1A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. 2GRAIN LENGTH AND AWN 1 negatively regulates grain size in rice显示文摘Grain size is an important factor determining yield in rice. Here, we identified a recessive mutant gene, grain length and awn 1 (gla1), which caused a significant increase in grain length and weight, and was associated with long awns. The gla1 mutation was mapped to a single-nucleotide polymorphism in a gene encoding a cytoplasmically-localized mitogen-activated protein kinase (MAPK) phosphatase. Overexpression of GLA1 caused a decrease in grain length, and the GLA1 protein interacted with OsMAPK6. 3Less and shrunken pollen 1(LSP1) encodes a member of the ABC transporter family required for pollen wall development in rice(Oryza sativa L.)显示文摘Pollen fertility is an agronomic trait that strongly influences rice yield. Recent studies have revealed that the development of the pollen wall is required for pollen fertility and is regulated by several genes. However, the mechanisms underlying pollen and pollen wall development in rice remain largely unknown. In the present study, a point mutation in a gene on chromosome 1 was identified that resulted in the production of less and shrunken pollen(LSP) and led to defects in pollen wall formation. This gene was named LSP1 and was found to encode a member of the adenosine triphosphate-binding cassette(ABC)transporter G subfamily, OsABCG3. Two other loss-of-function mutants of LSP1/OsABCG3,generated using CRISPR/Cas9 technology, showed the same male sterile phenotype. The LSP1/OsABCG3 gene showed a spatio-temporal expression pattern in the developing anthers, and is an ortholog of the Arabidopsis genes At ABCG1 and At ABCG16, which play an important role in pollen wall development. Mutation of LSP1/OsABCG3 affected the expression of several genes involved in pollen and pollen wall formation. 4Progress of power field effect transistor based on ultra-wide bandgap Ga_2O_3 semiconductor material显示文摘As a promising ultra-wide bandgap semiconductor, gallium oxide(Ga_2O_3) has attracted increasing attention in recent years. The high theoretical breakdown electrical field(8 MV/cm), ultra-wide bandgap(~ 4.8 eV) and large Baliga's figure of merit(BFOM) of Ga_2O_3 make it a potential candidate material for next generation high-power electronics, including diode and field effect transistor(FET). In this paper, we introduce the basic physical properties of Ga_2O_3 single crystal, and review the recent research process of Ga_2O_3 based field effect transistors. Furthermore, various structures of FETs have been summarized and compared, and the potential of Ga_2O_3 is preliminary revealed. 6Non-parallel double-crack propagation in rock-like materials under uniaxial compression显示文摘Coalescence among fractures would have influence on the stability of rock masses. Deep understanding of mechanical behavior of fractured rock masses is an important mean to identify failure mechanism of geological disaster. In this study, crack propagation processing was studied through loading pre-fractured specimens of concrete block, termed as rock-like material, in uniaxial compression tests. New non-parallel double-crack geometry was introduced to observe crack coalescence. The flaw combinations are different from the normally used flaw configurations. In addition, ultrasonic detection tests were performed on the test blocks. The stress and strain data of these tests and characteristic parameters of sound wave were recorded. The stress-strain curves of each test block under the uniaxial compression test were drawn, relations among deformation characteristics and crack angle of the crack specimens, and their overall strength were analyzed. It is found that strength of the specimen decreases as crack inclination increases under two crack inclinations. The highest uniaxial compressive strength is found in the specimen with the cracks at the same angle in different directions. Based on description of the crack initiation location, crack surface and the ultimate failure patterns, failure modes of eight subtype for test blocks are divided into three categories. 7An approach to wide-field imaging of linear rail ground-based SAR in high squint multi-angle mode显示文摘Ground-based synthetic aperture radar(GB-SAR) has been successfully applied to the ground deformation monitoring.However, due to the short length of the GB-SAR platform, the scope of observation is largely limited. The practical applications drive us to make improvements on the conventional linear rail GB-SAR system in order to achieve larger field imaging. First, a turntable is utilized to support the rotational movement of the radar.Next, a series of high-squint scanning is performed with multiple squint angles. Further, the high squint modulation phase of the echo data is eliminated. Then, a new multi-angle imaging method is performed in the wave number domain to expand the field of view. 9Self-Catalyzed Rechargeable Lithium-Air Battery by in situ Metal Ion Doping of Discharge Products: A Combined Theoretical and Experimental Study显示文摘Lithium-air battery has emerged as a viable electrochemical energy technology;yet a substantial overpotential is typically observed,due to the insulating nature of the discharge product Li_(2)O_(2) that hinders the reaction kinetics and device performance.Furthermore,finite solid–solid/-liquid interfaces are formed between Li_(2)O_(2) and catalysts and limit the activity of the electrocatalysts in battery reactions,leading to inadequate electrolytic efficiency.Herein,in-situ doping of Li_(2)O_(2) by select metal ions is found to significantly enhance the lithium-air battery performance,and Co^(2+)stands out as the most effective dopant among the series.This is ascribed to the unique catalytic activity of the resulting Co-O_(x) sites towards oxygen electrocatalysis,rendering the lithium-air battery self-catalytically active.Theoretical studies based on density functional theory calculations show that structural compression occurs upon Co^(2+)doping,which lowers the energy barrier of Li_(2)O_(2) decomposition.Results from this study highlight the significance of in situ electrochemical doping of the discharge product in enhancing the performance of lithium-air battery.Mengwei Yuan Zemin Sun Han Yang Di Wang Qiming Liu Caiyun Nan Huifeng Li Genban Sun Shaowei Chen 2023Energy & Environmental Materials2023,6,1:1

13Effect of Sea Spray on the Numerical Simulation of Super Typhoon ‘Ewiniar’显示文摘To study the potential effect of sea spray on the evolution of typhoons,two kinds of sea spray flux parameterization schemes developed by Andreas (2005) and Andreas and Wang (2006) and Fairall et al. (1994) respectively are incorporated into the regional atmospheric Mesoscale Model version 3.6 (MM5V3) of Pennsylvania State University/National Center for Atmospheric Research (PSU/NCAR) and the coupled atmosphere-sea spray modeling system is applied to simulate a Western Pacific super ty-phoon Ewiniar in 2006. The simulation results demonstrate that sea spray can lead to a significant increase in heat fluxes at the air-sea interface and the simulated typhoon’s intensity. Compared with the results without sea spray,the minimum sea level pressure reduces about 8hPa after taking account of sea spray by Fairall et al.’s parameterization (1994) and about 5hPa by Andreas’ (2005) and Andreas and Wang’s (2006) parameterization at the end of the model integration,while the maximum 10m wind speed increases about 17% and 15% on average,respectively,through the entire simulation time period. Taking sea spray into account also causes significant changes in Tropical Cyclone (TC) structure due to an enhancement of water vapor and heat transferred from the sea sur-face to the air; therefore,the center structure of the typhoon becomes more clearly defined and the wind speed around the typhoon eye is stronger in numerical experiments. 15Oxygen Reduction Reaction Catalyzed by Carbon-Supported Platinum Few-Atom Clusters:Significant Enhancement by Doping of Atomic Cobalt显示文摘Oxygen reduction reaction(ORR)plays an important role in dictating the performance of various electrochemical energy technologies.As platinum nanoparticles have served as the catalysts of choice towards ORR,minimizing the cost of the catalysts by diminishing the platinum nanoparticle size has become a critical route to advancing the technological development.Herein,first-principle calculations show that carbon-supported Pt9 clusters represent the threshold domain size,and the ORR activity can be significantly improved by doping of adjacent cobalt atoms.This is confirmed experimentally,where platinum and cobalt are dispersed in nitrogen-doped carbon nanowires in varied forms,single atoms,few-atom clusters,and nanoparticles,depending on the initial feeds.The sample consisting primarily of Pt_(2~7)clusters doped with atomic Co species exhibits the best mass activity among the series,with a current density of 4:16Amg^(-1)_(Pt)at+0.85V vs.RHE that is almost 50 times higher than that of commercial Pt/C.Bingzhang Lu Qiming Liu Forrest Nichols Rene Mercado David Morris Ning Li Peng Zhang Peng Gao Yuan Ping Shaowei Chen 2020Research2020,,1:1
16Analysis of top Chinese contractors' developmental tracks and evaluation of their strategic selection of international construction markets显示文摘The past decade has witnessed a rapid development of the international construction market. This research aims to investigate top Chinese contractors' developmental tracks and evaluate their strategic selection of international construction markets over the period from 2001 to 2006,based on the modified Boston Consulting Group Matrix (BCG). Results from 2001 to 2005 indicate that Asia and Africa,which were 'question marks' markets for top Chinese contractors (abbreviated as TCCs),have the most potential to become 'stars' markets; other question marks markets such as the Middle East and Europe are found not to be of potential; 'dogs' markets such as the U.S.,Canada and Latin America should be relinquished progressively unless there is renewed growth opportunity over the next two years. Compared to developmental tracks of top contractors from some advanced countries,TCCs' investment and development strategy in Africa seems to be sustainable and intensive; TCCs' exit strategy for the Middle East appears determined and swift,while their strategy in Asia seems to be lacking in persistence. Furthermore,the data of 2006 indicates that TCCs improve their performance in Asia and Africa,and Africa has become a 'stars' market for TCCs.This research sheds important lights and insights into the prevailing process of internationalization of TCCs and provides a strategic direction for their selection of international construction market. 17Hollow sphere of heterojunction(NiCu)S/NC as advanced anode for sodium-ion battery显示文摘Metal sulfide is considered as a potential anode for sodium-ion batteries(SIBs),due to the high theoretical capacity,strong thermodynamic stability and low-cost.However,their cycle capacity and rate performance are limited by the excessive expansion rate and low intrinsic conductivity.Herein,heterogeneous hollow sphere NiS-Cu_(9)S_(5)/NC(labeled as(NiCu)S/NC)based on Oswald ripening mechanism was prepared through a simple and feasible methodology.From a structural perspective,the hollow structure provides an expansion buffer and raises the electrochemical active area.In terms of electron/ion during the cycles,Na^(+)storage mechanism is optimized by NiS/Cu_(9)S_(5)heterogeneous interface,which increases the storage sites and shortens the migration path of Na^(+).The formation of built-in electric field strengthens the electron/ion mobility.Based on the first principle calculations,it is further proved the formation of heterogeneous interfaces and the direction of electron flow.As the anode for SIBs,the synthesized(NiCu)S/NC delivers high reverse capacity(559.2 mA h g^(-1)at 0.5 A g^(-1)),outstanding rate performance(185.3 mA h g^(-1)at 15 A g^(-1)),long-durable stability(342.6 mA h g^(-1)at 4 A g^(-1)after 1500cycles,150.0 m A h g^(-1)at 10 A g^(-1)after 20,000 cycles with 0.0025%average attenuation rate).The matching cathode electrode Na_(3)V_(2)(PO_(4))_(3)/C is assembled with(NiCu)S/NC for the full-battery that achieves high energy density(253.7 W h kg^(-1))and reverse capacity(288.7 mA h g^(-1)).The present work provides a distinctive strategy for constructing electrodes with excellent capacity and stability for SIBs.Hongyi Chen Pengfei Lv Pengfu Tian Shiyue Cao Shengjun Yuan Qiming Liu 2023Journal of Energy Chemistry2023,,7:0
18Blockade of integrin signaling reduces chemotherapy-induced premature senescence in collagen cultured bladder cancer cells显示文摘Background:Diminished sensitivity towards chemotherapy remains the major impediment to the clinical treatment of bladder cancer.However,the critical elements in control of chemotherapy resistance remain obscure.Methods:We adopted improved collagen gels and performed cytotoxicity analysis of doxorubicin(DOX)and mitomycin C(MMC)of bladder cancer cells in a 3D culture system.We then detected the expression of multidrug resistant gene ABCB1,dormancy-associated functional protein chicken ovalbumin upstream-transcription factor 1(COUPTF1),cell proliferation marker Ki-67,and cellular senescence marker senescence-associatedβ-galactosidase(SA-β-Gal)in these cells.We further tested the effects of integrin blockade or protein kinase B(AKT)inhibitor on the senescent state of bladder cancer.Also,we examined the tumor growth and survival time of bladder cancer mouse models given the combination treatment of chemotherapeutic agents and integrinα2β1 ligand peptide TFA(TFA).Results:Collagen gels played a repressive role in bladder cancer cell apoptosis induced by DOX and MMC.In mechanism,collagen activated the integrinβ1/AKT cascade to drive bladder cancer cells into a premature senescence state via the p21/p53 pathway,thus attenuating chemotherapy-induced apoptosis.In addition,TFA had the ability to mediate the switch from senescence to apoptosis of bladder cancer cells in xenograft mice.Meanwhile,TFA combined with chemotherapeutic drugs produced a substantial suppression of tumor growth as well as an extension of survival time in vivo.Conclusions:Based on our finding that integrinβ1/AKT acted primarily to impart premature senescence to bladder cancer cells cultured in collagen gel,we suggest that integrinβ1 might be a feasible target for bladder cancer eradication.Linghui Deng Kun Jin Xianghong Zhou Zilong Zhang Liming Ge Xingyu Xiong Xingyang Su Di Jin Qiming Yuan Chichen Zhang Yifan Li Haochen Zhao Qiang Wei Lu Yang Shi Qiu 2022Precision Clinical Medicine2022,5,2:0

20Genome-wide pedigree analysis of elite rice Shuhui 527 reveals key regions for breeding显示文摘Hybrid rice significantly contributes to the food supply worldwide. Backbone parents play important roles in elite hybrid rice breeding systems. In this study, we performed pedigree-based analysis of the elite backbone parent rice variety, namely, Shuhui 527(SH527, Oryza sativa), to exploit key genome regions during breeding. Twenty-four cultivars(including SH527, its six progenitors and 17 derived cultivars) were collected and analyzed with high-density single nucleotide polymorphism(SNP) array. Scanning all these cultivars with genome-wide SNP data indicated the unique contributions of progenitors to the SH527 genome and identified the key genomic regions of SH527 conserved within all its derivatives. These findings were further supported by known rice yield-related genes or unknown QTLs identified by genome-wide association study.
