| 2 | Enhancing the antihepatitis B virus immune response by adefovir dipivoxil and entecavir therapies显示文摘Chronicity of hepatitis B(CHB)infection is characterized by a weak immune response to the virus.Entecavir(ETV)and adefovir dipivoxil(ADV)are effective in suppressing hepatitis B virus(HBV)replication.However,the underlying immune mechanism in the antiviral response of patients treated with nucleoside or nucleotide analogs is not clearly understood.In this study,regulatory T cells(Tregs)and intracellular cytokines,including IL-2,interferon(IFN)-γ,tumor-necrosis factor(TNF)-α and IL-4,were measured prior to and at 12,24,36 and 48 weeks after treatment with ETV or ADV.The cytokines were increased from 24 to 48 weeks after treatment.Higher levels of Th1 cytokines were observed with ETV(n=29)versus ADV(n=28)treatment.By contrast,the numbers of Tregs in both groups were decreased.The altered cytokine profile and cellular component was accompanied by a decrease in HBV DNA levels in both groups,which may contribute to their therapeutic effect in CHB infection.Our findings suggest that the antiviral effect of the drugs may be attributed not only to their direct effect on virus suppression but also to their immunoregulatory capabilities. | Yanfang Jiang Wanyu Li Lei Yu Jingjing Liu Guijie Xin Hongqing Yan Pinghui Sun Hong Zhang Damo Xu Junqi Niu | 2011 | Cellular & Molecular Immunology2011,8,1: | 16 |

| 4 | Abnormal activation of the occipital lobes during emotion picture processing in major depressive disorder patients显示文摘A large number of studies have demonstrated that depression patients have cognitive dysfunction. With recently developed brain functional imaging, studies have focused on changes in brain function to investigate cognitive changes. However, there is still controversy regarding abnormalities in brain functions or correlation between cognitive impairment and brain function changes. Thus, it is important to design an emotion-related task for research into brain function changes. We selected positive, neutral, and negative pictures from the International Affective Picture System. Patients with major depressive disorder were asked to judge emotion pictures. In addition, functional MRI was performed to synchronously record behavior data and imaging data. Results showed that the total correct rate for recognizing pictures was lower in patients compared with normal controls. Moreover, the consistency for recognizing pictures for depressed patients was worse than normal controls, and they frequently recognized positive pictures as negative pictures. The consistency for recognizing pictures was negatively correlated with the Hamilton Depression Rating Scale. Functional MRI suggested that the activation of some areas in the frontal lobe, temporal lobe, parietal lobe, limbic lobe, and cerebellum was enhanced, but that the activation of some areas in the frontal lobe, parietal lobe and occipital lobe was weakened while the patients were watching positive and neutral pictures compared with normal controls. The activation of some areas in the frontal lobe, temporal lobe, parietal lobe, and limbic lobe was enhanced, but the activation of some areas in the occipital lobe were weakened while the patients were watching the negative pictures compared with normal controls. These findings indicate that patients with major depressive disorder have negative cognitive disorder and extensive brain dysfunction. | 5 | Inflammation-induced CD69^+ Kupffer cell feedback inhibits T cell proliferation via membrane-bound TGF-β1显示文摘Kupffer cells,tissue-resident macrophage lineage cell,are enriched in vertebrate liver.The mouse F4/80^+ Kupffer cells have been subclassified into two subpopulations according to their phenotype and function:CD68^+ subpopulation with potent reactive oxygen species(ROS) production and phagocytic capacities,and CD11b^+ subpopulation with a potent capacity to produce T helper 1 cytokines.In addition,CD11b^+ Kupffer cells/macrophages may be migrated from the bone marrow or spleen,especially in inflammatory conditions of the liver.For analyzing diverse Kupffer cell subsets,we infected mice with Listeria monocytogenes and analyzed the phenotype variations of hepatic Kupffer cells.During L.monocytogenes infection,hepatic CD69^+ Kupffer cells were significantly induced and expanded,and CD69^+ Kupffer cells expressed higher level of CD11 b,and particularly high level of membrane-bound TGF-β1(mTGF-β1) but lower level of F4/80.We also found that clodronate liposome administration did not eliminate hepatic CD69^+ Kupffer cell subset.We consider the hepatic CD69^+ Kupffer cell population corresponds to CD11b^+Kupffer cells,the bone marrow-derived population.Hepatic CD69^+ Kupffer cells suppressed Ag-nonspecific and OVA-specific CD4 T cell proliferation through mTGF-β1 both in vitro and in vivo,meanwhile,they did not interfere with activation of CD4 T cells.Thus,we have identified a new subset of inflammation-induced CD69^+ Kupffer cells which can feedback inhibit CD4 T cell response via cell surface TGF-β1 at the late stage of immune response against infection.CD69^+ Kupffer cells may contribute to protect host from pathological injure by preventing overactivation of immune response. | Xiang Zhang Zhengping Jiang Yan Gu Yanfang Liu Xuetao Cao Yanmei Han | 2016 | Science China(Life Sciences)2016,59,12: | 5 |
| 6 | Impact of biochar amendment on soil aggregation varied with incubation duration and biochar pyrolysis temperature显示文摘Soil aggregation is one of the crucial processes that facilitate carbon sequestration and maintain soil fertility.So far,the effect of biochar amendment on soil aggregation remains inconclusive.Here,we tested the hypothesis that the response of soil aggregation to biochar addition varied with incubation duration and biochar chemistry.A one year microcosm experiment of soil with biochar was conducted that included biochar produced at three different temperatures(300,450,and 600°C),and three biochar application rates,i.e.,0,1,and 3 wt%.It was observed that after one and three months,biochar mainly(>90%)distributed in the micro-aggregates,and slightly reduced aggregate stability and increased proportion of micro-aggregates,which was demonstrated to result from the mechanical mixture of amended biochar with soil.Contrastingly,when the duration was prolonged to six months and one year,a significant increase in macro-aggregates(6.6-38.5%)and aggregate stability(7.3-29.4%)was detected,with the increasing extent being apparently higher for low-temperature biochar.This was related to the comparatively strong interaction of biochar particles with soil minerals or microbes after long-time incubation.The strong interaction was directly supported by the significant increase in H/C,O/C ratios of isolated biochar from treated soils,the detection of typical soil mineral elements on the surface of isolated biochar,and the increase in microbial biomass carbon of treated soils.The findings of this study highlighted the role of biochar type and amendment duration in mediating the effect of biochar application on soil aggregation. | Lanfang Han Biao Zhang Liying Chen Yanfang Feng Yan Yang Ke Sun | 2021 | Biochar2021,3,3: | 4 |

| 13 | Expression of P-gp and miRNA-21 in Colorectal Cancer Tissues and the Clinical Significance显示文摘[Objectives]To study the expression of P-gp and miRNA-21 in colorectal cancer tissues and the relationship between the expression of P-gp and clinicopathological features,and to analyze the correlation between the expression of miRNA-21 and P-gp.[Methods]Immunohistochemical SP method was used to detect the expression level of P-gp in the tissues of 40 cases of colorectal cancer and the normal mucosal tissues 5 cm away from the corresponding cancer tissues,and the expression level of miRNA-21 in the paraffin tissues of the 40 cases was detected by RT-qPCR.[Results]The positive expression of P-gp was localized in the cell membrane,and the expression in colorectal cancer tissues was significantly higher than that in the corresponding adjacent tissues(P<0.05);the positive expression of P-gp was closely related to the degree of tumor differentiation and the depth of invasion(P<0.05),and the comparison results of P-gp expression in the differentiation groups of different cancer tissues showed that the expression level of P-gp in the moderately and poorly differentiated groups was significantly higher than that in the highly differentiated group(P<0.05),and the expression level of the moderately and poorly differentiated groups was similar(P>0.05).The expression of miRNA-21 in all cancer tissues was higher than that in the corresponding adjacent tissues(P<0.05),and the expression levels of miRNA-21 in differentiation groups of different cancer tissues were 1.286,1.445 and 1.992 times of those in the corresponding adjacent tissues,respectively;the comparison results of miRNA-21 between different differentiation groups showed that the expression level of miRNA-21 in the poorly differentiated group was significantly higher than that in the highly differentiated group and the moderately differentiated group(P<0.05),while the expression level of miRNA-21 in the highly differentiated group and the moderately differentiated group was similar(P>0.05).Spearman correlation analysis showed that there was a positive correlation on the expression between the two in colorectal cancer tissues(r=0.423,P<0.05).[Conclusions]The expression levels of miRNA-21 and P-gp are closely related to the differentiation degree of clinical colorectal cancer,the lower the differentiation degree,the higher the expression level of the two. | Xiaoli YAN Peng YAN Yanfang FAN Jing XUE Mingjuan WANG Jining ZHENG | 2020 | Medicinal Plant2020,11,6: | 1 |

| 19 | Crosstalk between CYP2E1 and PPARα substrates and agonists modulate adipose browning and obesity显示文摘Although the functions of metabolic enzymes and nuclear receptors in controlling physiological homeostasis have been established, their crosstalk in modulating metabolic disease has not been explored.Genetic ablation of the xenobiotic-metabolizing cytochrome P450 enzyme CYP2 E1 in mice markedly induced adipose browning and increased energy expenditure to improve obesity. CYP2 E1 deficiency activated the expression of hepatic peroxisome proliferator-activated receptor alpha(PPARa) target genes,including fibroblast growth factor(FGF) 21, that upon release from the liver, enhanced adipose browning and energy expenditure to decrease obesity. Nineteen metabolites were increased in Cyp2 e1-null mice as revealed by global untargeted metabolomics, among which four compounds, lysophosphatidylcholine and three polyunsaturated fatty acids were found to be directly metabolized by CYP2 E1 and to serve as PPARa agonists, thus explaining how CYP2 E1 deficiency causes hepatic PPARa activation through increasing cellular levels of endogenous PPARa agonists. Translationally, a CYP2 E1 inhibitor was found to activate the PPARa-FGF21-beige adipose axis and decrease obesity in wild-type mice, but not in liver-specific Pparanull mice.
