1FAT1, a direct transcriptional target of E2F1, suppresses cell proliferation, migration and invasion in esophageal squamous cell carcinoma显示文摘Objective: Growing evidence indicates that FAT atypical cadherin 1(FAT1) has aberrant genetic alterations and exhibits potential tumor suppressive function in esophageal squamous cell carcinoma(ESCC). However, the role of FAT1 in ESCC tumorigenesis remains not well elucidated. The aim of this study was to further investigate genetic alterations and biological functions of FAT1, as well as to explore its transcriptional regulation and downstream targets in ESCC.Methods: The mutations of FAT1 in ESCC were achieved by analyzing a combined study from seven published genomic data, while the copy number variants of FAT1 were obtained from an analysis of our previous data as well as of The Cancer Genome Atlas(TCGA) and Cancer Cell Line Encyclopedia(CCLE) databases using the cBioPortal. The transcriptional regulation of FAT1 expression was investigated by chromatin immunoprecipitation(ChIP) and the luciferase reporter assays. In-cell western, Western blot and reverse transcription-quantitative polymerase chain reaction(RT-qPCR) were used to assess the indicated gene expression. In addition, colony formation and Transwell migration/invasion assays were employed to test cell proliferation, migration and invasion.Finally, RNA sequencing was used to study the transcriptomes.Results: FAT1 was frequently mutated in ESCC and was deleted in multiple cancers. Furthermore, the transcription factor E2 F1 occupied the promoter region of FAT1, and depletion of E2 F1 led to a decrease in transcription activity and mRNA levels of FAT1. Moreover, we found that knockdown of FAT1 promoted KYSE30 and KYSE150 cell proliferation, migration and invasion;while overexpression of FAT1 inhibited KYSE30 and KYSE410 cell proliferation, migration and invasion. 2Animal diversities and characteristics of environmental change revealed by skeletons unearthed at Zhongba Site of Chongqing City,China显示文摘Nearly 200000 animal skeletons are unearthed in the Unit T0202 from Zhongba Site of Zhongxian County.According to the analysis of 129165 specimens,these skeletons may be classified into 5 kinds, namely,Mammalia,Osteichthys,Aves,Amphibian and Reptilia,which belong to 13 orders,28 families and 42 genera.In this paper,based on archeological dating and AMS 14 C data,through statistically analyzing the unearthed skeletons and studying the change of the smallest individual numbers,these research results detected the following:1)In almost all the time of 2370―200 BC,in Zhongba region, some animals distributed widely,such as Muntiacus sp,Elaphodus cephalophus and Muntiacus sp. inhabiting in glade and grassland,Scrofa sp.and Canis familiaris raised by the ancient people,rodentia rabbit and Rattus rattus,which suggested that a fairly good ecological environment of forest and grassland was preserved at that period and the predecessors began to raise domestic animals from 1750 to 1000 BC,which has lasted until today.2)Rhinoceros only lived during 2000―1750 BC,1000― 700 BC and 700―500 BC,which indicates that the ecological environment of grassland and wetland might be better in these phases.3)Macaca sp.and Ursus arctos appeared only after 1750―1000 BC, which may show that the forest condition is better for animals to live during 1750―200 BC.4)The smallest individual numbers of Bublus sp.,bos sp.and otters emerged during about 2370―1750 BC, which perhaps infers that water area during the early period was wider than that of the late period.5) Since skeletons of rhinoceros are discovered in strata of Zhongba Site during 2000―1750 BC and 1000―500 BC,according to the climate and ecology environment which rhinoceros live in now,the average annual temperature and precipitation during 2000―1750 BC and 1000―500 BC are supposed to be probably higher than that of today.Although Zhu Kezhen considered that the first low temperature period in the past 5000 years of China was between 1100 BC and 850 BC,massive pollen of Morus, Ulmus,Fagus,Quercus,Castanea,etc.were found in Dajiuhu peat at that time,which should indicate that the climate was still moderate for living things to live at least in Dajiuhu basin and Zhongba Site.6) Because skeletons of Gervus albirostris were unearthed during 310―200 BC at Zhongba Site,based onthe climate and the ecological environment those animals live in now,the average annual temperature and the average annual precipitation in Zhongba area between 310 BC and 200 BC should be lower than those of today,which is confirmed by the TOC research of Zhongba Site strata.ZHU Cheng MA ChunMei OUYANG Jie LI ZhongXuan YIN Qian SUN ZhiBin HUANG YunPing R.K.FLAD LI Lan LI YuMei 2008Chinese Science Bulletin2008,53,S1:4

4Pathological features of COVID-19-associated lung injury:a preliminary proteomics report based on clinical samples显示文摘The COVID-19 pandemic has emerged as a global health emergency due to its association with severe pneumonia and relative high mortality.However,the molecular characteristics and pathological features underlying COVID-19 pneumonia remain largely unknown.To characterize molecular mechanisms underlying COVID-19 pathogenesis in the lung tissue using a proteomic approach,fresh lung tissues were obtained from newly deceased patients with COVID-19 pneumonia.After virus inactivation,a quantitative proteomic approach combined with bioinformatics analysis was used to detect proteomic changes in the SARS-CoV-2-infected lung tissues.We identified significant differentially expressed proteins involved in a variety of fundamental biological processes including cellular metabolism,blood coagulation,immune response,angiogenesis,and cell microenvironment regulation.Several inflammatory factors were upregulated,which was possibly caused by the activation of NF-κB signaling.Extensive dysregulation of the lung proteome in response to SARS-CoV-2 infection was discovered.Our results systematically outlined the molecular pathological features in terms of the lung response to SARS-CoV-2 infection,and provided the scientific basis for the therapeutic target that is urgently needed to control the COVID-19 pandemic.Ling Leng Ruiyuan Cao Jie Ma Danlei Mou Yunping Zhu Wei Li Luye Lv Dunqin Gao Shikun Zhang Feng Gong Lei Zhao Bintao Qiu Haiping Xiang Zhongjie Hu Yingmei Feng Yan Dai Jiang Zhao Zhihong Wu Hongjun Li Wu Zhong 2020Signal Transduction and Targeted Therapy2020,5,1:4

6Production of human lysozyme-transgenic cloned porcine embryos by somatic nuclear transfer显示文摘Due to their physiology and organ size, pigs have significant potential as human disease models and as organ transplantation donors. Genetic modification of pigs could provide benefits for both agriculture and human medicine. In this study, five fetal pig fibroblast cell lines from two species (Wuzhishan and Landrace pigs) were transfected using double-marked human lysozyme (HLY) plasmids (pBC1-HLY-GFP-NEO) by a liposome-mediated method. The ratio of green fluorescent protein (GFP)-expressing cells was >95% in sw7, sw8, slw3 and slw6 cell lines, but only 49.3% in slw9 cells. Cells from the four highly transgenic lines were used as nuclear donors to construct embryos, which were then cultured after fusion and activation by electric stimulation. The rate of cleavage was 76.7%, 48 h after activation. After 7 days, 18.5% of cleaved eggs had developed to the blastocyst stage and 93.3% of blastocysts were GFP-positive. These results indicate that transgenic fetal pig flbroblast cell lines could be obtained by a liposome-mediated method, though the transfection efficiency varied between cell lines.
