| 6 | Highly stable femtosecond pulse generation from a MXene Ti_3C_2T_x(T = F, O, or OH) mode-locked fiber laser显示文摘Ultrafast fiber lasers are in great demand for various applications, such as optical communication, spectroscopy,biomedical diagnosis, and industrial fabrication. Here, we report the highly stable femtosecond pulse generation from a MXene mode-locked fiber laser. We have prepared the high-quality Ti_3C_2 T_x nanosheets via the etching method, and characterized their ultrafast dynamics and broadband nonlinear optical responses. The obvious intensity-and wavelength-dependent nonlinear responses have been observed and investigated. In addition, a highly stable femtosecond fiber laser with signal-to-noise ratio up to 70.7 dB and central wavelength of 1567.3 nm has been delivered. | 8 | Effects of Antiretroviral Therapy and HIV Exposure in Utero on Adverse Pregnancy and Infant Outcomes:A Prospective Cohort Study in Guangzhou, China显示文摘Objective This study aimed to evaluate the effects of in-utero exposure to HIV and ART on pregnancy outcome and early growth of children.Methods This cohort study enrolled 802 HIV-infected pregnant women between October 2009 and May 2018 in Guangzhou, China. The women were assigned to receive combination ART(c ART) or mono/dual ART or no treatment. The primary outcomes were the combined endpoints of any adverse pregnancy outcome [including ectopic pregnancy, spontaneous abortion, stillbirth, preterm birth, small for gestational age(SGA)] and adverse early growth outcome(including infant death, HIV infection of mother-to-child transmission, and underweight, wasting and stunting of infants at 4 weeks of age).Results Adverse pregnancy outcomes occurred in 202(35.1%) of all enrolled HIV-infected women, and121(31.3%) of all infants exhibited adverse effects on early growth at 4 weeks of age. The rates of adverse pregnancy outcomes, spontaneous abortion, ectopic pregnancy, stillbirth, infant death and perinatal HIV infection were higher among women not receiving ART, compared to those treated with c ART or mono/dual ART(P < 0.05). However, women treated with c ART had a higher rate of SGA,compared to untreated women(P < 0.05). | 11 | Construction and prokaryotic expression of the fusion protein Stx2B-IntiminC300 of EHEC O157: H7 and its immunoprophylactic potential显示文摘To construct and express the fusion protein Stx2B-IntiminC300 of EHEC 0157 :H7, and to further investigate its immunoprophyiactic potential, the gene of Stx2B (stx2b) from EHEC 0157:H7 chromosome was cloned into pMD18-T vector. Thereafter, the amplified gene was cloned into prokaryotic expression plasmid pET-28a (+)-eaeC300, which was constructed previously. The recombinant pasmid pET-28a(+)-stx2b-eaeC300 was transformed into E. coli BL21(DE3). After inducement, the protein Stx2B-IntiminC300 was successfully expressed and analyzed with sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), Western blotting and N-terminal amino acid residual sequencing. To evaluate its immunoprophyiactic potential, it was primarily purified by ion-exchange chromatography and injected into 30 BALB/c mice with Al(OH)3 in the subscapular region. Ten days after the last booster vaccination, 20 mice were attacked with EHEC 0157: H7 lysate and the protective efficacy was observed. In the present study, the gene of Stx2B-IntiminC300 was successfully cloned into pET-28a (+) vector. The results of SDS-PAGE and Western blotting assay showed that the fusion protein was successfully expressed in the inclusion body form, accounting for 25% of total expression products, and its molecular weight was about 43 kDa. The result of the N-terminal amino acid residual sequencing showed that it was identical to that of the molecular designed. The purity was about 75 % after primary purification. Animal tests revealed that the fusion protein Stx2B-IntiminC300 has elicited high titer of protective antibody relatively. | 14 | Repeat HIV Testing and Incident Rates among Individuals Attending Voluntary Counseling and Testing Clinics in Wuxi, China: A Retrospective Study显示文摘Objective We aimed to elucidate the rates of repeat HIV testing and incident HIV diagnosis,and baseline CD^(4+) T cell count among individuals attending HIV voluntary counseling and testing(VCT) clinics in Wuxi,China.Methods A repeat HIV testing within 12 months was recorded if individuals had their first test with negative results,during 2013-2014 and retested within 12 months.An incident HIV diagnosis was recorded if individuals had their first test with negative results,during 2013-2015 and had a subsequent positive result at any point by the end of 2015.Data on HIV testing and diagnosis among individuals attending 32 VCT clinics from 2013 to 2015 and HIV diagnosis from other clinical services in Wuxi,China,were retrieved.A multivariate logistic regression model was used to analyze factors associated with repeat HIV testing.Cox regression was used to evaluate factors associated with incident HIV diagnosis.Results From 2013 to 2014,11,504 individuals tested HIV negative at their first recorded test,with 655(5.7%) retesting within 12 months.Higher repeat HIV testing within 12 months was associated with male gender [adjusted odds ratio(a OR) = 1.7,95% confidence interval(CI): 1.4-2.2],risk behaviors [commercial heterosexual behaviors(a OR = 1.4,CI: 1.1-1.6),male-male sexual behaviors(a OR = 3.7,CI: 2.7-4.9)],injection drug use(a OR = 9.9,CI: 6.5-15.1),and having taken HIV tests previously(a OR = 2.0,CI: 1.6-2.4).From 2013 to 2015,1,088 individuals tested negative on HIV test at their visit and at ≥ 2 subsequent tests; of them 30 had incident HIV diagnosis.The overall rate of incident HIV diagnosis among all VCT individuals was 1.6(95% CI: 1.1-2.1) per 100 person-years.Incident HIV diagnosis was associated with male gender [adjusted hazard ratio(a HR) = 8.5,95% CI: 1.9-38.1],attending hospital-based VCT clinics(a HR = 7.8,95% CI: 1.1-58.3),and male-male sexual behavior(a HR = 8.4,95% CI: 1.5-46.7).Individuals diagnosed at VCT clinics had higher CD^(4+) T cell count compared with those diagnosed at other clinical services(median 407 vs.326 copies/mm^3,P = 0.003).Conclusion VCT individuals in Wuxi,China,had a low repeat HIV testing rate and high HIV incidence.VCT-clinic-based interventions aimed at increasing repeat HIV testing are needed to detect more cases at an earlier stage,especially among individuals at high risk for HIV infection such as men who have sex with men. | MENG Xiao Jun Andrew GRULICH WANG Xu Wen YIN Han Lu GU Jing ZHANG Xuan ZOU Hua Chun | 2018 | Biomedical and Environmental Sciences2018，31，1： | 1 |
