| 1 | Defined tumor antigen-specific T cells potentiate personalized TCR-T cell therapy and prediction of immunotherapy response显示文摘Personalized immunotherapy targeting tumor-specific antigens(TSAs)could generate efficient and safe antitumor immune response without damaging normal tissues.Although neoantigen vaccines have shown therapeutic effect in clinic trials,precise prediction of neoantigens from tumor mutations is still challenging.The host antitumor immune response selects and activates T cells recognizing tumor antigens.Hence,T cells engineered with T-cell receptors(TCRs)from these naturally occurring tumor antigen-specific T(Tas)cells in a patient will target personal TSAs in his/her tumor.To establish such a personalized TCR-T cell therapy,we comprehensively characterized T cells in tumor and its adjacent tissues by single-cell mRNA sequencing(scRNA-seq),TCR sequencing(TCR-seq)and in vitro neoantigen stimulation.Compared to bystander T cells circulating among tissues,Tas cells were characterized by tumor enrichment,tumor-specific clonal expansion and neoantigen specificity.We found that CXCL13 is a unique marker for both CD4^(+)and CD8^(+)Tas cells.Importantly,TCR-T cells expressing TCRs from Tas cells showed significant therapeutic effects on autologous patient-derived xenograft(PDX)tumors.Intratumoral Tas cell levels measured by CXCL13 expression precisely predicted the response to immune checkpoint blockade,indicating a critical role of Tas cells in the antitumor immunity.We further identified CD200 and ENTPD1 as surface markers for CD4^(^(+))and CD8^(^(+))Tas cells respectively,which enabled the isolation of Tas cells from tumor by Fluorescence Activating Cell Sorter(FACS)sorting.Overall,our results suggest that TCR-T cells engineered with Tas TCRs are a promising agent for personalized immunotherapy,and intratumoral Tas cell levels determine the response to immunotherapy. | Jingjing He Xinxin Xiong Han Yang Dandan Li Xuefei Liu Shuo Li Shuangye Liao Siyu Chen Xizhi Wen Kuai Yu Lingyi Fu Xingjun Dong Kaiyu Zhu Xiaojun Xia Tiebang Kang Chaochao Bian Xiang Li Haiping Liu Peirong Ding Xiaoshi Zhang Zhenjiang Liu Wende Li Zhixiang Zuo Penghui Zhou | 2022 | Cell Research2022,32,6: | 10 |
| 2 | A Lamb wave quantification model for inclined cracks with experimental validation显示文摘This paper investigates the influence of crack orientation on damage quantification using Lamb wave in plate structures. Finite element simulation is performed to acquire Lamb wave signal responses for different configurations of crack orientations and crack lengths. Two Lamb wave features, namely the normalized amplitude and the phase change, are used as damage sensitive features to develop a crack size quantification model. A hypothesis based on the geometrical influence on signal features is proposed, and the crack size quantification model incorporating the orientation angle is established using the hypothesis. An index of Probability of Reliable Quantification(PRQ) is proposed to evaluate the performance of the model. The index can be used to determine the sizing risk in terms of probabilities. A realistic aluminum plate is used to obtain the experimental data using piezoelectric(PZT) wafer-type sensors around a center through crack. The experimental data are used to validate the overall method. | 3 | Electronic-property dependent interactions between tetracycline and graphene nanomaterials in aqueous solution显示文摘Understanding the interactions between graphene nanomaterials(GNMs) and antibiotics in aqueous solution is critical to both the engineering applications of GNMs and the assessment of their potential impact on the fate and transport of antibiotics in the aquatic environment. In this study, adsorption of one common antibiotic, tetracycline, by graphene oxide(GO) and reduced graphene oxide(RGO) was examined with multi-walled carbon nanotubes(MWCNTs) and graphite as comparison. The results showed that the tetracycline adsorption capacity by the four selected carbonaceous materials on the unit mass basis followed an order of GO > RGO > MWCNTs > graphite. Upon normalization by surface area,graphite, RGO and MWCNTs had almost the same high tetracycline adsorption affinity while GO exhibited the lowest. We proposed π-electron-property dependent interaction mechanisms to explain the observed different adsorption behaviors. Density functional theory(DFT) calculations suggested that the oxygen-containing functional groups on GO surface reduced its π-electron-donating ability, and thus decreased the π-based interactions between tetracycline and GO surface. Comparison of adsorption efficiency at different p H indicated that electrostatic interaction also played an important role in tetracycline-GO interactions. | 5 | Corrosion Behavior of Anodic Oxidized TiO_2 Film in Seawater显示文摘TiO2 films were formed on metallic titanium substrates by the anodic oxidation method in H2SO4 solution under the 80V D.C..Phase component and microstructure were characterized by X-ray diffraction (XRD) and scanning electron microscopy (SEM).Water contact angles on titanium oxide film surface were measured under both dark and sunlight illumination conditions.Corrosion tests were carried out in seawater under different illumination conditions by electrochemistry impedance spectrum (EIS) and polarization curves.The result showed that the TiO2 film prepared by the anodic oxidation method was anatase with a uniform structure and without obvious pores or cracks on its surface.The average water contact angle of the film was 116.4? in dark, in contrast to an angle of 42.7? under the UV illumination for 2 hours, which demonstrates good hydrophobic property.The anti-corrosion behavior of the TiO2 film was declining with the extended immersion time.Under dark conditions, however, the hydrophobic TiO2 film retarded the water infiltrating into the substrate.The impedance changed slowly and the corrosion current density was 2 orders of magnitude lower than that with the film illuminated by sunlight.All of those mentioned above indicate that the TiO2 film possesses much better performance under dark condition, and it can be applied as an engineering material under dark seawater environment. | WANG Min WANG Wei HE Benlin SUN Mingliang YIN Yansheng LIU Lan ZOU Wuyuan XU Xuefei | 2010 | Journal of Ocean University of China2010,9,4: | 2 |

| 15 | Carbon fluxes and soil carbon dynamics along a gradient of biogeomorphic succession in alpine wetlands of Tibetan Plateau显示文摘Hydrological changes under climate warming drive the biogeomorphic succession of wetlands and may trigger substantial carbon loss from the carbon-rich ecosystems.Although many studies have explored the responses of wetland carbon emissions to short-term hydrological change,it remains poorly understood how the carbon cycle evolves with hydrology-driven wetland succession.Here,we used a space-for-time approach across hydrological gradients on the Tibetan Plateau to examine the dynamics of ecosystem carbon fluxes(carbon dioxide(CO_(2))and methane(CH4))and soil organic carbon pools during alpine wetland succession.We found that the succession from mesic meadow to fen changed the seasonality of both CO_(2) and CH4 fluxes,which was related to the shift in plant community composition,enhanced regulation of soil hydrology and increasing contribution of spring-thaw emission.The paludification caused a switch from net uptake of gaseous carbon to net release on an annual timescale but produced a large accumulation of soil organic carbon.We attempted to attribute the paradox between evidence from the carbon fluxes and pools to the lateral carbon input and the systematic changes of historical climate,given that the wetlands are spatially low-lying with strong temporal climate-carbon cycle interactions.These findings demonstrate a systematic change in the carbon cycle with succession and suggest that biogeomorphic succession and lateral carbon flows are both important for understanding the long-term dynamics of wetland carbon footprints. | Hao Wang Lingfei Yu Litong Chen Zhenhua Zhang Xuefei Li Naishen Liang Changhui Peng Jin-Sheng He | 2023 | Fundamental Research2023,3,2: | 1 |
