| 1 | Ecosystem carbon and nitrogen storage following farmland afforestation with black locust(Robinia pseudoacacia) on the Loess Plateau, China显示文摘Although afforestation of farmlands has been proposed as an effective method of carbon(C) sequestration, there remain uncertainties that deter us from developing a clear picture of C stocks in plantation ecosystems.This study investigated the dynamics of stand structure and plant diversity, and C and nitrogen(N) pools in trees,herbs, litter, and soil(0–100 cm depth) in black locust plantations aged 9, 17, 30, and 37 years, and in newly abandoned farmlands as pre-afforestation sites, on the Loess Plateau, China. Stand density decreased significantly, while tree diameter at breast height and height increased during stand development. The dominant species of the herb layer differed with age. Afforestation resulted in slight increases in tree C and N storage in plantations from 9 to 30 years of age, and then significantly increased from 30 to 37 years. Compared to pre-afforestation, C and N storage in soil decreased to minimum values in stands aged 17 and 9 years, respectively. The soil re-accumulated C and N during stand development, attaining equilibrium levels similar to those in pre-afforestation when stands reached about 30 years of age. Soil C and N storage in 37-year stands were 29 and 16% higher, respectively, than in pre-afforestation levels. However, C and N concentrations in the subsoil(20–40 cm) were still less than the preafforestation levels for stands of all ages(from 9 to 37 years). The relative contribution to the total ecosystem C and N pools increased in trees and decreased in soil during the observed period. Our results indicate that afforestation reduced soil C and N storage during the early stages of stand development. | 2 | Synthesis and pervaporation performance of high-reproducibility silicalite-1 membranes显示文摘High-reproducibility silicalite-1 membranes were synthesized on silica tubes by in-situ hydrothermal synthesis, and the relative standard deviation (R.S.D.) of average separation factor for eight membranes was only 7.5%. By using an effective method called 'solution-filling (SF)', the average flux of membranes prepared with SF method was improved by about 49% compared to membranes prepared without SF method, and the highest flux of membrane prepared with SF method towards ethanol/H2O mixture at 80℃ was 1.49 kg/(m2·h). | 4 | Disruption of crosstalk between LX-2 and liver cancer stem-like cells from MHCC97H cells by DFOG via inhibiting FOXM1显示文摘Hepatic stellate cell(HSC)line LX-2 is activated by liver cancer stem-like cells(LCSLCs)and produces various cytokines that make up most of the hepatocellular carcinoma(HCC)microenvironment.The new genistein derivative,7-difluoromethoxyl-5,4-dirboctylgenistein(DFOG),shows anticancer effects in multiple malignancies by controlling forkhead box M1(FOXM1).In this study,we aimed to assess whether DFOG disrupts the crosstalk between human HSC LX-2 cells and LCSLCs.Distinct gen erations of MHCC97H-derived spheres were obtained with the sec ond generation considered as LCSLCs which displayed enhanced self-renewal ability and elevated expression levels of CD133,CD44,and EpCAM proteins,as well as tumorigenicity,as revealed by colony formation assay in vitro and tumorigenicity assay in vivo.LX-2 and MHCC97H cells were co-cultured with/without DFOG(1,5,and 10 pM,respectively)using the transwell system.FOXM1 overexpression and/or knockdown were employed for mechanistic investigations.Our results suggested that Co-CM promoted LX-2 cell transformation into liver cancer-associated HSCs.Meanwhile,FOXM1 was up-regulated and the level of hepatocyte growth factor(HGF)was in creased in LX-2 cells and in the super nata nt after Co-CM stimulati on.Sphere and colony formation abilities in MHCC97H cells,and protein levels of CD133r CD44,and EpCAM,were also markedly elevated.DFOG dose-dependently inhibited the above effects,similar to FOXM1 knockdown in LX-2 cells.FOXM1 overexpression reversed the inhibitory effects of DFOG or FOXM1 knockdown or both on LX-2 cell activation and LCSLC feature induction in MHCC97H cells by LCSLC/LX-2 co-culture.This study demonstrated that DFOG disrupts the crosstalk between HSCs and LCSLCs to suppress LCSLC features via dowrrregulating FOXM1 expression and reducing HGF secretion in HSCs. | A Chen Chang Xu Yimin Luo Lihua Liu Kun Song Guangqi Deng Mengjie Yang Jianguo Cao Liming Yuan Xiang Li | 2019 | Acta Biochimica et Biophysica Sinica2019,51,12: | 2 |

| 9 | Synthesis and separation performance of silicalite-1 membranes on silica tubes显示文摘High-performance silicalite-1 membranes were synthesized on silica tubes by in-situ hydrothermal synthesis. By using the 'solution-filling (SF)' method, the average flux of membranes with the SF method was improved by about 25% compared to that of the membranes without using the SF method; the flux and the separation factor of the membranes prepared with the SF method for an ethanol/water mixture at 60 ℃ were 0.99 kg/(m2·h) and 73, respectively. | 10 | Generating homozygous mutant populations of barley microspores by ethyl methanesulfonate treatment显示文摘Induced mutations are important for genetic research and breeding.Mutations induced by physical or chemical mutagenesis are usually heterozygous during the early generations.However,mutations must be fixed prior to phenotyping or field trials,which requires additional rounds of self-pollination.Microspore culture is an effective method to produce double-haploid(DH)plants that are fixed homozygotes.In this study,we conducted ethyl methanesulfonate(EMS)-induced mutagenesis of microspore cultures of barley(Hordeum vulgare)cultivar‘Hua30’and landrace‘HTX’.The EMS concentrations were negatively correlated with the efficiency of callus induction and the frequency of mutant plant regeneration.The two genotypes showed different regeneration efficiencies.The phenotypic variation of the regenerated M1 plants and the presence of genome-wide nucleotide mutations,revealed by whole-genome sequencing,highlight the utility of EMS-induced mutagenesis of isolated microspore cultures for developing DH mutants.Genome-wide analysis of the mutation frequency in the regenerated plants revealed that a considerable proportion of mutations resulted from microspore culture(somaclonal variation)rather than EMS-induced mutagenesis.In addition to producing a population of 1972 homozygous mutant lines that are available for future field trials,this study lays the foundation for optimizing the regeneration efficiency of DH plants and the richness of mutations(mainly by fine-tuning the mutagen dosage). | Linli Huang Guangqi Gao Congcong Jiang Guimei Guo Qiang He Yingjie Zong Chenghong Liu Ping Yang | 2023 | aBIOTECH2023,4,3: | 0 |

| 14 | A novel and effective hyperthermia method for Schistosomiasis japonica prevention and treatment显示文摘The emerging resistance to schistosoma has been paid much attention and is in urgent need for a novel strategy to control the prevalent parasitic zoonosis. In this study, the efficiency of hyperthermia therapy was investigated by the skin hyperthermia device. The survival rate of cercariae decreased from 68.15 %(37 °C, 5 min) to 0(49 °C, 10 min) with the thermal dosages increased, which proved the preventing effect of hyperthermia therapy(P \ 0.05). Therapeutic effects were assessed in Schistosoma japonicum-infected BALB/c mice. When the cercarial contact region of the skin was treated at 45–49 °C for5 min within 8 h of infection, worm reduction rate(WRR)reached 74 %–83 %(P \ 0.01). The sensitivity of adultschistosoma to heat was also investigated using microwave intraperitoneal hyperthermia(thermal dosages 42–43 °C,20 min). The WRR, hepatic shift rates and egg reduction rates were 23.7 %, 40 % and 30 %, respectively, comparing with 80.2 %, 59.6 % and 53.9 % of praziquantel(PZQ)-treated group. Encouraging results have been obtained that hyperthermia can effectively kill schistosomula, especially with the appearance of cercarial dermatitis, while PZQ lacks efficacy against the cercariae.
