A Chromosome-Level Genome Assembly of Garlic (Allium sativum) Provides Insights into Genome Evolution and Allicin Biosynthesis显示文摘Garlic,an economically important vegetable, spice, and medicinal crop, produces highly enlarged bulbs and unique organosulfur compounds. Here,we report a chromosome-level genome assembly for garlic, with a total size of approximately 16.24 Gb,as well as the annotation of 57561 predicted protein-coding genes, making garlic the first Allium species with a sequenced genome.Analysis of this garlic genome assembly reveals a recent burst of transposable elements, explaining the substantial expansion of the garlic genome. We examined the evolution of certain genes associated with the biosynthesis of allicin and inulin neoseries-type fructans, and provided new insights into the biosynthesis of these two compounds.Furthermore,a large-scale transcriptome was produced to characterize the expression patterns of garlic genes in different tissues and at various growth stages of enlarged bulbs. The reference genome and large-scale transcriptome data generated in this study provide valuable new resources for research on garlic biology and breeding. | Xiudong Sun Siyuan Zhu Ningyang Li Yi Cheng Jing Zhao Xuguang Qiao Li Lu Shiqi Liu Yanzhou Wang Chan Liu Benping Li Wu Guo Shuang Gao Zemao Yang Fu Li Zheng Zeng Qing Tang Yupeng Pan Mengjiao Guan Jian Zhao Xiaomi ng Lu Huanwe n Meng Zhenlin Han Chun she ng Gao Wenkai Jiang Xing Zhao Shilin Tian Jianguang Su Zhihui Cheng Touming Liu | 2020 | Molecular Plant2020,13,9: | 14
Mitochondrion-targeted PENTATRICOPEPTIDE REPEAT5 is required for cis-splicing of nad4 intron 3 and endosperm development in rice显示文摘Endosperm as the storage organ of starch and protein in cereal crops largely determines grain yield and quality.Despite the fact that several pentatricopeptide repeat(PPR)proteins required for endosperm development have been identified in rice, the molecular mechanisms of many P-type PPR proteins in endosperm development remains unclear. Here,we isolated a rice floury endosperm mutant ppr5 that developed small starch grains and an abnormal aleurone layer, accompanied by decreased starch, protein, and amylose contents.Map-based cloning combined with a complementation test demonstrated that PPR5 encodes a P-type PPR protein that is localized to the mitochondria. The mutation in PPR5 caused reduced splicing efficiency of mitochondrial NADH dehydrogenase 4(nad4)gene intron 3 and reduced complex I assembly and activity.Loss of PPR5 function greatly upregulated expression of alternative oxidases(AOXs),reduced ATP production, and affected mitochondrial morphology. We demonstrate that PPR5,as a P-type PPR protein,is required for mitochondrial function and endosperm development by controlling the cis-splicing of mitochondrial nad4 intron 3. | Long Zhang Yanzhou Qi Mingming Wu Lei Zhao Zhichao Zhao Cailin Lei Yuanyuan Hao Xiaowen Yu Yinglun Sun Xin Zhang Xiuping Guo Yulong Ren Jianmin Wan | 2021 | The Crop Journal2021,9,2: | 2

Facile discovery of red blood cell deformation and compromised membrane/skeleton assembly in Prader—Willi syndrome显示文摘Prader—Willi syndrome(PWS)is a rare congenital disease with genetic alterations in chromosome 15.Although genetic disorders and DNA methylation abnormalities involved in PWS have been investigated to a significant degree, other anomalies such as those in erythrocytes may occur and these have not been clearly elucidated. In the present study,we uncovered slight anemia in children with PWS that was associated with increased red blood cell(RBC)distribution width(RDW)and contrarily reduced hematocrit(HCT)values.Intriguingly, the increased ratio in RDW to HCT allowed sufficient differentiation between the PWS patients from the healthy controls and, importantly, with individuals exhibiting conventional obesity.Further morphologic examinations revealed a significant deformity in erythrocytes and mild hemolysis in PWS patients.Comprehensive mechanistic investigations unveiled compromised membrane skeletal assembly and membrane lipid composition, and revealed a reduced F-actin/G-actin ratio in PWS patients. We ascribed these phenotypic changes in erythrocytes to the observed genetic defects, including DNA methylation abnormalities. Our collective data allowed us to uncover RBC deformation in children with PWS,and this may constitute an auxiliary indicator of PWS in early childhood. | Yashuang Yang Guimei Li Yanzhou Wang Yan Sun Chao Xu Zhen Wei Shuping Zhang Ling Gao Sijin Liu Jiajun Zhao | 2022 | Frontiers of Medicine2022,16,6: | 0
Memory characteristics of microcavity dielectric barrier discharge显示文摘The nonlinear resistance characteristics of microcavity dielectric barrier discharge are mainly studied in the paper. A simulation model of microcavity dielectric barrier discharge is herein built to study the relationship between voltage and current in the process of discharge, and thus its I–V characteristic curve can be obtained. The I–V characteristics of the memristor are analyzed and compared with the I–V characteristics of the dielectric barrier discharge; it can be found that the I–V characteristics of the microcavity dielectric barrier discharge are similar to the characteristics of the memristor by analyzing them. The memory characteristics of microcavity dielectric barrier discharge are further analyzed. | Yanzhou Sun Xiaoqian Liu Dati Su Huibin Yang | 2018 | Journal of Semiconductors2018,39,11: | 0
Design and Implementation of a Battery Big Data Platform Through Intelligent Connected Electric Vehicles显示文摘The development of a battery management algorithm is highly dependent on high-quality battery operation data, especially the data in extreme conditions such as low temperatures. The data in faults are also essential for failure and safety management research. This study developed a battery big data platform to realize vehicle operation, energy interaction and data management.First,we developed an electric vehicle with vehicle navigation and position detection and designed an environmental cabin that allows the vehicle to operate autonomously.Second, charging and heating systems based on wireless energy transfer were developed and equipped on the vehicle to investigate optimal charging and heating methods of the batteries in the vehicle.Third, the data transmission network was designed,a real-time monitoring interface was developed, and the self-developed battery management system was used to measure, collect, upload, and store battery operation data in real time. Finally, experimental validation was performed on the platform. Results demonstrate the efficiency and reliability of the platform.Battery state of charge estimation is used as an example to illustrate the availability of battery operation data. | Rui Xiong Baoqiang Zhu Kui Zhang Yanzhou Duan Fengchun Sun | 2023 | Chinese Journal of Mechanical Engineering2023,36,2: | 0
