| 3 | Relationship between the expression of iNOS,VEGF,tumor angiogenesis and gastric cancer显示文摘AIM:To in vestigate the relationship between the expression of inducible nitric oxide synthase(iNOS),vascular endothelial growth factor(VEGF),the microvascular density(MVD)and the pathological features and clinical staging of gastric cancer.METHODS:Immunohistochemical staining was used for detecting the expression of iNOS and VEGFin46resected specimens of gastric carcinoma;the monoclonal antibody against CD34 was used for displaying vascular endothelial cells,and MVD was detected by counting of CD34-positive vascular endothelial cells.RESULTS:Of 46resected specimens of gastric carcinoma,the rates of expressions of iNOS and VEGF were 58.70%and76.09%,respectively,and MVDaveraged55.59±19.39,Judged by the standard TNM criteria,the rate of expression of iNOS in stageⅣ(84.46%)was higher than those in stageⅠ,Ⅱ,Ⅲ(Fish exact probabilities test,P=0.019,0.023and 0.033,respectively);the rates of expression of VEGFin stage Ⅲ,Ⅳ(76.0%,92.31%,respectively)were higher than those in stageⅠ,Ⅱ(Fis exact probabilities test,P=0.031,0.017,0.022and0.019).MVDs in stageⅢ,Ⅳ(64.72±14.96,67.09±18.29,respectively)were higher than those in stageⅠ,Ⅱ(t\2.378,4.015,2.503and2.450,P<0.05,P<0.001,P<0.001,P<0.05,respectively),In37gastric carcinoma specimens with lymph node metastasis,MVD(68.69±18.07)and the rates of expression of iNOS and VEGF(70.27%,83.78%,respectively)were higher than those in the specimens with absence of metastasis(t=2.205,X^2=6.3587,X^2=6.2584,P<0.01,P<0.05,P<0.05,respectively),MVD and the expressions of iNOS and EGF were not correlated to the location,size or grade of tumor,nor with the depth of invasion of tumor;MVDs in the positive iNOS and VEGF specimens(59.88±18.02,58.39±17.73,repectively)were higher than those in the negative iNOS and VEGF specimens(X^2=6.3587and 6.1574,P<0.05,P<0.05,respectively);thus the expressions of iNOS and VEGF was correlated to MVD,but the expression of iNOS was not correlated to that of VEGF,In addition.of the 46 surviving patients,the 5-year survival rate of patients with positive iNOS or VEGF tumors was significantly less than that of patients with negative iNOS-or VEGF tumors(X^2=4.3842and 5.4073,P<0.05,P<0.05.respectively).CONCLUSION:The expressions of iNOS and VEGF are colosely related to tumor angiogenesis,and are involved in the advancement and the lymph node metastasis;thusMVD and the expressions of iNOS and EGF may serve indexes for evaluating staging of gastric carcinoma and forecasting its risk of metastasis,which will help establish a comprehensive therapeutical measure of post-operative patients and provide a new approach to tumor therapy. | Zheng-Jun Song Ping Gong Yu-E Wu Department of Gastroenterology,First Affiliated Hospital of Xi’an Jiaotong University,Xi’an 710061,ShaanXi Province,China | 2002 | World Journal of Gastroenterology2002,8,4: | 87 |

It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. | 5 | Assessment of duodenogastric reflux by combined continuous intragastric pH and bilirubin monitoring显示文摘AIM: To assess the diagnostic value of a combination of continuous intragastric pH and bilirubin monitoring in the detection of duodenogastric reflux (DGR), and the effects of diet on the bilirubin absorbance.METHODS: 30 healthy volunteers were divided into twogroups: standard diet group (Group 1) 18 cases, free diet group (Group 2) 12 cases. Each subjects were subjected to simultaneous 24 hour intragastric pH and spectrophotometric bilirubin concentration monitoring (Bilitec 2000).RESULTS: There was no difference of preprandial phasebilirubin absorbance between two groups. The absorbanceof postprandial phase was significantly increased in group 2than group 1. There was no difference between preprandialphase and postprandial phase absorbance in group 1.Postprandial phase absorbance was significantly higher ingroup 2. In a comparison of bile reflux with intragastric pHduring night time, there were 4 types of reflux:Simultaneous increase in absorbance and pH in only 19.6%, increase in bilirubin with unchanged pH 33. 3 %, pHincrease with unchanged absorbance 36. 3 %, and bothunchanged in 10. 8 %. Linear regression analysis showed nocorrelation between percertage total time of pH < 4 aridpercentage total time of absortance > 0. 14, r=0.068, P<0.05.CONCLUSION: Because of the dietary effect, highabsorbance fluids or foods should be avoided in detection.Intrsgastric pH and bilirubin monitoring separately predictthe presence of duodenal (and/or pancreatic) reflux and bilereflux. They can not substitute for each other. | 8 | Memetic computation based on regulation between neural and immune systems: the framework and a case study显示文摘Lamarckian learning has been introduced into evolutionary computation to enhance the ability of local search. The relevant research topic, memetic computation, has received significant amount of interest. In this study, a novel memetic computational framework is proposed by simulating the integrated regulation between neural and immune systems. The Lamarckian learning strategy of simulating the unidirectional regulation of neural system on immune system is designed. Consequently, an immune memetic algorithm based on the Lamarckian learning is proposed for numerical optimization. The proposed algorithm combines the advantages of immune algorithms and mathematical programming, and performs well in both global and local search. METHODS:Rats were divided into SAP group and sham-operation group as a control group(n=6). Rhein,chrysophanol,rheochrysidin,magnolol,hesperidin and naringin in DCQD were quantified in rat serum by high performance liquid chromatography tandem mass spectrometry for studying their pharmacokinetics. RESULTS:Early absorption of each DCQD component was tended to degrade in SAP group after treatment with DCQD by gavage.The Cmax(chrysophanol,P= 0.0059;rheochrysidin,P=0.0288;magnolol,P= 0.0487;hesperidin,P=0.0277;naringin,P=0.0023) and AUC(rhein,P=0.0186;chrysophanol,P=0.0013; magnolol,P=0.001;hesperidin,P=0.0081;naringin, P=0.0272)of DCQD component were obviously lower in SAP group than in control group.The T1/2α of chrysophanol and rheochrysidin(P=0.0467 and 0.0005,respectively)and Tmax of chrysophanol and rheochrysidin(P=0.0101 and 0.0037,respectively) lasted longer in SAP group than in control group. | 11 | Clinical value of preoperative methylated septin 9 in Chinese colorectal cancer patients显示文摘BACKGROUND The methylated septin 9(mSEPT9) assay was the first blood-based test approved by the United States Food and Drug Administration as a colorectal screening test.However, the diagnostic and prognostic role of preoperative mSEPT9 for colorectal cancer(CRC) in Chinese patients is still unknown.AIM To improve the understanding of diagnostic and prognostic factors, serum m SEPT9 was detected in Chinese CRC patients.METHODS A retrospective analysis of 354 cases, of which 300 had CRC and 54 were normal,was performed in China. Patients' characteristics, treatments, and laboratory data, including age, the date of surgery, Union for International Cancer Control(UICC) stages, distant metastasis(M), and so on, were collected. Methylation levels of SEPT9 were quantified by quantitative, methylation-specific polymerase chain reaction before surgery. In addition, the effects of mSEPT9 on the occurrence and prognosis of 330 CRC cases from The Cancer Genome Atlas(TCGA) database were evaluated using bioinformatics analyses. Potential prognostic factors for overall survival(OS) and progression-free survival(PFS)were evaluated by Kaplan-Meier univariate analysis.RESULTSIn Chinese CRC patients, positive mSEPT9 was strongly associated with advanced UICC stages, deeper invasion by the primary tumor, and more distant metastasis. Methylation levels of SEPT9 were stage-dependent and showed a stepwise increase in UICC stages(I–IV), primary tumor categories(T1–T4),regional node categories(N0–N2), and distant metastasis categories(M0–M1).The patients with positive mSEPT9 showed a tendency toward lower PFS. After analyzing TCGA clinical data, the high mSEPT9 group was found to be obviously correlated only with more distant metastasis. The patients with high mSEPT9 levels showed a tendency toward lower OS. Besides, nine meaningful mSEPT9 sites were found to provide guidance for the follow-up studies.CONCLUSION MSEPT9 analysis may add valuable information to current tumor staging. | 13 | Evaluation on the accuracy of digital elevation models显示文摘There is a growing interest in investigating the accuracy of digital elevation model (DEM). However people usually have an unbalanced view on DEM errors. They emphasize DEM sampling errors, but ignore the impact of DEM resolution and terrain roughness on the accuracy of terrain representation. This research puts forward the concept of DEM terrain representation error (Et) and then investigates the generation, factors, measurement and simulation of DEM terrain representation errors. A multi-resolution and multi-relief comparative approach is used as the major methodology in this research. The experiment reveals a quantitative relationship between the error and the variation of resolution and terrain roughness at a global level. Root mean square error (RMS Et) is regressed against surface profile curvature (V) and DEM resolution (R) at 10 resolution levels. It is found that the RMS Et may be expressed as RMS Et = (0.0061 ( V+ 0.0052) ( R - 0.022 ( V + 0.2415. | 14 | Spatial and temporal change patterns of net primary productivity and its response to climate change in the Qinghai–Tibet Plateau of China from 2000 to 2015显示文摘The vegetation ecosystem of the Qinghai–Tibet Plateau in China,considered to be the′′natural laboratory′′of climate change in the world,has undergone profound changes under the stress of global change.Herein,we analyzed and discussed the spatial-temporal change patterns and the driving mechanisms of net primary productivity(NPP)in the Qinghai–Tibet Plateau from 2000 to 2015 based on the gravity center and correlation coefficient models.Subsequently,we quantitatively distinguished the relative effects of climate change(such as precipitation,temperature and evapotranspiration)and human activities(such as grazing and ecological construction)on the NPP changes using scenario analysis and Miami model based on the MOD17A3 and meteorological data.The average annual NPP in the Qinghai–Tibet Plateau showed a decreasing trend from the southeast to the northwest during 2000–2015.With respect to the inter-annual changes,the average annual NPP exhibited a fluctuating upward trend from 2000 to 2015,with a steep increase observed in 2005 and a high fluctuation observed from 2005 to 2015.In the Qinghai–Tibet Plateau,the regions with the increase in NPP(change rate higher than 10%)were mainly concentrated in the Three-River Source Region,the northern Hengduan Mountains,the middle and lower reaches of the Yarlung Zangbo River,and the eastern parts of the North Tibet Plateau,whereas the regions with the decrease in NPP(change rate lower than–10%)were mainly concentrated in the upper reaches of the Yarlung Zangbo River and the Ali Plateau.The gravity center of NPP in the Qinghai–Tibet Plateau has moved southwestward during 2000–2015,indicating that the increment and growth rate of NPP in the southwestern part is greater than those of NPP in the northeastern part.Further,a significant correlation was observed between NPP and climate factors in the Qinghai–Tibet Plateau.The regions exhibiting a significant correlation between NPP and precipitation were mainly located in the central and eastern Qinghai–Tibet Plateau,and the regions exhibiting a significant correlation between NPP and temperature were mainly located in the southern and eastern Qinghai–Tibet Plateau.Furthermore,the relative effects of climate change and human activities on the NPP changes in the Qinghai–Tibet Plateau exhibited significant spatial differences in three types of zones,i.e.,the climate change-dominant zone,the human activity-dominant zone,and the climate change and human activity interaction zone.These research results can provide theoretical and methodological supports to reveal the driving mechanisms of the regional ecosystems to the global change in the Qinghai–Tibet Plateau. | GUO Bing ZANG Wenqian YANG Fei HAN Baomin CHEN Shuting LIU Yue YANG Xiao HE Tianli CHEN Xi LIU Chunting GONG Rui | 2020 | Journal of Arid Land2020,12,1: | 12 |
| 15 | Anti-HBV effect of TAT-HBV targeted ribonuclease显示文摘AIM: To prepare and purify TAT-HBV targeted ribonuclease fusion protein, evaluate its transduction activity and investigate its effect on HBV replication in 2.2.15 cells.METHODS: The prokaryotic expression vector pTAT containing TR gene was used in transforming E. coli BL21(DE3) LysS and TR was expressed with the induction of IPTG. The TAT-TR fusion protein was purified using Ni-NTA-agrose and PD-10 desalting columns, and analyzed by SDSPAGE. Transduction efficiency of TAT-TR was detected with immunofluorescence assay and the concentration of HBeAg in the supernatant of the 2.2.15 cells was determined via solid-phase radioimmunoassay (spRIA). MTT assay was used to detect the cytotoxicity of TAT-TR.RESULTS: The SDS-PAGE showed that the TAT-TR fusion protein was purified successfully, and the purity of TAT-TR was 90 %. | 17 | Dynamic resistance exercise increases skeletal muscle-derived FSTL1 inducing cardiac angiogenesis via DIP2A-Smad2/3 in rats following myocardial infarction显示文摘Purpose:The aim of this study was to investigate the potential of dynamic resistance exercise to generate skeletal muscle-derived follistatin like-1(FSTL1),which may induce cardioprotection in rats following myocardial infarction(MI)by inducing angiogenesis.Methods:Male,adult Sprague-Dawley rats were randomly divided into 5 groups(n=12 in each group):sham group(S),sedentary MI group(MI),MI+resistance exercise group(MR),MI+adeno-associated virus(AAV)-FSTL1 injection group(MA),and MI+AAV-FSTL1 injection+resistance exercise group(MAR).The AAV-FSTL1 vector was prepared by molecular biology methods and injected into the anterior tibialis muscle.The MI model was established by ligation of the left anterior descending coronary artery.Rats in the MR and MAR groups underwent 4 weeks of dynamic resistance exercise training using a weighted climbing-up ladder.Heart function was evaluated by hemodynamic measures.Collagen volume fraction of myocardium was observed and analyzed by Masson’s staining.Human umbilical vein vessel endothelial cells culture and recombinant human FSTL1 protein or transforming growth factor-b receptor 1(TGFbR1)inhibitor treatment were used to elucidate the molecular signaling mechanism of FSTL1.Angiogenesis,cell proliferation,and disco interacting protein 2 homolog A(DIP2A)location were observed by immunofluorescence staining.The expression of FSTL1,DIP2A,and the activation of signaling pathways were detected by Western blotting.Angiogenesis of endothelial cells was observed by tubule experiment.One-way analysis of variance and Student’s t test were used for statistical analysis.Results:Resistance exercise stimulated the secretion of skeletal muscle FSTL1,which promoted myocardial angiogenesis,inhibited pathological remodeling,and protected cardiac function in MI rats.Exercise facilitated skeletal muscle FSTL1 to play a role in protecting the heart.Exogenous FSTL1 promoted the human umbilical vein vessel endothelial cells proliferation and up-regulated the expression of DIP2A,while TGFbR1 inhibitor intervention down-regulated the phosphorylation level of Smad2/3 and the expression of vascular endothelial growth factor-A,which was not conducive to angiogenesis.FSTL1 bound to the receptor,DIP2A,to regulate angiogenesis mainly through the Smad2/3 signaling pathway.FSTL1-DIP2A directly activated Smad2/3 and was not affected by TGFbR1.Conclusion:Dynamic resistance exercise stimulates the expression of skeletal muscle-derived FSTL1,which could supplement the insufficiency of cardiac FSTL1 and promote cardiac rehabilitation through the DIP2A-Smad2/3 signaling pathway in MI rats. | Yue Xi Meili Hao Qiaoqin Liang Yongxia Li Da-Wei Gong Zhenjun Tian | 2021 | Journal of Sport and Health Science2021,10,5: | 11 |
