| 1 | Responses of the Toll-like receptor and melanoma differentiation-associated protein 5 signaling pathways to avian infectious bronchitis virus infection in chicks显示文摘Avian infectious bronchitis virus(IBV) is a Gammacoronavirus in the family Coronaviridae and causes highly contagious respiratory disease in chickens. Innate immunity plays significant roles in host defense against IBV. Here, we explored the interaction between IBV and the host innate immune system. Severe histopathological lesions were observed in the tracheal mucosa at 3–5days post inoculation(dpi) and in the kidney at 8 dpi, with heavy viral loads at 1–11 and 1–28 dpi,respectively. The expression of m RNAs encoding Toll-like receptor(TLR) 3 and TLR7 were upregulated at 3–8 dpi, and that of TIR-domain-containing adapter-inducing interferon(IFN) β(TRIF) was upregulated at 21 dpi in the trachea and kidney. Myeloid differentiation primary response protein 88(My D88) was upregulated in the trachea during early infection. Tumor necrosis factor receptor-associated factor(TRAF) 3 and TRAF6 were upregulated expression in both tissues.Moreover, melanoma differentiation-associated protein 5(MDA5), laboratory of genetics and physiology 2(LGP2), stimulator of IFN genes(STING), and mitochondrial antiviral signaling protein(MAVS), as well as TANK binding kinase 1(TBK1), inhibitor of kappa B kinase(IKK) ?, IKKα, IKKβ,IFN regulatory factor(IRF) 7, nuclear factor of kappa B(NF-κB), IFN-α, IFN-β, various interleukins(ILs), and macrophage inflammatory protein-1β(MIP-1β) were significantly upregulated in the trachea and downregulated in the kidney. These results suggested that the TLR and MDA5 signaling pathways and innate immune cytokine were induced after IBV infection. | 2 | Triple oxygen isotope constraints on the origin of ocean island basalts显示文摘Understanding the origin of ocean island basalts(OIB) has important bearings on Earth's deep mantle.Although it is widely accepted that subducted oceanic crust, as a consequence of plate tectonics, contributes material to OIB's formation, its exact fraction in OIB's mantle source remains ambiguous largely due to uncertainties associated with existing geochemical proxies. Here we show, through theoretical calculation, that unlike many known proxies, triple oxygen isotope compositions(i.e.D^(17 )O) in olivine samples are not affected by crystallization and partial melting. This unique feature, therefore, allows olivine D^(17 )O values to identify subducted oceanic crusts in OIB's mantle source. Furthermore, the fractions of subducted ocean sediments and hydrothermally altered oceanic crust in OIB's mantle source can be quantified using their characteristic D^(17 )O values. | 4 | A two-dimensional double layer-averaged model of hyperconcentrated turbidity currents with non-Newtonian rheology显示文摘Hyperconcentrated turbidity currents typically display non-Newtonian characteristics that influence sediment transport and morphological evolution in alluvial rivers.However,hydro-sedimentmorphological processes involving hyperconcentrated turbidity currents are poorly understood,with little known about the effect of the non-Newtonian rheology.The current paper extends a recent twodimensional double layer-averaged model to incorporate non-Newtonian constitutive relations.The extended model is benchmarked against experimental and numerical data for cases including subaerial mud flow,subaqueous debris flow,and reservoir turbidity currents.The computational results agree well with observations for the subaerial mud flow and independent numerical simulations of subaqueous debris flow.Differences between the non-Newtonian and Newtonian model results become more pronounced in terms of propagation distance and sediment transport rate as sediment concentration increases.The model is then applied to turbidity currents in the Guxian Reservoir planned for middle Yellow River,China,which connects to a tributary featuring hyperconcentrated sediment-laden flow.The non-Newtonian model predicts slower propagation of turbidity currents and more significant bed aggradation at the confluence between the tributary Wuding River and the Yellow River in the reservoir than its Newtonian counterpart.This difference in model performance could be of considerable importance when optimizing reservoir operation schemes. | Yining Sun Ji Li Zhixian Cao Alistair George Liam Borthwick | 2023 | International Journal of Sediment Research2023,38,6: | 0 |
| 5 | Mathematical Analysis of the Jin-Neelin Model of El Niño-Southern-Oscillation显示文摘The Jin-Neelin model for the El Nio–Southern Oscillation(ENSO for short) is considered for which the authors establish existence and uniqueness of global solutions in time over an unbounded channel domain. The result is proved for initial data and forcing that are sufficiently small. The smallness conditions involve in particular key physical parameters of the model such as those that control the travel time of the equatorial waves and the strength of feedback due to vertical-shear currents and upwelling; central mechanisms in ENSO dynamics.From the mathematical view point, the system appears as the coupling of a linear shallow water system and a nonlinear heat equation. Because of the very different nature of the two components of the system, the authors find it convenient to prove the existence of solution by semi-discretization in time and utilization of a fractional step scheme. The main idea consists of handling the coupling between the oceanic and temperature components by dividing the time interval into small sub-intervals of length k and on each sub-interval to solve successively the oceanic component, using the temperature T calculated on the previous sub-interval, to then solve the sea-surface temperature(SST for short) equation on the current sub-interval. | 6 | Facile and controllable synthesis of amino-modified carbon dots for efficient oil displacement显示文摘Zero-dimensional(0D)carbon dots exhibit excellent potential as a new oil-displacing agent for unconventional reservoir development.However,the difficulty in size/surface properties control and unclear mechanism hinder their further applications.In this study,amino-modified carbon dots(am-CDs)for oil displacement were facilely synthesized through the rapid polymerization of D-glucose(D-Glc)and 3-aminopropyltriethoxysilane(APTES).The size of am-CDs could be precisely controlled by the reaction condition and quenching achieved by adjusting the pH value to neutral.The surface amine groups endow am-CDs with excellent hydrophilicity and dispersion stability.The 0.30 wt.%nanofluid based on am-CDs with an average size of 2.6±0.040 nm showed remarkable oil recovery efficiency(54.09%)without the addition of surfactant.The oil recovery efficiency of am-CDs is much higher than those of water flooding(30.25%),nano-SiO_(2) flooding(36.45%),and amino-free carbon dots(af-CDs)flooding(37.80%).Experimental and theoretical results reveal that am-CDs can be favorably adsorbed on the core surface to modulate the micro-scale wettability,changing the surface from oil-wet to relatively uniform water-wet.Meanwhile,am-CDs can effectively reduce the adhesion force between alkanes and sandstone surfaces,contributing to oil droplets peeling off and oil displacement.This study provides a new strategy for developing efficient carbon dots-based nanofluids for enhanced oil recovery. | Yining Wu Lisha Tang Mengjiao Cao Li Li Kai Liu Demin Kong Qingshan Zhao Xu Jin He Liu Caili Dai | 2023 | Nano Research2023,16,5: | 0 |
| 7 | LncRNA ZFAS1 regulates cardiomyocyte differentiation of human embryonic stem cells显示文摘Background:Cardiomyocytes derived from human embryonic stem cells(hESCs)are regulated by complex and stringent gene networks during differentiation.Long non-coding RNAs(lncRNAs)exert critical epigenetic regulatory functions in multiple differentiation processes.However,the involvement of lncRNAs in the differentiation of hESCs into cardiomyocytes has not yet been fully elucidated.Here,we identified the key roles of ZFAS1(lncRNA zinc finger antisense 1)in the differentiation of cardiomyocytes from hESCs.Methods:A model of cardiomyocyte differentiation from stem cells was established using the monolayer differentiation method,and the number of beating hESCs-derived cardiomyocytes was calculated.Gene expression was analyzed by quantitative real-time PCR(qRTPCR).Immunofluorescence assays were performed to assess the expression of cardiac troponin T(cTnT)andα-actinin protein in cardiomyocytes.Results:qRT-PCR showed that ZFAS1 expression in the mesoderm was significantly higher than that in embryonic stem cells,cardiac progenitor cells,and cardiomyocytes.Knockdown of ZFAS1 inhibited cardiomyocyte differentiation from hESCs,which was characterized by reduced expression of the cardiac-specific markers cTnT,α-actinin,myosin heavy chain 6(MYH6),and myosin heavy chain 7(MYH7).In contrast,ZFAS1 overexpression remarkably increased the percentage of spontaneously beating cardiomyocytes.In terms of the mechanism,we found that ZFAS1 is an antisense lncRNA at the 5′end of the protein-coding gene ZNFX1.Knockdown of ZFAS1 could increase the mRNA expression level of ZNFX1.Furthermore,qRT-PCR demonstrated that the silencing of ZNFX1 led to an increase in cardiac-specific markers that predicted the promotion of cardiomyocyte differentiation.Conclusion:Altogether,these data suggest that lncRNA-ZFAS1 is required for cardiac differentiation by functionally inhibiting the expression of ZNFX1,which may provide a reference for the treatment of heart disease to a certain extent. | YANG CAO YINING LIU YANG YU XIAOFEI GUO XIUXIU WANG WENYA MA HANJING LI ZHONGYU REN XINLU GAO SIJIA LI HAOYU JI HONGYANG CHEN HONG YAN YANAN TIAN XIN WANG BENZHI CAI | 2023 | BIOCELL2023,47,6: | 0 |
| 8 | In-situ synthesis of high thermal stability and salt resistance carbon dots for injection pressure reduction and enhanced oil recovery显示文摘Carbon dots(CDs)show great potential as a new type of oil-displacing agent for unconventional oil and gas development.However,the instability and easy aggregation epitomize the challenges that accompany the application of CDs in high temperature and high salinity(HT/HS)reservoirs.In this research,novel benzene sulfonate-modified carbon dots(BS-CDs)with remarkable thermal stability and salt resistance were fabricated through an in-situ electrochemical exfoliation method.Molecular simulation verifies that the introduction of benzene sulfonate groups substantially strengthens the electrostatic repulsion between BS-CDs,leading to outstanding dispersibility and stability even at a temperature of 100℃ and salinity of 14×10^(4) mg/L.Core flooding tests show that 0.05 wt.%BS-CDs nanofluid can significantly reduce the water injection pressure by 50.00%and enhanced oil recovery(EOR)to 68.39%under HT/HS conditions.According to the atomic force microscopy(AFM)scanning results,the adhesion force between the core(after BS-CDs treatment)and oil decreased by 11.94 times,indicating that the hydrophilicity of the core surface was increased.In addition,the distribution of the adhesion force curve is more concentrated,which means that the micro-scale wettability of the core changes from oil-wet to more homogeneous water-wet.This study provides a feasible way for the development and application of good thermal stability and salt resistance CDs in unconventional reservoir development. | Yining Wu Lisha Tang Dayu Liu Demin Kong Liu Kai Mengjiao Cao Qingshan Zhao | 2023 | Nano Research2023,16,10: | 0 |
| 9 | Defective g-C_(3)N_(4)supported Ru_(3)single-cluster catalyst for ammonia synthesis through parallel reaction pathways显示文摘Designing catalyst to achieve ammonia synthesis at mild conditions is a meaningful challenge in catalysis community.Defective g-C_(3)N_(4)nanosheet supported single-cluster ruthenium and iron catalysts were investigated for their ammonia synthesis performance.Based on density functional theory(DFT)calculations and microkinetic simulations,Ru_(3)single-cluster anchored on defective g-C3N4 nanosheet(Ru_(3)/Nv-g-C_(3)N_(4))has a turnover frequency(TOF)5.8 times higher than the Ru(0001)step surface at industrial reaction conditions of 673 K and 100 bar for ammonia synthesis.In other words,similar TOFs could be achieved on Ru_(3)/Nv-g-C_(3)N_(4)at much milder conditions(623 K,30 bar)than on Ru(0001)(673 K,100 bar).Our computations reveal the reaction proceeds parallelly on Ru_(3)/Nv-g-C_(3)N_(4)through both dissociative and alternative associative mechanisms at typical reaction conditions(600–700 K,10–100 bar);N–N bond cleavage of*N2 and*NNH from the two respective pathways controls the reaction collectively.With increasing temperatures or decreasing pressures,the dissociative mechanism gradually prevails and associative mechanism recedes.In comparison,Fe_(3)/Nv-g-C_(3)N_(4)catalyst shows a much lower catalytic activity than Ru3/Nv-g-C_(3)N_(4)by two orders of magnitude and the reaction occurs solely through the dissociative pathway.The finding provides a prospective candidate and deepens the mechanistic understanding for ammonia synthesis catalyzed by single-cluster catalysts(SCCs). | Yining Zhang Sha Li Chao Sun Xiaorou Cao Xi Wang Jiannian Yao | 2023 | Nano Research2023,16,2: | 0 |

| 11 | Genetic and histological relationship between pheromone-secreting tissues of the musk gland and skin of juvenile Chinese forest musk deer(Moschus berezovskii Flerov,1929)显示文摘Background The musk glands of adult male Chinese forest musk deer(Moschus berezovskii Flerov,1929)(FMD),which are considered as special skin glands,secrete a mixture of sebum,lipids,and proteins into the musk pod.Together,these components form musk,which plays an important role in attracting females during the breeding season.However,the relationship between the musk glands and skin of Chinese FMD remains undiscovered.Here,the musk gland and skin of Chinese FMD were examined using histological analysis and RNA sequencing(RNA-seq),and the expression of key regulatory genes was evaluated to determine whether the musk gland is derived from the skin.Methods A comparative analysis of musk gland anatomy between juvenile and adult Chinese FMD was conducted.Then,based on the anatomical structure of the musk gland,skin tissues from the abdomen and back as well as musk gland tissues were obtained from three juvenile FMD.These tissues were used for RNA-seq,hematoxylin-eosin(HE)staining,immunohistochemistry(IHC),western blot(WB),and quantitative real-time polymerase chain reaction(qRT-PCR)experiments.Results Anatomical analysis showed that only adult male FMD had a complete glandular organ and musk pod,while juvenile FMD did not have any well-developed musk pods.Transcriptomic data revealed that 88.24%of genes were co-expressed in the skin and musk gland tissues.Kyoto Encyclopedia of Genes and Genomes(KEGG)signaling pathway analysis found that the genes co-expressed in the abdomen skin,back skin,and musk gland were enriched in biological development,endocrine system,lipid metabolism,and other pathways.Gene Ontology(GO)enrichment analysis indicated that the genes expressed in these tissues were enriched in biological processes such as multicellular development and cell division.Moreover,the Metascape predictive analysis tool demonstrated that genes expressed in musk glands were skin tissue-specific.qRT-PCR and WB revealed that sex-determining region Y-box protein 9(Sox9),Caveolin-1(Cav-1),and androgen receptor(AR)were expressed in all three tissues,although the expression levels differed among the tissues.According to the IHC results,Sox9 and AR were expressed in the nuclei of sebaceous gland,hair follicle,and musk gland cells,whereas Cav-1 was expressed in the cell membrane.Conclusions The musk gland of Chinese FMD may be a derivative of skin tissue,and Sox9,Cav-1,and AR may play significant roles in musk gland development. | Long LI Heran CAO Jinmeng YANG Tianqi JIN Yuxuan MA Yang WANG Zhenpeng LI Yining CHEN Huihui GAO Chao ZHU Tianhao YANG Yalong DENG Fangxia YANG Wuzi DONG | 2023 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,9: | 0 |
