2Anlotinib has good efficacy and low toxicity: a phase Ⅱ study of anlotinib in pre-treated HER-2 negative metastatic breast cancer显示文摘Objective:Anlotinib is a novel tyrosine kinase inhibitor blocking angiogenesis.This study was performed to assess the efficacy and safety of anlotinib in patients with metastatic breast cancer.Methods:Patients with HER2-negative breast cancer,who were pre-treated with anthracycline or taxanes in a neoadjuvant,adjuvant,or metastatic setting,and had treatment failure after at least one prior chemotherapy regimen in the metastatic setting were enrolled.Anlotinib was administered at 12 mg daily for 14 days in a 21-day cycle until disease progression or unacceptable toxicity occurred.Simultaneously,5–10 m L of venous blood was collected to perform circulating tumor DNA(ct DNA)testing every 2 treatment cycles.The primary endpoint was the objective response rate(ORR).Secondary endpoints included the disease control rate(DCR),progression-free survival(PFS),overall survival,safety,and biomarkers.Results:Twenty-six eligible patients were enrolled,with a median age of 56(30–75)years.The median follow-up time was 10.5 months.The ORR was 15.4%,the DCR was 80.8%,and the median PFS was 5.22 months(95%confidence interval 2.86–6.24).Fourteen(53.8%)patients survived for more than 10 months.The changes in the detectable ct DNA variant allele frequency were consistent with the tumor response.The most common treatment-related adverse events were hypertension(57.7%),thyroidstimulating hormone elevation(34.6%),and hand-foot syndrome(23.1%).Conclusion:Anlotinib showed objective efficacy with tolerable toxicity in heavily pre-treated,metastatic HER2-negative breast cancer.The dynamic changes in the ct DNA variant allele fraction may be predictive of the tumor response.Nanlin Hu Yiran Si Jian Yue Tingting Sun Xue Wang Zhuqing Jia Songlin Gao Qiao Li Yang Shao Jiayu Wang Yang Luo Fei Ma Binghe Xu Peng Yuan 2021Cancer Biology & Medicine2021,18,3:11
3Preparation of cationic polyacrylamide microsphere emulsion and its performance for permeability reduction显示文摘In this paper, cationic polyacrylamide microspheres(CPAM) were synthesized using acrylamide(AM) and methacryloyloxyethyl trimethyl ammonium chloride(TMAEMC) as monomers, ammonium sulfate as dispersant, poly(acryloyloxyethyl trimethyl ammonium chloride)(PAETAC) as dispersion stabilizer, and ammonium persulfate as initiator. The synthetic method was dispersion polymerization. The effects of monomer ratio(AM/TMAEMC), dispersant concentration, and dispersion stabilizer dosage on dispersion polymerization were systematically studied to determine the optimal preparation conditions. The structure and viscosity of the synthesized polymer were characterized by FTIR and capillary viscometry, respectively, and the particle sizes and distribution of the polymer microspheres were characterized by microscopy and dynamic light scattering, respectively. Finally, flow tests were conducted to measure the permeability reduction performance of the microspheres at various concentrations in sand packs with different permeability. 4Role of Microphysical Parameterizations with Droplet Relative Dispersion in IAP AGCM 4.1显示文摘Previous studies have shown that accurate descriptions of the cloud droplet effective radius(Re) and the autoconversion process of cloud droplets to raindrops(Ar) can effectively improve simulated clouds and surface precipitation, and reduce the uncertainty of aerosol indirect effects in GCMs. In this paper, we implement cloud microphysical schemes including two-moment Ar and Re considering relative dispersion of the cloud droplet size distribution into version 4.1 of the Institute of Atmospheric Physics' s atmospheric GCM(IAP AGCM 4.1), which is the atmospheric component of the Chinese Academy of Sciences' Earth System Model. Analysis of the effects of different schemes shows that the newly implemented schemes can improve both the simulated shortwave and longwave cloud radiative forcings, as compared to the standard scheme, in IAP AGCM 4.1. The new schemes also effectively enhance the large-scale precipitation, especially over low latitudes, although the influences of total precipitation are insignificant for different schemes. 6Increased mitochondrial fission drives the reprogramming of fatty acid metabolism in hepatocellular carcinoma cells through suppression of Sirtuin 1显示文摘Background:Mitochondria are dynamic organelles that constantly change their morphology through fission and fusion processes.Recently,abnormally increased mitochondrial fission has been observed in several types of can-cer.However,the functional roles of increased mitochondrial fission in lipid metabolism reprogramming in cancer cells remain unclear.This study aimed to explore the role of increased mitochondrial fission in lipid metabolism in hepa-tocellular carcinoma(HCC)cells.Methods:Lipid metabolism was determined by evaluating the changes in the expressions of core lipid metabolic enzymes and intracellular lipid content.The rate of fatty acid oxidation was evaluated by[PH]-labelled oleic acid.The mito-chondrial morphology in HCC cells was evaluated by fluorescent staining.The expression of protein was determined by real-time PCR,imnmunohistochemistry and Western blotting.Results:Activation of mitochondrial fission significantly promoted de novo fatty acid synthesis in HCC cells through upregulating the expression of lipogenic genes fatty acid synthase(FASN),acetyl-CoA carboxylasel(ACCI),and elonga-tion of very long chain fatty acid protein 6(ELOVL6),while suppressed fatty acid oxidation by downregulating carnitine palmitoyl transferase 1A(CPTIA)and acyl-CoA oxidase 1(ACOX1).Consistently,suppressed mitochondrial fission exhibited the opposite effects.Moreover,in vitro and in vivo studies revealed that mitochondrial fission-induced lipid metabolism reprogramming significantly promoted the proliferation and metastasis of HCC cells.Mechanistically,mito-chondrial fission increased the acetylation level of sterol regulatory element-binding protein 1(SREBPI)and peroxisome proliferator-activated receptor coaC-tivator 1 alpha(PGC-1a)by suppressing nicotinamide adenine dinucleotide(NAD+)/Sirtuin 1(SIRTI)signaling.The elevated SREBP1 then upregulated the expression of FASN,ACC1 and ELOVL6 in HCC cells,while PGC-1c/PPARa sup-pressed the expression of CPTIA and ACOXL Conclusions:Increased mitochondrial fission plays a crucial role in the repro-gramming of lipid metabolism in HCC cells,which provides strong evidence for the use of this process as a drug target in the treatment of this malignancy.Dan Wu Yi Yang Yiran Hou Zifeng Zhao Ning Liang Peng Yuan Tao Yang Jinliang Xing Jibin Li 2022Cancer Communications2022,42,1:4
7Building a model-based personalised recommendation approach for tourist attractions from geotagged social media data显示文摘When travelling,people are accustomed to taking and uploading photos on social media websites,which has led to the accumulation of huge numbers of geotagged photos.Combined with multisource information(e.g.weather,transportation,or textual information),these geotagged photos could help us in constructing user preference profiles at a high level of detail.Therefore,using these geotagged photos,we built a personalised recommendation system to provide attraction recommendations that match a user’s preferences.Specifically,we retrieved a geotagged photo collection from the public API for Flickr(Flickr.com)and fetched a large amount of other contextual information to rebuild a user’s travel history.We then created a model-based recommendation method with a two-stage architecture that consists of candidate generation(the matching process)and candidate ranking.In the matching process,we used a support vector machine model that was modified for multiclass classification to generate the candidate list.In addition,we used a gradient boosting regression tree to score each candidate and rerank the list.Finally,we evaluated our recommendation results with respect to accuracy and ranking ability.Compared with widely used memory-based methods,our proposed method performs significantly better in the cold-start situation and when mining‘long-tail’data.Xiaoyu Sun Zhou Huang Xia Peng Yiran Chen Yu Liu 2019International Journal of Digital Earth2019,12,6:4

9Boosting sulfur redox kinetics by a pentacenetetrone redox mediator for high-energy-density lithium-sulfur batteries显示文摘Lithium-sulfur(Li-S)battery is considered as a promising energy storage system due to its ultrahigh theoretical energy density of 2,600 Wh·kg^(−1).Redox mediation strategies have been proposed to promote the sluggish sulfur redox kinetics.Nevertheless,the applicability of redox mediators in practical high-energy-density Li-S batteries has seldomly been manifested.In this work,5,7,12,14-pentacenetetrone(PT)is proposed as an effective redox mediator to promote the sulfur redox kinetics under practical working conditions.A high initial specific discharge capacity of 993 mAh·g^(−1) is achieved at 0.1 C with high-sulfur-loading cathodes of 4.0 mgS·cm^(−2)and low electrolyte/sulfur(E/S)ratio of 5μL·mg_(S)^(−1).More importantly,practical Li-S pouch cells with the PT mediator realize an actual initial energy density of 344 Wh·kg^(−1)and cycle stably for 20 cycles wih a high capacity retention of 88%.This work proposes an effective redox mediator and further verifies the redox mediation strategy for practical high-energydensity Li-S batteries.Yan-Qi Peng Meng Zhao Zi-Xian Chen Qian Cheng Yiran Liu Xi-Yao Li Yun-Wei Song Bo-Quan Li Jia-Qi Huang 2023Nano Research2023,16,6:2
10News recommendation based on time factor and word embedding显示文摘Existing algorithms of news recommendations lack in depth analysis of news texts and timeliness. To address these issues, an algorithm for news recommendations based on time factor and word embedding(TFWE) was proposed to improve the interpretability and precision of news recommendations. First, TFWE used term frequency-inverse document frequency(TF-IDF) to extract news feature words and used the bidirectional encoder representations from transformers(BERT) pre-training model to convert the feature words into vector representations. By calculating the distance between the vectors, TFWE analyzed the semantic similarity to construct a user interest model. Second, considering the timeliness of news, a method of calculating news popularity by integrating time factors into the similarity calculation was proposed. Finally, TFWE combined the similarity of news content with the similarity of collaborative filtering(CF) and recommended some news with higher rankings to users. 11Carrier and microstructure tuning for improving the thermoelectric properties of Ag_(8)SnSe_(6)via introducing SnBr_(2)显示文摘The argyrodite compounds(2 A(12)/B X6 m n n^(m+)+--(Am+=Li^(+),Cu^(+),and Ag^(+);Bn^(+)=Ga^(3+),Si^(4+),Ge^(4+),Sn^(4+),P^(5+),and As^(5+);and X^(2−)=S^(2−),Se^(2−),or Te^(2−)))have attracted great attention as excellent thermoelectric(TE)materials due to their extremely low lattice thermal conductivity(κl).Among them,Ag_(8)SnSe_(6)-based TE materials have high potential for TE applications.However,the pristine Ag_(8)SnSe_(6)materials have low carrier concentration(<1017 cm^(−3)),resulting in low power factors.In this study,a hydrothermal method was used to synthesize Ag_(8)SnSe_(6)with high purity,and the introduction of SnBr_(2)into the pristine Ag_(8)SnSe_(6)powders has been used to simultaneously increase the power factor and decrease the thermal conductivity(κ).On the one hand,a portion of the Br−ions acted as electrons to increase the carrier concentration,increasing the power factor to a value of~698 mW·m^(−1)·K^(−2)at 736 K.On the other hand,some of the dislocations and nanoprecipitates(SnBr_(2))were generated,resulting in a decrease ofκl(~0.13 W·m^(−1)·K^(−1))at 578 K.As a result,the zT value reaches~1.42 at 735 K for the sample Ag8Sn1.03Se5.94Br0.06,nearly 30%enhancement in contrast with that of the pristine sample(~1.09).The strategy of synergistic manipulation of carrier concentration and microstructure by introducing halogen compounds could be applied to the argyrodite compounds to improve the TE properties.Zhonghai YU Xiuxia WANG Chengyan LIU Yiran CHENG Zhongwei ZHANG Ruifan SI Xiaobo BAI Xiaokai HU Jie GAO Ying PENG Lei MIAO 2022Journal of Advanced Ceramics2022,11,7:1

13SARS-CoV-2 Surveillance Through China Influenza Surveillance Information System—China,December 1,2022 to February 12,2023显示文摘Introduction:The World Health Organization(WHO)proposed using influenza surveillance systems to carry out coronavirus disease 2019(COVID-19)surveillance due to the similarity between the two diseases in some respiratory symptoms.To assess the prevalence of COVID-19,we analyzed the influenzalike illness(ILI)and positive rate of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)detections in ILI patients reported to the influenza Surveillance Information System(CNISIS)since late 2022.Methods:Data related to ILI were reported by national surveillance sentinel hospitals.Positive testing for SARS-CoV-2 and influenza viruses was conducted using real-time reverse transcription polymerase chain reaction(rRT-PCR)detection by the national influenza surveillance network laboratories.Surveillance data were reported to CNISIS.Results:Beginning on December 12,2022(Week 50),the ILI percentage increased dramatically,peaking in Week 51 at 12.1%.Subsequently,the ILI percentage began to decline rapidly from Week 52,2022,and by Week 6,2023(February 6-12),the ILI and ILI percentage had returned to the levels observed at the beginning of December 2022.From December 1,2022 to February 12,2023,115,844 specimens were tested for both SARS-CoV-2 and influenza virus.Of these,30,381(26.2%)were positive for SARSCoV-2 and 1,763(1.5%)were positive for influenza virus.The positive rate of SARS-CoV-2 tests peaked at 74.1%around December 23 and 25.Conclusions:Sentinel-based surveillance,previously established for influenza,is an effective way to track the circulation trend of SARS-CoV-2 during community-level epidemics.There was no coprevalence of SARS-CoV-2 and influenza virus during the outbreak of SARS-CoV-2,even during the winter influenza season.However,it is important to remain vigilant for the potential rise of influenza activities following the COVID-19 epidemic.Xiaoxu Zeng Yiran Xie Xiaokun Yang Zhibin Peng Jing Tang Lei Yang Shuxia Lin Jun Han Yanping Zhang Dayan Wang 2023China CDC weekly2023,5,7:1
14Association between homologous recombination deficiency and outcomes with platinum and platinum-free chemotherapy in patients with triple-negative breast cancer显示文摘Objective:The choice of chemotherapeutic regimen for triple-negative breast cancer(TNBC)remains controversial.Homologous recombination deficiency(HRD)has attracted increasing attention in informing chemotherapy treatment.This study was aimed at investigating the feasibility of HRD as a clinically actionable biomarker for platinum-containing and platinum-free therapy.Methods:Chinese patients with TNBC who received chemotherapy between May 1,2008 and March 31,2020 were retrospectively analyzed with a customized 3D-HRD panel.HRD positivity was defined by an HRD score≥30 or deleterious BRCA1/2 mutation.A total of 386 chemotherapy-treated patients with TNBC were screened from a surgical cohort(NCT01150513)and a metastatic cohort,and 189 patients with available clinical and tumor sequencing data were included.Results:In the entire cohort,49.2%(93/189)of patients were identified as HRD positive(40 with deleterious BRCA1/2 mutations and 53 with BRCA1/2 intact with an HRD score of≥30).In the first-line metastatic setting,platinum therapy was associated with longer median progression-free survival(mPFS)than platinum-free therapy[9.1 vs.3.0 months;hazard ratio(HR),0.43;95%confidence interval 0.22–0.84;P=0.01].Among HRD-positive patients,the mPFS was significantly longer in those treated with platinum rather than platinum-free therapy(13.6 vs.2.0 months;HR,0.11;P=0.001).Among patients administered a platinum-free regimen,HRD-negative patients showed a PFS significantly superior to that of HRD-positive patients(P=0.02;treatment-biomarker P-interaction=0.001).Similar results were observed in the BRCA1/2-intact subset.In the adjuvant setting,HRD-positive patients tended to benefit more from platinum chemotherapy than from platinum-free chemotherapy(P=0.05,P-interaction=0.02).Conclusions:HRD characterization may guide decision-making regarding the use of platinum treatment in patients with TNBC in both adjuvant and metastatic settings.Yimeng Chen Xue Wang Feng Du Jian Yue Yiran Si Xiaochen Zhao Lina Cui Bei Zhang Ting Bei Binghe Xu Peng Yuan 2023Cancer Biology & Medicine2023,20,2:1

16Traumatic brain injury stimulates sympathetic tone-mediated bone marrow myelopoiesis to favor fracture healing显示文摘Traumatic brain injury(TBI)accelerates fracture healing,but the underlying mechanism remains largely unknown.Accumulating evidence indicates that the central nervous system(CNS)plays a pivotal role in regulating immune system and skeletal homeostasis.However,the impact of CNS injury on hematopoiesis commitment was overlooked.Here,we found that the dramatically elevated sympathetic tone accompanied with TBI-accelerated fracture healing;chemical sympathectomy blocks TBIinduced fracture healing.TBI-induced hypersensitivity of adrenergic signaling promotes the proliferation of bone marrow hematopoietic stem cells(HSCs)and swiftly skews HSCs toward anti-inflammation myeloid cells within 14 days,which favor fracture healing.Knockout ofβ3-orβ2-adrenergic receptor(AR)eliminate TBI-mediated anti-inflammation macrophage expansion and TBIaccelerated fracture healing.RNA sequencing of bone marrow cells revealed that Adrb2 and Adrb3 maintain proliferation and commitment of immune cells.Importantly,flow cytometry confirmed that deletion ofβ2-AR inhibits M2 polarization of macrophages at 7th day and 14th day;and TBI-induced HSCs proliferation was impaired inβ3-AR knockout mice.Moreover,β3-andβ2-AR agonists synergistically promote infiltration of M2 macrophages in callus and accelerate bone healing process.Thus,we conclude that TBI accelerates bone formation during early stage of fracture healing process by shaping the anti-inflammation environment in the bone marrow.These results implicate that the adrenergic signals could serve as potential targets for fracture management.Weijian Liu Wei Chen Mao Xie Chao Chen Zengwu Shao Yiran Zhang Haiyue Zhao Qingcheng Song Hongzhi Hu Xin Xing Xianyi Cai Xiangtian Deng Xinyan Li Peng Wang Guohui Liu Liming Xiong Xiao Lv Yingze Zhang 2023Signal Transduction and Targeted Therapy2023,8,8:1

19Therapeutic potential of an anti-HER2 single chain antibody-DM1 conjugates for the treatment of HER2-positive cancer显示文摘Antibody–drug conjugates(ADCs)take the advantage of monoclonal antibodies to selectively deliver highly potent cytotoxic drugs to tumor cells,which have become a powerful measure for cancer treatment in recent years.To develop a more effective therapy for human epidermal growth factor receptor 2(HER2)-positive cancer,we explored a novel ADCs composed of anti-HER2 scFv–HSA fusion antibodies conjugates with a potent cytotoxic drug DM1.The resulting ADCs,T-SA1–DM1 and T-SA2–DM1(drug-to-antibody ratio in the range of 3.2–3.5)displayed efficient inhibition in the growth of HER2-positive tumor cell lines and the half-maximal inhibitory concentration on SKBR-3 and SKOV3 cells were both at the nanomolar levels in vitro.In HER2-positive human ovarian cancer xenograft models,T-SA1–DM1 and T-SA2–DM1 also showed remarkable antitumor activity.Importantly,three out of six mice exhibited complete remission without regrowth in the high-dose group of T-SA1–DM1.On the basis of the analysis of luminescence imaging,anti-HER2 scFv–HSA fusion antibodies,especially T-SA1,showed strong and rapid tumor tissue penetrability and distribution compared with trastuzumab.Collectively,the novel type of ADCs is effective and selective targeting to HER2-positive cancer,and may be a promising antitumor drug candidate for further studies.Hang Zhang Yuxi Wang Yangping Wu Xiaohua Jiang Yiran Tao Yuqin Yao Yujia Peng Xiangzheng Chen Yuyin Fu Lin Yu Ruixue Wang Qinhuai Lai Weirong Lai Wenting Li Yuhuan Kang Shuli Yi Ying Lu Lantu Gou Min Wu Jinliang Yang 2017Signal Transduction and Targeted Therapy2017,2,1:0
20A time-coded multi-concentration microfluidic chemical waveform generator for high-throughput probing suspension single-cell signaling显示文摘The cellular response to the complex extracellular microenvironment is highly dynamic in time and type of extracellular matrix.Accurately reconstructing this process and analyzing the changes in receptor conformation on the cell membrane surface and intracellular or intercellular signaling has been a major challenge in analytical chemistry and biophysical methodology.In this paper,a time-coded multiconcentration microfluidic chemical waveform generator was developed for the dynamic signaling probing with single-cell array of high temporal resolution,high throughput,and multi-concentration combination stimulation.Based on innovative microchannel structure,sophisticated external control methods and multiplexing technology,the system not only allowed for temporally sequential permutations of the four concentrations of stimuli(time code),but also generated pulsed and continuous waveforms at different frequencies in a highly controllable manner.Furthermore,the single-cell trap array was set up to efficiently capture cells in suspension,dramatically increasing throughput and reducing experiment preparation time.The maximum frequency of the platform was 1 Hz,and one cell could be stimulated at multiple frequencies.To show the ability of the system to investigate rapid biochemical events in high throughput,pulse stimulation and continuous stimulation of different frequencies and different time codes,combined with four concentrations of histamine(HA),were generated for probing G protein-coupled receptor(GPCR)signaling in He La cells.Then,statistical analysis was performed for the mean peak height and mean peak area of the cellular response.We believe that the time-coded multi-concentration microfluidic chemical waveform generator will provide a novel strategy for analytical chemistry,biophysics,cell signaling,and individualized medicine applications.Yiran Guo Peng Chen Zhaolong Gao Yiwei Li Shunji Li Xiaojun Feng Bi-Feng Liu 2022Chinese Chemical Letters2022,33,6:0
