2Safety and efficacy of tenecteplase versus alteplase in patients with acute ischaemic stroke (TRACE): a multicentre, randomised, open label, blinded-endpoint (PROBE) controlled phase II study显示文摘Background Tenecteplase(TNK)possesses several pharmacological characteristics superior to conventional alteplase(rt-PA),with well-established safety and efficacy profile in Caucasians.There exists controversy over the optimal dose of intravenous rt-PA for East Asians with acute ischaemic stroke(AIS).Current study aimed to determine the safety dose range of recombinant human TNK tissue-type plasminogen activator(rhTNK-tPA)for patients with AIS in China.Methods This multicentre,prospective,randomised,open-label,blinded end-point,phase II study compared three tiers of 0.1,0.25,0.32 mg/kg rhTNK-tPA(to a maximum of 40 mg)with standard 0.9 mg/kg rt-PA(to a maximum of 90 mg)in patients who were eligible for intravenous thrombolysis.The safety outcome were symptomatic intracranial haemorrhage(sICH)within 36 hours.Results Between May 2018 and February 2020,240 patients were randomly assigned to four group,4 of whom did not receive study treatment.The intention-to-treat analysis included 236 patients.There was no difference in the improvement on National Institutes of Health Stroke Scale at day 14 in the 3 tiers and control group(63.3%,77.2%,66.7%vs 62.7%).The number of sICH was 3 of 60(5.0%)in the 0.1 mg/kg group,none in the 0.25 mg/kg group,2 of 60(3.3%)in the 0.32 mg/kg group and 1(1.7%)of 59 in the rt-PA group.There were no significant between-group differences in severe adverse events.Conclusions Similar to the Caucasians,rhTNK-tPA was well tolerated in Chinese patients with AIS at all doses administered within 3 hours of symptom onset.The dose-efficacy profile of rhTNK-tPA needs to be established with future investigations.Shuya Li Yuesong Pan Ziran Wang Zhigang Liang Huisheng Chen Dong Wang Yi Sui Xingquan Zhao Yilong Wang WanLiang Du Huaguang Zheng Yongjun Wang 2022Stroke & Vascular Neurology2022,7,1:9
3Gastrointestinal bleeding during acute ischaemic stroke hospitalisation increases the risk of stroke recurrence显示文摘Objective Gastrointestinal(GI)bleeding in patients who had a stroke is strongly associated with a higher risk of death and loss of independence.However,it is unknown whether GI bleeding increases risk for recurrence of stroke.In this study,we assess the potential relationship between GI bleeding and stroke recurrence in patients within 12 months of an acute ischaemic stroke(AIS),using the China National Stroke Registry(CNSR).Methods This study included 22216 patients who had an ischaemic stroke included in the CNSR from 2007 to 2008.We analysed baseline patient characteristics,GI bleeding and outcomes of patients who had an AIS,specifically stroke recurrence at 3,6 and 12 months.We used multivariable logistic regression to evaluate a possible association between GI bleeding and stroke recurrence.results Of the 12415 patients included in our study,12.3%,15.5%and 17.7%had a stroke recurrence at 3,6 and 12 months,respectively.GI bleeding was an independent stroke recurrence risk factor in patients after ischaemic stroke at 3 months(adjusted OR 1.481,95%CI 1.118 to 1.962),6 months(adjusted OR 1.448,95%CI 1.106 to 1.896)and 12 months(adjusted OR 1.350;95%CI 1.034 to 1.763).Conclusion GI bleeding was associated with the increased risk of stroke recurrence after an AIS.Wanliang Du Xingquan Zhao Yilong Wang Yuesong Pan Gaifen Liu Anxin Wang Ruijun Ji Liping Liu Hongqiu Gu Kehui Dong Penglian Wang Yongjun Wang 2020Stroke & Vascular Neurology2020,5,2:7
4Lanthanum-doped Cu-Mn composite oxide catalysts for catalytic oxidation of toluene显示文摘CuMn mixed oxides catalysts doped with La were prepared following a co-precipitation method and used for the catalytic oxidation of toluene. Catalysts properties of the catalysts were investigated by X-ray diffraction, N_2 adsorption/desorption,scanning electron microscopy, H_2-temperature-programmed reduction(H_2-TPR), O_2-temperature-programmed desorption(O_2-TPD) and X-ray photoelectron spectroscopy techniques. Characterization data reveal that the phase change and decrease in crystallinity of the La-doped catalysts increase the number of oxygen vacancies. Improvements in reducibility and an increase in the amount of chemisorbed oxygen of the La-doped catalysts were also verified by H_2-TPR and O_2-TPD. The activity of the CuMn mixed oxides catalysts is significantly improved by the addition of a nominal amount of La. The CuMn/La-4 mol% catalyst exhibits the best catalytic activity, with a 90%conversion temperature of 255 ℃,attributed to a high Mn^(3+)ratio, superficial chemisorbed oxygen,and high surface area. 5Hexagonal boron nitride nanosheet for effective ambient N2 fixation to NH3显示文摘Industrial production of NH3 from N2 and H2 significantly relies on Haber-Bosch process,which suffers from high energy consume and CO2 emission.As a sustainable and environmentally-benign alternative process,electrochemical artificial N2 fixation at ambient conditions,however,is highly required efficient electrocatalysts.In this study,we demonstrate that hexagonal boron nitride nanosheet (h-BNNS) is able to electrochemically catalyze N2 to NH3.In acidic solution,h-BNNS catalyst attains a high NH3 formation rate of 22.4 μg·h-1·mg-1cat.and a high Faradic efficiency of 4.7% at-0.75 V vs.reversible hydrogen electrode,with excellent stability and durability.Density functional theory calculations reveal that unsaturated boron at the edge site can activate inert N2 molecule and significantly reduce the energy barrier for NH3 fonmation.Ya Zhang Huitong Du Yongjun Ma Lei Ji Haoran Guo Ziqi Tian Hongyu Chen Hong Huang Guanwei Cui Abdullah M. 6Staged angioplasty versus regular carotid artery stenting in patients with carotid artery stenosis at high risk of hyperperfusion:a randomised clinical trial显示文摘Background and purpose Hyperperfusion(HP)is a devastating complication associated with carotid artery stenting(CAS)or endarterectomy.The efficacy and safety of staged angioplasty(SAP)in patients with CAS at high risk of HP remains unclear.We sought to determine whether SAP is superior to regular CAS in patients with high risk of HP.Methods A randomised,multicentre open-label clinical trial with blinded outcome assessment(STEP)was conducted.Patients with severe carotid stenosis at high risk of HP were randomly assigned(1:1)to the SAP or regular CAS group.The primary endpoint was hyperperfusion syndrome(HPS)and intracerebral haemorrhage(ICH)within 30 days after the procedure.Results From November 2014 to January 2017,a total of 64 patients were enrolled in 11 centres.33 patients were allocated to the SAP group and 31 to the regular CAS group.At 30 days,the rate of primary endpoint was 0.0%(0/33)in the SAP group and 9.7%(3/31)in the regular CAS group(absolute risk reduction(ARR),9.7%;95%CI−20.1% to 0.7%;p=0.11).As one of the secondary endpoints,the incidence of HP phenomenon(HPP)was lower in the SAP group than the regular CAS group(0.0%vs 22.6%,ARR,−22.6%;95% CI−36.8%to−10.2%;p=0.04).Conclusion The rate of HPS and ICH was not significantly lower in SAP group;the extended secondary endpoint of HPP,however,significantly reduced,which suggested that SAP may be a safe and effective carotid revascularisation procedure to prevent HP.Dapeng Mo Baixue Jia Huaizhang Shi Yaxuan Sun Qingan Liu Chengzhe Fan Jianping Deng Jinglin Yuan Wei Wu Changchun Jiang Guilian Zhang Hanjun Du Ning Ma Feng Gao Xuan Sun Ligang Song Lian Liu Guangge Peng Yongjun Wang Yilong Wang Zhongrong Miao the STEP Study Group 2021Stroke & Vascular Neurology2021,6,1:3
7Significance of VEGF and NF-κB Expression in Thyroid Carcinoma显示文摘OBJECTIVE To investigate the significance of vascular endothelial growth factor (VEGF) and nuclear factor-kappaB (NF-κB) expression in thyroid carcinoma. METHODS The expression of NF-κB and VEGF was determined by im- munohistochemistry in formalin-fixed and paraffin-embedded specimens obtained from 10 normal thyroid tissues (NT), 12 cases of thyroid adeno- ma (TA) and 68 cases of thyroid carcinoma (TC).Differences in expres- sion between NT, TA and TC were statistically analyzed. In addition, in cases of TC, the relationship of NF-κB and VEGF expression with various clinicopathological factors, including histological typing, clinical staging and lymph node metastasis, as well as the correlation between N F - κ B and VEGF expression was examined. RESULTS In contrast to the negative immunoreactivity for VEGF in NT, there was a significantly higher positive incidence (PI) in TA (41.7%, P= 0.040) and TC (75.0%, P<0.001), and a significant difference between TA and TC (P=0.036). Immunoreactivity for NF-κB in NT was negative and significantly higher in TC (63.2%, P<0.001),but not in TA (16.7%, P= 0.481). However the PI difference between TA and TC (P=0.003) was significant. Between the histological types of TC, a significantly higher PI was found in undifferentiated thyroid carcinoma (UTC),namely,100% for VEGF and 90.0% for NF-κB. We also found significant positive relation- ships of VEGF and N F - κ B expression with the clinical stage and lymph node metastasis. Furthermore, a significant positive correlation between VEGF and NF-κB expression in TC was observed. CONCLUSION Our data showed that the expression of VEGF and NF- ΚB/P65 was greater in TC and UTC, and documented their significant positive correlations with the clinical stage and lymph node metastasis in TC. 8SOCS3 Attenuates GM-CSF/IFN-γ-Mediated Inflammation During Spontaneous Spinal Cord Regeneration显示文摘SOCS3,a feedback inhibitor of the JAK/STAT signal pathway,negatively regulates axonal regrowth and inflammation in the central nervous system(CNS).Here,we demonstrated a distinct role of SOCS3 in the injured spinal cord of the gecko following tail amputation.Severing the gecko spinal cord did not evoke an inflammatory cascade except for an injury-stimulated elevation of the granulocyte/macrophage colony-stimulating factor(GM-CSF) and interferon gamma(IFN-γ) cytokines.Simultaneously,the expression of SOCS3 was upregulated in microglia,and unexpectedly not in neurons.Enforced expression of SOCS3 was sufficient to suppress the GMCSF/IFN-γ-driven inflammatory responses through its KIR domain by attenuating the activities of JAK1 and JAK2.SOCS3 was also linked to GM-CSF/IFN-y-induced crosstolerance.Transfection of adenovirus overexpressing SOCS3 in the injured cord resulted in a significant decrease of inflammatory cytokines.These results reveal a distinct role of SOCS3 in the regenerating spinal cord,and provide new hints for CNS repair in mammals.Xuejie Zhang Bingqiang He Hui Li Yingjie Wang Yue Zhou Wenjuan Wang Tiancheng Song Nan Du Xingxing Gu Yi Luo Yongjun Wang 2020Neuroscience Bulletin2020,36,7:3

11Computational prediction and experimental verification of miRNAs in Panicum miliaceum L.显示文摘MicroRNAs (miRNAs) are a class of non-coding RNAs that play critical roles in post-transcriptional regulation. Their target genes are involved in a variety of biological processes such as development, metabolism, and stress response. Panicum miliaceum L. (Panicum) is an important grain crop, but, until now, no miRNAs have been identified in this plant. Using a homology search based on expressed sequence tag (EST) analysis and miRNA precursor secondary structure, a total of 43 new miRNAs were identified. The miRNAs were found to be unevenly distributed among 11 miRNA families. Target analysis using the plant small RNA target analysis server psRNATarget showed that the newly identified miRNAs can potentially regulate 68 target genes. Ten of the 11 miRNA families were annotated as involved in RNA regulation, suggesting they may play an essential role in post-transcriptional regulation in Panicum. 12Dynamic precipitation during multi-axial forging of an Mg-7Gd-5Y-1Nd-0.5Zr alloy显示文摘The dynamic precipitation behavior during multi-axial forging in an Mg-7Gd-5Y-1Nd-0.5Zr alloy has been investigated and compared with that in static precipitation treatment. The results indicated that dynamic precipitation does occur during multi-axial forging. The dynamic precipitate can be deduced as β phase with face-centered cubic crystal structure (a = 2.22 nm). Most of the β precipitates locate at the dynamic recrystallization grain boundaries. The morphology and orientation relationship is different from that of the β phase formed in the static precipitation treated alloys, although the crystal structure is the same. 14Moth sex pheromones affect interspecific competition among sympatric species and possibly population distribution by modulating pre-mating behavior显示文摘Premating behaviors mediated by pheromones play pivotal roles in animal mating choices.In natural populations of the striped stem borer Chilo suppressalis and the rice leaf roller Cnaphalocrocis medinalis in the rice field habitat,we discovered that Z11-16:Ald,a major component of the C.suppressalis pheromone,modulated the premating behavior of C.medinalis.Z11-16:Ald evoked a strong olfactory response in male antennae and strongly inhibited the sex pheromone trapping of male C.medinalis in the field.The functions of three C.medinalis sex pheromone receptor genes(CmedPR1–3)were verified through heterologous expression in Xenopus oocytes.CmedPR1 responded to Z11-18:OH and Z11-18:Ald,as well as the interspecific pheromone compound Z11-16:Ac of sympatric species;CmedPR2 responded to Z13-18:OH and Z13-18:Ald,as well as the sex pheromone compounds Z11-16:Ald and Z9-16:Ald of sympatric species;and CmedPR3 responded to Z11-18:OH and Z13-18:OH,as well as the interspecific pheromones Z11-16:OH,Z9-16:Ald,Z11-16:Ac,and Z11-16:Ald of sympatric species.Thus,CmedPR2 and CmedPR3 share the ligand Z11-16:Ald,which is not a component of the C.medinalis sex pheromone.Therefore,the sex pheromones of interspecific species affected the input of neural signals by stimulating the sex pheromone receptors on the antennae of male C.medinalis moths,thereby inhibiting the olfactory responses of the male moths to the sex pheromones.Our results demonstrate chemical communication among sympatric species in the rice field habitat,the recognition of intra-and interspecific sex pheromones by olfactory receptors,and how insect premating behaviors are modulated to possibly affect resource partitioning.Jianjun Cheng Qinghua Chen Qianshuang Guo Yongjun Du 2023Insect Science2023,30,2:1

18Simultaneous determination of nineteen major components in Qi She Pill by ultra-high-performance liquid chromatography–tandem mass spectrometry显示文摘Qi She Pill(QSP)is a traditional Chinese medicine(TCM)prescription that has been used in treating cervical spondylosis radiculopathy for many years.In this study,a simple and sensitive method using ultra-high-performance liquid chromatography–tandem mass spectrometry(UHPLC–MS/MS)on a reverse-phase C18 column was developed for the simultaneous determination of the 19 major components in QSP.We found that the optimum mobile phase for gradient elution was 0.1% formic acid and methanol.The correlation coefficients of all calibration curves were greater than 0.99.Recoveries measured at three concentration levels varied from 95.43%to 102.35%.Relative standard deviations of intra-and inter-day precisions were less than 4.45%.After successfully validating our method,we then applied it to the quantification of 19 components in QSP products to show that this method provides a new standard in quality assessment of TCM prescriptions containing multiple bioactive components.Zhongliang Zhang Qiang Li Qiufen Li Simiao Du Yongquan Zhou Chunming Lv Yan Zhao Yongjun Wang Ning Zhang 2014Acta Pharmaceutica Sinica B2014,4,5:1
19Near-infrared light-driven multifunctional metal ion(Cu^(2+))-loaded polydopamine nanomotors for therapeutic angiogenesis in critical limb ischemia显示文摘Most of the current nanomedicine-based treatments for critical limb ischemia(CLI)only aim at promoting angiogenesis,ignoring the negative influence on the therapeutic effects caused by the complex pathological micro-environment of ischemic tissue.Herein,near-infrared(NIR)light-driven metal ion(Cu^(2+))-loaded polydopamine(PDA)nanomotors(JMPN@Cu^(2+))is designed and prepared.Due to the good antioxidant and anti-inflammatory activities of PDA,JMPN@Cu^(2+)exhibits excellent biocompatibility and significantly improves the ischemic micro-environment.Additionally,based on superior photothermal conversion effect and jellyfish-like structure,the nanomotors are quickly propelled under NIR laser with low energy intensity to acquire the ability of movement and facilitate intracellular uptake of JMPN@Cu^(2+)by endothelial cells,resulting in the enhanced pro-angiogenic effect of Cu^(2+).Moreover,in vivo experimental findings show that JMPN@Cu^(2+)combined with NIR irradiation can successfully accelerate blood flow recovery and improve muscle repair.Taking these results together,this kind of nanomotor can promote angiogenesis along with ischemic micro-environment amelioration,holding great potential applications for the treatment of limb ischemia.Liang Gui Juju Huang Yi Xing Yongjun Li Junjie Zou Yingwei Zhu Xiao Liang Xiwei Zhang Qiang Xu Xin Du 2023Nano Research2023,16,4:0
20Nucleolus localization of SpyCas9 affects its stability and interferes with host protein translation in mammalian cells显示文摘The CRISPR/Cas9 system,originally derived from the prokaryotic adaptive immune system,has been developed as efficient genome editing tools.It enables precise gene manipulation on chromosomal DNA through the specific binding of programmable sgRNA to target DNA,and the Cas9 protein,which has endonuclease activity,will cut a double strand break at specific locus.However,Cas9 is a foreign protein in mammalian cells,and the potential risks associated with its introduction into mammalian cells are not fully understood.In this study,we performed pull-down and mass spectrometry(MS)analysis of Streptococcus pyogenes Cas9(SpyCas9)interacting proteins in HEK293T cells and showed that the majority of Cas9-associated proteins identified by MS were localized in the nucleolus.Interestingly,we further discovered that the Cas9 protein contains a sequence encoding a nucleolus detention signal(NoDS).Compared with wild-type(WT)Cas9,NoDS-mutated variants of Cas9(mCas9)are less stable,although their gene editing activity is minimally affected.Overexpression of WT Cas9,but not mCas9,causes general effects on transcription and protein translation in the host cell.Overall,identification of NoDS in Cas9 will improve the understanding of Cas9's biological function in vivo,and the removal of NoDS in Cas9 may enhance its safety for future clinical use.Renke Tan Wenhao Du Yiyang Liu Xiaoji Cong Meirong Bai Chenxiao Jiang Zengxia Li Minjia Tan Dengke K.Ma Qiang Huang Wei Jiang Yongjun Dang 2022Genes & Diseases2022,9,3:0
