| 1 | Essential roles of exosome and circRNA_101093 on ferroptosis desensitization in lung adenocarcinoma显示文摘Background:Resistance to ferroptosis,a regulated cell death caused by irondependent excessive accumulation of lipid peroxides,has recently been linked to lung adenocarcinoma(LUAD).Intracellular antioxidant systems are required for protection against ferroptosis.The purpose of the present studywas to investigate whether and how extracellular system desensitizes LUAD cells to ferroptosis.Methods:Established human lung fibroblasts MRC-5,WI38,and human LUAD H1650,PC9,H1975,H358,A549,and H1299 cell lines,tumor and matched normal adjacent tissues of LUAD,and plasma from healthy individuals and LUAD patients were used in this study.Immunohistochemistry and immunoblotting were used to analyze protein expression,and quantitative reverse transcription-PCR was used to analyze mRNA expression.Cell viability,cell death,and the lipid reactive oxygen species generationwere measured to evaluate the responses to ferroptosis.Exosomes were observed using transmission electron microscope.The localization of arachidonic acid(AA)was detected using click chemistry labeling followed by confocal microscopy.Interactions between RNAs and proteins were detected using RNA pull-down,RNA immunoprecipitation and photoactivatable ribonucleoside-enhanced crosslinking and immunoprecipitation methods.Proteomic analysis was used to investigate RNA-regulated proteins,and metabolomic analysis was performed to analyze metabolites.Cellderived xenograft,patient-derived xenograft,cell-implanted intrapulmonary LUAD mouse models and plasma/tissue specimens from LUAD patients were used to validate the molecular mechanism.Results:Plasma exosome from LUAD patients specifically reduced lipid peroxidation and desensitized LUAD cells to ferroptosis.A potential explanation is that exosomal circRNA_101093(cir93)maintained an elevation in intracellular cir93 in LUAD to modulate AA,a poly-unsaturated fatty acid critical for ferroptosisassociated increased peroxidation in the plasma membrane.Mechanistically,cir93 interacted with and increased fatty acid-binding protein 3(FABP3),which transported AA and facilitated its reaction with taurine.Thus,global AA was reduced,whereas N-arachidonoyl taurine(NAT,the product of AA and taurine)was induced.Notably,the role of NAT in suppressing AA incorporation into the plasma membrane was also revealed.In pre-clinical in vivo models,reducing exosome improved ferroptosis-based treatment.Conclusion:Exosome and cir93 are essential for desensitizing LUAD cells to ferroptosis,and blocking exosome may be helpful for future LUAD treatment. | Xiao Zhang Yunhua Xu Lifang Ma Keke Yu Yongjie Niu Xin Xu Yi Shi Susu Guo Xiangfei Xue Yikun Wang Shiyu Qiu Jiangtao Cui Hong Wang Xiaoting Tian Yayou Miao Fanyu Meng Yongxia Qiao Yongchun Yu Jiayi Wang | 2022 | Cancer Communications2022,42,4: | 10 |

| 4 | Crack-free single-crystalline Co-free Ni-rich LiNi_(0.95)Mn_(0.05)O_(2) layered cathode显示文摘The rapid growth in global electric vehicles(EVs)sales has promoted the development of Co-free,Ni-rich layered cathodes for state-of-the-art high energy-density,inexpensive lithium-ion batteries(LIBs).However,progress in their commercial use has been seriously hampered by exasperating performance deterioration and safety concerns.Herein,a robust single-crystalline,Co-free,Ni-rich LiNi_(0.95)Mn_(0.05)O_(2)(SC-NM95)cathode is successfully designed using a molten salt-assisted method,and it exhibits better structural stability and cycling durability than those of polycrystalline LiNi_(0.95)Mn_(0.05)O_(2) (PC-NM95).Notably,the SC-NM95 cathode achieves a high discharge capacity of 218.2 mAh g^(-1),together with a high energy density of 837.3 Wh kg^(-1) at 0.1 C,mainly due to abundant Ni^(2+)/Ni^(3+) redox.It also presents an outstanding capacity retention(84.4%)after 200 cycles at 1 C,because its integrated single-crystalline structure effectively inhibits particle microcracking and surface phase transformation.In contrast,the PC-NM95 cathode suffers from rapid capacity fading owing to the nucleation and propagation of intergranular microcracking during cycling,facilitating aggravated parasitic reactions and rocksalt phase accumulation.This work provides a fundamental strategy for designing high-performance singlecrystalline,Co-free,Ni-rich cathode materials and also represents an important breakthrough in developing high-safe,low-cost,and high-energy LIBs. | Lianshan Ni Ruiting Guo Susu Fang Jun Chen Jinqiang Gao Yu Mei Shu Zhang Wentao Deng Guoqiang Zou Hongshuai Hou Xiaobo Ji | 2022 | eScience2022,2,1: | 2 |

| 10 | Mefatinib as first-line treatment of patients with advanced EGFR-mutant non-small-cell lung cancer:a phase Ib/II efficacy and biomarker study显示文摘EGFR inhibitors have revolutionized the treatment of advanced non-small-cell lung cancer(NSCLC).Mefatinib is a novel,bioavailable,second-generation,irreversible pan-EGFR inhibitor.This phase Ib/II open-label,single-arm,multi-center study investigated the efficacy,safety,biomarker,and resistance mechanisms of mefatinib in the first-line treatment of patients with advanced EGFR-mutant NSCLC.This study included 106 patients with EGFR-mutant stage IIIB-IV NSCLC who received first-line mefatinib at a daily dose of either 60 mg(n=51)or 80 mg(n=55).The primary endpoint was progression-free survival(PFS).Secondary endpoints were overall response rate(ORR),disease control rate(DCR),overall survival(OS),and safety.The cohort achieved an ORR of 84.9%and DCR of 97.2%.The median PFS was 15.4 months and the median OS was 31.6 months.Brain metastasis was detected in 29%of patients(n=31)at diagnosis and demonstrated an ORR of 87.1%,PFS of 12.8 months,and OS of 25.2 months.Adverse events primarily involved skin and gastrointestinal toxicities,which were well-tolerated and manageable.Analyses of mutation profiles were performed using targeted sequencing of plasma samples at baseline,first follow-up 6 weeks from starting mefatinib therapy(F1),and at progression.Patients with concurrent TP53 mutations had comparable PFS as wild-type TP53(14.0 vs 15.4 months;p=0.315).Furthermore,circulating tumor DNA clearance was associated with longer PFS(p=0.040)and OS(p=0.002).EGFR T790M was the predominant molecular mechanism of mefatinib resistance(42.1%,16/38).First-line mefatinib provides durable PFS and an acceptable toxicity profile in patients with advanced EGFR-mutant NSCLC. | Pingli Wang Yuping Li Dongqing Lv Lingge Yang Liren Ding Jianya Zhou Wei Hong Youfei Chen Dongqing Zhang Susu He Jianying Zhou Kai Wang | 2021 | Signal Transduction and Targeted Therapy2021,6,12: | 1 |

| 16 | Efficient synthesis of cyclic carbonates under atmospheric CO_(2) by DMAP-based ionic liquids: the difference of inert hydrogen atom and active hydrogen atom in cation显示文摘The coupling reaction of carbon dioxide(CO_(2)) and epoxides is one of the most efficient pathways to achieve the carbon balance.However,to accomplish it under the mild conditions,especially under the atmospheric pressure,is still a perplexing problem.Three novel ionic liquids(ILs),[DMAPBrPC][TMGH],[DMAPBrPC][DBUH],and[DMAPBrPC][BTMA],are designed and synthesized.All of them display the excellent catalytic activity for the title reaction achieving the yield over 96.6% under the atmospheric CO_(2) pressure at 60℃.Interestingly,[DMAPBrPC][BTMA]with the inert hydrogen atom in cation exhibits the superior catalytic activity as compared to other two ILs with the protic hydrogen atom in cation along with the same anion.The active hydrogen atom in[DMAPBrPC][TMGH]and[DMAPBrPC][DBUH]would impede the–COO^(-)group to absorb CO 2,which is an unfavorable item for the reaction.Moreover,the strong hydrogen bond in[DMAPBrPC][TMGH]and[DMAPBrPC][DBUH]would lessen the nucleophilic ability of Br^(-) anion resulting in the inferior catalytic performance,which is further confirmed by the density functional theory(DFT)calculations.The cation without the active hydrogen atom could also be employed to design the ILs with the excellent catalytic feature when it is combined with the suitable anion. | Zhengkun Zhang Jinya Li Guanyao Yu Chao Zeng Menglong Wang Susu Huang Li Wang Jinglai Zhang | 2023 | Green Chemical Engineering2023,4,3: | 0 |
