| 1 | Gallic acid inhibits fibroblast growth and migration in keloids through the AKT/ERK signaling pathway显示文摘Keloids are a fibroproliferative disorder of the skin resulted from abnormal healing of injured or irritated skin and are characterized by the ability to spread beyond the original boundary of the wound.Here,we tested the effect of gallic acid (GA),a plant polyphenol with selective growth inhibitory effects in cancer,on the proliferation and invasion of keloid fibroblasts (KFs)isolated from patients undergoing surgery.GA inhibited KF proliferation,migration,and invasion in parallel with the downregulation of matrix metalloproteinase-1 and -3 and upregulation of tissue inhibitors of metalloproteinase-1.Flow cytometric analysis showed that GA inhibited cell cycle progression and induced apoptosis.The effects of GA on KFs occurred in parallel with the inhibition of AKT and ERK1/2,suggesting that GA acts by suppressing the AKT/ERK signaling pathway.In ex vivo explant cultures of keloid tissues,GA inhibited the migration of KFs to the wound area and suppressed the expression of angiogenic markers concomitant with the inhibition of collagen deposition.These results identify GA as a potential therapeutic agent for the treatment of keloids and suggest a potential mechanism underlying its protective effect. | Xiuxia Wang Ke Liu Mengying Ruan Jun Yang Zhen Gao | 2018 | Acta Biochimica et Biophysica Sinica2018,50,11: | 6 |
| 2 | Development of an Inactivated Iridovirus Vaccine Against Turbot Viral Reddish Body Syndrome显示文摘Turbot(Scophthalmus maximus L.) reddish body iridovirus(TRBIV) was propagated in turbot fin cells(TF cells) and inactivated as the TRBIV vaccine with its protection efficiency evaluated in this study.TF cells were cultured in 10% bovine calf serum(BCS)-containing MEM medium(pH7.0) at 22℃,in which TRBIV propagated to a titer as high as 105.6 TCID50 mL-1.The TRBIV was inactivated with 0.1% formalin and formulated with 0.5% aluminum hydroxide.The inactivated vaccine caused neither cytopathogenic effect(CPE) on TF cells nor pathogenic effect on turbots.After being administered with the vaccine twice via muscle injection,the turbot developed high-tittered TRBIV neutralizing antibodies in a dose-dependent manner.The vaccine protected the turbot from dying with an immunoprotection rate of 83.3% as was determined via subcutaneous vaccination in the laboratory and 90.5% via bath vaccination in turbot farms,respectively.The inactivated vaccine was very immunogenic,efficiently preventing tur-bot from death.It holds the potential of being applied in aquaculture. | FAN Tingjun HU Xiuzhong WANG Liyan GENG Xiaofen JIANG Guojian YANG Xiuxia YU Miaomiao | 2012 | Journal of Ocean University of China2012,11,1: | 5 |

| 4 | Obstacle avoidance method of three-dimensional obstacle spherical cap显示文摘Focusing on obstacle avoidance in three-dimensional space for unmanned aerial vehicle(UAV), the direct obstacle avoidance method in dynamic space based on three-dimensional velocity obstacle spherical cap is proposed, which quantifies the influence of threatening obstacles through velocity obstacle spherical cap parameters. In addition, the obstacle avoidance schemes of any point on the critical curve during the multi-obstacles avoidance are given. Through prediction, the insertion point for the obstacle avoidance can be obtained and the flight path can be replanned. Taking the Pythagorean Hodograph(PH) curve trajectory re-planning as an example, the three-dimensional direct obstacle avoidance method in dynamic space is tested. | 5 | Establishment of a Spleen Cell Line from Large Yellow Croaker Pseudosciaena crocea and its Primitive Application in Foreign Gene Transfection显示文摘A large yellow croaker,Pseudosciaena crocea,spleen(LYCS)cell line was established and the feasibility of using it for foreign gene transfection was evaluaed in this study.Primary culture of LYCS cells was initiated from spleen tissue pieces,which were cultured at 25℃ in Dulbecco's modiced Eagle medium/F12 medium(DMEM/F12,1:1)(pH7.2),supplemented with 20% fetal bovine serum,carboxymethyl chitosan,chondroitin sulfate,basic fibroblast growth factor(bFGF)and insulin-like growth factor-I(IGF-I).The cultured LYCS cells,in fibroblast shape,proliferated to 100% confluency 20 days later.Chromosome analyses indicated that the LYCS cells exhibited chromosomal aneuploidy with a modal chromosome number of 48 which displayed the normal diploid karyotype of P.crocea(6m+6sm+36t,NF=60).A LYCS cell line,with a population doubling time of 48.7 h at passage 60,has been established and subcultured to passage 70.Transgenic feasibility test demonstrated that positive green fluorescence protein(GFP)expression was observed in LYCS cells after pcDNA3.1-GFP plasmid transfection.In conclusion,a continuous foreign gene trans-fection feasible LYCS cell line has been established successfully.The cell line might serve as a valuable tool for studies of transgenic breeding and has potential applications for different kinds of cytotechnological studies. | SUN Ai FAN Tingjun YANG Xiuxia XU Bin XU Xiaohui GUO Xueyang | 2011 | Journal of Ocean University of China2011,10,4: | 3 |
| 6 | Recent developments and progress of inorganic photo-stimulated phosphors显示文摘Photo-stimulated luminescence(PSL) is the process in which trapped charges are released by photons and produce luminescence through recombination. The variegated optical characteristics of photostimulated phosphors(PSPs) have drawn increasing attention and a large body of work encompassing mechanism and application of PSPs has been addressed. The optical data storage capacity resulting from abundant defect states enables PSPs to be applied to information storage. Moreover, PSPs provide potential application for anti-counterfeiting, as color changes due to the tunneling process. Recently, near infrared(NIR) light PSPs have been developed, exhibiting enormous potential for in vivo bio-imaging, as the stable and high noise-signal ratio characteristic of PSL. | 7 | Preparation of electrolyte membranes for micro tubular solid oxide fuel cells显示文摘Yttria-stabilized zirconia (YSZ) micro tubular electrolyte membranes for solid oxide fuel cells (SOFCs) were prepared via the combined wet phase inversion and sintering technique. The as-derived YSZ mi- cro tubes consist of a thin dense skin layer and a thick porous layer that can serve as the electrode of fuel cells. The dense and the porous electrolyte layers have the thickness of 3-5 μm and 70-90 μm, respectively, while the inner surface porosity of the porous layer is higher than 28.1%. The two layers are perfectly integrated together to preclude the crack or flake of electrolyte film from the electrode. | 8 | Establishment and Characterization of a New Marine Fish Cell Line from Ovary of Barfin Flounder(Verasper moseri)显示文摘A novel continuous ovary cell line from barfin flounder(Verasper moseri)(BFO cell line) was established with its primitive application in transgenic expression demonstrated in this study. Primarily cultured cells grew well at 22℃ in Dulbecco's modified Eagle medium/F12 medium(DMEM/F12, 1:1; p H 7.2) supplemented with 20% fetal bovine serum(FBS), carboxymethyl chitooligosaccharide, basic fibroblast growth factor(b FGF) and insulin-like growth factor-I(IGF-I). The primary BFO cells in fibroblastic morphology proliferated into a confluent monolayer about 2 weeks later, and were able to be subcultured. Impacts of medium and temperature on the growth of the cells were examined. The optimum growth was found in DMEM/F12 with 20% FBS and at 22℃. The BFO cells can be continuously subcultured to Passage 120 steadily with a population doubling time of 32.7 h at Passage 60. Chromosome analysis revealed that 72% of BFO cells at Passage 60 maintained the normal diploid chromosome number(46) with a normal karyotype of 2st+44t. The results of gene transformation indicated that green fluorescence protein(GFP) positively expressed in these cells after being transformed with pc DNA3.1-GFP. | 17 | Development of a novel platform for recombinant protein production in Corynebacterium glutamicum on ethanol显示文摘Corynebacterium glutamicum represents an emerging recombinant protein expression factory due to its ideal features for protein secretion,but its applicability is harmed by the lack of an autoinduction system with tight regulation and high yield.Here,we propose a new recombinant protein manufacturing platform that leverages ethanol as both a delayed carbon source and an inducer.First,we reanalysed the native inducible promoter PICL from the acetate uptake operon and found that its limited capacity is the result of the inadequate translation initial architecture.The two strategies of bicistronic design and ribozyme-based insulator can ensure the high activity of this promoter.Next,through transcriptional engineering that alters transcription factor binding sites(TFBSs)and the first transcribed sequence,the truncated promoter PA256 with a dramatically higher transcription level was generated.When producing the superfolder green fluorescent protein(sfGFP)under 1%ethanol conditions,PA256 exhibited substantially lower protein accumulation in prophase but an approximately 2.5-fold greater final yield than the strong promoter PH36.This superior expression mode was further validated using two secreted proteins,camelid antibody fragment(VHH)and endoxylanase(XynA).Furthermore,utilizing CRISPRi technology,ethanol utilization blocking strains were created,and PA256 was shown to be impaired in the phosphotransacetylase(PTA)knockdown strains,indicating that ethanol metabolism into the tricarboxylic acid cycle is required for PA256 upregulation.Finally,this platform was applied to produce the“de novo design”protein NEO-2/15,and by introducing the N-propeptide of CspB,NEO-2/15 was effectively secreted with the accumulation 281 mg/L obtained after 24 h of shake-flask fermentation.To the best of our knowledge,this is the first report of NEO-2/15 secretory overexpression. | Xinyu Yu Xiuxia Liu Xiong Gao Xunxun Luo Yankun Yang Ye Li Chunli Liu Chong Zhang Zhonghu Bai | 2022 | Synthetic and Systems Biotechnology2022,7,2: | 1 |

| 19 | NICKEL(Ⅱ)ion-intercalated MXene membranes for enhanced H_(2)/CO_(2) separation显示文摘Hydrogen fuel has been embraced as a potential long-term solution to the growing demand for clean energy.A membrane-assisted separation is promising in producing high-purity H_(2).Molecular sieving membranes(MSMs)are endowed with high gas selectivity and permeability because their well-defined micropores can facilitate molecular exclusion,diffusion,and adsorption.In this work,MXene nanosheets intercalated with Ni^(2+) were assembled to form an MSM supported on Al_(2)O_(3) hollow fiber via a vacuum-assisted filtration and drying process.The prepared membranes showed excellent H_(2)/CO_(2) mixture separation performance at room temperature.Separation factor reached 615 with a hydrogen permeance of 8.35×10^(-8) mol·m^(-2)·s^(-1) ·Pa^(-1).Compared with the original Ti_(3)C_(2)T_(x)/Al_(2)O_(3) hollow fiber membranes,the permeation of hydrogen through the Ni^(2+)-Ti_(3)C_(2)T_(x)/Al_(2)O_(3) membrane was considerably increased,stemming from the strong interaction between the negatively charged MXene nanosheets and Ni^(2+).The interlayer spacing of MSMs was tuned by Ni^(2+).During 200-hour testing,the resultant membrane maintained an excellent gas separation without any substantial performance decline.Our results indicate that the Ni2+tailored Ti_(3)C_(2)T_(x)/Al_(2)O_(3) hollow fiber membranes can inspire promising industrial applications. | Yiyi Fan Jinyong Li Saidi Wang Xiuxia Meng Yun Jin Naitao Yang Bo Meng Jiaquan Li Shaomin Liu | 2021 | Frontiers of Chemical Science and Engineering2021,15,4: | 1 |
