1Genome architecture and tetrasomic inheritance of autotetraploid potato显示文摘Potato(Solanum tuberosum)is the most consumed non-cereal food crop.Most commercial potato cultivars are autotetraploids with highly heterozygous genomes,severely hampering genetic analyses and improvement.By leveraging the state-of-the-art sequencing technologies and polyploid graph binning,we achieved a chromosome-scale,haplotype-resolved genome assembly of a cultivated potato,Cooperation-88(C88).lntra-haplotype comparative analyses revealed extensive sequence and expression differences in this tetraploid genome.We identified haplotype-specific pericentromeres on chromosomes,suggesting a distinct evolutionary trajectory of potato homologous centromeres.Furthermore,we detected double reduction events that are unevenly distributed on haplotypes in 1021 of 1034 selfing progeny,a feature of autopolyploid inheritance.By distinguishing maternal and paternal haplotype sets in C88,we simulated the origin of heterosis in cultivated tetraploid with a survey of 3110 tetra-allelic loci with deleterious mutations,which were masked in the heterozygous condition bytwo parents.This study provides insights into the genomic architecture of autopolyploids and will guide their breeding.Zhigui Bao Canhui Li Guangcun Li Pei Wang Zhen Peng Lin Cheng Hongbo Li Zhiyang Zhang Yuying Li Wu Huang Mingwang Ye Daofeng Dong Zhukuan Cheng Peter VanderZaag Evert Jacobsen Christian W.B.Bachem Suomeng Dong Chunzhi Zhang Sanwen Huang Qian Zhou 2022Molecular Plant2022,15,7:4

3Drug resistance mutation profiles of the drug-nave and first-line regimen-treated HIV-1-infected population of Suzhou, China显示文摘Little is known about the prevalence of drug-resistant mutations in HIV-1-positive individuals in Suzhou, China. To elucidate the transmitted drug resistance(TDR) and acquired drug resistance mutation(ADR) profiles, we collected blood specimens from 127 drug-naive and 117 first-line drugtreated HIV-1-infected individuals sampled from 2014 to 2016 in Suzhou. We successfully amplified po/fragments from 100 drug-naive and 20 drug-treated samples. We then determined the drugresistant mutations to protease(PR) and reverse-transcriptase(RT) inhibitors according to the Stanford drug resistance database. Overall, 11 and 13 individuals had transmitted(drug-naive group) and acquired(treated group) resistance mutations, respectively. Six transmitted drugresistant mutations were found, including two mutations(L33F and L76V) in the protease region and four(K70N/E and V179D/E) in the RT region. Only L76 V was a major mutation, and K70N/E and V179D/E are known to cause low-level resistance to RT inhibitors. All 13 treated participants who had major drug resistance mutations demonstrated intermediate to high resistance to efavirenz and nevirapine, and six had a treatment duration of less than three months. No major mutations to RT inhibitors were found, implying that the epidemic of transmitted resistance mutations was not significant in this area. Our results suggest that more frequent virus load and drug resistance mutation tests should be conducted for individuals receiving antiretroviral treatment, especially for newly treated patients. 4XAFS study of Cu^(2+) in aqueous solution of CuBr_2显示文摘Copper ion is the essential microelement to many organisms. In this paper, the local structure of Cu2+ in CuBr2 aqueous solutions with different concentrations are investigated by using X-ray absorption fine structure (XAFS) technique. XANES (X-ray Absorption Near Edge Structure) spectra indicate that charge transfer from Br- to Cu2+ decreases with the solution concentration, which lead to a shift of the absorption edge. The shoulder appearing at the rising edge proves to be characteristic of a tetragonal distortion. The Fourier transform magnitudes of EXAFS (Extended X-ray absorption fine structure) data of Cu species suggest that more Cu-Br bonds may exist in high concentrations. A fivefold coordination configuration like a pyramid is used as the fitting parameters. From the analysis of the coordination numbers, the proportion of Cu-O and Cu-Br is 4:1 in the saturated solution. The Br atom is on the equatorial plane of the model. 5Amino-metalloporphyrin polymers derived Fe single atom catalysts for highly efficient oxygen reduction reaction显示文摘Recently,nitrogen-doped porous carbon supported single atom catalysts(SACs)have become one of the most promising alternatives to precious metal catalysts in oxygen reduction reaction(ORR)due to their outstanding performance,especially those derived from porphyrin-based materials.However,most of them involve other metal residuals,which would cause the tedious pre-and/or post-treatment,even mislead the mechanistic investigations and active-site identification.Herein,we report a precursor-dilution strategy to synthesize Fe SACs through the Schiff-based reaction via co-polycondensation of amino-metalloporphyrin,followed by pyrolysis at high temperature.Systematic characterization results provide the compelling evidence of the dominant presence of atomically dispersed Fe-Nxspecies.Our catalyst shows superior ORR performance with positive half-wave potential(E1/2=0.85 V vs.RHE)in alkaline condition and moderate activity(E1/2=0.68 V vs.RHE)under the acidic condition,excellent methanol tolerance and good long-term stability.All the results indicate Fe SACs would be a promising candidate for replacing the precious Pt in metal-air batteries and fuel cells.Qian He Yuying Meng Hao Zhang Ying Zhang Qingdi Sun Tao Gan Huajian Xiao Xiaohui He Hongbing Ji 2020Science China Chemistry2020,63,6:2
6Inducible CRISPRa screen identifies putative enhancers显示文摘Enhancers are critical cis-regulatory elements that regulate spatiotemporal gene expression and control cell fates.However,the identification of enhancers in native cellular contexts still remains a challenge.Here,we develop an inducible CRISPR activation(CRISPRa)system by transgenic expression of doxycycline(Dox)-inducible dCas9-VPR in mouse embryonic stem cells(iVPR ESC).With this line,a simple introduction of specific guide RNAs targeting promoters or enhancers allows us to realize the effect of CRISPRa in an inducible,reversible,and Dox concentration-dependent manner.Taking advantage of this system,we induce tiled CRISPRa across genomic regions(105 kilobases)surrounding T(Brachyury),one of the key mesodermal development regulator genes.Moreover,we identify several CRISPRa-responsive elements with chromatin features of putative enhancers,including a region the homologous sequence in which humans harbors a body height risk variant.Genetic deletion of this region in ESC does affect subsequent T gene activation and osteogenic differentiation.Therefore,our inducible CRISPRa ESC line provides a convenient platform for high-throughput screens of putative enhancers.Zhongye Dai Rui Li Yuying Hou Qian Li Ke Zhao Ting Li Mulin Jun Li Xudong Wu 2021Journal of Genetics and Genomics2021,48,10:1
7Thermal Performance of a Micro Heat Pipe Array for Battery Thermal Management Under Special Vehicle-Operating Conditions显示文摘The thermal management of battery systems is critical for maintaining the energy storage capacity,life span,and thermal safety of batteries used in electric vehicles,because the operating temperature is a key factor affecting battery performance.Excessive temperature rises and large temperature differences accelerate the degradation rate of such batteries.Currently,the increasing demand for fast charging and special on-vehicle scenarios has increased the heat dissipation requirements of battery thermal management systems.To address this demand,this work proposes a novel micro heat pipe array(MHPA)for thermal management under a broadened research scope,including high heat generation rates,large tilt angles,mild vibration,and distributed heat generation conditions.The experimental results indicate that the temperature difference is maintained 3.44°C at a large heat generation of 50 W for a limited range of tilt angles.Furthermore,a mild vehicle vibra-tion condition was found to improve temperature uniformity by 3.3°C at a heat generation of 10 W.However,the use of distributed heat sources results in a temperature variation of 3.88°C,suggesting that the heat generation distribution needs to be considered in thermal analyses.Understanding the effects of these special battery-operating conditions on the MHPA could significantly contribute to the enhancement of heat transfer capability and temperature uniformity improvement of battery thermal management systems based on heat pipe technologies.This would facilitate the realization of meeting the higher requirements of future battery systems.Chengning Yao Dan Dan Yangjun Zhang Yueqi Wang Yuping Qian Yuying Yan Weilin Zhuge 2020Automotive Innovation2020,3,4:1

10A MIXED FE-BE METHOD FOR COUPLED VIBRATION OF GRAVITY DAM-RESERVOIR SYSTEM WITH VARIABLE WATER DEPTH显示文摘To solve the coupled vibration of a gravity dam-reservoir system with variable waterdepth by using a hybrid element method,the fluid region with variable water depth needs to be dis-cretized by FE meshes.However,such a method asks for a great computational cost owing to the ex-cessive unknowns,especially when the fluid region with variable water depth is relatively large.Toovercome the shortcoming,a refined boundary element method is proposed to analyze the fluid field,in which only the discretization for the boundary of the variable depth region is required.But as a basisof this approach,it is necessary to construct a new Green’s function corresponding to an infinite stripregion.The problem is solved as the first step in this paper by employing Fridman’s operator functiontheory,and then a mixed FE-BE formulation for analyzing the free vibration of the gravity dam-reservoir system is derived by means of the coupling conditions on the dam-reservoir interface.Final-ly,a numerical example is provided to illustrate a great improvement of the method developed hereinover the hybrid element method.Jin Tao Huang Yuying Qian Qin 1998Acta Mechanica Solida Sinica1998,11,2:0
11Sodium titanate nanowires for Na^(+) -based hybrid energy storage with high power density显示文摘It is crucial to enhance the rate capability of the titanium-based materials for fulfilling their promising potential as the anode materials of sodium-ion bat-teries(SIBs).Herein,Mn-doped sodium titanate(Mn-NTO)nanowires with homogeneously distributed ultrathin carbon nanosheets(Mn-NTO@C)are syn-thesized by a one-step salt-template-assisted method,showing much-enhanced power density.The as-prepared Mn-NTO@C demonstrates the realization of hybrid energy storage,which reconciles the diffusion-controlled behavior with the pseudocapacitive-controlled behavior.It has been revealed that the Mn heteroatoms can raise the proportion of Na_(2)Ti_(3)O_(7) phase with the expanded crystal lattice,facilitating the diffusion-controlled insertion/extraction process of sodium ions.Meanwhile,the hybrid morphology of Mn-NTO nanowires and carbon nanosheets provides a promoted structure stability.Asaresult,theassem-bled Na||Mn-NTO@C half-cells work well at an extreme current density of 24 A g^(−1) for 10000 cycles with a capacity retention of 95.2%.Moreover,the Mn-NTO@C||Na _(3) V_(2)(PO_(4))_(3)(NVP)full cells exhibit an attenuation of only 0.0015%per cycle at 20 A g^(−1) for over 10000 cycles,and the energy density and power density of the full cells reachan ultrahigh level of 262 Wh kg−1 and 16.3 kW kg−1,respectively.Zuo Jinghan Liu Zeyu Jiang Huaning Chen Qian Yang Zhilin Gu Xiaokang Jiao Yuying Gong Yongji 2022SusMat2022,2,6:0
12PKM2-mediated neuronal hyperglycolysis enhances the risk of Parkinson's disease in diabetic rats显示文摘Epidemiological and animal studies indicate that pre-existing diabetes increases the risk of Parkinson's disease(PD).However,the mechanisms underlying this association remain unclear.In the present study,we found that high glucose(HG)levels in the cerebrospinal fluid(CSF)of diabetic rats might enhance the effect of a subthreshold dose of the neurotoxin 6-hydroxydopamine(6-OHDA)on the development of motor disorders,and the damage to the nigrostriatal dopaminergic neuronal pathway.In vitro,HG promoted the 6-OHDA-induced apoptosis in PC12 cells differentiated to neurons with nerve growth factor(NGF)(NGF-PC12).Metabolomics showed that HG promoted hyperglycolysis in neurons and impaired tricarboxylic acid cycle(TCA cycle)activity,which was closely related to abnormal mitochondrial fusion,thus resulting in mitochondrial loss.Interestingly,HG-induced upregulation of pyruvate kinase M2(PKM2)combined with 6-OHDA exposure not only mediated glycolysis but also promoted abnormal mitochondrial fusion by upregulating the expression of MFN2 in NGF-PC12 cells.In addition,we found that PKM2 knockdown rescued the abnormal mitochondrial fusion and cell apoptosis induced by HGþ6-OHDA.Furthermore,we found that shikonin(SK),an inhibitor of PKM2,restored the mitochondrial number,promoted TCA cycle activity,reversed hyperglycolysis,enhanced the tolerance of cultured neurons to 6-OHDA,and reduced the risk of PD in diabetic rats.Overall,our results indicate that diabetes promotes hyperglycolysis and abnormal mitochondrial fusion in neurons through the upregulation of PKM2,leading to an increase in the vulnerability of dopaminergic neurons to 6-OHDA.Thus,the inhibition of PKM2 and restoration of mitochondrial metabolic homeostasis/pathways may prevent the occurrence and development of diabetic PD.Ya Zhao Yanwei Wang Yuying Wu Cimin Tao Rui Xu Yong Chen Linghui Qian Tengfei Xu Xiaoyuan Lian 2023Journal of Pharmaceutical Analysis2023,13,2:0

14Facile immobilization of heparin on bioabsorbable iron via mussel adhesive protein(MAPs)显示文摘Motivated by adhesive proteins in mussels, strategies using dopamine to modified surface have become particularly attractive. In the present work, we developed a novel and convenient method to modify the biodegradable Fe plates with heparin. Iron was first treated by a facile one-step p H-induced polymerization of dopamine, and then a high density heparin was successfully grafted onto the surface via coupling with polydopamine(PDA) active layer. Heparin immobilization contributed much longer blood clotting coagulation time than the pure Fe sample, and hence reduced the risk of thrombosis. Cell viability tests suggested that the heparin modified Fe plates were more favorable to the proliferation of ECV304 cells. 15Plant-derived squalene supplementation improves growth performance and alleviates acute oxidative stress-induced growth retardation and intestinal damage in piglets显示文摘Piglets are particularly susceptible to oxidative stress,which causes inferior growth performance and intestinal damage.Squalene(SQ),a natural bioactive substance enriched in shark liver oil,shows excellent antioxidant properties and can currently be obtained at a low cost from deodorizer distillate during the production of plant oil.This study aimed to evaluate the effects of plant-derived SQ supplementation on the growth performance of piglets and explore the beneficial roles of SQ against oxidative stress and intestinal injury in diquat-challenged piglets.Forty piglets were randomly divided into five groups and fed a basal diet supplemented with SQ at 0,500,1000,or 2000 mg/kg for 5 weeks.Acute oxidative stress was induced in the piglets with diquat(10 mg/kg BW)at the fourth week of the experiment,followed by a 7-d recovery period.Results showed that before the diquat challenge,SQ supplementation significantly improved growth performance(average daily gain and feed conversion ratio)and serum antioxidant status,and after the diquat challenge,SQ supplementation significantly mitigated diquat-induced growth arrest,intestinal villous atrophy,intestinal epithelial cell apoptosis,intestinal hyperpermeability,and deficiency of intestinal epithelial tight junction proteins(zonula occludens-1,occludin,and claudin-3).Under oxidative stress induced by diquat,SQ supplementation consistently improved the antioxidant status of the small intestine,liver,and muscle.In vitro,SQ was shown to alleviate hydrogen peroxide(H2O2)-induced increase of the levels of intracellular reactive oxygen species and apoptosis of porcine intestinal epithelial cells.Taken together,SQ supplementation improves growth performance and effectively alleviates acute oxidative stress-induced growth retardation and intestinal injury via improving antioxidant capacity in piglets.Our findings may provide an efficient strategy for alleviating oxidative stress-induced inferior growth performance and intestinal damage in piglets.Junquan Tian Qian Jiang Xuetai Bao Fan Yang Yuying Li Haihui Sun Kang Yao Yulong Yin 2023Animal Nutrition2023,,4:0
