1Celastrol suppresses colorectal cancer via covalent targeting peroxiredoxin 1显示文摘As a terpenoids natural product isolated from the plant Thunder God Vine,Celastrol is widely studied for its pharmacological activities,including anti-tumor activities.The clinical application of Celastrol is strictly limited due to its severe side effects,whereas previously revealed targets and mechanism of Celastrol seldom reduce its in vivo toxicity via structural optimization.Target identification has a far-reaching influence on the development of innovative drugs,and omics data has been widely used for unbiased target prediction.However,it is difficult to enrich target of specific phenotype from thousands of genes or proteins,especially for natural products with broad promising activities.Here,we developed a text-mining-based web-server tool to enrich targets from omics data of inquired compounds.Then peroxiredoxin 1(PRDX1)was identified as the ROS-manipulating target protein of Celastrol in colorectal cancer.Our solved high-resolution crystal structure revealed the unique covalent binding mode of Celastrol with PRDX1.New derivative compound 19-048 with improved potency against PRDX1 and selectivity towards PRDX2~PRDX6 were synthesized based on crystal structure analysis.Both Celastrol and 19-048 effectively suppressed the proliferation of colorectal cancer cells.The anti-tumor efficacy of Celastrol and 19-048 was significantly diminished on xenograft nude mice bearing PRDX1 knock-down colorectal cancer cells.Several downstream genes of p53 signaling pathway were dramatically up-regulated with Celastrol or 19-048 treatment.Our findings reveal that the side effects of Celastrol could be reduced via structural modification,and PRDX1 inhibition is promising for the treatment of colorectal cancer.Heng Xu Hongfang Zhao Chunyong Ding Defang Jiang Zijie Zhao Yang Li Xiaoyu Ding Jing Gao Hu Zhou Cheng Luo Guoqiang Chen Ao Zhang Ying Xu Hao Zhang 2023Signal Transduction and Targeted Therapy2023,8,3:3
2Compact and broadband multimode waveguide bend by shape-optimizing with transformation optics显示文摘Multimode waveguide bend is one of the key components for realizing high-density mode-division multiplexing systems on chip.However,the reported multimode waveguide bends are either large,bandwidth-limited or fabrication-complicated,which hinders their applications in future high-density multimode photonic circuits.Here we propose a compact multimode waveguide bend supporting four TE modes simply by shape-optimizing with transformation optics.The shape of the waveguide is optimized in the virtual space with gradient distribution of the refractive index,so that the scattering loss and intermode cross talk are well suppressed.After conformal mapping back into the physical space,a compact(effective radius of 17μm)multimode bending waveguide is obtained.Simulations show that the proposed multimode waveguide bend has little loss(<0.1 dB)and low cross talk(<−20 dB)throughout an ultrabroad wavelength range of 1.16–1.66μm.We also fabricated the shape-optimized multimode bending waveguide on a silicon-on-insulator wafer.At 1550 nm wavelength,the measured excess losses for the four lowest-order TE modes are less than 0.6 dB,and the intermode cross talks are all below−17 dB.Our study paves the way for realizing high-density and large-scale multimode integrated optical circuits for optical interconnect.SHUYI LI LIFENG CAI DINGSHAN GAO JIANJI DONG JIN HOU CHUNYONG YANG SHAOPING CHEN XINLIANG ZHANG 2020Photonics Research2020,8,12:3

8Tectonics and Seismicity of the Yarlung Tsangpo Grand Canyon Region显示文摘The Yarlung Tsangpo Grand Canyon region is located in the frontal zone of the eastern Himalayan syntaxis, where neo-tectonics and seismicity are intensive and closely related to each other. In the region, two sets of fault structures have developed, striking NNE-NE and NWW-NW, respectively. Investigation shows that they differ markedly in terms of scope, property, active times and intensity. The NWW-NW trending faults are large in size, and most are thrust and thrust strike-slip faults, formed in earlier times. The NEE-NE-strike faults are relatively small in size individually, with concentrated distribution, constituting the NNE-trending shear extensional fault zone, which is relatively younger with evident late Quaternary activities. Strong earthquakes occur mainly in the areas or zones of intensive differential movement of the Himalayas, e.g. along the deep and large fault zones around the crustal blocks. Most earthquakes of M≥7.0 are closely related to tectonics, where large-scale Holocene active faults are distributed with complicated fault geometry, or the faults of multiple directions intersect. 9Anaerobic Fungal Diversity in the Rumen of Gayals (Bos frontalis) in Yunnan Province, China显示文摘To investigate the anaerobic fungal diversity in the rumen of gayals(Bos frontalis), Yunnan yellow cattle, yaks, and Tibetan yellow cattle, the ITS region of ruminal anaerobic fungi was sequenced. The anaerobic fungi found in four types of cattle could be divided into 16 groups(Neocallimastix, Anaeromyces, Orpinomyces, Caecomyces, Cyllamyces and unknown groups 1-11). There were 10 anaerobic fungal groups in the rumen of gayals, with three belonging to known genera(Orpinomyces, Neocallimastix, and Caecomyces), which accounted for 2.8% of sequences of gayals. There were fewer known anaerobic fungi in gayals than in Yunnan yellow cattle(Orpinomyces), yaks(Anaeromyces, Orpinomyces, and Cyllamyces) and Tibetan yellow cattle(Anaeromyces, Orpinomyces, Caecomyces, and Cyllamyces), accounting for 30.7%, 93.9%, and 35% of each of the relevant cattle species. The other seven unknown groups(NG1, NG4, NG6, NG7, NG9, NG10, and NG11) accounted for 91.6% of sequences from gayals, which was higher than Yunnan yellow cattle [69.3%(NG2, NG8 and NG9)], Tibetan yellow cattle [35%(NG4, NG5 and NG9)], and yaks [6.1%(NG3)]. Furthermore, NG1, NG6, NG7, NG10, and NG11 were only found in gayals,whereas NG3 and NG5 were found only in Tibetan yellow cattle and yak, respectively. Piromyces was not found in any of the four types of cattle. The anaerobic fungi abundance in the rumen of gayals was significantly higher than that of the other three types of cattle. 11A generic approach yields organic solar cells with enhanced efficiency and thermal stability显示文摘The use of deuterium are critical for promoting the fundamental understanding of aggregate materials and their new functions.Particularly,the solution structure of conjugated polymers can be hardly resolved without deuteration.However,studies about the isotopic effects of casting solvents on the aggregated structures of photovoltaic polymers and their bulk-heterojunction blends are deficient.Here,the impact of deuterated solvents on the thermal behavior,aggregated structures,and device performance of photovoltaic polymers is clearly delineated for the first time by multiple techniques.The enhanced π-π stacking order of photovoltaic polymers is highly relevant to their relatively poor miscibility with deuterated solvents.Benefiting from higher crystallinity and optimized morphology of deuterated solvents processed films,the devices are able to achieve better efficiency and notable improvement in thermal stability.Our results highlight the isotopic effects of solvents on the aggregated structure of conjugated polymer systems and reveal the potential of innovative approaches to fabricate thermally stable high-efficiency solar cells.Mengyuan Gao Kai Zhang Chunyong He Hanqiu Jiang Xiong Li Qingchun Qi Kangkang Zhou Yu Chen Wenchao Zhao Long Ye 2023Aggregate2023,4,2:0

13Cloning Sequencing and Bioinformatics Analysis of GRX2 and TRX2 from Puerpiao chicken and Tengchongxue chicken显示文摘[Objective] The study was to explore whether glutaredoxin( GRX2) and thioredoxin( TRX2) genes vary between Puerpiao chicken and Tengchongxue chicken in Yunnan Province. [Methods] Using Sanhuang chicken as the control,the GRX2 and TRX2 genes were cloned from skin tissues of Puerpiao chicken and Tengchongxue chicken and then subjected to bioinformatics analysis. [Results] The sequencing results of both the GRX2 and TRX2 genes were completely consistent among Puerpiao chicken,Tengchongxue chicken and Sanhuang chicken. And the sequences of GRX2 and TRX2 genes share a 100% homology with the results accessed in Gen Bank,and no any mutation was observed. 14The role of aromatic residues in controlling the supramolecular chirality of short amphiphilic peptides显示文摘Although the relationship between molecular and supramolecular chirality remains elusive,the existing results have demonstrated the vital role of hydrophilic motifs in controlling the supramolecular handedness of peptide nanofibrils compared with hydrophobic ones.However,unlike conventional hydrophobic residues,we speculate that aromatic hydrophobic residues are mostly likely to play a unique role in regulating the supramolecular handedness because theπ–πstacking interactions of their side chains are directional like hydrogen bonding and can direct high levels of self-assembly due to the geometric confining of aromatic rings.To confirm this hypothesis,we here design a series of amphiphilic short peptides,with their hydrophobic motifs being composed of aromatic residues.Their short lengths not only favor their structural stability,synthesis,and sequence variation but also enable us to readily link their molecular and supramolecular structures.Through the combination of experiments and theoretical simulations,we demonstrate that the peptides containing L-form aromatic residues form left-handed nanofibrils while those containing D-form aromatic residues assemble into right-handed ones,irrespective of the chirality of their C-terminal hydrophilic residue.Theoretical calculations revealed that the stacking of aromatic side chains betweenβ-strands directed the twisting direction of theβ-sheets formed,with L-and D-form phenylalanine side chains stacking in a clockwise and anti-clockwise way,and more ordered and stronger aromatic stacking for homochiral peptides facilitated the formation of nanofibrils with a marked tubular feature.This study has bridged the knowledge gap in our understanding of how aromatic residues affect the supramolecular chirality of short peptides.Hao Qi Kai Qi Jie Li Chunyong He Mingrui Liao Xuzhi Hu Yurong Zhao Yubin Ke Chunqiu Zhang Jun Zhang Jiqian Wang Jian R.Lu Hai Xu 2023Nano Research2023,16,10:0
