2Analysis of F_1 hybrid and BC_1 monosomic alien addition line plants from Brassica oleracea × Sinapis alba by GISH显示文摘Sterile and semi-fertile F_1 plants were obtained by intergeneric sexual hybridization between paternal Brassica oleracea var. alboglabra (genome CC, 2n=18) and maternal Sinapis alba (genome SS, 2n=24), BC_1 plants were obtained by backcrossing between paternal B. oleracea and ma-ternal semi-fertile F_1 plants. Genomic in situ hybridization (GISH) combined with dual-colour fluorescence in situ hybridization (dcFISH) showed that sterile F_1 plants contained 21 chromosomes consist-ing of one B. oleracea chromosome set and one S. alba chromosome set, belonging to expected hybrids, and semi-fertile F_1 plants contained 30 chromosomes consisting of two B. oleracea chromosome sets and one S. alba chromosome set. It is obvious that the semi-fertile F_1 plants belong to unexpected hybrids. 1―3 trivalents were detected at meiotic metaphase I of semi-fertile F_1 pollen mother cells (PMCs). Different separation ratios of S chromosomes were detected at anaphase I. A monosomic alien addition line (MAAL) was identified by GISH-dcFISH from BC_1 plants; it contained 19 chromosomes consisting of 18 C chromosomes and 1 S chromosome. At meiotic metaphase I, 9 divalents from B. oleracea and one univalent from S. alba could be detected. Sometimes, one putative C-S trivalent could also be detected. 3Sawdust discharge rate from aerated hoppers显示文摘This paper presents a numerical and experimental study of the discharge rate of sawdust from an aerated hopper as an important parameter in many industrial processes involving the handling of other granular materials. Numerical experiments are conducted by means of an Eulerian-Eulerian approach coupled with the kinetic theory of granular fiow(KTGF) . Emphasis is given to the effects of particle size,hopper outlet width,hopper half angle,aeration height and air fiow rate. The results show that the discharge rate is significantly affected by hopper outlet width,particle size and air fiow rate,but is not sensitive to the hopper half angle and aeration height:increasing hopper outlet width or air fiow rate increases dis charge rate,while increasing particle size decreases discharge rate. 5Development and Evaluation of a MAb-Based ELISA for Detection of Chlamy- dophila pneumoniae Infection with Variable Domain 2 and 3 of the Major Outer Membrane protein显示文摘Objective This paper aims to develop a monoclonal antibodies (MAbs)- based ELISA for detecting Chlamydophila pneumoniae (C. pneumoniae) antigens in humans with the variable domains (VD) 2 and 3 of the major outer membrane protein (MOMP VD2-VD3 ) and to assess its sensitivity and specificity by comparing with a widely used MAb that is able to recognize the elementary bodies of C. pneumoniae. Methods MOMP VD2-VD3 were overexpressed in Escherichia coli and purified by affinity chromatography. Mice were immunized with the recombinant antigen, and hybridomas secreting MAbs were screened. Three stable hybridomas clones were selected and named 5D6, 7G3, and 8C9. The MAbs-based ELISA was scrutinized for species-specific recognition with a number of human throat swab samples from Group I (156 patients with typical respiratory illness clinically confirmed before) and Group II (57 healthy donors). Results In Group I, 55 positive cases were detected by anti-EB MAb-based ELISA, 51 cases were positive by MAbs 5D6-based ELISA, and 33 and 38 cases were positive by MAb 8C9 and 7G3-based ELISA respectively. Of the 57 samples from Group II 'healthy donors', 5 were positive and 52 were negative with both anti-EB and 5D6-based tests, while 2 and 3 positive cases were identified by the other two MAb-based ELISAs respectively. 10Generation of Hematopoietic Stem Cells from Purified Embryonic Endothelial Cells by a Simple and Efficient Strategy显示文摘Recent progress by versatile approaches supports the new hypothesis that multi-potent hematopoietic stem cells(HSCs) are directly formed from a rare population of endothelial cells in mid-gestation mouse embryos.This process is therefore known as the endothelial-tohematopoietic transition(EHT).Nevertheless,there is no functional evidence that documents the HSC transition from purified endothelial cells.In this study,we developed an OP9-DL1-based co-culture system that was able to facilitate the HSC specification and/or expansion in vitro of mouse embryonic day 10.5(E10.5) Tie2^+ cells remarkably.Then,the immunophenotypically defined endothelial cells were harvested by a combination of surface markers(Flk1^+CD31^+CD41^-CD45^-Ter119^-) from the caudal half of E10.0-E11.0 mouse cmbryos.The transplantation of the endothelia/OP9-DL1 co-cultures led to long-term,high-level,multi-lineage,and multi-organ hematopoietic reconstitution in the irradiated adult recipients.The induced HSC activity was initially observed at E10.5,and a significant increase was detected at E11.0,which suggests a temporally specific regulation.Taken together,for the first time,we provide functional evidence showing the HSC potential of purified embryonic endothelial cells,which is indispensable for the emerging EHT concept. 14Targeted Micellar Phthalocyanine for Lymph Node Metastasis Homing and Photothermal Therapy in an Orthotopic Colorectal Tumor Model显示文摘Tumor lymph node(LN)metastasis seriously affects the treatment prognosis.Studies have shown that nanoparticles with size of sub-50 nm can directly penetrate into LN metastases after intravenous administration.Here,we speculate through introducing targeting capacity,the nanoparticle accumulation in LN metastases would be further enhanced for improved local treatment such as photothermal therapy.Trastuzumabtargeted micelles(<50 nm)were formulated using a unique surfactantstripping approach that yielded concentrated phthalocyanines with strong near-infrared absorption.Targeted micellar phthalocyanine(T-MP)was an effective photothermal transducer and ablated HT-29 cells in vitro.A HER2-expressing colorectal cancer cell line(HT-29)was used to establish an orthotopic mouse model that developed metastatic disease in mesenteric sentinel LN.T-MP accumulated more in the LN metastases compared to the micelles conjugated with control IgG.Following surgical resection of the primary tumor,minimally invasive photothermal treatment of the metastatic LN with T-MP,but not the control micelles,extended mouse survival.Our findings demonstrate for the first time that targeted small-sized nanoparticles have potential to enable superior paradigms for dealing with LN metastases.Hai‑Yi Feng Yihang Yuan Yunpeng Zhang Hai‑Jun Liu Xiao Dong Si‑Cong Yang Xue‑Liang Liu Xing Lai Mao‑Hua Zhu Jue Wang Qin Lu Quanjun Lin Hong‑Zhuan Chen Jonathan F.Lovell Peng Sun Chao Fang 2021Nano-Micro Letters2021,13,11:1
15Application of UAV in Road Safety in Intelligent Areas显示文摘With the continuous development of remote sensing(RS)technology,the surface information can be collected conveniently and quickly by using the popular unmanned aerial vehicle(UAV).The application of UAV low altitude RS technology in road safety in intelligent area has certain practical significance.It can provide safety warning for most drivers,and provide auxiliary decision-making for the road supervision department.Through the collection,processing,calculation and analysis of the road image,the UAV can find out the road obstacles with potential safety hazards,identify the road pit,calculate the radius and depth of the road pit through the digital mapping system,predict the accident risk according to different speed and provide scientific basis for the road safety monitoring.At the same time,UAV can provide repair scheme for damaged roads,estimate the quantity of materials needed for repair,and achieve the target of resource saving and efficiency improvement.The experimental results show that the UAV can not only provide scientific prediction information for driving safety,but also provide relatively accurate material consumption for road repair.Yanan Xu Jianxin Qin Pengcheng He Zhuan Chen 2019Journal of Geographical Research2019,2,4:0

172-D/2-D heterostructured biomimetic enzyme by interfacial assembling Mn_(3)(PO_(4))_(2) and MXene as a flexible platform for realtime sensitive sensing cell superoxide显示文摘It is critical for fabricating flexible biosensors with both high sensitivity and good selectivity to realize real-time monitoring superoxide anion(O_(2)^(·−)),a specific reactive oxygen species that plays critical roles in various biological processes.This work delicately designs a Mn_(3)(PO_(4))_(2)/MXene heterostructured biomimetic enzyme by assembling two-dimensional(2-D)Mn_(3)(PO_(4))_(2) nanosheets with biomimetic activity and 2-D MXene nanosheets with high conductivity and abundant functional groups.The 2-D nature of the two components with strong interfacial interaction synergistically enables the heterostructure an excellent flexibility with retained 100%of the response when to reach a bending angle up to 180°,and 96%of the response after 100 bending/relaxing cycles.It is found that the surface charge state of the heterostructure promotes the adsorption of O_(2)^(·−),while the high-energy active site improves electrochemical oxidation of O_(2)^(·−).The Mn_(3)(PO_(4))_(2)/MXene as a sensing platform towards O2•−achieves a high sensitivity of 64.93µA·µM^(−1)·cm^(−2),a wide detection range of 5.75 nM to 25.93µM,and a low detection limit of 1.63 nM.Finally,the flexible heterostructured sensing platform realizes real-time monitoring of O_(2)^(·−)in live cell assays,offering a promising flexible biosensor towards exploring various biological processes.Shen Fei Zhao Fang Xin Hu Zhuan Zhuan Shi Jing Jing Fu Yue Chen Fang Yin Dai Chun Xian Guo Chang Ming Li 2021Nano Research2021,14,3:0

19CD98-induced CD147 signaling stabilizes the Foxp3 protein to maintain tissue homeostasis显示文摘Regulatory T cell(Treg)stability is necessary for the proper control of immune activity and tissue homeostasis.However,it remains unclear whether Treg stability must be continually reinforced or is established during development under physiological conditions.Foxp3 has been characterized as a central mediator of the genetic program that governs Treg stability.Here,we demonstrate that to maintain Foxp3 protein expression,Tregs require cell-to-cell contact,which is mediated by the CD147-CD98 interaction.As Tregs are produced,CD147,which is expressed on their surface,is stimulated by CD98,which is widely expressed in the physiological environment.As a result,CD147’s intracellular domain binds to CDK2 and retains it near the membrane,leading to Foxp3 dephosphorylation and the prevention of Foxp3 degradation.In addition,the optimal distribution of Foxp3+Tregs under both pathological and physiological conditions depends on CD98 expression.Thus,our study provides direct evidence that Foxp3-dependent Treg stability is reinforced in the periphery by the interaction between CD147 and CD98 in the surrounding environment.More importantly,Tregs with high CD147 expression effectively inhibit inflammatory responses and maintain Foxp3 stability,which has guiding significance for the application of Tregs in immunotherapy.JieJie Geng Ruo Chen Feng-fan Yang Peng Lin Yu-meng Zhu Xianghui Fu Ke Wang Zhuan Feng Jiao Wu Hai Zhang Qi-jing Li Zhi-Nan Chen Ping Zhu 2021Cellular & Molecular Immunology2021,18,12:0
20Developmental Strategies of Betel Nut Industry in Hainan Province显示文摘The developmental status of the betel nut industry is introduced, including cultivation situation, processing and development situation, and consumer market situation. The result shows that the cultivation of betel nut in Hainan Province develops rapidly and the primary processing of betel nut has formed certain scale, but the deep processing of betel nut is backward. Except for the mino amount of the betel nuts consumed by Hainan Island, a large majority of the betel nuts are processed into dry fruit to sell to Hunan Province to reprocess. The problems exist in the development of betel nut industry in Hainan Province are analyzed, covering blind cultivation, extensive management, backward processing, lagged new product development and the single and concentrated consumer market. The strategic choice of developing the betel nut industry of Hainan Province is analyzed as well.
