1Influence of cosmic radiation on lymphocyte micronucleus,serum lipid peroxide and antioxidation capacity in aircrew members显示文摘This study looks into the influence of cosmic radiation at high altitudes on human bodies. Results reveal that the cytokinesis-block micronuclei (CBMN) and conventional cultured micronuclei in peripheral blood lymphocytes, serum levels of lipid peroxide, superoxide dismutase, and the total antioxidation capacity by chemical colorimetry all increased significantly in aircrew members. There exists a linear relationship between the CBMN and the average annual effective doses of radiation received or the average annual flying hours. With both of them, a trend shows that the serum lipid peroxide levels increase as well. Either the lipid peroxide or CBMN can sensitively reflect the recent changes in flight load. 2Comprehensive evaluation of the metabolic effects of porcine CRTC3 overexpression on subcutaneous adipocytes with metabolomic and transcriptomic analyses显示文摘Background:Meat quality is largely driven by fat deposition,which is regulated by several genes and signaling pathways.The cyclic adenosine monophosphate(cAMP)-regulated transcriptional coactivator 3(CRTC3)is a coactivator of cAMP response element binding protein(CREB)that mediates the function of protein kinase A(PKA)signaling pathway and is involved in various biological processes including lipid and energy metabolism.However,the effects of CRTC3 on the metabolome and transcriptome of porcine subcutaneous adipocytes have not been studied yet.Here,we tested whether porcine CRTC3 expression would be related to fat deposition in Heigai pigs(a local fatty breed in China)and Duroc×Landrace×Yorkshire(DLY,a lean breed)pigs in vivo.The effects of adenovirus-induced CRTC3 overexpression on the metabolomic and transcriptomic profiles of subcutaneous adipocytes were also determined in vitro by performing mass spectrometry-based metabolomics combined with RNA sequencing(RNA-seq).Results:Porcine CRTC3 expression is associated with fat deposition in vivo.In addition,CRTC3 overexpression increased lipid accumulation and the expression of mature adipocyte-related genes in cultured porcine subcutaneous adipocytes.According to the metabolomic analysis,CRTC3 overexpression induced significant changes in adipocyte lipid,amino acid and nucleotide metabolites in vitro.The RNA-seq analysis suggested that CRTC3 overexpression alters the expression of genes and pathways involved in adipogenesis,fatty acid metabolism and glycerophospholipid metabolism in vitro.Conclusions:We identified significant alterations in the metabolite composition and the expression of genes and pathways involved in lipid metabolism in CRTC3-overexpressing adipocytes.Our results suggest that CRTC3 might play an important regulatory role in lipid metabolism and thus affects lipid accumulation in porcine subcutaneous adipocytes.Jiaqi Liu Jie Li Wentao Chen Xintao Xie Xingang Chu Teresa G.Valencak Yizhen Wang Tizhong Shan 2021Journal of Animal Science and Biotechnology2021,12,3:2

7Failure analysis and control technology of intersections of large‑scale variable cross‑section roadways in deep soft rock显示文摘In deep underground mining,achieving stable support for roadways along with long service life is critical and the complex geological environment at such depths frequently presents a major challenge.Owing to the coupling action of multiple factors such as deep high stress,adjacent faults,cross-layer design,weak lithology,broken surrounding rock,variable cross-sections,wide sections up to 9.9 m,and clusters of nearby chambers,there was severe deformation and breakdown in the No.10 intersection of the roadway of large-scale variable cross-section at the−760 m level in a coal mine.As there are insufcient examples in engineering methods pertaining to the geological environment described above,the numerical calculation model was oversimplifed and support theory underdeveloped;therefore,it is imperative to develop an efective support system for the stability and sustenance of deep roadways.In this study,a quantitative analysis of the geological environment of the roadway through feld observations,borehole-scoping,and ground stress testing is carried out to establish the FLAC 3D variable cross-section crossing roadway model.This model is combined with the strain softening constitutive(surrounding rock)and Mohr–Coulomb constitutive(other deep rock formations)models to construct a compression arch mechanical model for deep soft rock,based on the quadratic parabolic Mohr criterion.An integrated control technology of bolting and grouting that is mainly composed of a high-strength hollow grouting cable bolt equipped with modifed cement grouting materials and a high-elongation cable bolt is developed by analyzing the strengthening properties of the surrounding rock before and after bolting,based on the Heok-Brown criterion.As a result of on-site practice,the following conclusions are drawn:(1)The plastic zone of the roof of the cross roadway is approximately 6 m deep in this environment,the tectonic stress is nearly 30 MPa,and the surrounding rock is severely fractured.(2)The deformation of the roadway progressively increases from small to large cross-sections,almost doubling at the largest cross-section.The plastic zone is concentrated at the top plate and shoulder and decreases progressively from the two sides to the bottom corner.The range of stress concentration at the sides of the intersection roadway close to the passageway is wider and higher.(3)The 7 m-thick reinforced compression arch constructed under the strengthening support scheme has a bearing capacity enhanced by 1.8 to 2.3 times and increase in thickness of the bearing structure by 1.76 times as compared to the original scheme.(4)The increase in the mechanical parameters c andφof the surrounding rock after anchoring causes a signifcant increase inσt;the pulling force of the cable bolt beneath the new grouting material is more than twice that of ordinary cement grout,and according to the test,the supporting stress feld shows that the 7.24 m surrounding rock is compacted and strengthened in addition to providing a strong foundation for the bolt(cable).On-site monitoring shows that the 60-days convergence is less than 30 mm,indicating that the stability control of the roadway is successful.Shengrong Xie Yiyi Wu Dongdong Chen Ruipeng Liu Xintao Han Qiucheng Ye 2022International Journal of Coal Science & Technology2022,9,2:1

10Identification of Dynamic Active Sites Among Cu Species Derived from MOFs@CuPc for Electrocatalytic Nitrate Reduction Reaction to Ammonia显示文摘Direct electrochemical nitrate reduction reaction(NITRR)is a promising strategy to alleviate the unbalanced nitrogen cycle while achieving the electrosynthesis of ammonia.However,the restructuration of the high-activity Cu-based electrocatalysts in the NITRR process has hindered the identification of dynamical active sites and in-depth investigation of the catalytic mechanism.Herein,Cu species(single-atom,clusters,and nanoparticles)with tunable loading supported on N-doped TiO_(2)/C are successfully manufactured with MOFs@CuPc precursors via the pre-anchor and post-pyrolysis strategy.Restructuration behavior among Cu species is co-dependent on the Cu loading and reaction potential,as evidenced by the advanced operando X-ray absorption spectroscopy,and there exists an incompletely reversible transformation of the restructured structure to the initial state.Notably,restructured CuN_(4)&Cu_(4) deliver the high NH_(3) yield of 88.2 mmol h^(−1)g_(cata)^(−1) and FE(~94.3%)at−0.75 V,resulting from the optimal adsorption of NO_(3)^(−) as well as the rapid conversion of^(*)NH_(2)OH to^(*)NH_(2) intermediates originated from the modulation of charge distribution and d-band center for Cu site.This work not only uncovers CuN_(4)&Cu_(4) have the promising NITRR but also identifies the dynamic Cu species active sites that play a critical role in the efficient electrocatalytic reduction in nitrate to ammonia.Xue‑Yang Ji Ke Sun Zhi‑Kun Liu Xinghui Liu Weikang Dong Xintao Zuo Ruiwen Shao Jun Tao 2023Nano-Micro Letters2023,15,8:1
11Effects of Ovarian Preservation Time on in vitro Maturation and in vitro Fertilization of Oocytes from Slaughtered Sheep显示文摘[Objectives] The aim was to study the effects of ovarian preservation time on in vitro fertilization of oocytes from slaughtered sheep. [Methods] The collected ovaries were randomly and evenly divided into four groups. They were preserved in physiological saline containing penicillin( 100 IU/ml) and streptomycin( 100 μg/ml) at 15-20 ℃ for 0( Control),6,12 and 18 h,respectively. Then,the oocytes were subjected to in vitro fertilization. [Results]The maturation rates,cleavage rates and blastocyst rates of the oocytes preserved for 6 and 12 h showed no significant differences compared with those of the oocytes preserved for0 h( 72. 03%,70. 87% vs. 73. 68%; 74. 12%,72. 60% vs. 74. 49%; 22. 22%,20. 75% vs. 23. 29%)( P > 0. 05). There were also no significant differences in maturation rate,cleavage rate or blastocyst rate between the oocytes preserved for 18 and 0 h( P > 0. 05). 19Understanding the superior mechanical properties of Mg-3Al-Zn alloy sheets:Role of multi-type unique textures显示文摘Mg-3Al-1Zn(AZ31)sheets were produced by transverse gradient extrusion(TGE)process.The flow behavior and dynamic recrystallization during extrusion were systematically analyzed.The microstructures,textures,and mechanical behavior of extruded AZ31 sheet were also analyzed and compared with conventional extruded(CE)sheet.The results showed that fine grain structure and multi-type unique textures were formed in TGE sheet because of the generation of extra flow velocity along transverse direction(TD)and flow velocity gradient along extrusion direction(ED)during extrusion.The basal poles gradually deviated away normal direction(ND)from edge to center of the TGE sheet along TD,and the largest inclination angle at center region reached around 65°.Furthermore,the basal poles inclined from ED to TD 40°-63°,except for the center region of TGE sheet.The TGE sheet presented higher ductility and strain hardening exponent(n-value),but lower yield strength and Lankford value(r-value)in comparison with the CE sheet.Both the basalslip and tensile twins were easy to be activated during deformation,and the largest elongation of 41%and the lowest yield strength of 86.5 MPa were obtained for the ED-center sample in the TGE sheet.Jun Xu Jun Zhao Bin Jiang Wenjun Liu Hong Yang Xintao Li Yuehua Kang Nan Zhou Kaihong Zheng Fusheng Pan 2023International Journal of Minerals,Metallurgy and Materials2023,30,6:0
20Dual-engineered cartilage-targeting extracellular vesicles derived from mesenchymal stem cells enhance osteoarthritis treatment via miR-223/NLRP3/pyroptosis axis:Toward a precision therapy显示文摘Osteoarthritis(OA)is the most common disabling joint disease with no effective disease modifying drugs.Extracellular vesicles released by several types of mesenchymal stem cells could promote cartilage repair and ameliorate OA pathology in animal models,representing a novel therapeutic strategy.In this study,we demonstrated that extracellular vesicles derived from human umbilical cord mesenchymal stem cells(hUC-EVs)could maintain chondrocyte homeostasis and alleviate OA,and further revealed a novel molecular mechanism of this therapeutic effect.miR-223,which could directly bind with the 3′UTR of NLRP3 mRNA,was found to be a key miRNA for hUC-EVs to exert beneficial effects on inflammation inhibiting and cartilage protecting.For enhancing the effect on mitigating osteoarthritis,exogenous miR-223 was loaded into hUC-EVs by electroporation,and a collagen II-targeting peptide(WYRGRL)was modified onto the surface of hUC-EVs by genetic engineering to achieve a more targeted and efficient RNA delivery to the cartilage.The dual-engineered EVs showed a maximal effect on inhibiting the NLRP3 inflammasome activation and chondrocyte pyroptosis,and offered excellent results for the treatment of OA.This study provides a novel theoretical basis and a promising therapeutic strategy for the application of engineered extracellular vesicles in OA treatment.Weixuan Liu Anqi Liu Xujun Li Ziyang Sun Zhenghua Sun Yaru Liu Gang Wang Dan Huang Hao Xiong Shiyang Yu Xintao Zhang Cunyi Fan 2023Bioactive Materials2023,,12:0
