1A new gain-of-function OsGS2/GRF4 allele generated by CRISPR/Cas9 genome editing increases rice grain size and yield显示文摘Grain size is one of the most important factors affecting rice grain quality and yield,and attracts great attention from molecular biologists and breeders.In this study,we engineered a CRISPR/Cas9 system targeting the miR396 recognition site of the rice GS2 gene,which encodes growth-regulating factor 4(OsGRF4)and regulates multiple agronomic traits including grain size,grain quality,nitrogen use efficiency,abiotic stress response,and seed shattering.In contrast to most previous genome editing efforts in which indel mutations were chosen to obtain null mutants,a mutant named GS2^(E) carrying an in-frame 6-bp deletion and 1-bp substitution within the miR396-targeted sequence was identified.GS2^(E) plants showed increased expression of GS2 in consistent with impaired repression by miR396.As expected,the gain-of-function GS2^(E) mutant exhibited multiple beneficial traits including increased grain size and yield and bigger grain length/width ratio.Thousand grain weight and grain yield per plant of GS2^(E) plants were increased by 23.5%and 10.4%,respectively.These improved traits were passed to hybrids in a semidominant way,suggesting that the new GS2^(E) allele has great potential in rice improvement.Taken together,we report new GS2 germplasm and describe a novel gene-editing strategy that can be widely employed to improve grain size and yield in rice.This trait-improvement strategy could be applied to other genes containing miRNA target sites,in particular the conserved miR396-GRF/GIF module that governs plant growth,development and environmental response.Wenshu Wang Weipeng Wang Yanlin Pan Chao Tan Hongjing Li Ya Chen Xingdan Liu Jing Wei Nian Xu Yu Han Han Gu Rongjian Ye Qi Ding Chonglie Ma 2022The Crop Journal2022,10,4:3
2Evolution and modeling of mine water inflow and hazard characteristics in southern coalfields of China:A case of Meitanba mine显示文摘In this paper,the hydrogeological characteristics in the southern coalfields of China are first briefly outlined.Then,taking the Meitanba mine as an example,the evolution and modeling of mine water inflow are studied.Finally,the hazard characteristics related to mine water and mud inrush are analyzed.The results show that the main mine water sources in the Meitanba mine area are groundwater,surface water and precipitation.The evolution of mine water inflow with time indicates that the water inflow is closely related to the development of karst structures,the amount of water from rainfall infiltration,and the scope of groundwater depression cone.The mine water inflow increases with time due to the increase in mining depth and the expansion of groundwater depression cone.Using the big well method and following the potential superposition principle,a hydrogeological model considering multi-well interactions has been developed to predict the mine water inflow.Based on the monitored data in the Meitanba mine area over a period of nearly 60 years,it is found that with increasing mining depth,the number of water and mud inrush points tended to decrease.However,the average water and mud flow rate per point tended to increase.Jinhai Liu Yanlin Zhao Tao Tan Lianyang Zhang Sitao Zhu Fangyan Xu 2022International Journal of Mining Science and Technology2022,32,3:2

7Bovine HSD17B8 gene and its relationship with growth and meat quality traits显示文摘17 b-Hydroxysteroid dehydrogenase type 8(HSD17B8) is an important regulator of lipid and steroid metabolism. In the present study, we aimed to assess the effects of HSD17B8 on growth and meat quality traits in cattle. Transcription profile analysis showed that HSD17B8 was primarily expressed in the salpinx, liver, and testis.Meanwhile, we identified three SNPs(SNP1: intron1-A91G; SNP2: exon 1-A90G; and SNP3: intron 8-A86G)of the bovine HSD17B8 gene and investigated its haplotype frequencies and linkage disequilibrium. The detected SNPs were found associated with growth traits(body weight,body length, height at withers, heart girth, hip width, and average daily gain) in native cattle populations(Nanyangand Jiaxian) as well as the meat quality traits(Warner–Bratzler shear force, rib area, dressing percentage, carcass weight, and backfat thickness) in commercial breeds(Angus, Hereford, Limousin, Luxi, Simmental, and Jinnan).Our results provided evidence that polymorphisms in the HSD17B8 gene were associated with growth traits and meat quality traits. 9Anti-Overturning Fully Symmetrical Triboelectric Nanogenerator Based on an Elliptic Cylindrical Structure for All-Weather Blue Energy Harvesting显示文摘Triboelectric nanogenerators(TENGs) have shown promising potential for large-scale blue energy harvesting. However, the lack of reasonable designs has largely hindered TENG from harvesting energy from both rough and tranquil seas. Herein, a fully symmetrical triboelectric nanogenerator based on an elliptical cylindrical structure(EC-TENG) is proposed for all-weather blue energy harvesting. The novel elliptical cylindrical shell provides a unique selfstability, high sensitivity to wave triggering, and most importantly, an anti-overturning capability for the EC-TENG. Moreover, benefiting from its internal symmetrical design, the EC-TENG can produce energy normally, even if it was overturned under a rude oscillation in the rough seas, which distinguishes this work from previous reported TENGs. The working mechanism and output performance are systematically studied. The as-fabricated EC-TENG is capable of lighting 400 light-emitting diodes and driving small electronics. More than that, an automatic monitoring system powered by the EC-TENG can also monitor the water level in real-time and provide an alarm if necessary. 12Multiscale design of stiffening and ROS scavenging hydrogels for the augmentation of mandibular bone regeneration显示文摘Although biomimetic hydrogels play an essential role in guiding bone remodeling,reconstructing large bone defects is still a significant challenge since bioinspired gels often lack osteoconductive capacity,robust mechanical properties and suitable antioxidant ability for bone regeneration.To address these challenges,we first engineered molecular design of hydrogels(gelatin/polyethylene glycol diacrylate/2-(dimethylamino)ethyl methacrylate,GPEGD),where their mechanical properties were significantly enhanced via introducing trace amounts of additives(0.5 wt%).The novel hybrid hydrogels show high compressive strength(>700 kPa),stiff modulus(>170 kPa)and strong ROS-scavenging ability.Furthermore,to endow the GPEGD hydrogels excellent osteoinductions,novel biocompatible,antioxidant and BMP-2 loaded polydopamine/heparin nanoparticles(BPDAH)were developed for functionalization of the GPEGD gels(BPDAH-GPEGD).In vitro results indicate that the antioxidant BPDAH-GPEGD is able to deplete elevated ROS levels to protect cells viability against ROS damage.More importantly,the BPDAH-GPEGD hydrogels have good biocompatibility and promote the osteo differentiation of preosteoblasts and bone regenerations.At 4 and 8 weeks after implantation of the hydrogels in a mandibular bone defect,Micro-computed tomography and histology results show greater bone volume and enhancements in the quality and rate of bone regeneration in the BPDAH-GPEGD hydrogels.Thus,the multiscale design of stiffening and ROS scavenging hydrogels could serve as a promising material for bone regeneration applications.Yanlin Wu Xuan Li Yimin Sun Xiujun Tan Chenglin Wang Zhenming Wang Ling Ye 2023Bioactive Materials2023,,2:0
