1Cardamonin from a medicinal herb protects against LPS-induced septic shock by suppressing NLRP3 inflammasome显示文摘Aberrant activation of NLRP3 inflammasome has been implicated in the pathogenesis of diverse inflammation-related diseases, and pharmacological molecules targeting NLRP3 inflammasome are of considerable value to identifying potential therapeutic interventions. Cardamonin(CDN), the major active ingredient of the traditional Chinese medicinal herb Alpinia katsumadai, has exerted an excellent anti-inflammatory activity, but the mechanism underlying this role is not fully understood. Here, we show that CDN blocks canonical and noncanonical NLRP3 inflammasome activation triggered by multiple stimuli. Moreover, the suppression of CDN on inflammasome activation is specific to NLRP3, not to NLRC4 or AIM2 inflammasome. Besides, the inhibitory effect is not dependent on the expression of NF-κB-mediated inflammasome precursor proteins. We also demonstrate that CDN suppresses the NLRP3 inflammasome through blocking ASC oligomerization and speckle formation in a dose-dependent manner.Importantly, CDN improves the survival of mice suffering from lethal septic shock and attenuates IL-1βproduction induced by LPS in vivo, which is shown to be NLRP3 dependent. 2Transcriptome analysis reveals long noncoding RNAs involved in fiber development in cotton (Gossypium arboreum)显示文摘Long noncoding RNAs(lnc RNAs)play important roles in various biological regulatory processes in yeast,mammals,and plants.However,no systematic identification of lnc RNAs has been reported in Gossypium arboreum.In this study,the strand-specific RNA sequencing(ss RNA-seq)of samples from cotton fibers and leaves was performed,and lnc RNAs involved in fiber initiation and elongation processes were systematically identified and analyzed.We identified 5,996 lnc RNAs,of which 3,510 and 2,486 can be classified as long intergenic noncoding RNAs(linc RNAs)and natural antisense transcripts(lnc NAT),respectively.Linc RNAs and lnc NATs are similar in many aspects,but have some differences in exon number,exon length,and transcript length.Expression analysis revealed that 51.9%of linc RNAs and 54.5%of lnc NATs transcripts were preferentially expressed at one stage of fiber development,and were significantly highly expressed than protein-coding transcripts(21.7%).During the fiber and rapid elongation stages,rapid and dynamic changes in lnc RNAs may contribute to fiber development in cotton.This work describes a set of lnc RNAs that are involved in fiber development.The characterization and expression analysis of lnc RNAs will facilitate future studies on their roles in fiber development in cotton.Changsong Zou Qiaolian Wang Cairui Lu Wencui Yang Youping Zhang Hailiang Cheng Xiaoxu Feng Mtawa Andrew Prosper Guoli Song 2016Science China(Life Sciences)2016,59,2:9
3Expression Patterns of ABA and GA Metabolism Genes and Hormone Levels during Rice Seed Development and Imbibition: A Comparison of Dormant and Non-Dormant Rice Cultivars显示文摘Seed dormancy is an important agronomic trait in cereals. Using deep dormant(N22), medium dormant(ZH11), and non-dormant(G46B)rice cultivars, we correlated seed dormancy phenotypes with abscisic acid(ABA) and gibberellin(GA) metabolism gene expression profiles and phytohormone levels during seed development and imbibition. A time course analysis of ABA and GA content during seed development showed that N22 had a high ABA level at early and middle seed developmental stages, while at late developmental stage it declined to the level of ZH11; however, its ABA/GA ratio maintained at a high level throughout seed development. By contrast, G46 B had the lowest ABA content during seed development though at early developmental stage its ABA level was close to that of ZH11, and its ABA/GA ratio peaked at late developmental stage that was at the same level of ZH11. Compared with N22 and G46 B, ZH11 had an even and medium ABA level during seed development and its ABA/GA ratio peaked at the middle developmental stage. Moreover, the seed development time-point having high ABA/GA ratio also had relatively high transcript levels for key genes in ABA and GA metabolism pathways across three cultivars. These indicated that the embryo-imposed dormancy has been induced before the late developmental stage and is determined by ABA/GA ratio. A similar analysis during seed imbibition showed that ABA was synthesized in different degrees for the three cultivars. 4Assessment on alien herbicide-resistant gene flow among crucifers by sexual compatibility显示文摘Cross-pollination and high cross-compatibility are frequently found in many cruciferous plants, which im-plies that there might be potential risk of alien gene flow among crucifers. In this work, the alien glyphosate-resistant gene flow between GM (Genetically modified) rapeseed vari-ety Q3 (Brassica napus) and 6 varieties from 5 cruciferous species was assessed by sexual compatibility through aniline blue fluorescence observation, manual cross, mentor pollina-tion with the mixture pollen and DNA identification. The results indicated that the compatibility indices of two B. napus varieties, B. juncea var. multiceps Tsen et Lee and Chinese cabbage (B. chinensis) (as female) with Q3 (as male) were 16.15, 12.77, 2.345 and 0.85, respectively. F1 plants were examined by PCR and all of these samples were confirmed as positive. Crossing between Orychophragmus violaceus, Des-curainia sophia (as female) and Q3 (as male) expressed high cross-incompatibility. Fluorescence microscope observation indicated that growing of pollen tube terminated at the upper 1/3 part of the style of O. violaceus and on the stigma surface of D. sophia. The mentor plants were further analyzed by PCR and all were shown to be negative. Under the neighboring growth and natural pollination conditions, the rates of gene flow to two B. napus varieties were 2.3278% and 2.1487%, respectively, B. juncea var. multiceps Tsen et Lee and Chinese cabbage were 1.0157% and 0.9236%, re-spectively. No gene flow was observed in O. violaceus and D. sophia. Frequency of gene flow was highly correlated with sexual compatibility. 5Clinical significance of MLH1/MSH2 for stage Ⅱ/Ⅲ sporadic colorectal cancer显示文摘BACKGROUND The development of colorectal cancer(CRC) is a complicated multistep process that involves an accumulation of mutations in tumor suppressor genes and oncogenes.In the process of DNA replication, base mismatch often occurs due to various factors leading to abnormal expression of mismatch repair genes(MMR),among which MLH1 and MSH2 are the most important.Recently, numerous studies indicated that MLH1/MSH2 phenotype is associated with CRC.We wanted to elucidate the role of MLH1/MSH2 in the prediction and prognosis of CRC through long-term clinical observation.AIM To evaluate the prognostic and predictive significance of MLH1/MSH2 in patients with stage Ⅱ-Ⅲ CRC using immunohistochemical analysis and GeneScan.METHODS Specimens from 681 patients with CRC(395 stage Ⅱ and 286 stage Ⅲ, 387 males and 294 females) who underwent curative surgical resection from 2013 to 2016 were tested.Immunohistochemistry was used to analyze MMR status and the microsatellite status of 133 patients was determined by GeneScan analysis.RESULTS Five hundred and fifty(80.76%) patients were MLH1/MSH2 positive and 131(19.24%) were negative by immunohistochemistry.MLH1/MSH2-positive tumors were significantly more frequent in the colon than in the rectum, and had poor differentiation and less mucin production(P < 0.05).Patients of different groups did not differ in terms of age, gender, tumor size, tumor stage, lymphocytic infiltration, or circumscribed margin.MLH1/MSH2-negative patients had a more favorable OS than MLH1/MSH2-positive patients(P < 0.001).Univariate and multivariate analyses demonstrated MLH1/MSH2 expression as an independent prognostic and predictive factor for stage Ⅱ/Ⅲ CRC.MLH1/MSH2 expression was a strong prognostic factor in all patients [P < 0.001, hazard ratio(HR) = 4.064,95%CI: 2.241–7.369].Adjuvant chemotherapy had a greater correlation with survival advantage in MLH1/MSH2-negative patients with stage Ⅲ disease(P <0.001, HR = 7.660, 95%CI: 2.974–15.883).However, patients with stage Ⅱ disease or MLH1/MSH2-positive patients with stage Ⅲ disease did not benefit from adjuvant chemotherapy.GeneScan analysis demonstrated that among 133 patients, 105(78.95%) were microsatellite stable, and 28(21.05%) had microsatellite instability(MSI), including 18(13.53%) with high MSI and 10(7.52%) with low MSI.This is consistent with the immunohistochemical results.CONCLUSION MLH1/MSH2 phenotype constitutes a pathologically and clinically distinct subtype of sporadic CRC.MLH1/MSH2 is an independent prognostic and predictive factor for outcome of stage Ⅱ-Ⅲ CRC.Shui-Ming Wang Bin Jiang Youping Deng Shu-Liang Huang Ming-Zhi Fang Yu Wang 2019World Journal of Gastrointestinal Oncology2019,11,11:5
6Phenolic Composition Analysis and Gene Expression in Developing Seeds of Yellow-and Black-seeded Brassica napus显示文摘Breeders have focused on yellow-seeded Brassica napus (rapeseed) for its better quality com- pared with the black-seeded variety. Moreover, flavonoids have been associated with this kind of rapeseed. In this study, we applied lipid chromatography-electrospray ionization mass spec- trometry (LC-ESI-MSn ) to compare flavonoids in developing seeds of natural black-seeded B. na- pus and yellow-seeded introgression lines selected from progenies of B. napus-Sinapis alba so- matic hybrids. Aside from the most abundant phenolic compounds (sinapine and sinapic acid) and 1, 2-disinapoylglucose, 16 different flavonoids were identified and quantified, including (-)- epicatechin, five monocharged oligomers of (-)-epicatechin ([DP 2]- , [DP 3]- , [DP 4] [DP 2]- B2 and [DP 2]- B5), quercetin, kaempferol, isorhamnetin-dihexoside, kaempferol-sinapoyl-trihexoside, isorhamnetin- sinapoyl-trihexoside, isorhamnetin-hexoside-sulfate, and isorhamnetin-3-O-glucoside. Most of the flavonoids accumulated with seed development, whereas some rapidly decreased during maturation. The content of these flavonoids was lower in the yellow-seeded materials than in the black seeds. In addition, variations of insoluble procyanidin oligomers and soluble phenolic acids were observed among both rapeseed varieties. Transcriptome changes of genes participating in the flavonoid pathway were discovered by quantitative reverse transcription polymerase chain reaction analysis. 9UHPLC-MS/MS analysis of cAMP and cGMP in rat plasma as potential biomarkers of Yin-Yang disharmony in traditional Chinese medicine显示文摘Cyclic 3’,5’-adenosine monophosphate(cAMP)and cyclic 3’,5’-guanosine monophosphate(cGMP)are considered as potential biomarkers for Yin-Yang disharmony in traditional Chinese medicine.However,phosphodiesterase-mediated ex vivo degradation of these molecules in biological samples may result in their underestimation.In the present study,a ultra-high performance liquid chromatography-tandem mass spectrometry(UHPLC-MS/MS)method was developed for determination of cAMP and cGMP in rat plasma,with special consideration of their stability ex vivo.Following precipitation of proteins from plasma samples with 0.4 M perchloric acid,the analytes were chromatographed on a Shimadzu Shimpack-XR-ODS II column with 2.5 mM ammonium acetate and methanol in gradient mode.The MS/MS detection was performed using multiple reaction monitoring in the positive electrospray ionization mode.The lower limit of quantification was 0.27 ng/mL for cAMP and 0.37 ng/mL for cGMP.The method was used to determine the plasma cAMP and cGMP levels in normal and Yin deficiency diabetic rats treated with or without Rehmannia glutinosa.The developed method may be useful for evaluating the regulatory effects of Chinese herbal medicine on the levels of cAMP and cGMP in the body.Xin Wang Yue Du Cuiting Wu Ming Xu Youping Liu Xin Di 2021Journal of Pharmaceutical Analysis2021,11,4:3

12TSC characteristics of AC aged ZnO varistors显示文摘The AC aging characteristics and mechanism of ZnO varistors have been widely concerned for a long time.In this paper,accelerated aging test of ZnO varistors was carried out based on the 'ten degrees and a half' rule,power loss and thermally stimulated current(TSC) of samples were tested during each aging stage.The test results show that with the increasing of aging time,the power loss increases,the trapped charges increase and the trap level becomes deeper.In addition,the increase of power loss and the increase of trapped charge with the aging time have the same trend.A preliminary analysis shows that the migration of interstitial zinc ions will change the space charge distribution and decrease the Schottky barrier height to cause the change of the trap level.TU YouPing WANG Qian HE Jie LI Xiao DING LiJian 2013Chinese Science Bulletin2013,58,7:2

14Flashover Behavior of Cone-type Spacers with Inhomogeneous Temperature Distribution in SF_(6)/N_(2)-filled DC-GIL Under Lightning Impulse with DC Voltage Superimposed显示文摘Before designing DC gas insulated transmission lines(GIL),the insulation characteristics of spacers under a lightning impulse(LI)voltage with a superimposed DC voltage must be clarified.Many experiments have been performed to guide the development of the DC spacer.However,the impacts of thermal stresses on their insulation performance under combined voltage are always neglected,and little attention has been given to the insulation characteristics of spacers in an eco-friendly gas filled DC-GIL.To solve these problems,an experiment setup with the abilities to load the thermal field and the DC voltage with LI voltage superimposed on the cone-type spacer are built.After that,the insulation characteristics of spacers under the combined voltage of DC with lightning impulse at 0.1 MPa SF_(6)/N_(2) gas mixtures are investigated,under different thermal stress conditions.The experiment’s results indicate that:1)The flashover voltage of spacers under the same polarity superimposed voltages is slightly higher than that under LI alone.Conversely,the flashover voltage under reverse polarity superimposed voltages is lower than that under LI alone.2)The longer the duration of the foregoing DC voltage,the differences between the flashover voltage under superposed voltage and that under LI alone are larger.3)The combined flashover voltages are reduced under thermal stress.To ensure the insulation performance of newly designed DC spacers,the insulation test is suggested to be applied under thermal stress and combined voltage of the DC with lightning impulse,especially for superimposed reverse polarity.Guoming Ma Hongyang Zhou Yuan Wang Hanchi Zhang Shijie Lu Youping Tu Jian Wang Chengrong Li 2020CSEE Journal of Power and Energy Systems2020,6,2:2
15Feasibility of C_(3)F_(7)CN/CO_(2)gas mixtures in high-voltage DC GIL:a review on recent advances显示文摘With the rise of renewable energy resources,especially offshore wind power plant,the DC transmission concept has attracted more and more attention.The selection of eco-friendly insulating gas,as most critical insulating part of gas-insulated equipment is top priority for developing highly reliable system.Moreover,it is acknowledged that eco-friendly C_(3)F_(7)CN/CO_(2) gas mixtures have become a potential alternative to SF_(6) gas due to its excellent performance.This study reviews the basic physical properties of C_(3)F_(7)CN gas/gas mixtures and insulation properties including the gas gap breakdown and surface flashover performance at DC,as well as lightning impulse voltage.Investigation about the gas stability is presented,including the compatibility of gas with solid materials in gas-insulated transmission line(GIL),and the decomposition characteristics under long-term electrical and thermal stresses.The important progress on the charge accumulation characteristics of gas-solid interface in the novel gas environment under DC voltage stress is analysed.Finally,the key issues that need to be paid attention to and further followed through regarding the application of this novel gas in GIL are summarised and put forward.This study hopefully can provide a complete reference for the development of eco-friendly DC gas insulation equipment with C_(3)F_(7)CN/CO_(2) gas mixtures.Youping Tu Geng Chen Cong Wang Yuming Shao Yujing Tong Chuanyang Li Guoming Ma Tohid Shahsavarian 2020High Voltage2020,5,4:2
16Relationship between space charge and nonlinear characteristics of ZnO varistor显示文摘In order to reveal the conduction mechanism of ZnO varistor, the pulsed electroacoustic (PEA) measurement is employed to investigate the space charge distribution characteristics of ZnO varistors under different current densities in this paper. Experimental results show that the relationship between the space charge and the current density is consistent with the voltage-current characteristics, and can reflect the nonlinear characteristics of ZnO varistor. The space charge increases linearly with the increase of the electric field strength and the current density in the small current region, this reflects the decrease of the surface states of the grain boundary and changes of the Schottky barrier. There exists a transition region between the small current and middle current regions, where the current is composed of the current by the emission of thermally activated electrons and the tunneling current. 17Comparative proteomics analysis of OsNAS1 transgenic Brassica napus under salt stress显示文摘Salt stress is one of the major abiotic stresses in agricultural plants worldwide.We used proteomics to analyze the differential expression of proteins in transgenic OsNAS1 and non-transformant Brassica napus treated with 20 mmol/L Na2CO3.Total protein from the leaves was extracted and separated through a high-resolution and highly repetitive two-dimensional electrophoresis(2-DE)technology system.Twelve protein spots were reproducibly observed to be upregulated by more than 2-fold between transgenic and non-transformant B.napus.These 12 spots were digested in-gel with trypsin and characterized by matrix-assisted laser-desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS)to obtain the peptide mass fingerprints.Protein database searching revealed that 5 of these proteins are involved in salt tolerance:dehydrogenase,glutathione S-transferase,peroxidase,20S proteasome beta subunit,and ribulose-1,5-bisphosphate carboxylase/oxygenase.The potential functions of these identified proteins in substance and energy metabolism,stress tolerance,protein degradation,and cell defense are discussed.The salt tolerance of the transgenic rapeseed was significantly improved by the introduction of the OsNAS1 gene from Brazilian upland rice of Oryza sativa(cv.IAPAR 9).KONG Fang MAO ShanJing DU Kun WU Mian ZHOU XiaoYan CHU ChengCai WANG YouPing 2011Chinese Science Bulletin2011,56,22:2
