| 1 | Identification and primary genetic analysis of Arabidopsis stomatal mutants in response to multiple stresses显示文摘In response to variable environmental conditions, guard cells located in the leaf epidermis can integrate and cope with a multitude of complicated stimuli, thereby making stomata in an appro- priate state. However, many signaling components in guard cell signaling remain elusive. In our laboratory, a tool for non-invasive remote infrared thermal images was used to screen an ethyl methane sulfonate-mutagenized population for Arabidopsis stomatal response mutants under multiple stresses (ABA, H2O2, CO2, etc.). More than forty 'hot' or 'cold' mutants were isolated (above or below 0.5℃ in con- trast to normal plantlets). Identification and primary genetic analysis of these mutants show that they are monogenic recessive mutations and there exist distinct difference in stomata apertures compared to wild type. | 5 | The Chinese herbal formula Tongluo Jiunao promotes expression of brain-derived neurotrophic factor/tropomyosin-related kinase B pathways in a rat model of ischemic brain injury显示文摘The neurotrophin-Trk receptor pathway is an intrinsic pathway to relieve damage to the central nervous system. The present study observed the effects of Tongluo Jiunao (TLJN), which comprises Panax Notoginseng and Gardenia Jasminoides, on expression of brain-derived neurotrophic factor (BDNF) and tropomyosin-related kinase B (TrkB) in a rat model of focal cerebral ischemic injury. Xue Sai Tong (XST), comprising Panax Notoginseng, served as the positive control. Mechanisms of neuroprotection were analyzed following TLJN injection. Following establishment of the middle cerebral artery occlusion models, TLJN and XST were intraperitoneally injected, and 2, 3, 5-triphenyltetrazolium chloride staining results revealed that TLJN injection reduced infarct volume, suggesting that TLJN exerted a neuroprotective effect. Enzyme-linked immunosorbent assay results showed that TLJN elevated BDNF and growth associated protein-43 expression in ischemic brain tissues, as well as serum BDNF levels. Reverse-transcription polymerase chain reaction and western blot results showed that TLJN injection did not affect TrkB expression in the ischemic brain tissues of rats. These results suggested that TLJN injection reduced damage to ischemic brain tissues and increased BDNF expression. | 6 | Development of a double antibodies sandwich ELISA for the detection of avian leukosis virus subgroup J based on monoclonal antibodies against gp85显示文摘Avian leukosis virus(ALV)belongs to the genus Alpharetrovirus of the family Retroviridae and can cause various types of neoplastic diseases,even mortality in infected chickens[1].ALV is categorized into seven subgroups,including A,B,C,D,E,J,and K,based on the pathogenicity and antibody neutralization characteristics[2].The subgroup E is endogenous and transmitted by Mendelian inheritance.The subgroups A,B,C,D,J,and K are exogenous and oncogenic,transmitted horizontally and vertically and caused different pathological lesions[3].Among them,subgroups A and B were most common in commercial poultry and subgroup J is the major cause for myeloid leukosis and nephromas in broilers.In 1980,ALV subgroup J(ALV-J)was first isolated and identified from infected broilers in Britain.It caused significant economic losses in the poultry industry because of increased myelocytomas and other tumors[4].In China,ALV-J was first detected in broilers in 1999 and,subsequently,spread to other chicken types.Compared to other ALV subgroups,ALV-J has been a serious issue in China poultry industry since 2007.Currently,there is no effective vaccine and drugs against ALV-J infection;thus,early detection and elimination of ALV-positive breeder chickens are key measures to block vertical and horizontal ALV-J transmission.Given the importance of ALV to the poultry industry,many ALV detection methods,including virus isolation,immunofluorescence assays,neutralization tests,reverse transcription polymerase chain reaction(PCR)or real-time fluorescent quantitative PCR-based molecular diagnosis ways,and enzyme-linked immunosorbent assays(ELISA)have been developed.Among them,ELISA is widely used for its efficiency,convenience,and sensitivity.Yun et al.[5]developed an antigen-capture ELISA for the detection of ALV based on highly conserved antigen p27.Yu et al.[6]exploited a colloidal gold test strip specific for the detection of ALV p27 antigen.These antigen capture-based serological detection methods share obviously simple,accurate,and rapid advantages in comparison with conventional time-consuming methods of indirect immunofluorescence assay or neutralization tests.However,it is difficult to identify different subgroups of ALV using p27-coated antibodies. | Lili Cao Panpan Zhao He Ding Nan Wang Xue Zhang Shuxian Yuan Hang Dong Yanbing Guo Xinhua Yao Qinlei Yu Hongze Shao Pengtao Gong | 2021 | Acta Biochimica et Biophysica Sinica2021,53,10: | 1 |
| 7 | Colon hydrotherapy plus Traditional Chinese Medicine to treat non-alcoholic fatty liver disease:a pilot study显示文摘OBJECTIVE: To demonstrate the potential to treat non-alcoholic fatty liver disease(NAFLD) with colon hydrotherapy(CHT) plus Traditional Chinese Medicine(TCM).METHODS: A total of 20 patients were enrolled into the study and received CHT with TCM for 2 weeks.Body mass index(BMI) and levels of serum triglycerides(TG) and total cholesterol(TC) were compared between pre-treatment and post-treatment.RESULTS: Two-week treatment with CHT plus TCM significantly lowered BMI and reduced blood lipids.BMI decreased from 29.5 ± 4.3 to 25.4 ± 1.0, while mean TG levels decreased by 0.70 mmol/L on average from baseline and mean TC levels decreased by0.37 mmol/L. Forty-five percent of patients exhibited TC decreasing by more than 10% from baseline and25%ofpatientsexhibited TCdecreasingbymore than 20%. Sixty percent of patients exhibited TG decreasing by more than 20% and 20% of patients exhibited TG decreasing by more than 40%. However,high-density and low-density lipoprotein cholester-ol levels did not change significantly after intervention. The calculation results indicated that different spinel phases were produced after the interactions between the slag and three types of refractory materials( 10Cr2O3- 90Al2O3,50Cr2O3- 50Al2O3 and 90Cr2O3- 10Al2O3) at 1 500℃. Increased contents of Al2O3 and Cr2O3in the remained liquid slag after interaction implied the dissolution of refractory. Thermodynamic modeling predicted that the process of dissolution might lead to much deeper penetration into the refractory materials by the present compositional slag. | 15 | In vitro leishmanicidal activity of antimicrobial peptide KDEL against Leishmania tarentolae显示文摘Leishmaniasis,caused by the intracellular protozoan parasite Leishmania,remains an important neglected tropical infectious disease.Infection may be lethal if untreated.Currently,the available drugs for the disease are limited by high toxicity and drug resista nee.There is an urgent n eed to develop novel anti-leishmanial strategies.Antimicrobial peptides(AMPs)have been described as the first-line immune defense against pathogenic microbes and are being developed as emerging anti-parasitic therapies.In the present study,we showed the anti-leishmanial activity of the synthetic 4-amino acid peptide lysine,aspartic acid,glutamic acid,and leucine(KDEL),the endoplas?mic reticulum retention sequence,against Leishmania tarentolae promastigote and amastigote.Different concentrations of KDEL peptides were incubated with promastigotes,MTT viability assay,and promastigote assay were carried out.Macrophages infected with GFP-transfected L.tarentolae promastigotes were incubated with KDEL peptides,and the anti-amastigote activity of the KDEL peptides was measured by fluoresce nee microscopy.The damage of L.tarentolae was observed by light microscopy and electron microscopy.The cell apoptosis was analyzed using the Annexin V-FITC/PI apoptosis detection kit and mitochondrial membrane potential assay kit and by flow cytometry.Results showed that L.tarentolae was susceptible to KDEL peptides in a dosedepende nt manner,and KDEL peptides disrupted the surface membrane integrity and caused cell apoptosis.In our study,we found for the first time an AMP KDEL from Pseudomonas aeruginosa and proved its significant therapeutic potential as a novel anti-leishmanial drug. | Lili Cao Weina Jiang Songgao Cao Panpan Zhao Juan Liu Hang Dong Yanbing Guo Quan Liu Pengtao Gong | 2019 | Acta Biochimica et Biophysica Sinica2019,51,12: | 0 |
| 16 | Structural,physicochemical,antioxidant and in vitro digestibility properties of banana flours from different banana varieties(Musa spp.)显示文摘Green bananas occupy an important position in the diet of Tanzanians with potential beneficial health effects.Banana flours prepared from five Tanzania banana varieties(Mzuzu,Malindi,Mshale,Bukoba,and Moshi)were compared for the proximate composition,structure,physicochemical properties,antioxidant activities,and in vitro digestibility properties.The total starch content of banana flours was 58.01%-68.74%,and the amylose content was 16.67%-23.11%.All the samples exhibited B-type crystals.Bukoba had the highest pasting temperature and peak viscosity,and Malindi had the lowest breakdown.The peak temperature(Tp)and the enthalpy change(ΔH)of samples differed from 65.99◦C to 67.25℃ and from 7.98 to 12.59 J/g,respectively.The total polyphenol content and total flavonoid content in banana flour were positively correlated with DPPH and FRAP values,respectively.Bukoba and Moshi presented the stronger antioxidant activities among the varieties.Banana flours with the high resistant starch(RS)content(89.20%-90.86%of total starch)were highly resistant to enzyme digestion.Banana flour can be considered as a potential source of bioactive components.The varying physicochemical properties of banana flours from different varieties could broaden their application as formulations in food and nonfood industries and promote the rational utilization of banana resources of Tanzania. | Lei Chang Min Yang Ning Zhao Fei Xie Pengtao Zheng Jonatha Simbo Xiuzhu Yu Shuang-kui Du | 2022 | Food Bioscience2022,47,3: | 0 |
| 17 | Hot deformation behavior of a novel bimetal consisting of BTW1 and Q345R characterized by processing maps显示文摘Only a few studies have been conducted on the flow behavior of the novel BTW1/Q345R bimetal,which is widely used in coal equipment.In this work,compression tests were conducted on BTW1/Q345R bimetal at a temperature range of 950°C–1200°C and strain rates of 0.05,0.5,5,and 15 s^−1 by using a Gleeble-3800 thermomechanical simulator.A constitutive equation was validated by referring to the Arrhenius equation during the characterization of hot workability.The computed apparent activation energy of the BTW1/Q345R bimetal was 360 kJ/mol,and processing maps under different strain conditions were drawn.Analysis of the stress-strain relationship revealed that work hardening exerted a dominant effect on the thermal deformation of the BTW1/Q345R bimetal.The processing maps predicted that the optimal processing interval will increase with strain.Results showed that thermal deformation of the BTW1/Q345R bimetal should proceed when the temperature range varies from 1182°C to 1200°C and the strain rate interval is from 4.2 to 15 s^−1. | Pengtao LIU Lifeng MA Weitao JIA Tao WANG Guanghui ZHAO | 2019 | Frontiers of Mechanical Engineering2019,14,4: | 0 |
