1Preliminary report on methane emissions from the Three Gorges Reservoir in the summer drainage period显示文摘Recently reported summertime methane (CH4) emissions (6.7 ± 13.3 mg CH4/(m2·hr)) from newly created marshes in the drawdown area of the Three Gorges Reservoir (TGR), China have triggered broad concern in academic circles and among the public. The CH4 emissions from TGR water surfaces and drawdown areas were monitored from 3rd June to 16th October 2010 with floating and static chambers and gas chromatography. The average CH4 emission flux from permanently flooded areas in Zigui, Wushan and Yunyang Counties was (0.33 ± 0.09) mg CH4/(m2·hr). In half of these hottest months of the year, the wilderness, cropland and deforested drawdown sites were aerobic and located above water level, and the CH4 emissions were very small, ranging from a sink at 0.12 mg CH4/(m2·hr) to a source at 0.08 mg CH4/(m2·hr) except for one mud-covered site after flood. Mean CH4 emission in flooded drawdown sites was 0.34 mg CH4/(m2·hr). The emissions from the rice paddy sites in the drawdown area were averaged at (4.86 ± 2.31) mg CH4/(m2·hr). Excepting the rice-paddy sites, these results show much lower emission levels than previously reported. Our results indicated considerable spatial and temporal variation in CH4 emissions from the TGR. Human activities and occasional events, such as flood, may also affect emission levels. 4Performance of the Autism Spectrum Rating Scale and Social Responsiveness Scale in Identifying Autism Spectrum Disorder Among Cases of Intellectual Disability显示文摘The Autism Spectrum Rating Scale(ASRS) and the Social Responsiveness Scale(SRS) have been widely used for screening autism spectrum disorder(ASD) in the general population during epidemiological studies, but studies of individuals with intellectual disability(ID) are quite limited. Therefore, we recruited the parents/caregivers of 204 ASD cases, 71 ID cases aged 6–18 years from special education schools, and 402 typically developing(TD) children in the same age span from a communitybased population to complete the ASRS and SRS. The results showed that the ID group scored significantly lower on total and subscale scores than the ASD group on both scales(P \ 0.05) but higher than TD children(P \ 0.05).Receiver operating characteristic analyses demonstrated a similar fair performance in discriminating ASD from ID with the ASRS(area under the curve(AUC) = 0.709,sensitivity = 77.0%, specificity = 52.1%, positive predictive value(PPV) = 82.2%) and the SRS(AUC = 0.742,sensitivity = 59.8%, specificity = 77.5%, PPV = 88.4%).The results showed that individuals with ID had clear autistic traits and discriminating ASD from ID cases was quite challenging, while assessment tools such as ASRS and SRS, help to some degree.Chunpei Li Hao Zhou Tianqi Wang Shasha Long Xiaonan Du Xiu Xu Weili Yan Yi Wang 2018Neuroscience Bulletin2018,34,6:5

7The origin and evolution of the diosgenin biosynthetic pathway in yam显示文摘Diosgenin,mainly produced by Dioscorea species,is a traditional precursor of most hormonal drugs in the pharmaceutical industry.The mechanisms that underlie the origin and evolution of diosgenin biosynthesis in plants remain unclear.After sequencing the whole genome of Dioscorea zingiberensis,we revealed the evolutionary trajectory of the diosgenin biosynthetic pathway in Dioscorea and demonstrated the de novo biosynthesis of diosgenin in a yeast cell factory.First,we found that P450 gene duplication and neofunctionalization,driven by positive selection,played important roles in the origin of the diosgenin biosynthetic pathway.Subsequently,we found that the enrichment of diosgenin in the yam lineage was regulated by CpG islands,which evolved to regulate gene expression in the diosgenin pathway and balance the carbon flux between the biosynthesis of diosgenin and starch.Finally,by integrating genes fromplants,animals,and yeast,weheterologously synthesized diosgenin to 10mg/l in genetically-engineered yeast.Our study not only reveals the origin and evolutionary mechanisms of the diosgenin biosynthetic pathway in Dioscorea,but also introduces an alternative approach for the production of diosgenin through synthetic biology.Jian Cheng Jing Chen Xiaonan Liu Xiangchen Li Weixiong Zhang Zhubo Dai Lina Lu Xiang Zhou Jing Cai Xueli Zhang Huifeng Jiang Yanhe Ma 2021Plant Communications2021,2,1:2
8A time domain induced polarization relaxation time spectrum inversion method based on a damping factor and residual correction显示文摘Relaxation time spectra(RTS) derived from time domain induced polarization data(TDIP) are helpful to assess oil reservoir pore structures. However, due to the sensitivity to the signal-to-noise ratio(SNR), the inversion accuracy of the traditional singular value decomposition(SVD) inversion method reduces with a decrease of SNR. In order to enhance the inversion accuracy and improve robustness of the inversion method to the SNR, an improved inversion method, based on damping factor and spectrum component residual correction, is proposed in this study. The numerical inversion results show that the oscillation of the RTS derived from the SVD method increased with a decrease of SNR, which makes it impossible to get accurate inversion components. However, the SNR has little influence on inversion components of the improved method, and the RTS has high inversion accuracy and robustness. Moreover, RTS derived from core sample data is basically in accord with the pore-size distribution curve, and the RTS derived from the actual induced polarization logging data is smooth and continuous, which indicates that the improved method is practicable.Liu Xiaonan Kong Li Zhou Kaibo Zhang Pu 2014Petroleum Science2014,11,4:2

11The level of bile salt-stimulated lipase in the milk of Chinese women and its association with maternal BMI显示文摘This study aimed to investigate the bile salt-stimulated lipase(BSSL) concentration in the milk of Chinese women and its correlation with maternal body mass index(BMI), gestational diabetes mellitus(GDM) and gestational hypertensive disorder(GHD). The BSSL levels in the milk samples were measured by enzyme-linked immunosorbent assay(ELISA). BSSL level in colostrum milk of mothers with full-term infants was positively correlated with pregnancy week and negatively correlated with maternal pre-pregnancy BMI and BMI late in pregnancy. Moreover, the BSSL concentration in mature milk was positively correlated with BMI gain during pregnancy. The BSSL concentration in colostrum milk was lower in GDM mothers than in normal mothers. The BSSL helps infants digest fat in early life and its level was associated with lactation. 17PXR activation impairs hepatic glucose metabolism partly via inhibiting the HNF4α-GLUT2 pathway显示文摘Drug-induced hyperglycemia/diabetes is a global issue. Some drugs induce hyperglycemia by activating the pregnane X receptor(PXR), but the mechanism is unclear. Here, we report that PXR activation induces hyperglycemia by impairing hepatic glucose metabolism due to inhibition of the hepatocyte nuclear factor 4-alpha(HNF4 a)-glucose transporter 2(GLUT2) pathway. The PXR agonists atorvastatin and rifampicin significantly downregulated GLUT2 and HNF4 a expression, and impaired glucose uptake and utilization in HepG2 cells. Overexpression of PXR downregulated GLUT2 and HNF4 a expression, while silencing PXR upregulated HNF4 a and GLUT2 expression.Silencing HNF4 a decreased GLUT2 expression, while overexpressing HNF4 a increased GLUT2 expression and glucose uptake. Silencing PXR or overexpressing HNF4 a reversed the atorvastatininduced decrease in GLUT2 expression and glucose uptake. In human primary hepatocytes, atorvastatin downregulated GLUT2 and HNF4 a mRNA expression, which could be attenuated by silencing PXR. Silencing HNF4 a downregulated GLUT2 mRNA expression. These findings were reproduced with mouse primary hepatocytes. Hnf4 a plasmid increased Slc2 a2 promoter activity. Hnf4 a silencing or pregnenolone-16 a-carbonitrile(PCN) suppressed the Slc2 a2 promoter activity by decreasing HNF4 a recruitment to the Slc2 a2 promoter. Liver-specific Hnf4 a deletion and PCN impaired glucose tolerance and hepatic glucose uptake, and decreased the expression of hepatic HNF4 a and GLUT2. In conclusion, PXR activation impaired hepatic glucose metabolism partly by inhibiting the HNF4 aGLUT2 pathway. 20Generation and expression analysis of BAC humanized mice carrying HLA-DP401 haplotype显示文摘Background:Human leukocyte antigen(HLA)-DP is much less studied than other HLA class Ⅱ antigens,that is,HLA-DR and HLA-DQ,etc.However,the accumulating data have suggested the important roles of DP-restricted responses in the context of cancer,allergy,and infectious disease.Lack of animal models expressing these genes as authentic cis-haplotypes blocks our understanding for the role of HLA-DP haplotypes in immunity.Methods:To explore the potential cis-acting control elements involved in the tran-scriptional regulation of the HLA-DPA1/DPB1 gene,we performed the expression analysis using bacterial artificial chromosome(BAC)-based transgenic humanized mice in the C57BL/6 background,which carried the entire HLA-DP401 gene locus.We further developed a mouse model of Staphylococcus aureus pneumonia in HLA-DP401 humanized transgenic mice,and performed the analysis on the expres-sion pattern of HLA-DP401 and immunological responses in the model.Results:In this study,we screened and identified a BAC clone spanning the entire HLA-DP gene locus.DNA from this clone was analyzed for integrity by pulsed-field gel electrophoresis and then microinjected into fertilized mouse oocytes to produce transgenic founder animals.Nine sets of PCR primers for regional markers with an average distance of 15 kb between each primer were used to confirm the integrity of the transgene in the five transgenic lines carrying the HLA-DPA1/DPB1 gene.Transgene copy numbers were determined by real-time PCR analysis.HLA-DP401 gene expression was analyzed at the mRNA and protein level.Although infection with S aureus Newman did not alter the percentage of immune cells in the spleen and thymus from the HLA-DP401-H2-Aβ1 humanized mice.Increased expression of HLA-DP401 was observed in the thymus of the humanized mice infected by S aureus.Conclusions:We generated several BAC transgenic mice,and analyzed the expres-sion of HLA-DPA1/DPB1 in those mice.A model of S aureus-induced pneumonia in the HLA-DP401-H2-Aβ1^(-/-)��humanized mice was further developed,and S aureus in-fection upregulated the HLA-DP401 expression in thymus of those humanized mice.These findings demonstrate the potential of those HLA-DPA1/DPB1 transgenic humanized mice for developing animal models of infectious diseases and MHC-associated immunological diseases.Feng Li Meng-min Zhu Bo-wen Niu Ling-ling Liu Xiu-hua Peng Hua Yang Bo-yin Qin Meixiang Wang Xiaonan Ren Xiaohui Zhou 2021Animal Models and Experimental Medicine2021,4,2:1
