1Numerical simulation of sandy bed erosion by 2D vertical jet显示文摘A two-dimensional numerical model of vertical jet scour was developed based on the turbulent flow theory and jet scour mechanism. In this model,drag force acts as the main reference variable and the critical Shields number acts as the incipient motion criteria of sediment. The morphological change in the bed caused by bed-load transport is simulated using the moving mesh method and the suspended-load is not considered. An experiment investigating vertical clear water jet scour was conducted in the laboratory,and some effective experimental results,such as flow patterns and distribution characteristics of scour pits,were obtained. Numerical simulation of the 2D jet scour was conducted using the same parameters as were used in the flume experiment. The evolution process of the jet scour observed in the experiment was simulated by the new model;validation of the numerical model and the algorithm was conducted. 4An antibody against Siglec-15 promotes bone formation and fracture healing by increasing TRAP^(+)mononuclear cells and PDGF-BB secretion显示文摘Osteoporosis(OP)is a common age-related disease characterized by a deterioration of bone mass and structure that predisposes patients to fragility fractures.Pharmaceutical therapies that promote anabolic bone formation in OP patients and OP-induced fracture are needed.We investigated whether a neutralizing antibody against Siglec-15 can simultaneously inhibit bone resorption and stimulate bone formation.We found that the multinucleation of osteoclasts was inhibited in SIGLEC-15 conditional knockout mice and mice undergoing Siglec-15 neutralizing antibody treatment.The secretion of platelet-derived growth factor-BB(PDGF-BB),the number of tartrate-resistant acid phosphatase-positive(TRAP+)mononuclear cells,and bone formation were significantly increased in the SIGLEC-15 conditional knockout mice and antibody-treated mice.The anabolic effect of the Siglec-15 neutralizing antibody on bone formation was blunted in mice with Pdgfb deleted in TRAP-1'cells.These findings showed that the anabolic effect of the Siglec-15 neutralizing antibody was mediated by elevating PDGF-BB production of TRAP4 mononuclear cells.To test the therapeutic potential of the Siglec-15 neutralizing antibody,we injected the antibody in an ovariectomy-induced osteoporotic mouse model,which mimics postmenopausal osteoporosis in women,and in two fracture healing models because fracture is the most serious health consequence of osteoporosis.The Siglec-15 neutralizing antibody effectively reduced bone resorption and stimulated bone formation in estrogen deficiency-induced osteoporosis.Of note,the Siglec-15 neutralizing antibody promoted intramembranous and endochondral ossification at the damaged area of cortical bone in fracture healing mouse models.Thus,the Siglec-15 neutralizing antibody shows significant translational potential as a novel therapy for OP and bone fracture.Gehua Zhen Yang Dan Ruomei Wang Ce Dou Qiaoyue Guo Melissa Zarr Linda N.Liu Lieping Chen Ruoxian Deng Yusheng Li Zengwu Shao Xu Cao 2021Bone Research2021,9,4:6

6Kindlin-2 inhibits Nlrp3 inflammasome activation in nucleus pulposus to maintain homeostasis of the intervertebral disc显示文摘Intervertebral disc(IVD) degeneration(IVDD) is the main cause of low back pain with major social and economic burdens;however, its underlying molecular mechanisms remain poorly defined. Here we show that the focal adhesion protein Kindlin-2 is highly expressed in the nucleus pulposus(NP), but not in the anulus fibrosus and the cartilaginous endplates, in the IVD tissues. Expression of Kindlin-2 is drastically decreased in NP cells in aged mice and severe IVDD patients. Inducible deletion of Kindlin-2 in NP cells in adult mice causes spontaneous and striking IVDD-like phenotypes in lumbar IVDs and largely accelerates progression of coccygeal IVDD in the presence of abnormal mechanical stress. Kindlin-2 loss activates Nlrp3 inflammasome and stimulates expression of IL-1β in NP cells, which in turn downregulates Kindlin-2. This vicious cycle promotes extracellular matrix(ECM) catabolism and NP cell apoptosis. Furthermore, abnormal mechanical stress reduces expression of Kindlin-2, which exacerbates Nlrp3 inflammasome activation, cell apoptosis, and ECM catabolism in NP cells caused by Kindlin-2 deficiency. In vivo blocking Nlrp3 inflammasome activation prevents IVDD progression induced by Kindlin-2 loss and abnormal mechanical stress. Of translational significance, adeno-associated virus-mediated overexpression of Kindlin-2 inhibits ECM catabolism and cell apoptosis in primary human NP cells in vitro and alleviates coccygeal IVDD progression caused by mechanical stress in rat. 7Multi-objective evolutionary design of selective triple modular redundancy systems against SEUs显示文摘To improve the reliability of spaceborne electronic systems,a fault-tolerant strategy of selective triple modular redundancy(STMR)based on multi-objective optimization and evolvable hardware(EHW)against single-event upsets(SEUs)for circuits implemented on field programmable gate arrays(FPGAs)based on static random access memory(SRAM)is presented in this paper.Various topologies of circuit with the same functionality are evolved using EHW firstly.Then the SEU-sensitive gates of each circuit are identified using signal probabilities of all the lines in it,and each circuit is hardened against SEUs by selectively applying triple modular redundancy(TMR)to these SEU-sensitive gates.Afterward,each circuit hardened has been evaluated by SEU Simulation,and the multi-objective optimization technology is introduced to optimize the area overhead and the number of functional errors of all the circuits.The proposed fault-tolerant strategy is tested on four circuits from microelectronics center of North Carolina(MCNC)benchmark suite.The experimental results show that it can generate innovative trade-off solutions to compromise between hardware resource consumption and system reliability.The maximum savings in the area overhead of the STMR circuit over the full TMR design is 58%with the same SEU immunity.Yao Rui Chen Qinqin Li Zengwu Sun Yanmei 2015Chinese Journal of Aeronautics2015,28,3:6
8Comparison of lymph node number and prognosis in gastric cancer patients with perigastric lymph nodes retrieved by surgeons and pathologists显示文摘Objective: To compare the numbers of positive and total lymph nodes and prognosis in gastric cancer patients whose perigastric lymph node retrieval was performed by surgeons and pathologists.Methods: We conducted a retrospective analysis of clinical and follow-up data from 1,056 patients who underwent gastric cancer D2 radical lymph node resection between January 2008 and December 2010 in the Gastrointestinal Surgery Department of Yantai Yuhuangding Hospital.The follow-up ended in December 2015.Patients were divided into two groups according to the specialty of physicians who performed the postoperative perigastric lymph node retrieval: the surgeon group(475 cases) and the pathologist group(581 cases).The numbers of positive and total perigastric lymph nodes and the 3- and 5-year survival were compared between gastric cancer patients in the two groups overall and stratified by TNM stage(AJCC 7th Edition).Results: Overall, the numbers of positive and total lymph nodes were significantly higher in the surgeon group than in the pathologist group(6.53±4.07 vs.4.09±3.70, P=0.021; 29.64±11.50 vs.20.71±8.56, P<0.001).Further analysis showed that the total number of lymph nodes in stage Ⅰ patients(19.40±9.62 vs.15.45±8.59, P=0.011) and the numbers of positive and total lymph nodes in stage Ⅱ(1.38±1.08 vs.0.87±1.55, P=0.031; 25.35±10.80 vs.16.75±8.56, P<0.001) and stage Ⅲ patients(8.11±6.91 vs.6.66±5.12, P=0.026; 32.34±12.55 vs.25.45±8.31, P<0.001)were significantly higher in the surgeon group than in the pathologist group.The survival analysis showed that the3- and 5-year survival of stage Ⅱ and Ⅲ patients was significantly higher in the surgeon group than in the pathologist group(82.0% vs.73.1%, 69.5% vs.61.2%, P=0.038; 49.2% vs.38.9%, 36.3% vs.28.0%; P=0.045).Conclusions: Compared with retrieval performed by pathologists, postoperative perigastric lymph node retrieval performed by surgeons was associated with significant increase in the total lymph node number of stage Ⅰ patients, the numbers of positive and total lymph nodes of stage Ⅱ and Ⅲ patients, and the survival of stage Ⅱ and stage Ⅲ gastric cancer patients.Lixin Jiang Zengwu Yao Yifei Zhang Jinchen Hu Dawei Zhao Huiyuan Zhai Xixun Wang Zhenbin Zhang Dong Wang 2016Chinese Journal of Cancer Research2016,28,5:5

10Retrieval of Aerosol Optical Depth for Chongqing Using the HJ-1 Satellite Data显示文摘Aerosol optical depth(AOD) is a common indicator applied in monitoring aerosols in the atmosphere. The hilly landscape and rapid economic growth of the megacity Chongqing have facilitated increased aerosol concentration,and it is meaningful to accurately retrieve AOD over Chongqing. The HJ-1A/B satellite of China carries a sensor/camera called the Charge Coupled Device(CCD), the spatial resolution of which meets the requirement for retrieving high resolution AOD. In this paper, analysis of the AOD retrievals from different methods using the HJ-1satellite data revealed the most suitable algorithm. Through comparison with the AOD product of Moderate Resolution Imaging Spectroradiometer(MODIS), the AOD retrieval results using enhanced vegetation index(EVI) to estimate dark pixels showed the highest correlation. The continental aerosol model was used to build a lookup table that was able to facilitate a good AOD retrieval for both city and rural areas. Finally, the algorithm that combined dark pixels, buffer areas, and the deep blue algorithm was found to be most suitable for AOD retrieval. The AOD retrieval results based on the HJ-1 data were consistent with MODIS products, and our algorithm yields reasonable results in most cases. The results were also compared with ground-based PM10 measurements synchronized with the overpass time of the HJ-1 satellite, and high correlation was found. 12Three-dimensional biofabrication of an aragoniteenriched self-hardening bone graft substitute and assessment of its osteogenicity in vitro and in vivo显示文摘A self-hardening three-dimensional(3D)-porous composite bone graft consisting of 65 wt%hydroxyapatite(HA)and 35 wt%aragonite was fabricated using a 3D-Bioplotter®.New tetracalcium phosphate and dicalcium phosphate anhydrous/aragonite/gelatine paste formulae were developed to overcome the phase separation of the liquid and solid components.The mechanical properties,porosity,height and width stability of the end products were optimised through a systematic analysis of the fabrication processing parameters including printing pressure,printing speed and distance between strands.The resulting 3D-printed bone graft was confirmed to be a mixture of HA and aragonite by X-ray diffraction,Fourier transform infrared spectroscopy and energy dispersive X-ray spectroscopy.The compression strength of HA/aragonite was between 0.56 and 2.49 MPa.Cytotoxicity was assessed using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay in vitro.The osteogenicity of HA/aragonite was evaluated in vitro by alkaline phosphatase assay using human umbilical cord matrix mesenchymal stem cells,and in vivo by juxtapositional implantation between the tibia and the anterior tibialis muscle in rats.The results showed that the scaffold was not toxic and supported osteogenic differentiation in vitro.HA/aragonite stimulated new bone formation that bridged host bone and intramuscular implants in vivo.We conclude that HA/aragonite is a biodegradable and conductive bone formation biomaterial that stimulates bone regeneration.Since this material is formed near 37°C,it will have great potential for incorporating bioactive molecules to suit personalised application;however,further study of its biodegradation and osteogenic capacity is warranted.The study was approved by the Animal Ethical Committee at Tongji Medical School,Huazhong University of Science and Technology(IACUC No.738)on October 1,2017.Yunsong Shi Ruijun He Xiangyu Deng Zengwu Shao Davide Deganello Chunze Yan Zhidao Xia 2020Biomaterials Translational2020,1,1:2
13Proper animal experimental designs for preclinical research of biomaterials for intervertebral disc regeneration显示文摘Low back pain is a vital musculoskeletal disease that impairs life quality,leads to disability and imposes heavy economic burden on the society,while it is greatly attributed to intervertebral disc degeneration(IDD).However,the existing treatments,such as medicines,chiropractic adjustments and surgery,cannot achieve ideal disc regeneration.Therefore,advanced bioactive therapies are implemented,including stem cells delivery,bioreagents administration,and implantation of biomaterials etc.Among these researches,few reported unsatisfying regenerative outcomes.However,these advanced therapies have barely achieved successful clinical translation.The main reason for the inconsistency between satisfying preclinical results and poor clinical translation may largely rely on the animal models that cannot actually simulate the human disc degeneration.The inappropriate animal model also leads to difficulties in comparing the efficacies among biomaterials in different reaches.Therefore,animal models that better simulate the clinical charateristics of human IDD should be acknowledged.In addition,in vivo regenerative outcomes should be carefully evaluated to obtain robust results.Nevertheless,many researches neglect certain critical characteristics,such as adhesive properties for biomaterials blocking annulus fibrosus defects and hyperalgesia that is closely related to the clinical manifestations,e.g,low back pain.Herein,in this review,we summarized the animal models established for IDD,and highlighted the proper models and parameters that may result in acknowledged IDD models.Then,we discussed the existing biomaterials for disc regeneration and the characteristics that should be considered for regenerating different parts of discs.Finally,well-established assays and parameters for in vivo disc regeneration are explored.Yizhong Peng Xiangcheng Qing Hongyang Shu Shuo Tian Wenbo Yang Songfeng Chen Hui Lin Xiao Lv Lei Zhao Xi Chen Feifei Pu Donghua Huang Xu Cao Zengwu Shao 2021Biomaterials Translational2021,2,2:2

15Influence on lacustrine source rock by hydrothermal fluid: a case study of the Chang 7 oil shale, southern Ordos Basin显示文摘Hydrothermal fluid activity during sedimentation of the Triassic Yanchang Formation in the Ordos Basin and the impact of said activity on formation and preservation conditions of source rocks have received little attention. Oil yield, major element, trace element, rare earth element, and total sulfur(TS) data from the oil shale within the Yanchang are here presented and discussed in the context of hydrothermal influence. Oil shale samples returned relatively high total organic carbon(TOC), in the range of 4.69%–25.48%. A high correlation between TS and TOC suggests TS in the oil shale is dominated by organic sulfur and affected by organic matter. The low Al/Si ratio of oil shale samples implies quartz is a major mineralogical component. Si/(Si + Al + Fe) values suggest close proximity of the oil shale to a terrigenous source.d Eu; Fe versus Mn versus(Cu + Co + Ni) 9 10; and Si O2/(K2 O + Na2 O) versus Mn O/Ti O2, Fe/Ti, and(Fe + Mn)/Ti are evidence of hydrothermal fluid activity during oil shale sedimentation, and d U and U/Th of the oil shale indicate reducing conditions. The Sr/Ba of oil shale samples suggests fresh-water deposition. The high correlations of Fe/Ti and(Fe + Mn)/Ti with d U, U/Th, and TS demonstrate that hydrothermal fluid activity promotes reducing conditions. Sr/Ba ratios had low correlation with Fe/Ti and(Fe + Mn)/Ti, implying that hydrothermal fluid activity had little impact on paleosalinity. 16Elemental characteristics of lacustrine oil shale and its controlling factors of palaeo-sedimentary environment on oil yield: a case from Chang 7 oil layer of Triassic Yanchang Formation in southern Ordos Basin显示文摘As an important unconventional resource, oil shale has received widespread attention. The oil shale of the Chang 7 oil layer from Triassic Yanchang Formation in Ordos Basin represents the typical lacustrine oil shale in China. Based on analyzing trace elements and oil yield from boreholes samples, characteristics and paleo-sedimentary environments of oil shale and relationship between paleo-sedimentary environment and oil yield were studied. With favorable quality, oil yield of oil shale varies from 1.4% to 9.1%. Geochemical data indicate that the paleo-redox condition of oil shale's reducing condition from analyses of V/Cr, V/(V + Ni), U/Th, d U, and authigenic uranium. Equivalent Boron, Sp, and Sr/Ba illustrate that paleosalinity of oil shale is dominated by fresh water.The paleoclimate of oil shale is warm and humid by calculating the chemical index of alteration and Sr/Cu. Fe/Ti and(Fe + Mn)/Ti all explain that there were hot water activities during the sedimentary period of oil shale. In terms of Zr/Rb, paleohydrodynamics of oil shale is weak.By means of Co abundance and U/Th, paleo-water-depth of oil shale is from 17.30 to 157.26 m, reflecting sedimentary environment which is mainly in semi deep–deep lake facies.
