| 2 | Adipocyte-derived SFRP5 inhibits breast cancer cells migration and invasion through Wnt and epithelial-mesenchymal transition signaling pathways显示文摘Objective:Obesity is closely associated with metastasis in breast cancer patients.Secreted frizzled-related protein 5(SFRP5),one of the novel adipokines with anti-inflammatory properties,is associated with obesity.This study aims to study the role of SFRP5 in the crosstalk between obesity and breast cancer metastasis and identify the underlying mechanism.Methods:3T3-L1 pre-adipocytes were differentiated to mature adipocytes and a hypertrophic adipocyte model was induced with palmitic acid(PA).Cell motility was measured in MDA-MB-231 and MCF-7 breast cancer cells co-cultured with adipocytes conditioned medium(CM)with or without SFRP5 protein.Wnt and epithelialmesenchymal transition(EMT)signal pathways were investigated by western blot.Circulating SFRP5 level in 218 breast cancer patients and the association with clinicopathologic characteristics of breast cancer were further determined.Online databases ENCORI and PREDICT Plus were used to exam the link between SFRP5 and prognosis.Results:Reduced SFRP5 level was detected in the hypertrophic adipocyte model.Recombinant SFRP5 protein inhibited MDA-MB-231 and MCF-7 cells invasion and migration induced by PA-treated adipocyte CM,and SFRP5 inhibition by specific antibody reversed the effect of SFRP5.Furthermore,SFRP5 significantly inhibited Wnt and downstream EMT in breast cancer cells.Low circulating SFRP5 level correlated with body mass index(BMI),lymph node(LN)metastasis,TNM stage and high Ki67 expression in breast cancer patients.Increased SFRP5 level was associated with favorable predicted survival.Kaplan-Meier curves showed high SFRP5 level in tumor tissue was associated with better outcome of breast cancer patients.Conclusions:Our findings demonstrated SFRP5 is a vital adipokine that mediates the crosslink between obesity and the metastatic potential of breast cancer.Promotion of SFRP5 expression in the adipose microenvironment may represent a novel approach for preventing breast cancer metastasis. | Wenzhong Zhou Chunmiao Ye Liang Li Liyuan Liu Fei Wang Lixiang Yu Fei Zhou Yujuan Xiang Yongjiu Wang Gengshen Yin Zhongbing Ma Qinye Fu Qiang Zhang Dezong Gao Shuya Huang Zhigang Yu | 2020 | Chinese Journal of Cancer Research2020,32,3: | 5 |
| 3 | Genome sequencing and phylogenetic analysis of allotetraploid Salix matsudana Koidz显示文摘Polyploidy is a common phenomenon among willow species.In this study,genome sequencing was conducted for Salix matsudana Koidz(also named Chinese willow),an important greening and arbor tree species,and the genome of this species was compared with those of four other tree species in Salicaceae.The total genome sequence of S.matsudana was 655.72 Mb in size,with repeated sequences accounting for 45.97%of the total length.In total,531.43 Mb of the genome sequence could be mapped onto 38 chromosomes using the published genetic map as a reference.The genome of S.matsudana could be divided into two groups,the A and B genomes,through homology analysis with the genome of Populus trichocarpa,and the A and B genomes contained 23,985 and 25,107 genes,respectively.4DTv combined transposon analysis predicted that allotetraploidy in S.matsudana appeared~4 million years ago.The results from this study will help reveal the evolutionary history of S.matsudana and lay a genetic basis for its breeding. | Jian Zhang Huwei Yuan Yujuan Li Yanhong Chen Guoyuan Liu Meixia Ye Chunmei Yu Bolin Lian Fei Zhong Yuna Jiang Jichen Xu | 2020 | Horticulture Research2020,7,1: | 3 |

| 5 | Efficacy of needling Baihui(GV20),Neiguan(PC6),Shenmen(HT7) and Taichong(LR3) on cerebral cortical blood oxygen level in rats with insomnia显示文摘OBJECTIVE:To evaluate the effect of needling Baihui(GV20),Neiguan(PC6),Shenmen(HT7) and Taichong(LR3) on cerebral cortical blood oxygen level in rats with insomnia identified as liver-Qi stagnation pattern in terms of Traditional Chinese Medicine.METHODS:Sixty Wistar rats were randomly divided into blank group(n = 10),the rest of the rats were clamped tail as stimulation combined with injected intraperitoneally p-chlorophenylalanine(PCPA) to establish sleep deprivation model.After the successful replication of the model,they were randomly divided into model group,grabbing group,Western Medicine group,acupuncture group and sham acupuncture group,with 10 rats in each group.The model group was given normal saline;The grasping group was grasped in the same way as the two treatment groups;Western medicine group was given estazolam solution;The acupuncture group was treated with the acupuncture method of 'soothing the liver Qi and regulating mind' needling Baihui(GV20),Neiguan(PC6),Shenmen(HT7) and Taichong(LR3);The sham acupuncture group were needled with four nonacupoint sites.After 7 d of treatment of rats in each group,sodium pentobarbital sleep experiment was used to evaluate the sleep latency(SL) and sleep time(ST).The percentage of rats entering the open arm(OE%) and the percentage of time entering the open arm(OT%) in each group were monitored by elevated cross maze,and the vertical score,horizontal crossing times,central grid score and modification times were monitored by open field test,functional near infrared spectroscopy(f NIRs) was used to monitor the changes of oxygenated hemoglobin(Oxy-Hb),deoxyhemoglobin(Deoxy-Hb) and total hemoglobin(Total-Hb) in the cerebral cortex of rats in each group under light stimulation and dark stimulation,the statistically significant channel combinations were selected from 8 light sources and 12 detectors(S-D).According to the position of the light source detector covering the cerebral cortex,Preliminarily determine the key brain areas of insomnia(the preliminary experiment found that 6S-8D and 7S-9D were the key channels of insomnia under light stimulation,and the corresponding brain areas were prefrontal lobe and occipital lobe respectively;7S-7D channel under dark stimulation,and the corresponding brain area was occipital lobe);The hemodynamic map of cerebral cortex was constructed through the absolute value of whole brain blood oxygen level.Further identify key brain regions for insomnia.RESULTS:Compared with the blank group,ST,OE%,OT%,the vertical score,horizontal crossing times,central grid score,the concentration of Deoxy-Hb in prefrontal and occipital lobes were decreased significantly(P < 0.01),and the concentrations of SL,modification times,Oxy-Hb and Total-Hb were increased significantly(P < 0.01);There was no difference in the above indexes between the model group and the grabbing group(P > 0.05);After treatment,ST,OE%,OT%,the vertical score,horizontal crossing times,central grid score and DeoxyHb concentration in acupuncture group and Western medicine group increased significantly,while SL,modification times,Oxy-Hb and Total-Hb concentrations were decreased significantly(P < 0.01,P < 0.05),Compared with the Western Medicine group,the values of OE% and OT% were more significantly higher in the acupuncture group(P < 0.05),while the rest of the indexes were not significantly different between the two groups(P > 0.05);Compared with the acupuncture group,ST,OE%,OT%,the vertical score,horizontal crossing times,central grid score and the concentration of deoxy Hb in the sham acupuncture group decreased significantly(P < 0.01),and the concentrations of SL,modification times,Oxy-Hb and Total-Hb increased significantly(P < 0.01).CONCLUSIONS:The needling method for soothing the liver Qi and regulating mind could improve the abnormal behavior of insomnia rats with liver Qi stagnation,and its effect on the improvement of abnormal mood caused by insomnia with liver Qi stagnation is better than that of Western Medicine,and the mechanism of action may be related to the regulation of blood oxygen metabolism in the prefrontal and occipital lobes of the cerebral cortex by acupuncture. | YE Yujuan WEI Yuting JIA Jing YAN Xingke | 2023 | Journal of Traditional Chinese Medicine2023,43,3: | 1 |

| 8 | Real-world effectiveness and safety of OBT/PTV/r and dasabuvir for patients with chronic HCV genotype 1b infection in China:A multicenter prospective observational study显示文摘Background and aim:Real-world data on the effectiveness and safety of treatment with the direct-acting antiviral agent-based regimen are limited on the Chinese mainland.The aim of this study was to conduct a multicenter,prospective,real-world study of ombitasvir/paritaprevir/ritonavir(OBT/PTV/r)combined with dasabuvir(DSV)in hepatitis C virus(HCV)genotype 1b-infected non-cirrhotic or compensated cirrhotic Chinese adult patients.Materials and methods:Genotype 1b-infected patients were enrolled at eight sites in China.Patients received 25/150/100 mg of OBT/PTV/r once daily combined with 250 mg of DSV twice daily for 8 weeks or 12 weeks.Sustained virological response at 12 weeks post-treatment(SVR12)and the incidence of adverse events were assessed.We have also evaluated the effect of intensive questioning of patients who were overdue for SVR12 testing.Intention-to-treat(ITT)and modified ITT(mITT)populations were used in the analysis.Results:One hundred forty patients were included,among whom 90.0%(126/140)were newly diagnosed,9.3%(13/140)had compensated cirrhosis,92.9%(130/140)received 12 weeks of treatment,and 7.1%(10/140)received 8 weeks of treatment.In the mITT population,the virological response rate at week 4 was 96.4%(108/112),and at the end of treatment was 100%(102/102).Among these patients,139 patients completed 12 weeks of treatment,and 73 patients were followed-up.All followed-up patients achieved SVR12.There was no adverse event-related discontinuation.Serious adverse events during treatment were reported in two(1.4%)patients,and none were considered to be drug-related.Sixty-six(47.1%)patients did not return to receive the HCV RNA test at 12 weeks post-treatment.Conclusions:The rate of SVR12 was consistent with Phase III clinical studies.OBT/PTV/r combined with DSV showed effectiveness in Chinese adult patients,and both tolerability and safety profile were favorable.However,patient compliance should be further improved in the real world. | Yusheng Jie Chaoshuang Lin Jing Yuan Zhixin Zhao Yujuan Guan Yuanping Zhou Xiaohui Zhou Bihui Zhong Yinong Ye Lihua Zhang Ling Tao Jianping Li Xiaohong Zhang Yutian Chong | 2020 | Liver Research2020,4,3: | 0 |
