1Characterization of Hand,Foot,and Mouth Disease in China between 2008 and 2009显示文摘Objective To investigate the epidemiological and clinical features of hand,foot and mouth disease (HFMD) since several outbreaks of HFMD caused by enteroviruses were documented in China between 2007 and 2008.Methods HFMD cases reported to the National Infectious Disease Information Management System database between May 2008 and April 2009 were assessed.Clinical features in some of the severe and fatal cases were analyzed the etiology of the outbreaks was investigated.Results 89.1% of reported HFMD cases were found in children<5 year‐old with an age‐specific incidence rate of 834.1/100 000 in the first year as the notifiable disease in China from May 2008 to April 2009.The incidence,mortality and percentage of severe cases were studied for three regions of China and found to be highest in the central region.The incidence of severe cases and mortality in rural population were significantly higher than those in urban population.Among the laboratory confirmed EV17 positive cases there were 52.6% mild,83.5% severe,and 96.1% fatal cases.More myoclonic jerks were found in the severe case group than in group that died.Tachypnea,lip purpling,pink foaming and low limb temperature occurred more frequently in the fatal cases than in the severe cases.Conclusion The epidemic of HFMD in China was characterized predominantly by EV71 infections,had relatively high mortality rates especially in the central region,and was most prevalent in young,rural populations.ZHANG Jing SUN JunLing CHANG ZhaoRui ZHANG WeiDong WANG ZiJun FENG ZiJian 2011Biomedical and Environmental Sciences2011,24,3:74
2BH3-only proteins target BCL-xL/MCL-1,not BAX/BAK,to initiate apoptosis显示文摘It has been widely accepted that mitochondria-dependent apoptosis initiates when select BH3-only proteins(BID,BIM,etc.)directly engage and allosterically activate effector proteins BAX/BAK.Here,through reconstitution of cells lacking all eight pro-apoptotic BH3-only proteins,we demonstrate that all BH3-only proteins primarily target the anti-apoptotic BCL-2 proteins BCL-xL/MCL-1,whose simultaneous suppression enables membrane-mediated spontaneous activation of BAX/BAK.BH3-only proteins*apoptotic activities correlate with affinities for BCL-xL/MCL1 instead of abilities to directly activate BAX/BAK.Further,BID and BIM do not distinguish BAX from BAK or accelerate BAX/BAK activation following inactivation of BCL-xL/MCL-1.Remarkably,death ligandinduced apoptosis in cells lacking BH3-only proteins and MCL-1 is fully restored by BID mutants capable of neutralizing BCL-xL,but not direct activation of BAX/BAK.Taken together,our findings provide a'Membrane-mediated Permissive'model,in which the BH3-only proteins only indirectly activate BAX/BAK by neutralizing the anti-apoptotic BCL-2 proteins,and thus allowing BAX/BAK to undergo unimpeded,spontaneous activation in the mitochondrial outer membrane milieu,leading to apoptosis initiation.Kai Huang Katelyn L.O'Neill Jian Li Wei Zhou Na Han Xiaming Pang Wei Wu Lucas Struble Gloria Borgstahl Zhaorui Liu Liqiana Zhang Xu Luo 2019Cell Research2019,29,11:16

4肝癌乙型肝炎病毒X抗原相关基因的克隆显示文摘目的 观察HepG2细胞因乙型肝炎病毒X抗原 (HBx)转染导致的基因表达差异。 方法①用逆转录病毒感染的方法建立表达乙型肝炎病毒X抗原 (HepG2X)及氯霉素乙酰转移酶 (HepG2Cat)的细胞模型。②用抑制差减杂交的方法做HepG2X和对照细胞 (HepG2Cat)的cDNA文库差示筛选。③用3 2 P随机引物标记法标记探针做NorthernBlot杂交及地高辛缺口翻译标记法做原位分子杂交以筛选基因探针。结果 经cDNA文库差减杂交及抑制性PCR扩增共得到 10个cDNA探针 ,其中 8个被HBx所启动 ,2个因HBx而表达下降。经检测 ,这些基因在HepG2X和对照细胞中的表达均有差异。在被HBx所抑制的 2个差示表达基因中 ,1个与人的蛋白翻译起始因子同源 ,同时发现 ,此基因在正常组织中表达较强 ,而在肿瘤组织中表达被抑制。结论 HBx能够改变宿主细胞的基因表达 。刘杰 LIAN Zhaorui PAN Jinbo Clayton M 胡家露 朱明华 窦科峰 丁杰 吴开春 樊代明 Feitelson M 2000中华消化杂志2000,20,3:7

7Characteristics and risk assessment of agricultural meteorological disasters based on 30 years’disaster data from Heilongjiang Province of China显示文摘To determine the influence of agricultural meteorological disasters on agriculture in Heilongjiang Province,the disaster areas associated with different types of disasters and their variation characteristics were analyzed based on the statistical data of agricultural disasters from 1983 to 2013 in Heilongjiang Province,China.The moving average and the Mann-Kendall test were applied to identify the variation trends of drought,flooding,hailstorms and freezing(based on the disaster ratio and the disaster intensity index).Then,the Morlet wavelet analysis method was used to identify the periodicity of these four kinds of agricultural meteorological disasters.Finally,a fuzzy comprehensive evaluation method was adopted to analyze the degrees of agricultural loss induced by these disasters.The following results were obtained:1)The disaster ratio and disaster intensity index for drought exhibited increasing trends;the disaster ratio and disaster intensity index for flooding exhibited decreasing trends;for hailstorms,the disaster ratio exhibited no obvious trend of change,whereas the disaster intensity index exhibited an increasing trend;and for freezing,the disaster ratio also exhibited no obvious trend of change,whereas the disaster intensity index exhibited a decreasing trend.2)Mutation points were observed in the disaster ratio series for drought,flooding and hailstorms,whereas no mutation point was evident in the disaster ratio series for freezing.3)Multiple time-scale characteristics were observed in the disaster ratio series for all four types of agricultural meteorological disasters.Furthermore,the disaster ratio series for the different types of disasters had different main periodicities.4)From the perspective of the degree of agricultural loss induced by each type of disaster,drought was identified as the most severe type of agricultural meteorological disaster,followed by flooding,freezing,and hailstorms.The degree of agricultural loss caused by each type of disaster was different during different periods.Finally,based on the results,several strategies were identified for mitigating the effect of agricultural meteorological disasters in Heilongjiang Province.Xing Zhenxiang Yang Zhaorui Fu Qiang Li Heng Gong Xinglong Wu Jingyan 2017International Journal of Agricultural and Biological Engineering2017,10,6:4
8Compatibility of rubber stoppers for recombinant antitumor-antivirus protein injection by gas chromatography-mass spectrometry显示文摘A simple, rapid, and sensitive gas chromatography-mass spectrometry (GC-MS) method was developed and validated for the simultaneous determination of two fatty acids, methyl hexadecanoate (MH) and methyl stearate (MS), to allow the evaluation of packaging-drug compatibility. The two migrants were quantified in selective ion-monitoring (SIM) mode, with limits of detection (LOD) of 0.0030 μg/mL and 0.0121 μg/mL. Linear calibration curves for MH and MS were obtained in the concentration ranges of 0.1011–5.0570 μg/mL and 0.2015–10.0740 μg/mL, respectively. The developed method was successfully applied to estimate the safety of the injection of recombinant antitumor-antivirus protein (RAAP). The results showed that the possible maximum daily intake was 3.0 ng and 12.1 ng for MH and MS, respectively. 9High expression of human serum albumin in milk of trans-genie mice directed by the goat β-casein gene promoter region显示文摘We have constructed a mammary gland expression vector that contained the goat β-casein gene pro-moter, 5’upstream regulatory region, exons 1, 2, intron 1 as well as the human serum albumin (hALB) mini-gene (including the full-long sequences of hALB cDNA and its intron 1). Injection of the vector into mouse tail veins showed that the recombinant construct was expressed only in mammary glands. The vector was microinjected into the mouse fertilized eggs, followed by transferring the eggs into the foster mice. 33 F0 mice were obtained. Of the 33, 8 mice (5 , 3 ) were transgenic with hALB gene integration identified by PCR as well as Southern blot hybridization. The integration rate was 24.2% (8/33). Western blot analysis showed that 3 female transgenic mice had hALB expression in their milk. 10Anodic Oxidation on Structural Evolution and Tensile Properties of Polyacrylonitrile Based Carbon Fibers with Different Surface Morphology显示文摘Polyacrylonitrile (PAN) based carbon fibers with different surface morphology were electrochemically treated in 3 wt% NH4HCO3 aqueous solution with current density up to 3.47 A/m 2 at room temperature, and surface structures, surface morphology and residual mechanical properties were characterized. The crystallite size (La) of carbon fibers would be interrupted due to excessive electrochemical etching, while the crystallite spacing (d(002)) increased as increasing current density. The disordered structures on the surface of carbon fiber with rough surface increased at the initial oxidation stage and then removed by further electrochemical etching, which resulting in continuous increase of the extent of graphitization on the fiber surface. However, the electrochemical etching was beneficial to getting ordered morphology on the surface for carbon fiber with smooth surface, especially when the current density was lower than 1.77 A/m 2 . The tensile strength and tensile modulus could be improved by 17.27% and 5.75%, respectively, and was dependent of surface morphology. 12Reactivity of hydrogen species on oxide surfaces显示文摘Hydrogen species on oxides are widely involved in oxides-catalyzed reactions such as H_2/hydrocarbon oxidation, hydrogenation/dehydrogenation, water-gas shift, and water-splitting reactions. Thus identifications of hydrogen species on oxide surfaces and their reactivity are important for fundamental understanding of these oxides-catalyzed reactions. In this Feature Article, we briefly review our research progress on the reactivity of various hydrogen species on oxides, including surface hydroxyl species,hydride species and hydrated protons. We have successfully developed effective strategies of using gas-phase atomic H to controllably create oxygen vacancies and prepare various hydrogen species on oxide model catalysts under ultra-high vacuum(UHV) conditions and using well-defined oxide nanocrystals with different surface structures and oxygen vacancy concentrations to study the H_2-oxide interaction under ambient or even higher H_2 pressures. Reactivity of various hydrogen species on oxide surfaces has been identified, including local oxygen vacancy-controlled reactivity of OH species, oxygen vacancystabilized hydride species, homolytic dissociation of H2 at oxygen vacancies of reduced oxide surfaces into hydride species accompanied by surface oxidation, photoexcited holes-stimulated desorption of hydride species, electron-stimulated desorption of hydride and OH species, and photoexcited electrons-stimulated desorption of hydrated protons. 13Faceted Kurdjumov-Sachs interface-induced slip continuity in the eutectic high-entropy alloy,AlCoCrFeNi_(2.1)显示文摘Recently,the eutectic high-entropy alloy(EHEA),AlCoCrFeNi_(2.1),can reach a good balance of strength and ductility.The dual-phase alloy exhibits a eutectic lamellar microstructure with large numbers of interfaces.However,the role of the interfaces in plastic deformation have not been revealed deeply.In the present work,the orientation relationship(OR)of the interfaces has been clarified as the Kurdjumov-Sachs(KS)interfaces presenting〈111〉_(B2) 〈110〉_(FCC)and {110} _(B2){111}_(FCC) independent of their morphologies.There exist three kinds of interfaces in the EHEA,namely,The dominating interface and the secondary interface are both non-slip planes and atomistic-scale faceted,facilitating the nucleation and slip transmission of the dislocations.The formation mechanism of the preferred interfaces is revealed using the atomistic geometrical analysis according to the criteria of the low interfacial energy based on the coincidence-site lattice(CSL)theory.In particular,the ductility of the dual-phase alloy originates from the KS interface-induced slip continuity across interfaces,which provides a high slip-transfer geometric factor.Moreover,the strengthening effect can be attributed to the interface resistance for the dislocation transmission due to the mismatches of the moduli and lattice parameters at the interfaces.Ting Xiong Wenfan Yang Shijian Zheng Zhaorui Liu Yiping Lu Ruifeng Zhang Yangtao Zhou Xiaohong Shao Bo Zhang Jun Wang Fuxing Yin Peter K.Liaw Xiuliang Ma 2021Journal of Materials Science & Technology2021,,6:2
14The Mesenchymal Stem Cells Derived from Transgenic Mice Carrying Human Coagulation Factor Ⅷ Can Correct Phenotype in Hemophilia A Mice显示文摘Hemophilia A(HA)is an inherited X-linked recessive bleeding disorder caused by coagulant factor Ⅷ(FⅧ)deficiency.Previous studies showed that introduction of mesenchymal stem cells(MSCs)modified by FⅧ-expressing retrovirus may result in phenotypic correction of HA animals.This study aimed at the investigation of an alternative gene therapy strategy that may lead to sustained FⅧ transgene expression in HA mice.B-domain-deleted human FⅧ(hFⅧBD)vector was microinjected into single-cell embryos of wild-type mice to generate a transgenic mouse line,from which hFⅧBD-MSCs were isolated,followed by transplantation into HA mice.RT-PCR and real-time PCR analysis demonstrated the expression of hFⅧBD in multi-organs of recipient HA mice.Immunohistochemistry showed the presence of hFⅧBD positive staining in multi-organs of recipient HA mice.ELISA indicated that plasma hFⅧBD level in recipient mice reached its peak(77 ng/mL)at the 3rd week after implantation,and achieved sustained expression during the 5-week observation period.Plasma FⅧ activities of recipient HA mice increased from 0%to 32%after hFⅧBD-MSCs transplantation.APTT(activated partial thromboplastin time)value decreased in hFⅧBD-MSCs transplanted HA mice compared with untreated HA mice(45.5 s vs.91.3 s).Our study demonstrated an effective phenotypic correction in HA mice using genetically modified MSCs from hFⅧBD transgenic mice.Qing Wang Xiuli Gong Zhijuan Gong Xiaoyie Ren Zhaorui Ren Shuzhen Huang Yitao Zeng 2013Journal of Genetics and Genomics2013,40,12:2
15Expression of biologically active human clotting factor Ⅸ(hFⅨ) in the mammary gland of transgenic mice显示文摘The DNA of human factor Ⅸ (hFⅨ) gene vector pMCⅨm, which had been proven to be able to express in in vitro and living cells, was introduced into 586 zygotes of Kunming White Mice by positive pressure microinjection technique with manual operation. The 499 survival embryos after microinjection were then transferred into pseudopregnant recipient mice and 216 F 0 pups were born. The analysis of PCR and Southern blot hybridization showed that, of the 216, 6 (2 females and 4 males) were integrated with foreign DNA in their genomes, giving an integration frequency of 3% (6/216). Two F\-0 female transgenic mice could express hFⅨ protein in their milk and the content was over 100 ng/mL as measured with ELISA. 16Study on gas–liquid flow characteristics in stirred tank with dual-impeller based on CFD-PBM coupled model显示文摘Study on gas–liquid flow in stirred tank with two combinations of dual-impeller(six-bent-bladed turbine(6BT)+six-inclined-blade down-pumping turbine(6 ITD),the six-bent-bladed turbine(6BT)+six-inclinedblade up-pumping turbine(6ITU))was conducted using computational fluid dynamics(CFD)and population balance model(PBM)(CFD-PBM)coupled model.The local bubble size was captured by particle image velocimetry(PIV)measurement.The gas holdup,bubble size distribution and gas–liquid interfacial area were explored at different conditions through numerical simulation.The results showed that the 4 mm bubbles accounted for the largest proportion of 33%at the gas flow rates Q=0.76 m^(3)·h^(-1) and 22%at Q=1.52 m^(3)·h^(-1) for combined impeller of 6BT+6ITU,while the bubbles of 4.7 mm and 5.5 mm were the largest proportion for 6BT+6ITD combination,i.e.25%at Q=0.76 m^(3)·h^(-1) and 22%at Q=1.52 m^(3)·h^(-1),respectively,which indicated that 6BT+6ITU could reduce bubble size effectively and promote gas dispersion.In addition,the gas holdup around impellers was increased obviously with the speed compared with gas flow rate.So it was concluded that 6ITU impeller could be more conductive to the bubble dispersion with more uniform bubble size,which embodied the advantages of 6BT+6ITU combination in gas–liquid mixing.Songsong Wang Qiuxiang Bu Deyu Luan Ying Zhang Longbin Li Zhaorui Wang Wenhao Shi 2021Chinese Journal of Chemical Engineering2021,34,10:2
