2Sintilimab versus docetaxel as second-line treatment in advanced or metastatic squamous non-small-cell lung cancer:an open-label,randomized controlled phase 3 trial(ORIENT-3)显示文摘Background:Treatment options for Chinese patients with locally advanced or metastatic squamous-cell non-small-cell lung cancer(sqNSCLC)after failure of first-line chemotherapy are limited.This study(ORIENT-3)aimed to evaluate the efficacy and safety of sintilimab versus docetaxel as second-line treatment in patients with locally advanced or metastatic sqNSCLC.Methods:ORIENT-3 was an open-label,multicenter,randomized controlled phase 3 trial that recruited patients with stage IIIB/IIIC/IV sqNSCLC after failure with first-line platinum-based chemotherapy.Patients were randomized in a 1:1 ratio to receive either 200 mg of sintilimab or 75 mg/m^(2) of docetaxel intravenously every 3 weeks,stratified by the Eastern Cooperative Oncology Group performance status.The primary endpoint was overall survival(OS)in the full analysis set(FAS).Secondary endpoints included progression-free survival(PFS),objective response rate(ORR),disease control rate(DCR),duration of response(DoR)and safety.Results:Between August 25,2017,and November 7,2018,290 patients were randomized.For FAS,10 patients fromthe docetaxel armwere excluded.Themedian OS was 11.79(n=145;95%confidence interval[CI],10.28-15.57)months with sintilimab versus 8.25(n=135;95%CI,6.47-9.82)months with docetaxel(hazard ratio[HR]:0.74;95%CI,0.56-0.96;P=0.025).Sintilimab treatment significantly prolonged PFS(median 4.30 vs.2.79 months;HR:0.52;95%CI,0.39-0.68;P<0.001)and showed higher ORR(25.50%vs.2.20%,P<0.001)and DCR(65.50%vs.37.80%,P<0.001)than the docetaxel arm.The median DoRwas 12.45(95%CI,4.86-25.33)months in the sintilimab arm and 4.14(95%CI,1.41-7.23)months in the docetaxel arm(P=0.045).Treatment-related adverse events of grade≥3were reported in 26(18.1%)patients in the sintilimab arm and 47(36.2%)patients in the docetaxel arm.Exploratory biomarker analysis showed potential predictive values of expression levels of two transcription factors,including OVOL2(HR:0.35;P<0.001)and CTCF(HR:3.50;P<0.001),for sintilimab treatment.Conclusions:Compared with docetaxel,sintilimab significantly improved the OS,PFS,and ORR of Chinese patients with previously treated locally advanced or metastatic sqNSCLC.Yuankai Shi Lin Wu Xinmin Yu Puyuan Xing Yan Wang Jianying Zhou Airong Wang Jianhua Shi Yi Hu Ziping Wang Guangyu An Yong Fang Sanyuan Sun Caicun Zhou Changli Wang Feng Ye Xingya Li Junye Wang Mengzhao Wang Yunpeng Liu Yanqiu Zhao Ying Yuan Jifeng Feng Zhendong Chen Jindong Shi Tao Sun Gang Wu Yongqian Shu Qisen Guo Yi Zhang Yong Song Shucai Zhang Yuan Chen Wei Li Hongrui Niu Wenwei Hu Lijun Wang Jianan Huang Yang Zhang Ying Cheng Zhengdong Wu Bo Peng Jiya Sun Christoph Mancao Yanqi Wang Luyao Sun 2022Cancer Communications2022,42,12:8
3Dominant Gene cplsr1 Corresponding to Premature Leaf Senescence Resistance in Cotton (Gossypium hirsutum L.)显示文摘Cotton (Gossypium hirsutum L.) premature leaf senescence-resistant inbred XLZ33 and senescence-susceptible inbred lines XLZ13 were selected and crossed to produce F1, F1-reciprocal, F2 and BC1 generations for evaluation of leaf senescence process and inheritance. The results showed that leaf senescence processes for XLZ13 and XLZ33 were obviously different and leaf senescence traits could be distinguished between the two parents at particular periods of cotton growth. Inheritance anlysis for the cotton premature leaf senescence resistant trait further showed that the segregation in the F2 fit a 3:1 ratio inheritance pattern, with resistance being dominant. The backcross of F1 to the susceptible parent produced a 1:1 ratio, confirming that cotton premature leaf senescence resistant trait was from a single gene. 4Pollen-based Holocene quantitative temperature reconstruction on the eastern Tibetan Plateau using a comprehensive method framework显示文摘Quantitative climate reconstruction on long timescales can provide important insights for understanding the climate variability and providing valuable data for simulations. Unfortunately, the credibility of some attempts was hampered by incomplete reconstruction procedures. We here establish a comprehensive framework resting on high-quality Chinese modern pollen database, including modern pollen data screening, calibration set selection, major climate factor analysis, appropriate model selection, strict statistical assessment of results and ecological interpretation. The application of this framework to three high-resolution pollen records from the eastern Tibetan Plateau allows accurate quantitative inferences of Holocene temperature changes, which is the major control of regional vegetation. The results show that the mean warmest month temperature(MTwa)during the early Holocene was ca. 10.4℃ and reached the highest value at 8.5–6 ka BP(ca. 11℃). The early and mid-Holocene(11–5 ka BP) warmth was followed by 1.2℃ temperature decrease, culminating in the coolest temperatures of the Holocene during the Neoglacial cooling. Superimposing on the general cooling trend, MTwareveals a significant 500-yr periodicity with varying intensities through time, showing that warm(cold) intervals are in phase with solar maxima(minima) periods. 5Ischemic postconditioning enhances glycogen synthase kinase-3β expression and alleviates cerebral ischemia/reperfusion injury显示文摘The present study established global brain ischemia using the four-vessel occlusion method.Following three rounds of reperfusion for 30 seconds,and occlusion for 10 seconds,followed by reperfusion for 48 hours,infarct area,the number of TUNEL-positive cells and Bcl-2 expression were significantly reduced.However,glycogen synthase kinase-3β activity,cortical Bax and caspase-3 expression significantly increased,similar to results following ischemic postconditioning.Our results indicated that ischemic postconditioning may enhance glycogen synthase kinase-3β activity,a downstream molecule of the phosphatase and tensin homolog deleted on chromosome 10/phosphatidylinositol 3-kinase/protein kinase B signaling pathway,which reduces caspase-3 expression to protect the brain against ischemic injury.Bo Zhao Wenwei Gao Jiabao Hou Yang Wu Zhongyuan Xia 2012Neural Regeneration Research2012,7,19:2

7An Impedance-based Parameter Design Method for Active Damping of Load Converter Station in MTDC Distribution System显示文摘To achieve the efficient application of impedance analysis in the stability assessment and enhancement of multiterminal DC distribution systems, this paper proposes the DCside reduced-order impedance models with power control and AC voltage control, respectively, by taking the load converter station as the object. By using the DC-side current as the feedforward state, the active compensator applied to the load converter station with two control modes is also derived as well as the corresponding reduced-order impedance models. Combined with the reduced-order impedance models, a method based on damping factor sensitivity is further proposed to design the parameters of the derived active compensators. 8nvasive Spartina and reduced sediments: Shanghai’s dangerous silver bullet显示文摘Aims This synthesis paper is developed to provide a summary of ecological,socioeconomic challenges facing the estuarine wetlands within the Yangtze River delta.Methods We combined literature review of the estuarine wetlands and ground measurements of sedimentation,vegetation,and carbon fluxes to illustrate the foreseeable crises in managing these wetlands that play a critical role in Shanghai’s urban development.Where the Yangtze River meets the Pacific Ocean,4.153108 mg/year of suspended sediments are deposited along mainland and island shorelines of the 40000 km2 delta-resulting in an average growth rate of land outwards 64 m/year since 1951.However,completion of the Three Gorges Dam in 2003,and earlier dam projects,reduced the rates of sedimentation and growth of the islands.To meet the increasing demands for lands and agriculture,policymakers have attempted to enlarge the islands by diking coastal areas and introducing Spartina alterniflora-a grass native to tidal salt marshes of the southeastern USA but exotic to China.Spartina is one of the 16 greatest invasive species listed by the State Environmental Protection Administration of China.Successful plantations and rapid spread of this species have increased the production and fertility of the coast,but at the cost of native ecosystems.We outline the social,economic,and ecological controversies related to this land management strategy in the context of global warming.Important findings Combinations of these changes,including sea level rise,and alterations to storm patterns and long-shore currents,with the continued spread of Spartina,human population growth,and river flow and sediment reduction will make current management untenable.Jiquan Chen Bin Zhao Wenwei Ren Sari C.Saunders Zhijun Ma Bo Li Yiqi Luo Jiakuan Chen 2008Journal of Plant Ecology2008,1,2:2

18Therapeutic safety evaluation of sodium glycididazole combined with ^(131)I radiotherapy for differentiated thyroid carcinoma显示文摘To evaluate safety and therapeutic efficacy of sodium glycididazole (CMNa) combined with 131I radiotherapy for differentiated thyroid carcinoma (DTC),the 60 patients of DTC therapeutic protocols were selected and divided into 3 groups of the DTC 4.44 GBq 131I,DTC 3.70 GBq 131I,and combination of DTC 3.70 GBq 131I with CMNa,and the 20 patients of Graves' Disease were selected as the control group.Peripheral blood was sampled at 131I pre-treatment of 1 day,and 131I post-treatment of 7,91,and 182 days,thus analyzing lymphocyte micronucleus scores and karyotyping profiles.Compared with the control group,the lymphocyte micronucleus and chromosome mutation rates in 131I treated DTC increased after post-treatment of 7 days,recovered after post-treatment of 91 days,and did not bounce after post-treatment of 182 days.The micronucleus and chromosome mutation rates in the combination of DTC 3.70 GBq 131I with CMNa showed less significant variation than other treated DTC groups.Our results demonstrate that micronucleus assay and karyotyping analysis are favorable to evaluate 131I radiotherapy for DTC.The combination of the CMNa with 131I radiotherapy was safe for DTC patients without affecting the long-term therapeutic outcomes.SUN Wenwei GU Xinquan GAO Shi BAI Lin CUI Qu MA Qingjie Wang Qing Zhao Guoqing 2011Nuclear Science and Techniques2011,22,4:0
19Optimal operating strategy of short turning lines for the battery electric bus system显示文摘Operating short turning line is an efficient strategy to satisfy the unevenly distributed demand during peak periods while reducing operational cost.However,for the battery electric bus(BEB)system,the application of the strategy is challenging due to the disadvantages of BEBs,such as limited driving mileage and long charging time.Improper vehicle configuration and charging scheduling may dramatically increase the operational cost and cut the benefits of these strategies.In this work,we propose a general framework to design an effective short turning strategy for the BEB system at a tactical planning level.First,the trade-off relationship between the battery capacity and the average trip time is identified by modeling the BEBs operations.Second,a microeconomic model is formulated to jointly optimize the frequencies and charging schedules of the whole bus line and the short turning line,to effectively minimize passengers’waiting time and operational cost.Finally,numerical experiments have been carried out for an illustrative linear line to demonstrate the potential benefits of the sub-line operating strategy compared with the normal operation.Wenwei Zhang Hui Zhao Min Xu 2021Communications in Transportation Research2021,1,1:0
20Expression of Invertase Genes in Virus-free Sugarcane显示文摘Virus-free sugarcane seedlings have improved biomass and sucrose content compared with ordinary seedlings,and sucrose invertases are key enzymes regulating sugarcane growth and sucrose accumulation.In this study,the differences in the expression levels of 3 invertase genes,CWI,SAI and NI,between virusfree and ordinary sugarcane seedlings were analyzed.Compared with ordinary sugarcane plants,the expression of CWI was mainly up-regulated in immature leaves and stems at elongation stage and leaves and immature internodes at maturation stage,and especially,greatly up-regulated in immature internodes at maturation stage of virus-free plants.The expression of SAI and NI were mainly up-regulated in leaves and immature internodes of virus-free plants at maturation stage,which might be beneficial to sugar accumulation and rapid utilization of monosaccharide in the stalks of virus-free plants.It is further indicated that virus-free treatment could significantly improve the expression of sucrose invertases at late growth period,and might facilitate the increase of plant biomass.Jungang WANG Tingting ZHAO Benpeng YANG Wenwei CAI Cuilian FENG Jun ZENG Guoru XIONG Shuzhen ZHANG 2017Agricultural Biotechnology2017,6,4:0
