Erianin,a novel dibenzyl compound in Dendrobium extract, inhibits lung cancer cell growth and migration via calcium/calmodulin-dependent ferroptosis显示文摘Ferroptosis,a novel form of programmed cell death,is characterized by iron-dependent lipid peroxidation and has been shown to be involved in multiple diseases, including cancer.Stimulating ferroptosis in cancer cells may be a potential strategy for cancer therapy.Therefore,ferroptosis-inducing drugs are attracting more attention for cancer treatment. Here,we showed that erianin,a natural product isolated from Dendrobium chrysotoxum Lindl, exerted its anticancer activity by inducing cell death and inhibiting cell migration in lung cancer cells.Subsequently,we demonstrated for the first time that erianin induced ferroptotic cell death in lung cancer cells, which was accompanied by ROS accumulation, lipid peroxidation, and GSH depletion. The ferroptosis inhibitors Fer-1 and Lip-1 but not Z-VAD-FMK,CQ,or necrostatin-1 rescued erianin-induced cell death, indicating that ferroptosis contributed to erianin-induced cell death.Furthermore,we demonstrated that Ca^(2+)/CaM signaling was a critical mediator of erianin-induced ferroptosis and that blockade of this signaling significantly rescued cell death induced by erianin treatment by suppressing ferroptosis.Taken together, our data suggest that the natural product erianin exerts its anticancer effects by inducing Ca^(2+)/CaMdependent ferroptosis and inhibiting cell migration, and erianin will hopefully serve as a prospective compound for lung cancer treatment.Peng Chen Qibiao Wu Jiao Feng Lili Yan Yitian Sun Shuiping Liu Yu Xiang Mingming Zhang Ting Pan Xiaying Chen Ting Duan Lijuan Zhai Bingtao Zhai Wengang Wang Ruonan Zhang Bi Chen Xuemeng Han Yicong Li Liuxi Chen Ying Liu Xingxing Huang Ting Jin Wenzheng Zhang Hong Luo Xiaohui Chen Yongqiang Li Qiujie Li Guohua Li Qin Zhang Lvjia Zhuo Zuyi Yang Huifen Tang Tian Xie Xiaoping Ouyang Xinbing Sui 2020Signal Transduction and Targeted Therapy2020,5,1:42

Bushen Qiangjin capsule(补肾强筋胶囊) inhibits the Wnt/β-catenin pathway to ameliorate papain-induced knee osteoarthritis in rats显示文摘OBJECTIVE:To evaluate the molecular mechanism underlying the beneficial effect of Bushen Qiangjin capsule(补肾强筋胶囊,BSQJ),a Traditional Chinese Medicine,on knee osteoarthritis(KOA).METHODS:In the present study,32 female Sprague-Dawley rats were randomly divided into four groups:control,KOA,high-dose BSQJ(H-BSQJ),and low-dose BSQJ(L-BSQJ).After successfully establishing the KOA model by intra-articular injection of papain,H-BSQJ and L-BSQJ groups were intragastrically administered 0.243 and 0.122 g/kg BSQJ,respectively, daily for 6 weeks. At the end of the experiment, knee articular cartilage tissues of rats were collected for evaluation by hematoxylin and eosin staining,Safranin O-Fast Green staining, and terminal deoxynucleotidyl transferase-mediated d UTP nick-end labeling assay.Serum interleukin-1βand tumor necrosis factor-αlevels of rats were detected with an enzyme-linked immunosorbent assay method.Gene expression of Wnt-4,β-catenin,Frizzled-2,glycogen synthase kinase-3β(GSK-3β),cysteinyl aspartate-specific proteinases 3 and 9(caspases 3 and 9),collagen typeⅡalpha 1(Col2 a1),and matrix metalloproteinases 1 and 13(MMP-1 and MMP-3)of rat knee articular cartilage was quantified by reverse transcription-quantitative polymerase chain reaction analysis.Wnt-4,β-catenin,Frizzled-2,GSK-3β,cleaved caspase-3,and cleaved caspase-9 protein expression in rat knee articular cartilage was determined by western blot analysis.RESULTS:BSQJ obviously reduced pathological damage and matrix degradation of articular cartilage in KOA rats.Compared with the KOA group,H-BSQJ rats exhibited downregulated m RNA and protein expression of Wnt-4,β-catenin,Frizzled-2,and caspase-3,as well as upregulated m RNA and protein expression of GSK-3β.In addition,H-BSQJ significantly increased m RNA expression of Col2 a1 and decreased m RNA expression of MMP-1 and MMP-13.CONCLUSION:BSQJ exerted a beneficial effect on KOA by a mechanism involving downregulation of the Wnt/β-catenin pathway, which inhibited both cartilage extracellular matrix degradation and chondrocyte apoptosis to ameliorate KOA in rats.YAO Nan CHEN Guocai LU Yanyan XU Xuemeng ZHAO Chuanxi HUANG Xuejun LIU Wengang PENG Sha WU Huai 2021Journal of Traditional Chinese Medicine2021,41,6:5
Deformation of the Most Recent Co-seismic Surface Ruptures Along the Garzê–Yushu Fault Zone(Dangjiang Segment)and Tectonic Implications For the Tibetan Plateau显示文摘The Garzê–Yushu strike-slip fault in central Tibet is the locus of strong earthquakes(M > 7). The deformation and geometry of the co-seismic surface ruptures are reflected in the surface morphology of the fault and depend on the structure of the upper crust as well as the pre-existing tectonics. Therefore, the most recent co-seismic surface ruptures along the Garzê–Yushu fault zone(Dangjiang segment) reveal the surface deformation of the central Tibetan Plateau. Remote sensing images and field investigations suggest a 85 km long surface rupture zone(striking NW-NWW), less than 50 m wide, defined by discontinuous fault scarps, right-stepping en echelon tensional cracks and left-stepping mole tracks that point to a left-lateral strike-slip fault. The gullies that cross fault scarps record systematic left-lateral offsets of 1.8 m to 5.0 m owing to the most recent earthquake, with moment magnitude of about M 7.5, in the Dangjiang segment. Geological and geomorphological features suggest that the spatial distribution of the 1738 co-seismic surface rupture zone was controlled by the pre-existing active Garzê–Yushu fault zone(Dangjiang segment). We confirm that the Garzê–Yushu fault zone, a boundary between the Bayan Har Block to the north and the Qiangtang Block to the south, accommodates the eastward extrusion of the Tibetan Plateau and generates strong earthquakes that release the strain energy owing to the relative motion between the Bayan Har and Qiangtang Blocks.WU Jiwen HUANG Xuemeng XIE Furen 2017Acta Geologica Sinica(English Edition)2017,91,2:3
Giant nonlinear Hall effect in twisted bilayer WSe_(2)显示文摘The recently discovered nonlinear Hall effect(NHE)in a few non-interacting systems provides a novel mechanism for generating second-harmonic electrical Hall signals under time-reversal-symmetric conditions. Here,we introduce a new approach to engineering an NHE by using twisted moiréstructures. We found that the twisted WSe_(2)bilayer exhibited an NHE when the Fermi level was tuned to the moiréflat bands. When the first moiréband was half-filled,the nonlinear Hall signal exhibited a sharp peak with a generation efficiency that was at least two orders of magnitude greater than those obtained in previous experiments. We discuss the possible origins of the diverging generation efficiency in twisted WSe_(2)based on resistivity measurements, such as moiré-interface-induced correlation effects and mass-diverging-type continuous Mot transition. This study demonstrates not only how interaction effects can combine with Berry curvature dipoles to produce novel quantum phenomena, but also the potential of NHE measurements as a new tool for studying quantum criticality.Meizhen Huang Zefei Wu Jinxin Hu Xiangbin Cai En Li Liheng An Xuemeng Feng Ziqing Ye Nian Lin Kam Tuen Law Ning Wang 2023National Science Review2023,10,4:0
Oxidized organic molecules in the tropical free troposphere over Amazonia显示文摘New particle formation(NPF) in the tropical free troposphere(FT) is a globally important source of cloud condensation nuclei, affecting cloud properties and climate. Oxidized organic molecules(OOMs)produced from biogenic volatile organic compounds are believed to contribute to aerosol formation in the tropical FT, but without direct chemical observations. We performed in situ molecular-level OOMs measurements at the Bolivian station Chacaltaya at 5240 m above sea level, on the western edge of Amazonia. For the first time, we demonstrate the presence of OOMs, mainly with 4–5 carbon atoms, in both gas-phase and particle-phase(in terms of mass contribution) measurements in tropical FT air from Amazonia. These observations, combined with air mass history analyses, indicate that the observed OOMs are linked to isoprene emitted from the rainforests hundreds of kilometers away. Based on particle-phase measurements, we find that these compounds can contribute to NPF, at least the growth of newly formed nanoparticles, in the tropical FT on a continental scale. Thus, our study is a fundamental and significant step in understanding the aerosol formation process in the tropical FT.Qiaozhi Zha Diego Aliaga Radovan Krejci Victoria ASinclair Cheng Wu Giancarlo Ciarelli Wiebke Scholz Liine Heikkinen Eva Partoll Yvette Gramlich Wei Huang Markus Leiminger Joonas Enroth Otso Perakyla Runlong Cai Xuemeng Chen Alkuin Maximilian Koenig Fernando Velarde Isabel Moreno Tuukka Petaja Paulo Artaxo Paolo Laj Armin Hansel Samara Carbone Markku Kulmala Marcos Andrade Douglas Worsnop Claudia Mohr Federico Bianchi 2024National Science Review2024,11,1:0
Target-responsive DNAzyme hydrogel for portable colorimetric detection of lanthanide(Ⅲ)ions显示文摘Lanthanide elements(Ln)play an important role in industry and agriculture. As a result of the increasing consumption of lanthanides, environmental emission of Ln has become detrimental to the health of flora and fauna.Current methods for trace lanthanides detection mainly rely on sophisticated instruments. In this article,a Ln^(3+)dependent DNAzyme was incorporated into a hydrogel to generate Ln^(3+)sensitive DNAzyme hydrogel for portable colorimetric detection. The enzyme strand and its substrate strand act as crosslinker and functional unit of the hydrogel with polyacrylamide chains as the scaffold and gold nanoparticles(AuNPs)as the indicator of hydrogel stability.Any ions in the Ln^(3+)series can trigger the cleavage of substrate strand by activating the enzyme strand, thereby decreasing the crosslink ratio and leading to collapse of the hydrogel. The release of the encapsulated AuNPs turns the supernatant wine red.Using this colorimetric method,Ln^(3+)can be detected with high sensitivity, with a limit of detection(LOD)of 20 nM for Ce^(3+).The hydrogel responds specifically to any Ln^(3+)ion and works well with the spiked lake sample without the need of instruments and skilled operators. Our results suggest that the lanthanide responsive hydrogel can be used for portable and sensitive detection of Ln^(3+)contamination in the field.Yishun Huang Xuemeng Wu Tian Tian Zhi Zhu Hui Lin Chaoyong Yang 2017Science China Chemistry2017,60,2:0
Zearalenone induces apoptosis and autophagy by regulating endoplasmic reticulum stress signalling in porcine trophectoderm cells显示文摘Zearalenone(ZEA),a mycotoxin produced mainly by fungi belonging to Fusarium species in foods and feeds, causes a serious hazard to humans and animals.Numerous studies have revealed that ingesting ZEA can disrupt the reproductive function and impair the reproductive process in animals. This experiment was to investigate the toxicological effect and the mechanism of ZEA exposure on reproduction in pigs during early stages of pregnancy. In the present study,we treated with 0 to 80μmol/L ZEA for 12 or 24 h in trophoblast ectoderm(pTr)cells. The results showed that ZEA had significantly decreased cell proliferation(P<0.05),which was accompanied by DNA damage-related cell cycle arrest at G2/M phase, activation of the apoptosis and endoplasmic reticulum(ER)stress,as well as impairment of barrier function(P<0.05).Western blot analysis and transmission electron microscopy(TEM)showed that exposure to ZEA can activation of autophagy in pTr cells.Importantly, pretreatment with chloroquine(CQ)or 3-methyladenine(3-MA)led to increased apoptosis in pTr cells.Interestingly,pTr cells pretreated with 4-phenylbutyric acid(4-PBA),an inhibitor of ER stress, resulted in reduced cell death in pTr cells, indicating a critical role for ER stress in the activation of autophagy. In conclusion, these results reveal that ZEA-triggered ER stress is critical for the cell fate decision of pTr cells during early porcine embryonic development.Application of small molecules with ability of blocking ER stress might be therapeutic option to reduce the deleterious effect of ZEA in pregnant animals.Jun Bai Jun Li Ning Liu Hai Jia Xuemeng Si Yusong Zhou Zhian Zhai Ying Yang Fazheng Ren Zhenlong Wu 2023Animal Nutrition2023,,1:0
Selection of specific nanobodies against peanut allergen through unbiased immunization strategy and the developed immuno-assay显示文摘Peanut allergy is considered to be a major health issue with global effects. To date,no effective curative approach has been applied for the therapy of the anaphylaxis resulting from the peanut allergens. The accurate and effective detection methods for the surveillance of allergens in food are still the primary strategies to avoid allergic diseases. In this study, nanobodies(Nbs)derived from the Heavy-Chain only Antibodies(HCAbs)were selected against the general peanut protein extract through the unbiased strategy to facilitate the development of the sandwich ELISA for the detection and surveillance of peanut allergen contamination. The target antigen of the selected Nb was identified as peanut allergen Ara h 3,and a cross-reaction was observed with the member of Gly 1 from the Ara h 3 family. The applicability of the self-paired Nb P43 on the establishment of the immuno-assay was verified.A sandwich ELISA against peanut allergen was developed, which reached a linear range of 0.2-10.6μg/mL,and a limit of detection of 53.13 ng/mL.Yaozhong Hu Chuan Zhang Jing Lin Yi Wang Sihao Wu Ying Sun Bowei Zhang Huan Lv Xuemeng Ji Yang Lu Shuo Wang 2023Food Science and Human Wellness2023,12,3:0
Cellularmetabolism: A key player in cancer ferroptosis显示文摘Cellular metabolism is the fundamental process by which cells maintain growth and self-renewal.It produces energy, furnishes raw materials, and intermedi-ates for biomolecule synthesis, and modulates enzyme activity to sustain normal cellular functions.Cellular metabolism is the foundation of cellular life processes and plays a regulatory role in various biological functions, including pro-grammed cell death.Ferroptosis is a recently discovered form of iron-dependent programmed cell death. The inhibition of ferroptosis plays a crucial role in tumorigenesis and tumor progression. However, the role of cellular metabolism, particularly glucose and amino acid metabolism,in cancer ferroptosis is not well understood. Here,we reviewed glucose, lipid, amino acid, iron and sele-nium metabolism involvement in cancer cell ferroptosis to elucidate the impact of different metabolic pathways on this process.Additionally,we provided a detailed overview of agents used to induce cancer ferroptosis. We explained that the metabolism of tumor cells plays a crucial role in maintaining intracellu-lar redox homeostasis and that disrupting the normal metabolic processes in these cells renders them more susceptible to iron-induced cell death, resulting in enhanced tumor cell killing. The combination of ferroptosis inducers and cel-lular metabolism inhibitors may be a novel approach to future cancer therapy and an important strategy to advance the development of treatments.Xianjie Jiang Qiu Peng Mingjing Peng Linda Oyang Honghan Wang Qiang Liu Xuemeng Xu Nayiyuan Wu Shiming Tan Wenjuan Yang Yaqian Han Jinguan Lin Longzheng Xia Yanyan Tang Xia Luo Jie Dai Yujuan Zhou Qianjin Liao 2024Cancer Communications2024,44,2:0
