to cementation solution(VRBC),particle sizes of the aggregates,and the number of grouting batches on the mechanical properties of MGB are studied.The experimental results show that MGB strength increased,up to a peak value,as CSC was increased,before decreasing as CSC was increased further.The results also show that MGB strength increased,up to a peak value,as VRBC decreased,before decreasing as the VRBC was decreased further.The peak strength was achieved at a CSC of 2 mol/L and a VRBC of 1:9.The strength of the MGB also increased as the number of grouting batches increased.Graded MGB samples showed the highest UCS,25.12 MPa,at particle sizes of 0.2 to 0.8 mm,while full(non-graded)MGB samples displayed mean UCS values ranging from1.56 MPa when the maximum particle size was 0.2 mm,up to 13 MPa when the maximum particle size was 1.2 mm.MGB samples are consolidated by the calcium carbonate that is precipitated during microbial metabolism,and the strength of MGB increases linearly as calcium carbonate content increases.The calcium carbonate minerals produced in MGB materials are primarily calcite,with secondary amounts of vaterite.Xuejie Deng Yu Li Fei Wang Xiaoming Shi Yinchao Yang Xichen Xu Yanli Huang Benjamin de Wit 2022International Journal of Mining Science and Technology2022,32,2:7

13Effect of Preparation Process on Compressive Strength and Hydrogenation Performance of Raney-Ni/Al2O3 Catalyst显示文摘In this study, Raney-Ni/Al_2O_3 catalysts were prepared from precursors which were calcined in different atmosphere(argon and air) and leached for different time. XRD, SEM, H_2-TPR and BET techniques were introduced to characterize the catalysts, and hydrogenation of unsaturated hydrocarbons, including vinyltoluene, dicyclopentadiene and indene, was used as probe reaction to evaluate the catalytic hydrogenation performance of the catalysts. The results demonstrated that the calcination atmosphere had noticeable effects on the compressive strength and hydrogenation performance of Raney-Ni/Al_2O_3. The catalyst calcined in argon exhibited better compressive strength and catalytic performance than the one calcined in air. The compressive strength and activity stability of catalyst were also determined by the leaching time. A too longer leaching time was not conducive to improving the compressive strength and catalytic performance of catalysts. When the precursor calcined in argon was treated in a 15%(by wt.) sodium hydroxide solution at 343 K for 5 h, a catalyst with higher compressive strength(at 26.10 N/mm) was obtained and the conversion of vinyltoluene, dicyclopentadiene(DCPD), and indene could amount to more than 87.5%, 99% and even 100%, respectively, under the reaction conditions covering a temperature of 493 K, a hydrogen pressure of 2.0 MPa, a LSHV of 3.0 h^(-1), and a hydrogen/oil ratio of 200:1.Luo Guohua Wang Yanli Sun Daming Xu Xin Jin Haibo 2017China Petroleum Processing & Petrochemical Technology2017,19,2:5
14A Comparative Study on Hydrodynamic Performance of Double Deflector Rectangular Cambered Otter Board显示文摘In the present work,the hydrodynamic performance of the double deflector rectangular cambered otter board was studied using wind tunnel experiment,flume tank experiment and numerical simulation.Results showed that the otter board had a good hydrodynamic performance with the maximum lift-to-drag ratio(K_(MAX) = 3.70).The flow separation occurred when the angle of attack(AOA) was at 45?,which revealed that the double deflector structure of the otter board can delay the flow separation.Numerical simulation results showed a good agreement with experiment ones,and could predict the critical AOA,which showed that it can be used to study the hydrodynamic performance of the otter board with the advantage of flow visualization.However,the drag coefficient in flume tank was much higher than that in wind tunnel,which resulted in a lower lift-to-drag ratio.These may be due to different fluid media between flume tank and wind tunnel,which result in the big difference of the vortexes around the otter board.Given the otter boards are operated in water,it was suggested to apply both flume tank experiment and numerical simulation to study the hydrodynamic performance of otter board.XU Qingchang FENG Chunlei HUANG Liuyi XU Jiqiang WANG Lei ZHANG Xun LIANG Zhenlin TANG Yanli ZHAO Fenfang WANG Xinxin 2018Journal of Ocean University of China2018,17,5:5
15Omics-based integrated analysis identified ATRX as a biomarker associated with glioma diagnosis and prognosis显示文摘Objective:ATRX is a multifunctional protein that is tightly regulated by and implicated in transcriptional regulation and chromatin remodeling.Numerous studies have shown that genetic alterations in ATRX play a significant role in gliomas.This study aims to further determine the relationship between ATRX and glioma prognosis and identify possible mechanisms for exploring the biological significance of ATRX using large data sets.Methods:We used The Cancer Genome Atlas(TCGA)database and 130 immunohistochemical results to confirm the difference in ATRX mutations in high-and low-grade gliomas.An online analysis of the TCGA glioma datasets using the cBioPortal platform was performed to study the relationship between ATRX mutations and IDH1,TP53,CDKN2 A and CDKN2 B mutations in the corresponding TCGA glioma dataset.In combination with clinical pathology data,the biological significance of the relationships were analyzed.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analyses and annotations of all adjacent genes in the network were performedin the Database for Annotation,Visualization and Integrated Discovery(DAVID)and R language.A protein-protein interaction(PPI)network was constructed,and the interactions of all adjacent nodes were analyzed by the String database and using Cytoscape software.Results:In the selected TCGA glioma datasets,a total of 2,228 patients were queried,21%of whom had ATRX alterations,which co-occurred frequently with TP53 and IDH1 mutations.ATRX alterations are associated with multiple critical molecular events,which results in a significantly improved overall survival(OS)rate.In low-grade gliomas,ATRX mutations are significantly associated with multiple important molecular events,such as ZNF274 and FDXR at mRNA and protein levels.A functional cluster analysis revealed that these genes played a role in chromatin binding and P53,and a link was observed between ATRX and IDH1 and TP53 in the interaction network.ATRX and TP53 are important nodes in the network and have potential links with the blood oxygen imbalance.Conclusions:ATRX mutations have clinical implications for the molecular diagnosis of gliomas and can provide diagnostic and prognostic information for gliomas.ATRX is expected to serve as a new therapeutic target.Yingbin Xie Yanli Tan Chao Yang Xuehao Zhang Can Xu Xiaoxia Qiao Jianglong Xu Shaohui Tian Chuan Fang Chunsheng Kang 2019Cancer Biology & Medicine2019,16,4:5
16The Bethesda System for Reporting Thyroid Cytopathology(TBSRTC):A report of 2,781 cases in a Chinese population显示文摘Objective:To evaluate the diagnostic utility of The Bethesda System for Reporting Thyroid Cytology(TBSRTC)at Peking University Cancer Hospital,the incidence of noninvasive follicular thyroid neoplasms with papillary-like nuclear features(NIFTP),and the impact of reclassification on cytopathological outcomes.Methods:We performed a retrospective study of thyroid fine-needle aspiration(FNA)cases between April 2014 and March 2019.The FNA results were classified according to TBSRTC.Post-surgery histological findings were followed up.Results:A total of 2,781 thyroid FNAs were performed.The incidences of the 6 diagnostic categories(DCs I-VI)were 14.8%,17.1%,15.8%,2.3%,11.6%and 38.5%,respectively.A total of 1,122 patients(40.3%)had corresponding histological results.NIFTP accounted for 0.4%of papillary thyroid carcinoma(PTC)cases,and there was no significant difference in the risk of malignancy(ROM)for each TBSRTC DC between'NIFTP=carcinoma(Ca)'and'NIFTP≠Ca'.When'NIFTP=Ca',the sensitivity,specificity,positive predictive value(PPV),negative predictive value(NPV),and accuracy of TBSRTC were 98.0%,84.0%,99.4%,58.3%,and97.5%,respectively.When'NIFTP≠Ca',the sensitivity,specificity,PPV,NPV and accuracy of the TBSRTC were 98.1%,81.5%,99.3%,61.1%,and 97.5%,respectively.Conclusions:TBSRTC is effective in the preoperative diagnosis of thyroid nodules in Peking University Cancer Hospital.The impact of the reclassification of NIFTP on cytopathological outcomes is limited because of its low incidence,and the revised ROMs are not suitable for Asian patients.Yanli Zhu Yuntao Song Guohui Xu Zhihui Fan Wenhao Ren 2020Chinese Journal of Cancer Research2020,32,2:4
17Genetic testing and PGD for unexplained recurrent fetal malformations with MAGEL2 gene mutation显示文摘Birth defects are caused by multiple factors,such as chromosome abnormality,environmental factors,and maternal factors.In this study,we focused on exploring the genetic causes of a non-consanguineous couple who suffered from four times of unsuccessful pregnancy due to unexplained recurrent fetal malformations with similar symptoms and normal chromosome copy number variations.Using trio-whole exome sequencing(trio-WES) for this couple and one of the affected fetuses,we found a mutation,c.1996 delC on the maternal imprinted gene MAGEL2 that was carried by the affected fetus and husband,leading to Schaaf-Yang syndrome.To screen this mutation,we further performed preimplantation genetic diagnosis(PGD) strategy followed by a gene pedigree validation and pathogenicity analysis.After the transfer of a PGD-screened embryo,a normal newborn without previous abnormal symptoms was born(February 15,2019).We present the first data that identified a pathogenic gene(MAGEL2 c.1996 delC) in a fetus with Schaaf-Yang syndrome in the EAS(East Asian) database and overcame this genetic defect by using processed PGD for this couple based on the WES results.Wei Guo Yanli Nie Zhiqiang Yan Xiaohui Zhu Yuqian Wang Shuo Guan Ying Kuo Wenxin Zhang Xu Zhi Yuan Wei Liying Yan Jie Qiao 2019Science China(Life Sciences)2019,62,7:4
18Assessment of Heavy Metals in Grain Fields with Long-term Treatment of Agrochemicals and Organic Manure in Hailun City of Northeast China显示文摘Heavy metals were evaluated in the soils of grain fields in Hailun City Experimental Research Station--Chinese Academy of Sciences (CAS). Four categories of the soil samples were chosen from the fields after 19 years of treatment with chemicals, no fertilizer, organic manure, and organic manure combined with chemical fertilizer. The study revealed a prevalence of significantly high concentrations of Cu, Zn and Pb in organic manure combined with chemical fertilizer in the grain fields of black soils. The concentrations of Cu and Zn in no fertilizer treatment, chemical treatment, and organic treatment also exceeded the permissible limits. 20Anti-tumor effect of the extract from Qingyihuaji formula on pancreatic cancer by down-regulating Notch-4 and Jagged-1显示文摘OBJECTIVE: To investigate, in terms of Notch signaling pathway, the effect on pancreatic cancer of the extract of an anti-tumor prescription — Qingyihuaji formula(QYHJ) — from Traditional Chinese Medicine(TCM).METHODS: Nude mice were implanted subcutaneously with human pancreatic cancer cell line SW1990 and then randomly divided into four groups: Control, QYHJ extract, Gemcitabine, and Combination of QYHJ extract and gemcitabine.Treatments were given for 21 days and tumor growth was evaluated simultaneously. Then, expression of Notch receptors(Notch-1, Notch-2,Notch-3, and Notch-4) and their Jagged ligands(Jagged-1 and Jagged-2) in dissected tumor tissuewere detected by real-time quantitative reverse transcription-polymerase chain reaction and Western blot. Finally, immunohistochemistry was performed to detect CD133, a marker of pancreatic cancer stem cells(CSCs), to evaluate the impact of QYHJ extract on pancreatic CSCs.RESULTS: QYHJ extract treatment effectively inhibited the tumor growth in nude mice. The expression of both Notch-4 and Jagged-1 were decreased significantly in QYHJ treatment groups(P < 0.05), while gemcitabine alone had no significant effect in down-regulating Jagged-1(P > 0.05).No significant difference was observed in the expression of Notch-1, Notch-2, Notch-3, and Jagged-2 between three treatment groups and control group(P > 0.05). Moreover, immunohistochemical analysis showed that the number of CD133 positive cells was significantly reduced by QYHJ treatment(P < 0.05), and the combined treatment was more effective than gemcitabine alone(P < 0.05).CONCLUSION: The role of the extract in pancreatic cancer treatment was associated with down-regulation of Notch-4 and Jagged-1 in Notch signaling pathway.
