2Obtained transgenic wheat expressing pac1 mediated by Agrobacterium is resistant against Barley yellow dwarf virus-GPV显示文摘In fission yeast (Schizosaccharomyces pombe), pac1 gene was cloned with 99.3% nucleo- tide sequence similarity with published pac1 in GenBank. In pET-5α expression system, the expres- sion product of cloned pac1 in E. coli showed activity to degrade the double-strand RNA. Harboring the binary vector pBI121, which contains pac1 gene, Agrobacterium tumefaciens strain LBA4404 was used to transform the wheat immature embryos pre- cultured 7―10 d. After preregeneration, regeneration and selection culture stage, totally 41 G418 resistant plants were obtained, in which 25 lines were proved to integrate with transgene and express transgene normally by PCR, Dot blot, RT-PCR and ELISA de- tection. Antivirus test carried out on 25 positive lines with high dose of Barley yellow dwarf virus-GPV re- vealed that 12 lines had resistance to BVDV-GPV in low level, another 12 lines had resistance to BVDV- GPV in middle level, and 1 line showed resistance to BVDV-GPV in high level. 3Effects of Cu^(2+)and humic acids on degradation and fate of TBBPA in pure culture of Pseudomonas sp.strain CDT显示文摘Soil contamination with tetrabromobisphenol A(TBBPA) has caused great concerns;however, the presence of heavy metals and soil organic matter on the biodegradation of TBBPA is still unclear. We isolated Pseudomonas sp. strain CDT, a TBBPA-degrading bacterium, from activated sludge and incubated it with ^(14)C-labeled TBBPA for 87 days in the absence and presence of Cu^(2+)and humic acids(HA). TBBPA was degraded to organic-solvent extractable(59.4% ± 2.2%) and non-extractable(25.1% ± 1.3%) metabolites,mineralized to CO_2(4.8% ± 0.8%), and assimilated into cells(10.6% ± 0.9%) at the end of incubation. When Cu^(2+)was present, the transformation of extractable metabolites into non-extractable metabolites and mineralization were inhibited, possibly due to the toxicity of Cu^(2+)to cells. HA significantly inhibited both dissipation and mineralization of TBBPA and altered the fate of TBBPA in the culture by formation of HA-bound residues that amounted to 22.1% ± 3.7% of the transformed TBBPA. The inhibition from HA was attributed to adsorption of TBBPA and formation of bound residues with HA via reaction of reactive metabolites with HA molecules, which decreased bioavailability of TBBPA and metabolites in the culture. When Cu^(2+)and HA were both present, Cu^(2+)significantly promoted the HA inhibition on TBBPA dissipation but not on metabolite degradation. 4Early Cretaceous Palynological Assemblages of the Shuangliao Fault Depression, Songliao Basin and their Geological Implications显示文摘The pollen and spores found in the Early Cretaceous strata of two cores from the Shuangliao Fault Depression were studied systematically, and two palynological assemblages have been recognized. Assemblage Ⅰ from the Yingcheng Formation is named Paleoconiferus-Pinuspollenites; the assemblage is characterized by an extremely high content of Paleoconifer pollen. Assemblage Ⅱ from Member 1 of the Denglouku Formation is named Cicatricosisporites-Cyathidites-Pinuspollenites; the assemblage is characterized by the abundant presence of Laevigati spores, but less Paleoconifer pollen. According to the palynological data and zircon U-Pb dating, the geological age of the Yingcheng Formation is thought to be Aptian-Albian, and that of the Denglouku Formation is thought to be Early Albian. On the basis of the composition of each assemblage, we can infer that during the depositional period of the Yingcheng Formation, the paleovegetation was mainly conifer forest, the paleoclimate was consistent with the temperate climate of today, and the paleoenvironment was humid feature. During the depositional period of Member 1 of the Denglouku Formation, the paleovegetation was mainly conifer forest and shrubs, the paleoclimate was consistent with the subtropical climate of today, and the paleoenvironment was humid. 7Fecal-associated microbiome differences between traditional Chinese medicine qi-deficiency and balanced constitutions显示文摘Objective:To explore the structural and functional characteristics of the fecal-associated microbiome(FAM)in a traditional Chinese medicine(TCM)qi-deficiency constitution(QDC)by comparing with balanced constitution(BC)and screen the related biomarkers.Methods:In this cross-sectional study,the TCM constitutions of subjects were determined based on published the Classification and Determination of constitution in TCM and further confirmed by a TCM clinician.Clinical characteristics were recorded,and fecal samples were collected for 16S rDNA sequencing using the Illumina Miseq platform.The FAM structure was described using alpha-diversity indexes,beta-diversity indexes,and the relative abundances of the dominant taxa.Differences in the FAM distribution and function were analyzed with a Wilcoxon rank-sum test,MetagenomeSeq,and LEfSe analysis,after which a receiver operating characteristic curve based on the specific operational taxonomic units(OTUs)was constructed to calculate the area under the curve.Results:Our study population was composed of 22 BCs and 9 QDCs.There were no significant differences between the two groups in the distribution of clinical characteristics or alpha-diversity indexes,except for the sweets preference and blood glucose level.In principal coordinate analysis and partial least squares discriminant analysis,the bacterial communities in the BC group samples and QDC group samples clustered separately.Notably,there were 214 OTUs significantly distributed between groups in the MetagenomeSeq analysis,200 OTUs identified by the Wilcoxon rank-sum test,and 6 OTUs found by the LEfSe analysis.Predicted function analysis revealed that six metabolic pathways were distinctly distributed between the two groups.The area under the curve for the receiver operating characteristic curve based on the four specific OTUs was 0.88.Conclusion:Unique FAM structural and related functional characteristics are displayed in individuals with a QDC,and four specific OTUs could be used as QDC biomarkers to assist in clinical diagnosis.Jianhua Zhen Pengfei Zhao Guangrui Huang Yini Li Lu Zhao Yuhang Zhang Anlong Xu 2020Journal of Traditional Chinese Medical Sciences2020,7,2:1

9Quick Phase Identification for Dense Seismic Array with Aid from Long Term Phase Records of Co-located Sparse Permanent Stations显示文摘The phase identification and travel time picking are critical for seismic tomography,yet it will be challenging when the numbers of stations and earthquakes are huge.We here present a method to quickly obtain P and S travel times of pre-determined earthquakes from mobile dense array with the aid from long term phase records from co-located permanent stations.The records for 1768 M≥2.0 events from 2011 to 2013 recorded by 350 ChinArray stations deployed in Yunnan Province are processed with an improved AR-AIC method utilizing cumulative envelope and rectilinearity.The reference arrivals are predicted based on phase records from 88 permanent stations with similar spatial coverage,which are further refined with AR-AIC.Totally,718573 P picks and 512035 S picks are obtained from mobile stations,which are 28 and 22 times of those from permanent stations,respectively.By comparing the automatic picks with manual picks from 88 permanent stations,for M≥3.0 events,81.5%of the P-pick errors are smaller than 0.5 second and 70.5%of S-pick errors are smaller than1 second.For events with a lower magnitude,76.5%P-pick errors fall into 0.5 second and 69.5%S-pick errors are smaller than 1 second.Moreover,the Pn and Sn phases are easily discriminated from directly P/S,indicating the necessity of combining traditional auto picking and integrating machine learning method.CHEN Yini LI Jun WANG Zhenjie ZHAO Mengqi YU Tiehong LI Danning YU Junyi WANG Weitao 2020Earthquake Research in China2020,34,3:1
10Enhanced chemoselectivity of a plant cytochrome P450 through protein engineering of surface and catalytic residues显示文摘Cytochrome P450s(P450s)are the most versatile catalysts utilized by plants to produce structurally and functionally diverse metabolites.Given the high degree of gene redundancy and challenge to functionally characterize plant P450s,protein engineering is used as a complementarystrategy to study the mechanisms of P450-mediated reactions,or to alter their functions.We previously proposed an approach of engineering plant P450s based on combining high accuracy homology models generated by Rosetta combined with data-driven design using evoluti onary information of these enzymes.With this strategy,we repurposed a multi-functional P450(CYP87D20)into a monooxygenase after red esigning its active site.Since most plant P450s are membrane-anchored proteins that are adapted to the micro-environments of plant cells,expressing them in heterologous hosts usually results in problems of expression or activity.Here,we applied compu-tational design to tackle these issues by simultaneous optimization of the protein surface and active site.After screening 17 variants,effective su bstitutions of surface residues were observed to improve both expression and activity of CYP87D20.In addition,the identified substitutions were additive and by com-bining them a highly eficient C11 hydroxylase of cucurbitadienol was created to participate in the mogrol biosynthesis.This study shows the importance of considering the interplay between surface and active site residues for P450 engineering.Our integrated strategy also opens an avenue to create more tai loring enzymes with desired functions for the metabolic engineering of high-valued compounds like mogrol,the precursor of natural sweetener mogrosides.Xiaopeng Zhang Wei Luo Yinying Yao Xuming Luo Chao Han Yang Zhong Bo Zhang Dawei Li Lida Han Sanwen Huang Per Greisen Yi Shang 2021aBIOTECH2021,2,3:1
11Systematic screening reveals synergistic interactions that overcome MAPK inhibitor resistance in cancer cells显示文摘Objective:Effective adjuvant therapeutic strategies are urgently needed to overcome MAPK inhibitor(MAPKi)resistance,which is one of the most common forms of resistance that has emerged in many types of cancers.Here,we aimed to systematically identify the genetic interactions underlying MAPKi resistance,and to further investigate the mechanisms that produce the genetic interactions that generate synergistic MAPKi resistance.Methods:We conducted a comprehensive pair-wise sgRNA-based high-throughput screening assay to identify synergistic interactions that sensitized cancer cells to MAPKi,and validated 3 genetic combinations through competitive growth,cell viability,and spheroid formation assays.We next conducted Kaplan-Meier survival analysis based on The Cancer Genome Atlas database and conducted immunohistochemistry to determine the clinical relevance of these synergistic combinations.We also investigated the MAPKi resistance mechanisms of these validated synergistic combinations by using co-immunoprecipitation,Western blot,qRTPCR,and immunofluorescence assays.Results:We constructed a systematic interaction network of MAPKi resistance and identified 3 novel synergistic combinations that effectively targeted MAPKi resistance(ITGB3+IGF1R,ITGB3+JNK,and HDGF+LGR5).We next analyzed their clinical relevance and the mechanisms by which they sensitized cancer cells to MAPKi exposure.Specifically,we discovered a novel protein complex,HDGF-LGR5,that adaptively responded to MAPKi to enhance cancer cell stemness,which was up-or downregulated by the inhibitors of ITGB3+JNK or ITGB3+IGF1R.Conclusions:Pair-wise sgRNA library screening provided systematic insights into elucidating MAPKi resistance in cancer cells.ITGB3-+IGF1R-targeting drugs(cilengitide+linsitinib)could be used as an effective therapy for suppressing the adaptive formation of the HDGF-LGR5 protein complex,which enhanced cancer stemness during MAPKi stress.Yu Yu Minzhen Tao Libin Xu Lei Cao Baoyu Le Na An Jilin Dong Yajie Xu Baoxing Yang Wei Li Bing Liu Qiong Wu Yinying Lu Zhen Xie Xiaohua Lian 2022Cancer Biology & Medicine2022,19,2:1

13Degradation of FAK-targeting by proteolytic targeting chimera technology to inhibit the metastasis of hepatocellular carcinoma显示文摘Liver cancer is a prevalent malignant cancer,ranking third in terms of mortality rate.Metastasis and recurrence primarily contribute to the high mortality rate of liver cancer.Hepatocellular carcinoma(HCC)has low expression of focal adhesion kinase(FAK),which increases the risk of metastasis and recurrence.Nevertheless,the efficacy of FAK phosphorylation inhibitors is currently limited.Thus,investigating the mechanisms by which FAK affects HCC metastasis to develop targeted therapies for FAK may present a novel strategy to inhibit HCC metastasis.This study examined the correlation between FAK expression and the prognosis of HCC.Additionally,we explored the impact of FAK degradation on HCC metastasis through wound healing experiments,transwell invasion experiments,and a xenograft tumor model.The expression of proteins related to epithelial-mesenchymal transition(EMT)was measured to elucidate the underlying mechanisms.The results showed that FAK PROTAC can degrade FAK,inhibit the migration and invasion of HCC cells in vitro,and notably decrease the lung metastasis of HCC in vivo.Increased expression of E-cadherin and decreased expression of vimentin indicated that EMT was inhibited.Consequently,degradation of FAK through FAK PROTAC effectively suppressed liver cancer metastasis,holding significant clinical implications for treating liver cancer and developing innovative anti-neoplastic drugs.XINFENG ZHANG SHUANG LI MEIRU SONG YUE CHEN LIANGZHENG CHANG ZHERUI LIU HONGYUAN DAI YUTAO WANG GANGQI YANG YUN JIANG YINYING LU 2024Oncology Research2024,32,4:0

18Structure and function of the fecal-associated microbiome in qi stagnation constitution显示文摘Objective:To clarify the structural and functional characteristics of the gut microbiota in individuals with qi stagnation constitution(QSC)and identify the potential biomarkers related to QSC.Methods:This cross-sectional study involved individuals with QSC and balanced constitution(BC)confirmed by TCM clinicians.The clinical features were recorded,and fecal samples were collected for 16S rDNA sequencing.The structure of the fecal-associated microbiome(FAM)was described by the alpha-diversity indexes,beta-distances,and relative abundances of dominant taxa.The differences in FAM distribution were analyzed by the Wilcoxon rank-sum test,MetagenomeSeq,and LEfSe analysis.The 16S rDNA gene sequences were assigned to the KEGG dataset to predict the functional information of bacterial metabolic pathways by using PICRUSt.Differences in functional pathways between groups were assessed with the Wilcoxon rank-sum test.The ROC curve based on specific operational taxonomic units(OTUs)was constructed,and the AUC was calculated.Results:Twenty-two individuals with BC and 8 with QSC were recruited.Significant differences between the two groups were found in body mass index,health status,and low-density lipoprotein,etc.There was no significant difference in the alpha-diversity index.PCoA showed no evident clustering of bacterial communities according to constitutions.Bacteroidaceae,Lachnospiraceae,Ruminococcaceae,and Prevotellaceae were the four common bacteria with high abundances.Notably,MetagenomeSeq,LEfSe analysis,and the Wilcoxon rank-sum test identified significantly different distributions of 66,42,and 36 OTUs,respectively.Predictive function analysis showed that 13 metabolic pathways were significantly differentially distributed,including those related to fatty acid synthesis.Five specific OTUs were selected as potential biomarkers of a QSC,and the AUC was 0.94.Conclusion:Individuals with QSC have unique FAM structure and related functional characteristics.Five specific OTUs were identified to serve as potentially effective biomarkers related to QSC.Lu Zhao Pengfei Zhao Jianhua Zhen Guangrui Huang Yini Li Anlong Xu 2021Journal of Traditional Chinese Medical Sciences2021,8,2:0
19m^(6)A demethylase ALKBH5 is required for antibacterial innate defense by intrinsic motivation of neutrophil migration显示文摘Neutrophil migration into the site of infection is necessary for antibacterial innate defense,whereas impaired neutrophil migration may result in excessive inflammation and even sepsis.The neutrophil migration directed by extracellular signals such as chemokines has been extensively studied,yet the intrinsic mechanism for determining neutrophil ability to migrate needs further investigation.N6-methyladenosine(m^(6)A)RNA modification is important in immunity and inflammation,and our preliminary data indicate downregulation of RNA m^(6)A demethylase alkB homolog 5(ALKBH5)in neutrophils during bacterial infection.Whether m^(6)A modification and ALKBH5 might intrinsically modulate neutrophil innate response remain unknown.Here we report that ALKBH5 is required for antibacterial innate defense by enhancing intrinsic ability of neutrophil migration.We found that deficiency of ALKBH5 increased mortality of mice with polymicrobial sepsis induced by cecal ligation and puncture(CLP),and Alkbh5-deficient CLP mice exhibited higher bacterial burden and massive proinflammatory cytokine production in the peritoneal cavity and blood because of less neutrophil migration.Alkbh5-deficient neutrophils had lower CXCR2 expression,thus exhibiting impaired migration toward chemokine CXCL2.Mechanistically,ALKBH5-mediated m^(6)A demethylation empowered neutrophils with high migration capability through altering the RNA decay,consequently regulating protein expression of its targets,neutrophil migration-related molecules,including increased expression of neutrophil migration-promoting CXCR2 and NLRP12,but decreased expression of neutrophil migration-suppressive PTGER4,TNC,and WNK1.Our findings reveal a previously unknown role of ALKBH5 in imprinting migrationpromoting transcriptome signatures in neutrophils and intrinsically promoting neutrophil migration for antibacterial defense,highlighting the potential application of targeting neutrophil m^(6)A modification in controlling bacterial infections.Yang Liu Renjie Song Lu Zhao Zhike Lu Yini Li Xinyi Zhan Fengjiao Lu Jiang Yang Yamei Niu Xuetao Cao 2022Signal Transduction and Targeted Therapy2022,7,7:0
20Relationship of Microchannels and Plaque Erosion in Patients with ST-Segment Elevation Myocardial Infarction: An Optical Coherence Tomography Study显示文摘Objective:Microchannels are associated with the progression of atherosclerotic vulnerable plaques.However,in patients with culprit optical coherence tomography(OCT)-defined plaque erosion,the knowledge of microchannels and culprit lesion vulnerability is limited.The aim of this study was to investigate culprit lesion characteristics in patients with ST-segment elevated myocardial infarction(STEMI)caused by plaque erosion with and without microchannels using OCT.Methods:In all,348 STEMI patients with plaque erosion who underwent OCT of the culprit lesion at the 2nd Affiliated Hospital of Harbin Medical University(Harbin,China)from August 2014 to December 2017 were included and divided into the microchannel group(n=116,33.3%)and no-microchannel group(n=232,66.7%).The clinical characteristics and OCT-derived plaque features were compared between both groups.Results:Among the 348 STEMI patients with plaque erosion,culprit lesions with microchannels had higher incidence of lipid plaque(59.5%vs.45.3%,P=0.012);calcification(41.4%vs.24.6%,P=0.002);spotty calcification(30.2%vs.18.1%,P=0.014);macrophages accumulation(72.4%vs.45.7%,P<0.001);and cholesterol crystals(32.8%vs.14.2%,P<0.001)than those without microchannels.In addition,minimal lumen area was smaller((1.9±0.9)mm2 vs.(2.8±2.3)mm2,P<0.001)and lumen area stenosis was greater((71.3%±13.4%)vs.(65.3%±19.3%),P=0.001)in the microchannel group than in the no-microchannel group.Conclusion:In patients with STEMI caused by plaque erosion,one-third manifested typical microchannel characteristics,and those with microchannels were associated with more severe luminal stenosis and more vulnerable plaque features than those without microchannels.Senqing Jiang Junchen Guo Yanwei Yin Chao Fang Jifei Wang Yidan Wang Fangmeng Lei Sibo Sun Xueying Pei Ruyi Jia Shaotao Zhang Lulu Li Yini Wang Lei Xing Huai Yu Huimin Liu Maoen Xu Xuefeng Ren Lijia Ma Guo Wei Jingbo Hou Jiannan Dai Bo Yu 2022Cardiology Discovery2022,2,2:0
