| 1 | Suppression or knockout of SaF/SaM overcomes the Sa-mediated hybrid male sterility in rice显示文摘Hybrids between the indica and japonica subspecies of rice(Oryza sativa) are usually sterile, which hinders utilization of heterosis in the inter-subspecific hybrid breeding. The complex locus Sa comprises two adjacently located genes, SaF and SaM, which interact to cause abortion of pollen grains carrying the japonica allele in japonica-indica hybrids. Here we showed that silencing of SaF or SaM by RNA interference restored male fertility in indica-japonica hybrids with heterozygous Sa. We further used clustered regularly interspaced short palindromic repeats(CRISPR)/Cas9-based genome editing to knockout the SaF and SaM alleles, respectively, of an indica rice line to create hybrid-compatible lines. The resultant artificial neutral alleles did not affect pollen viability and other agricultural traits, but did break down the reproductive barrier in the hybrids. We found that some rice lines have natural neutral allele Sa-n, which was compatible with the typical japonica or indica Sa alleles in hybrids. Our results demonstrate that SaF and SaM are required for hybrid male sterility, but are not essential for pollen development. | 8 | Relationship between peritumoral lymphatic microvessel density and the clinical and pathological characteristics of invasive breast cancer显示文摘Objective The aim of the study was to determine the morphological characteristics of lymphatic microvessels and the relationship between lymphatic microvessel density(LMVD) and clinical and pathological characteristics of invasive breast cancer.Methods Tissue specimens and clinical pathological data of 51 cases of female breast cancer were collected in the General Hospital of Shenyang Military Region(Shenyang, China) from January 2007 to October 2011. Another 20 breast fibroadenoma tissue samples were used as controls. All specimens were cut into 4-μm slices, and immunohistochemically stained using streptomycin-resistant avidin peroxidase antibody D2-40. SPSS 17.0 for Windows was used to perform all analyses.Results A total of 38 breast cancer tissue specimens showed varied staining with monoclonal antibody D2-40. The rate of positive staining was in these tissues was 74.5%(38/51), which is significantly higher than that observed in breast fibroadenoma tissues(chi-square = 35.197, P = 0.000). The average LMVD in 38 cases of breast cancer was(26.46 ± 10.06) microvessels/100× magnification field, which was higher than that in the control group(t = 10.74, P = 0.000). Microvessels in peritumoral tissues were abundant, with an average LMVD of(38.42 ± 11.38) microvessels/100× magnification field. Based on layered analysis, the expression level of peritumoral LMVD was correlated with metastasis of lymph nodes, tumor size, and the expression of C-erb B-2(P < 0.05); however, there was no correlation with age or expression of estrogen receptors or progesterone receptors(P > 0.05).Conclusion Lymphatic microvessels detected using D2-40 antibody are mainly present in the peritumoral region of breast cancer tissues, and LMVD showed a correlation with lymph node metastasis and the expression of C-erb B-2. Positive lymphatic vessels, especially in the peritumoral region, may provide a path for lymphatic metastasis in breast cancer. | 9 | Experimental and numerical analysis of the hydraulic and thermal performances of the gradually-varied porous volumetric solar receiver显示文摘A gradually-varied porous structure is designed to increase the thermal performance of the porous volumetric solar receiver.Based on the replica method and multilayer recoating technique, the silicon carbide porous ceramic with linear-changed geometrical parameters is fabricated. The performances of the uniform and gradually-varied porous volumetric solar receivers are studied by both experiment and numerical simulation. An optimization method combining genetic algorithm and computational fluid dynamics analysis is applied to determine the optimum porosity distribution. The results present that porous volumetric solar receiver with linear-changed geometrical parameters exhibits better thermal performance than the uniform porous volumetric solar receivers, especially when the thickness of the receiver is small. Larger porosity in the front is beneficial for increasing the solar radiation penetration depth, which limits the reflectance and thermal radiative losses. Smaller porosity in the rear traps more solar radiation and increases the convective heat transfer. When the receiver’s thickness is larger, the performance of the gradually-varied volumetric solar receiver is nearly identical to that of the uniform receiver with largest porosity. The double-layer configuration is found to be the optimized structure of the gradually-varied porous volumetric solar receiver. | 12 | Clinical observation of capecitabine monotherapy in elderly patients with advanced breast cancer显示文摘Objective The aim of the study was to evaluate the safety and efficacy of capecitabine mono-chemotherapy in elderly patients with advanced breast cancer. Methods The data from 36 cases of capecitabine monotherapy in elderly patients with advanced breast cancer were retrospectively analyzed. Oral administration of capecitabine 2000 mg/m2 twice daily(D1–14) for 21 days constituted a cycle. The effect of the disease and main adverse reactions were evaluated every 2 cycles. Results The data from 36 elderly patients were studied. The median number of chemotherapy cycles was 4. The total effective rate was 30.6%(11/36) and the disease control rate was 72.2%(26/36). The number of patients with clinical complete remission was 2, clinical partial response was 9, stable disease was 15, and progressive disease was 10. Where treatment was effective, the median time to progression was 6 months and the median overall survival was 9.5 months. The main adverse events were gastrointestinal reactions, bone marrow suppression, and oral mucositis; most of the reactions were grade 1 to 2. | 15 | Effect of dendritic cell/cytokine-induced killer cell immunobiological cancer therapy combined with adjuvant chemotherapy in patients with triple-negative breast cancer显示文摘Objective The aim of the present study was to investigate the effect of dendritic cell(DC)/cytokine-induced killer cell(CIK) immunobiological cancer therapy in patients with triple-negative breast cancer(TNBC) who underwent adjuvant chemotherapy. Methods From January 2010 to October 2013, 120 patients with postoperative TNBC were recruited and included in the study. Patients were enrolled in one of two groups according to whether they accepted DC/CIK immunobiological cancer therapy during adjuvant chemotherapy; the patients in the DC/CIK group underwent adjuvant chemotherapy combined with DC/CIK immunobiological cancer therapy, and the control group underwent adjuvant chemotherapy alone. When six cycles of adjuvant chemotherapy and six cycles of DC/CIK immunobiological cancer therapy had been completed, differences between the two groups with regard to quality of life(Qo L), immunological indicators(CD3, CD4, CD8, and NK cell levels), disease-free survival(DFS), and side effects of chemotherapy and DC/CIK treatment were evaluated.Results In the DC/CIK group, the proportion of NK cells and CD3+ and CD4+ T-cell subgroups significantly increased, and the proportion of CD8+ cells decreased when they were compared before and after DC/CIK therapy(P < 0.05). However, there were no significant changes in the control group. By the final follow-up, DFS of the treatment group and the control group was 38.4 and 34.2 months, respectively. The Qo L improved in the patients treated with chemotherapy plus DC/CIK therapy compared with the patients treated with chemotherapy alone, and the difference between groups was significant(P < 0.05). The side effects of two groups were tolerable and not significantly different between the two groups.Conclusion The DC/CIK treatment had potential benefits for patients with TNBC compared with the control group, and was not associated with any obvious side effects. Methods: The data of 72 cases of neoadjuvant chemotherapy(DX) in locally advanced breast cancer after 4 cycles were retrospectively analyzed. Docetaxel 75 mg/m2 by infusion 1 h on d1, capecitabine 2000 mg/m2 by oral for twice daily on d1–14, 21 days was a cycle. Results: All 72 patients were assessed for efficacy and adverse events. The total effective rate was 80.5%(58/72), including pathological complete response(p CR) was 7(9.7%), clinical complete remission(c CR) was 15(20.8%), clinical partial response(PR) was 43(59.7%), stable disease(SD) was 8(11.1%) and progressive disease(PD) was 6(8.3%). The main adverse events were gastrointestinal reactions and bone marrow suppression. The 3 to 4 degrees of adverse reactions including granulocytopenia in 7 patients(20.6%), hand-foot syndrome in 6 patients(15.2%). | 17 | 依托泊苷胶囊联合顺铂或卡铂治疗老年小细胞肺癌的临床观察(英文)显示文摘Objective: We aimed to explore the efficacy and safety of etoposide capsule combined with cisplatin or carboplatin in the treatment of elderly patients with small cell lung cancer(SCLC). Methods: From October 2011 to November 2013, 32 elderly patients(71–79 years old) with histopathologically confirmed SCLC in General Hospital of Shenyang Military Region(China) were enrolled in the research. The patients were administrated with lastet capsule 150–175 mg, d1–5, combined with cisplatin 20 mg/m2 d1–3 or carboplatin AUC = 5, applied over 2 days. Twenty-one days were 1 treatment cycle. Results: After treatments, 2 cases acquired complete response(CR), 19 cases acquired partial response(PR), 8 cases acquired stable disease(SD), and 3 cases had progression of disease(PD). The objective response rate was 65.6%(21/32), disease control rate was 90.6%(29/32). The median time of progression-free survival(PFS) was 6.9 months, the median survival time was 14.0 months, and 1 year survival rate was 62.4%. The main adverse reactions of I/II leukopenia and gastrointestinal reaction were observed. | 18 | Effect of pore size and porosity distribution on radiation absorption and thermal performance of porous solar energy absorber显示文摘In this paper, experimental and numerical study were both conducted to investigate the effect of pore size and porosity distribution on radiation absorption and thermal performance of porous solar energy absorber. Ultraviolet-visible-near infrared(UV-Vis-NIR) spectrophotometer was used to measure the transmittance of porous media to reflect its radiation absorption capabilities. Numerical model was established based on the assumption of thermal nonequilibrium condition as well as using P1 model to consider the radiation heat transfer. The UV-Vis-NIR spectrophotometer measurement showed that:(1) With smaller pore size, the spectral transmittance of the porous media would be lower and the solar radiation absorption would be better;(2)Among the materials with different pore size distributions, pore-size-decreased combo and pore-size-increased combo have almost equal absorption coefficient which are higher than that of uniform structure. Methods: The expression of VEGF-C mRNA in 51 cases of human breast cancer was assessed by hybridization in situ. The expressions of C-erbB-2 was assessed by immunohistochemistry. Results: The positive rate of VEGF-C mRNA was 54.9% in 51 cases of breast cancer. The transcription level had correlation with tumor size and status of lymph nodes(P < 0.05). The expression of VEGF-C mRNA had a positive correlation with the expression of C-erbB-2(P < 0.05). Conclusion: The up-expression of VEGF-C has a significant correlation with the malignancy level and clinical stage of breast cancer.
