1Cropland redistribution to marginal lands undermines environmental sustainability显示文摘Cropland redistribution to marginal land has been reported worldwide;however,the resulting impacts on environmental sustainability have not been investigated sufficiently.Here we investigated the environmental impacts of cropland redistribution in China.As a result of urbanization-induced loss of high-quality croplands in south China(~8.5 t ha^(-1)),croplands expanded to marginal lands in northeast(~4.5 t ha^(-1)) and northwest China(~2.9 t ha^(-1)) during 1990-2015 to pursue food security.However,the reclamation in these low-yield and ecologically vulnerable zones considerably undermined local environmental sustainability,for example increasing wind erosion(+3.47%),irrigation water consumption(+34.42%),fertilizer use(+20.02%) and decreasing natural habitats(-3.11%).Forecasts show that further reclamation in marginal lands per current policies would exacerbate environmental costs by 2050.The future cropland security risk will be remarkably intensified because of the conflict between food production and environmental sustainability.Our research suggests that globally emerging reclamation of marginal lands should be restricted and crop yield boost should be encouraged for both food security and environmental benefits.Wenhui Kuang Jiyuan Liu Hanqin Tian Hao Shi Jinwei Dong Changqing Song Xiaoyong Li Guoming Du Yali Hou Dengsheng Lu Wenfeng Chi Tao Pan Shuwen Zhang Rafiq Hamdi Zherui Yin Huimin Yan Changzhen Yan Shixin Wu Rendong Li Jiuchun Yang Yinyin Dou Wenbin Wu Liqiao Liang Bao Xiang Shiqi Yang 2022National Science Review2022,9,1:13
2Distinct immune signatures discriminate between asymptomatic and presymptomatic SARS-CoV-2^(pos)subjects显示文摘Increasing numbers of SARS-CoV-2-positive(SARS-CoV-2^(pos))subjects are detected at silent SARS-CoV-2 infection stage(SSIS).Yet,SSIS represents a poorly examined time-window wherein unknown immunity patterns may contribute to the fate determination towards persistently asymptomatic or overt disease.Here,we retrieved blood samples from 19 asymptomatic and 12 presymptomatic SARS-COV-2^(pos)subjects,47 age/gender-matched patients with mild or moderate COVID-19 and 27 normal subjects,and interrogated them with combined assays of 44-plex CyTOF,RNA-seq and Olink.Notably,both asymptomatic and presymptomatic subjects exhibited numerous readily detectable immunological alterations,while certain parameters including more severely decreased frequencies of CD107a^(low)classical monocytes,intermediate monocytes,non-classical monocytes and CD62L^(hi)CD8^(+)Tnaive cells,reduced plasma STC1 level but an increased frequency of CD4^(+)NKT cells combined to distinguish the latter.Intercorrelation analyses revealed a particular presymptomatic immunotype mainly manifesting as monocytic overactivation and differentiation blockage,a likely lymphocyte exhaustion and immunosuppression,yielding mechanistic insights into SSIS fate determination,which could potentially improve 5ARS-CoV-2 management.Shanhe Yu Caixia Di Shijun Chen Mingquan Guo Jiayang Yan Zhaoqin Zhu Li Liu Ruixue Feng Yinyin Xie Ruihong Zhang Juan Chen Mengxi Wang Dong Wei Hai Fang Tong Yin Jinyan Huang Saijuan Chen Hongzhou Lu Jiang Zhu Jieming Qu 2021Cell Research2021,31,11:4
3Internal Loads and Bioavailability of Phosphorus and Nitrogen in Dianchi Lake,China显示文摘Sediments have a significant influence on the cycling of nutrient elements in lake environments. In order to assess the distribution characteristics and estimate the bioavailability of phosphorus and nitrogen in Dianchi Lake, organic and inorganic phosphorus and nitrogen forms were analysed. The 210 Pb radiometric dating method was employed to study temporal changes in the phosphorus and nitrogen pools in Dianchi Lake. The result show that the total phosphorus(TP) and total nitrogen(TN) were both at high concentrations, ranging from 697.5–3210.0 mg/kg and 1263.7–7155.2 mg/kg, respectively. Inorganic phosphorus(IP) and total organic nitrogen(TON) were the main constituents, at percentages of 59%–78% and 74%–95%, respectively, in the sediments. Spatially, there was a decreasing trend in phosphorus and nitrogen contents from the south and north to the lake centre, which is related to the distribution pattern of local economic production. The burial rates of the various phosphorus and nitrogen forms increased in same spatially and over time. Particularly in the past two decades, the burial rates doubled, with that TN reached to 1.287 mg/(cm^2·yr) in 2014. As the most reactive forms, nitrate nitrogen(NO_3-N) and ammonia nitrogen(NH_4-N) were buried more rapidly in the south region, implying that the potential for releasing sedimentary nitrogen increased from north to south. Based on their concentrations and burial rates, the internal loads of phosphorus and nitrogen were analysed for the last century. A TP pool of 71597.6 t and a TN pool of 81191.7 t were estimated for Dianchi Lake. 5A Global Analysis of the Relationship Between Urbanization and Fatalities in Earthquake-Prone Areas显示文摘Urbanization can be a challenge and an opportunity for earthquake risk mitigation.However,little is known about the changes in exposure(for example,population and urban land)to earthquakes in the context of global urbanization,and their impacts on fatalities in earthquake-prone areas.We present a global analysis of the changes in population size and urban land area in earthquake-prone areas from 1990 to 2015,and their impacts on earthquake-related fatalities.We found that more than two thirds of population growth(or 70%of total population in 2015)and nearly three quarters of earthquake-related deaths(or 307,918 deaths)in global earthquake-prone areas occurred in developing countries with an urbanization ratio(percentage of urban population to total population)between 20 and 60%.Holding other factors constant,population size was significantly and positively associated with earthquake fatalities,while the area of urban land was negatively related.The results suggest that fatalities increase for areas where the urbanization ratio is low,but after a ratio between 40 and 50%occurs,earthquake fatalities decline.This finding suggests that the resistance of building and infrastructure is greater in countries with higher urbanization ratios and highlights the need for further investigation.Our quantitative analysis is extended into the future using Shared Socioeconomic Pathways to reveal that by 2050,more than 50%of the population increase in global earthquake-prone areas will take place in a few developing countries(Pakistan,India,Afghanistan,and Bangladesh)that are particularly vulnerable to earthquakes.To reduce earthquake-induced fatalities,enhanced resilience of buildings and urban infrastructure generally in these few countries should be a priority.Chunyang He Qingxu Huang Xuemei Bai Derek TRobinson Peijun Shi Yinyin Dou Bo Zhao Jubo Yan Qiang Zhang Fangjin Xu James Daniell 2021International Journal of Disaster Risk Science2021,12,6:1
6Regulation of P450 TleB catalytic flow for the synthesis of sulfur-containing indole alkaloids by substrate structure-directed strategy and protein engineering显示文摘Indole alkaloids have attracted considerable attention from synthetic chemists and biochemists for their structural diversity and important biological activities.Compared with traditional organic synthesis methods,the strategy of using cytochrome P450s'extraordinary abilities to selectively activate carbon-hydrogen bonds to assist in the synthesis of various indole alkaloids has the characteristics of short synthetic route,mild conditions and high atomic economy.Here,we utilized P450 monooxygenases HinD and TleB to synthesize a novel 6/5/8 tricyclic product from(S)-N-((S)-1-(4-fluoro-1H-indol-3-yl)-3-hydroxypropan-2-yl)-2-mercapto-3-methylbutanamide through the substrate structure-directed strategy.TleB was more effective in catalyzing C–S coupling,and was used to synthesize a series of 6/5/8 tricyclic indololactam derivatives to provide drug candidates.Interestingly,the S–S coupling product was observed in HinD catalysis,which was a minor product in the wild-type TleB catalysis.With the help of protein engineering,we accurately regulated the catalytic flow and reversed the selectivity of TleB to obtain the S–S coupling product.At the same time,the reaction mechanism was reasonably speculated by means of site blocking and protein-substrate complex analysis.Xinying Ge Yan Long Jun Wang Bo Gu Zixuan Yang Yinyin Feng Shuo Zheng Yingying Li Wupeng Yan Heng Song 2023Science China Chemistry2023,66,11:0
7Intra-heterogeneity in transcription and chemoresistant property of leukemia-initiating cells in murine Setd2^(-/-) acutemyeloid leukemia显示文摘Background:Heterogeneity of leukemia-initiating cells(LICs)is a major obstacle in acute myeloid leukemia(AML)therapy.Accumulated evidence indicates that the coexistence of multiple types of LICs with different pathogenicity in the same individual is a common feature in AML.However,the functional heterogeneity including the drug response of coexistent LICs remains unclear.Therefore,this study aimed to clarify the intra-heterogeneity in LICs that can help predict leukemia behavior and develop more effective treatments.Methods:Spleen cells from the primary Setd2^(-/-)-AML mouse were transplanted into C57BL/6 recipient mice to generate a transplantable model.Flow cytometry was used to analyze the immunophenotype of the leukemic mice.Whole-genome sequencing was conducted to detect secondary hits responsible for leukemia transformation.A serial transplantation assay was used to determine the self-renewal potential of Setd2^(-/-)-AML cells.A limiting-dilution assay was performed to identify the LIC frequency in different subsets of leukemia cells.Bulk and single-cell RNA sequencing were performed to analyze the transcriptional heterogeneity of LICs.Small molecular inhibitor screening and in vivo drug treatment were employed to clarify the difference in drug response between the different subsets of LICs.Results:In this study,we observed an aged Setd2^(-/-)mouse developing AML with co-mutation of Nras^(G12S) and Braf^(K520E).Further investigation identified two types of LICs residing in the c-Kit^(+)B220^(+)Mac-1^(-)and c-Kit^(+)B220^(+)Mac-1^(+)subsets,respectively.In vivo transplantation assay disclosed the heterogeneity in differentiation between the coexistent LICs.Besides,an intrinsic doxorubicinresistant transcriptional signature was uncovered in c-Kit^(+)B220^(+)Mac-1^(+)cells.Indeed,doxorubicin plus cytarabine(DA),the standard chemotherapeutic regimen used in AML treatment,could specifically kill c-Kit^(+)B220^(+)Mac-1^(−)cells,but it hardly affected c-Kit^(+)B220^(+)Mac-1^(+)cells.Transcriptome analysis unveiled a higher activation of RAS downstream signaling pathways in c-Kit^(+)B220^(+)Mac-1^(+)cells than in c-Kit^(+)B220^(+)Mac-1^(-)cells.Combined treatmentwithDAand RAS pathway inhibitors killed both c-Kit^(+)B220^(+)Mac-1^(−)and c-Kit^(+)B220^(+)Mac-1^(+)cells and attenuated disease progression.Conclusions:This study identified two cell subsets enriched for LICs inmurine Setd2^(-/-)-AML and disclosed the transcriptional and functional heterogeneity of LICs,revealing that the coexistence of different types of LICs in thismodel brings about diverse drug response.Jiachun Song Longting Du Ping Liu Fuhui Wang Bo Zhang Yinyin Xie Jing Lu Yi Jin Yan Zhou Gang Lv Jianmin Zhang Saijuan Chen Zhu Chen Xiaojian Sun Yuanliang Zhang Qiuhua Huang 2021Cancer Communications2021,41,9:0
8Fabrication of NiCoP decorated TiO_(2)/polypyrrole nanocomposites for the effective photocatalytic degradation of tetracycline显示文摘Due to the massive discharge of antibiotics in water,it is an urgent matter to remove antibiotics from waste water.The photocatalysts with high stability and activity have attracted extensive attention from researchers.By an in-situ polymerization method,polypyrrole(PPy)was modified on the surface of TiO_(2)(named as TiO_(2)/PPy).By one-step reduction method,NiCoP was grafted on the surface of TiO_(2)/PPy(named as TiO_(2)/PPy/NiCoP)to synthesize the photocatalyst of TiO_(2)/PPy/NiCoP for degradation of tetra-cycline(TC)antibiotic.The characterization results revealed that NiCoP was deposited on the surface of TiO_(2)/PPy successfully.The photocatalytic experiment results illustrated that 83.2%of TC could be de-graded at natural pH with 20 mg of TiO_(2)/PPy/NiCoP in 50 mL of TC solution(10 mg/L)under visible light irradiation.The high catalytic activity is attributed to the attachment of NiCoP on the surface of TiO_(2)/PPy which can enlarge the light response range of TiO_(2)effectively.Scavenger studies revealed that the degra-dation of TC was dominated by•O_(2)−and h+.The photodegradation efficiency of TC with TiO_(2)/PPy/NiCoP still reached over 74%after 5 consecutive cycles,indicating the potential applications in practical wastew-ater.Yalu Wu Yinyin Xu Yan Zhang Jingbo Feng Yuanyuan Li Jiaying Lan Xiuwen Cheng 2022Chinese Chemical Letters2022,33,5:0
9Visualization of the Oncolytic Alphavirus M1 Life Cycle in Cancer Cells显示文摘Oncolytic alphavirus M1 has been shown to selectively target and kill cancer cells,but cytopathic morphologies induced by M1 virus and the life cycle of the M1 strain in cancer cells remain unclear.Here,we study the key stages of M1 virus infection and replication in the M1 virus-sensitive HepG2 liver cancer cell line by transmission electron microscopy,specifically examining viral entry,assembly,maturation and release.We found that M1 virus induces vacuolization of cancer cells during infection and ultimately nuclear marginalization,a typical indicator of apoptosis.Specifically,our results suggest that the endoplasmic reticulum participates in the assembly of nucleocapsids.In the early and late stage of infection,three kinds of special cytopathic vacuoles are formed and appear to be involved in the replication,maturation and release of the virus.Taken together,our data displayed the process of M1 virus infection of tumor cells and provide the structural basis for the study of M1 virus-host interactions.Jia Dan Lin Nie Xudong Jia Cuiying Xu Jing Cai Yuan Lin Jun Hu Wenbo Zhu Yinyin Li Dong Chen Ying Liu Cheng Hu Guangmei Yan Jiankai Liang Qinfen Zhang 2021Virologica Sinica2021,36,4:0
