| 2 | Generation of seed lipoxygenase-free soybean using CRISPR-Cas9显示文摘Beany flavor induced by three lipoxygenases(LOXs, including LOX1, LOX2, and LOX3)restricts human consumption of soybean. It is desirable to generate lipoxygenase-free new mutant lines to improve the eating quality of soybean oil and protein products. In this study, a pooled clustered regularly interspaced short palindromic repeats(CRISPR)-CRISPRassociated protein 9(Cas9) strategy targeting three GmLox genes(GmLox1, GmLox2, and GmLox3) was applied and 60 T_0 positive transgenic plants were generated, carrying combinations of sg RNAs and mutations. Among them, GmLox-28 and GmLox-60 were gmlox1 gmlox2 gmlox3 triple mutants and GmLox-40 was a gmlox1 gmlox2 double mutant.Sequencing of T_1 mutant plants derived from GmLox-28, GmLox-60, and GmLox-40 showed that mutation in the GmLox gene was inherited by the next generation. Colorimetric assay revealed that plants carrying different combinations of mutations lost the corresponding lipoxygenase activities. Transgene-free mutants were obtained by screening the T_2 generation of lipoxygenase-free mutant lines(GmLox-28 and GmLox-60). The essence of catalytic activity is revealed to relate with electronic coupling between catalysts and charged species. Based on adsorption energy, some activity descriptors such as d-band center, e_g-electron number and charge-transfer capacity are further defined by electronic properties of catalysts that directly affect interaction between catalysts and charged species. | 6 | Estimation of tree transpiration and response of tree conductance to meteorological variables in desert-oasis system of Northwest China显示文摘Transpirations of three dominated tree species, namely Mongol Scotch Pine (Pinus sylvestris var. mongolica Litvin), White elm (Ulmus pumila) and Gansu Poplar (Populus gansuensis Wang et Yang) in oasis shelter forest (Linze site) and of two dominated tree species,namely Euphrates Poplar (Populus euphratica Oliv.) and Russia olive (Elaeagnus angustifolia Linn.) in lowland desert (Erjinaqi site) have been estimated using measured sapflow in summer,autumn and winter, 2002 and in spring, 2003. An ENVIS System was used for each site to measure microclimate variables, soil moisture and sapflow every half an hour, and the study time scale is one day. In the 104 days of observation during the growing season at the Linze site, the average daily sapflow of Gansu Poplar is 9.93L·d-1,and the average transpiration per unit leaf area is 1.99mm·d-1.For White elm tree,the daily average sapflow is 4.08L·d-1,while the daily average transpiration per unit leaf area is 0.49mm·d-1.The values for Mongol Scotch Pine are 3.91L·d-1 and 0.25mm·d-1,respectively.In the total 73 days of observation during the growing season at the Erjinaqi site, the daily average sapflows of Russia olive and Euphrates Poplar are 12.1 and 20.97L·d-1,respectively,and the average transpirations per unit leaf area are 0.22 amd 0.31mm·d-1,respectively.In the observation period of the growing season,tree conductances of Mongol Scotch Pine, White elm, Gansu Poplar or Russia olive show an exponential relationship with the daily average air temperature or vapour pressure deficit, but the relationship is not so obvious between tree conductance and global radiation. The transpiration process of each tree species is affected by all the observed four environmental variables. The response of tree conductance to different climatic factors changes with tree species. The effect of the same factor to the same tree species is also variable in different growing stages. The sapflow of every tree species is relatively large in later spring to early summer, and low in summer, and then reaches its largest value in later September. In the mid-November, the sapflow is relatively large, especially the deciduous tree species. | 7 | The Bethesda System for Reporting Thyroid Cytopathology(TBSRTC):A report of 2,781 cases in a Chinese population显示文摘Objective:To evaluate the diagnostic utility of The Bethesda System for Reporting Thyroid Cytology(TBSRTC)at Peking University Cancer Hospital,the incidence of noninvasive follicular thyroid neoplasms with papillary-like nuclear features(NIFTP),and the impact of reclassification on cytopathological outcomes.Methods:We performed a retrospective study of thyroid fine-needle aspiration(FNA)cases between April 2014 and March 2019.The FNA results were classified according to TBSRTC.Post-surgery histological findings were followed up.Results:A total of 2,781 thyroid FNAs were performed.The incidences of the 6 diagnostic categories(DCs I-VI)were 14.8%,17.1%,15.8%,2.3%,11.6%and 38.5%,respectively.A total of 1,122 patients(40.3%)had corresponding histological results.NIFTP accounted for 0.4%of papillary thyroid carcinoma(PTC)cases,and there was no significant difference in the risk of malignancy(ROM)for each TBSRTC DC between'NIFTP=carcinoma(Ca)'and'NIFTP≠Ca'.When'NIFTP=Ca',the sensitivity,specificity,positive predictive value(PPV),negative predictive value(NPV),and accuracy of TBSRTC were 98.0%,84.0%,99.4%,58.3%,and97.5%,respectively.When'NIFTP≠Ca',the sensitivity,specificity,PPV,NPV and accuracy of the TBSRTC were 98.1%,81.5%,99.3%,61.1%,and 97.5%,respectively.Conclusions:TBSRTC is effective in the preoperative diagnosis of thyroid nodules in Peking University Cancer Hospital.The impact of the reclassification of NIFTP on cytopathological outcomes is limited because of its low incidence,and the revised ROMs are not suitable for Asian patients. | Yanli Zhu Yuntao Song Guohui Xu Zhihui Fan Wenhao Ren | 2020 | Chinese Journal of Cancer Research2020,32,2: | 4 |
| 8 | Ectopic expression of VRT-A2 underlies the origin of Triticum polonicum and Triticum petropavlovskyi with long outer glumes and grains显示文摘Polish wheat (Triticum polonicum) is a unique tetraploid wheat species characterized by an elongated outer glume. The genetic control of the long-glume trait by a single semi-dominant locus, P1 (from Polish wheat), was established more than 100 years ago, but the underlying causal gene and molecular nature remain elusive. Here, we report the isolation of VRT-A2, encoding an SVP-clade MADS-box transcription factor, as the P1 candidate gene. Genetic evidence suggests that in T. polonicum, a naturally occurring sequence rearrangement in the intron-1 region of VRT-A2 leads to ectopic expression of VRT-A2 in floral organs where the long-glume phenotype appears. Interestingly, we found that the intron-1 region is a key ON/OFF molecular switch for VRT-A2 expression, not only because it recruits transcriptional repressors, but also because it confers intron-mediated transcriptional enhancement. Genotypic analyses using wheat accessions indicated that the P1 locus is likely derived from a single natural mutation in tetraploid wheat, which was subsequently inherited by hexaploid T. petropavlovskyi. | 10 | Spatial structure and seasonal changes of nekton assemblages in the Taiwan Strait显示文摘The Taiwan Strait is a transition zone between the East China Sea and the South China Sea with unique hydrology and a geographical environment that creates special marine community features.To analyze the spatial structure and seasonal changes of the nekton assemblages in the Taiwan Strait,seasonal experimental trawl surveys were conducted during 2006–2007.The results showed that there were two assemblages in the area with different sets of species,and the average similarity within each group varies between 39.38% and 74.20%.By using multivariate statistical analysis and analyzing the distribution of dominant species,we found that the structures of the assemblages had obvious seasonal variation.The middle region from the Putian transect to the Xiamen transect could be considered a mixing area for the two assemblages.The analysis of the relationship between species assemblages and environmental factors indicated that temperature was the most important factor affecting the community structure in cold seasons,and 22.5°C and 17°C could be considered dividing lines for spring and winter,respectively.In warm seasons,the most important factor was water depth,but the relationship with depth was not as significant,with a correlation between 0.264 and 0.399.The seasonal changes of nekton assemblages basically reflected the dynamic currents in the Taiwan Strait.The south coastal assemblage extended to nearly the entire area of the Taiwan Strait along with a strong and intense warm current that shrinks in spring and winter when the southward intrusion of the cold Zhejiang-Fujian coastal current becomes stronger.The impact of shortterm and long-term environmental changes,such as extreme weather,global warming and human activity on nekton assemblages,had been recognized but need further research.Our study on nekton assemblages could be used as a baseline for measuring future changes. | SONG Puqing LIN Longshan LI Yuan ZHONG Zhihui | 2016 | Acta Oceanologica Sinica2016,35,6: | 3 |
