| 2 | Expression analysis of a Na^+/H^+ antiporter gene PeNHX1 from Populus euphratica显示文摘Na+/H+ antiporters play an important role in the salt tolerance of a wide variety of plants.Using the rapid amplification of cDNA ends method,a Na+/H+ antiporter gene (PeNHX1) was isolated from Populus euphratica.The deduced amino acid sequence contained 528 amino acid residues with a conserved amiloride-binding domain (77LFFIYLLPPI86) and shared more than 68% identity with that of AtNHX1 from Arabidopsis thaliana.PeNHX1 can confer resistance to Na+,as well as Li+,to (EP432) an Escherichia coli strain deficient in both nhaA and nhaB,thus proving that it is a functional Na+/H+ antiporter.PeNHX1 expression profile in EP432 reflected pH independent manner.PeNHX1 expression was regulated by salt at the transcriptional level.Meanwhile,results demonstrated that transcripts of PeNHX1 in P.euphratica calli showed a salt dependent response,and thus provide a valuable tool for studying signaling and biochemical pathways involved in salt recognition and response in P.euphratica. | YuXia Wu JianQuan Liu | 2009 | Research in Cold and Arid Regions2009,1,6: | 2 |
| 3 | Prediction of VIGS efficiency by the Sfold program and its reliability analysis in Gossypium hirsutum显示文摘Genetic transformation in some plant species,including cotton(Gossypium hirsutum), is hampered by laborious and time-consuming processes and often unachievable. Virus-induced gene silencing(VIGS) by double-stranded RNAs can serve as a reverse-genetics tool to determine gene function. However, knockdown levels vary greatly when using a tobacco rattle virus-based vector that carries different c DNA fragments of a gene. How to choose the optional target fragment for high interference efficiency is very challenging. Addressing this challenge requires increasing the efficacy of small interference RNA(si RNA) in target fragment. Here, we describe a method to assess VIGS efficiency by comparing the following parameters of si RNA in target sequence: the disruption energy of the target(DGdisruption), the differential stability of si RNA duplex ends(DSSE), and the internal stability at positions 9–14 of the si RNA antisense strand(AIS), which are calculated by Sfold program(http://gffzz01a2382fbe924301h9w5660knc0k56pu6.ffgz.tsg.suse.edu.cn). We find that the si RNAs with low DGdisruption, high DSSE and high AIS have high activity and easily result in high VIGS efficiency by experimentally testing the actual knockdown levels of the four target genes, Gh PDS,Gh CLA1, Gh AOS1, and Gh CXE1 via choosing different target sequences for each gene. | 5 | Zhang Heng's Seismoscope( 1):Its Emergence显示文摘The emergence of Zhang Heng's seismoscope has a profound historical background. This paper intends to explore the formation and material basis of Zhang Heng's scientific thought. It appears that the simple intention of 'observing visions and distinguishing good fortune or misfortune'was his motivation to develop the seismoscope. Tracing its historical background,the authors proposed in this paper that the development of this seismoscope started around 128 A. D., and based on perceptual knowledge, two ideas about earthquakes came into being,i. e.,earthquake location could be judged according to the instrument shaking direction, and the earthquake and earth split were essentially different. In the instrument manufacture,Zhang Heng advocated a principle of 'making utensils to imitate cosmos'. By this principle,the following three problems were readily solved: the seismoscope imitated the response of the suspension system to earthquakes( i. e. natural seismoscope). Technically,the trigger mechanism of the latch was used for reference,and it recorded the physical evidence of earthquake occurrence by an ingenious way of a ball dropping from a dragon's mouth. The solution of these three key problems lead to the emergence of the seismoscope in 132 A. D. and the successful detection of an earthquake in 134 A. D. | 6 | Associations of muscle mass,strength,and quality with all-cause mortality in China:a population-based cohort study显示文摘Background It remains unclear about the association of muscle mass,strength,and quality with death in the general Chinese population of diverse economical and geographical backgrounds.The present study aimed to comprehensively examine such associations across different regions in China.Methods Based on the China Kadoorie Biobank study,the present study included 23,290 participants who were aged 38 to 88 years and had no prevalent cardiovascular diseases or cancer.Muscle mass and grip strength were measured using calibrated instruments.Arm muscle quality was defined as the ratio of grip strength to arm muscle mass.Low muscle mass,grip strength,and arm muscle quality were defined as the sex-specific lowest quintiles of muscle mass index,grip strength,and arm muscle quality,respectively.Cox proportional hazards models yielded hazard ratios(HRs)and 95%confidence intervals(CIs)for risks of all-cause mortality in relation to muscle mass,strength,and quality.Results During a median follow-up of 3.98 years,739 participants died.The HR(95%CI)of all-cause mortality risk was 1.28(1.08–1.51)for low appendicular muscle mass index,1.38(1.16–1.62)for low total muscle mass index,1.68(1.41–2.00)for low grip strength,and 1.41(1.20–1.66)for low arm muscle quality in models adjusted for sociodemographic characteristics,lifestyle factors,and medical histories.Conclusion Low muscle mass,grip strength,and arm muscle quality are all associated with short-term increased risks of mortality,indicating the importance of maintaining normal muscle mass,strength,and quality for general Chinese adults. | Man Wu Yuxia Wei Jun Lv Yu Guo Pei Pei Jiachen Li Huaidong Du Ling Yang Yiping Chen Xiaohui Sun Hua Zhang Junshi Chen Zhengming Chen Canqing Yu Liming Li 无 | 2022 | Chinese Medical Journal2022,,11: | 2 |
| 7 | Identification of Kalidium species(Chenopodiaceae)by DNA barcoding显示文摘DNA barcoding is an increasingly prevalent molecular biological technology which uses a short and conserved DNA fragment to facilitate rapid and accurate species identification. Kalidium species are distributed in saline soil habitat throughout Southeast Europe and Northwest Asia, and used mainly as forage grass in China. The discrimination of Kalidium species was based only on morphology-based identification systems and limited to recognized species. Here, we tested four DNA candidate loci, one nuclear locus(ITS, internal transcribed spacer) and three plastid loci(rbc L, mat K and ycf1b), to select potential DNA barcodes for identifying different Kalidium species. Results showed that the best DNA barcode was ITS locus, which displayed the highest species discrimination rate(100%), followed by mat K(33.3%), ycf1b(16.7%), and rbc L(16.7%). Meanwhile, four loci clearly identified the variant species, Kalidium cuspidatum(Ung.-Sternb.) Grub.var. sinicum A. J. | 8 | Differential expression of PeSOS1, PeNhaD1 and PeNHX1 genes between the salt-tolerant Populus euphratica and salt-sensitive P. hopeiensis calli显示文摘Populus euphratica Oliv. is of high salinity tolerance and used as a model species for investigating molecular mechanisms of trees' responses to salt stress. In the work presented here we found that calli of P. euphratica grew more rapidly and accumulated less Na+, but more K+, under salt stress than those of salt-sensitive poplar, Populus hopeiensis. Different types of Na+/H+ antiporters (SOS1, NhaD1 and NHX1) were isolated from P. euphratica; all of these genes have been shown to play important roles in plant salt tolerance mechanism in previous studies. Expression profiles of these three genes were compared between P. hopeiensis and P. euphratica in the presence and absence of salt stress by real-time PCR. The three genes were induced in both P. euphratica and P. hopeiensis by salt. Transcript levels of PeNHX1 were lower in P. euphratica than in P. hopeiensis under 150 mM NaCl stress. In addition, transcript levels of PeNhaD1 were lower, while PeSOS1 were higher in P. euphratica than in P. hopeiensis under both stressed and unstressed conditions. The results indicated that P. euphratica up-regulates different genes and consistently maintains both effluxes of Na+ and high K+ levels. | 9 | Tall gastrodis tuber combined with antiepileptic drugs repairs abnormal perfusion foci in focal epilepsy显示文摘One hundred patients with focal epilepsy were recruited for the present study and their seizures controlled with antiepileptic drugs. The patients then orally received a capsule of tall gastrodis tuber powder, a traditional Chinese drug, and underwent single photon emission computed tomography, long-term electroencephalogram, and CT/MRI. Blood drug levels were monitored throughout the study. Before treatment with tall gastrodis tuber, 35 of the 100 cases had abnormal CT/MRI scans; 79 cases had abnormal single photon emission computed tomography images; 86 cases had abnormal electroencephalogram; and a total of 146 abnormal perfusion foci were observed across the 100 subjects. After treatment, the number of patients with normal single photon emission computed tomography images increased by 12; normal electroencephalogram was observed in an additional 27 cases and the number of patients with epileptiform discharge decreased by 29 (34% of 86); the total number of abnormal perfusion foci decreased by 52 (36%) and changes in abnormal foci were visible in 65 patients. These changes indicate that the administration of tall gastrodis tuber in combination with antiepileptic drugs repairs abnormal perfusion foci in patients with focal epilepsy. | 13 | P53单克隆抗体的制备及其临床应用(英文)显示文摘Objective: The aim of this study was to prepare monoclonal antibody against P53, a kind of tumor suppressor protein,and use the antibody initially in clinical immunoassay. Methods: Monoclonal antibody was prepared and identified via the classic protocol of monoclonal antibody preparation. Identified monoclonal antibodies were purified by affinity chromatography. Antibody titer was determined by enzyme linked immunosorbent assay(ELISA). The specific binding activity of antibody was detected by Western blotting and immunohistochemistry. Results: Three strains of monoclonal antibodies named 1P15, 2P37 and 3P40 were obtained and purified by affinity chromatography. The purity of antibodies was higher than90%. The titers of antibodies were more than 1: 6000. Western blot and immunohistochemistry assay showed that the specific antibody can combine with endogenous P53 protein in the tumor cell lines and determine the expression of P53 in tumor tissue.
