2Determination of DNA single-strand breaks by low-energy heavy ion and analysis of dose-effect curves显示文摘Calf thymus DNA was exposed to low-energy heavy ions (N+) and 60Co-γ-rays, and the dose-effect on DNA single-strand breaks (SSB) has been investigated. The results indicate that the dose-effect curve by N+ irradiation is different from that of conventional ionizing radiation. While the curve from γ-irradiation follows exponential type, the effect curve produced by N+ ion is of 'saddle type'. The yield of DNASSB per dose unit per DNA unit remained at a certain level under different doses of γ-rays. 5Solid-state intramolecular motions in continuous fibers driven by ambient humidity for fluorescent sensors显示文摘One striking feature of molecular rotors is their ability to change conformation with detectable optical signals through molecular motion when stimulated.However,due to the strong intermolecular interactions,synthetic molecular rotors have often relied on fluid environments.Here,we take advantage of the solid-state intramolecular motion of aggregation-induced emission(AIE)molecular rotors and one-dimensional fibers,developing highly sensitive optical fiber sensors that respond to ambient humidity rapidly and reversibly with observable chromatic fluorescence change.Moisture environments induce the swelling of the polymer fibers,activating intramolecular motions of AIE molecules to result in red-shifted fluorescence and linear response to ambient humidity.In this case,polymer fiber provides a process-friendly architecture and a physically tunable medium for the embedded AIE molecules to manipulate their fluorescence response characteristics.Assembly of sensor fibers could be built into hierarchical structures,which are adaptive to diverse-configuration for spatial-temporal humidity mapping,and suitable for device integration to build light-emitting sensors as well as touchless positioning interfaces for intelligence systems.Yunmeng Jiang Yanhua Cheng Shunjie Liu Haoke Zhang Xiaoyan Zheng Ming Chen Michidmaa Khorloo Hengxue Xiang Ben Zhong Tang Meifang Zhu 2021National Science Review2021,8,4:3

13A critical review of on-site inorganic arsenic screening methods显示文摘Approximately 94 to 220 million people worldwide are at risk of drinking well water containing arsenic > 10 μg/L, the WHO guideline value. To identify non-compliant domestic wells, assess health risks and reduce exposure, accurate and rapid on-site inorganic arsenic screening methods are desirable because all domestic wells worldwide need to be tested.Here, the principles, advantages and limitations of commonly used colorimetry, electrochemistry, and biosensing methods are critically reviewed, with the performance compared with laboratory-based benchmark methods. Most commercial kits are based on the classic Gutzeit reaction. Despite being semi-quantitative, the more recent and more expensive products display improved and acceptable accuracy and shorter testing time (~10 min). Carried out by trained professionals, electrochemical methods are also feasible for on-site analysis, although miniaturization is desirable yet challenging. Biosensing using whole bacterial cells or bio-engineered materials such as aptamers is promising, if incorporated with function specific nanomaterials and biomaterials. Since arsenic is frequently found as arsenite in reducing groundwater and subject to oxidation during sampling, transportation and storage, on-site separation and sample preservation are feasible but the specific methods should be chosen based on sample matrix and tested before use. To eliminate arsenic exposure among hundreds of millions of mostly rural residents worldwide, we call for concerted efforts in research community and regulatory authority to develop accurate, rapid, and affordable tests for on-site screening and monitoring of arsenic in drinking water. 16Multiomics approach reveals the ubiquitination-specific processes hijacked by SARS-CoV-2显示文摘The Coronavirus Disease 2019(COVID-19)caused by Severe Acute Respiratory Syndrome Coronavirus 2(SARS-CoV-2)is a global pandemic that seriously threatens health and socioeconomic development,but the existed antiviral drugs and vaccines still cannot yet halt the spread of the epidemic.Therefore,a comprehensive and profound understanding of the pathogenesis of SARS-CoV-2 is urgently needed to explore effective therapeutic targets.Here,we conducted a multiomics study of SARS-CoV-2-infected lung epithelial cells,including transcriptomic,proteomic,and ubiquitinomic.Multiomics analysis showed that SARS-CoV-2-infected lung epithelial cells activated strong innate immune response,including interferon and inflammatory responses.Ubiquitinomic further reveals the underlying mechanism of SARS-CoV-2 disrupting the host innate immune response.In addition,SARS-CoV-2 proteins were found to be ubiquitinated during infection despite the fact that SARS-CoV-2 itself didn’t code any E3 ligase,and that ubiquitination at three sites on the Spike protein could significantly enhance viral infection.Further screening of the E3 ubiquitin ligases and deubiquitinating enzymes(DUBs)library revealed four E3 ligases influencing SARS-CoV-2 infection,thus providing several new antiviral targets.This multiomics combined with high-throughput screening study reveals that SARS-CoV-2 not only modulates innate immunity,but also promotes viral infection,by hijacking ubiquitination-specific processes,highlighting potential antiviral and anti-inflammation targets.Gang Xu Yezi Wu Tongyang Xiao Furong Qi Lujie Fan Shengyuan Zhang Jian Zhou Yanhua He Xiang Gao Hongxiang Zeng Yunfei Li Zheng Zhang 2022Signal Transduction and Targeted Therapy2022,7,10:1

19Single-cell transcriptomic profiling reveals the tumor heterogeneity of small-cell lung cancer显示文摘Small-cell lung cancer(SCLC)is the most aggressive and lethal subtype of lung cancer,for which,better understandings of its biology are urgently needed.Single-cell sequencing technologies provide an opportunity to profile individual cells within the tumor microenvironment(TME)and investigate their roles in tumorigenic processes.Here,we performed high-precision single-cell transcriptomic analysis of~5000 individual cells from primary tumors(PTs)and matched normal adjacent tissues(NATs)from 11 SCLC patients,including one patient with both PT and relapsed tumor(RT).The comparison revealed an immunosuppressive landscape of human SCLC.Malignant cells in SCLC tumors exhibited diverse states mainly related to the cell cycle,immune,and hypoxic properties.Our data also revealed the intratumor heterogeneity(ITH)of key transcription factors(TFs)in SCLC and related gene expression patterns and functions.The non-neuroendocrine(non-NE)tumors were correlated with increased inflammatory gene signatures and immune cell infiltrates in SCLC,which contributed to better responses to immune checkpoint inhibitors.These findings indicate a significant heterogeneity of human SCLC,and intensive crosstalk between cancer cells and the TME at single-cell resolution,and thus,set the stage for a better understanding of the biology of SCLC as well as for developing new therapeutics for SCLC.Yanhua Tian Qingqing Li Zhenlin Yang Shu Zhang Jiachen Xu Zhijie Wang Hua Bai Jianchun Duan Bo Zheng Wen Li Yueli Cui Xin Wang Rui Wan Kailun Fei Jia Zhong Shugeng Gao Jie He Carl MGay Jianjun Zhang Jie Wang Fuchou Tang 2022Signal Transduction and Targeted Therapy2022,7,11:1
