| 1 | 晚期非小细胞肺癌免疫检查点抑制剂新型生物标志物:血液PD-L1动态检测显示文摘背景免疫检查点抑制剂(immune checkpoint inhibitors,ICIs)具有良好疗效已成为恶性肿瘤治疗领域备受瞩目的翘楚。然而,仅有小部分患者获得长期临床获益。如何选择生物标志物来筛查ICIs治疗优势人群仍是一大热点。方法收集51例晚期恶性肿瘤患者的肿瘤组织和配对血样本,进行PD-L1表达的相关性分析。40例晚期非小细胞肺癌(non-small cell lung cancer,NSCLC)患者接受ICIs治疗,2个月时检测血液PD-L1(bloodPD-L1,bPD-L1)表达的动态变化,包括PD-L1 mRNA、外泌体PD-L1(exosomalPD-L1,exoPD-L1)蛋白和可溶性PD-L1(solublePD-L1,sPD-L1)。应用R软件计算最佳截断值,用于预测最佳客观缓解(best objective response,b OR)、无进展生存期(progression-free survival,PFS)和总生存期(overall survival,OS)。结果51例不同恶性肿瘤患者中,组织PD-L1(tissuePD-L1,tPD-L1)表达阳性患者的PD-L1mRNA表达水平显著升高。40例接受ICIs治疗的晚期NSCLC患者中,bPD-L1 mRNA上调≥2.04的患者b OR、PFS和OS更佳;21例接受ICIs治疗的晚期NSCLC患者中,exoPD-L1上调≥1.86的患者临床获益更大。sPD-L1动态变化与ICIs疗效无显著相关性。PD-L1 mRNA和exoPD-L1联合检测更有利于预测ICIs治疗疗效。结论bPD-L1表达与tPD-L1表达存在正相关;晚期NSCLC患者ICIs治疗初期PD-L1 mRNA和(或)exoPD-L1表达升高,可作为评估临床获益的新型生物标志物。 | Qiao YANG Mingjing CHEN Jiaoyang GU Kai NIU Xianlan ZHAO Linpeng ZHENG Zihan XU Yongxin YU Feng LI Lingxin MENG Zhengtang CHEN Wenlei ZHUO Luping ZHANG Jianguo SUN 杨峤(译) 孙建国(译) | 2021 | 中国肺癌杂志2021,24,12: | 10 |
| 2 | Clinically relevant factors associated with quantitative optical coherence tomography angiography metrics in deep capillary plexus in patients with diabetes显示文摘Background:To test clinically relevant factors associated with quantitative artifact-free deep capillary plexus(DCP)metrics in patients with diabetes mellitus(DM).Methods:563 eligible eyes(221 with no diabetic retinopathy[DR],135 with mild DR,130 with moderate DR,and 77 with severe DR)from 334 subjects underwent optical coherence tomography-angiography(OCT-A)with a swept-source OCT(Triton DRI-OCT,Topcon,Inc.,Tokyo,Japan).Strict criteria were applied to exclude from analysis those DCP images with artifacts and of poor quality,including projection artifacts,motion artifacts,blurriness,signal loss,B-scan segmentation error,or low-quality score.A customized MATLAB program was then used to quantify DCP morphology from the artifact-free DCP images by calculating three metrics:foveal avascular zone(FAZ),vessel density(VD),and fractal dimension(FD).Results:166(29.5%)eyes were excluded after quality control,leaving in the analysis 397 eyes(170 with no DR,101 with mild DR,90 with moderate DR,36 with severe DR)from 250 subjects.In the multiple regression models,larger FAZ area was associated with more severe DR(β=0.687;p=0.037),shorter axial length(AL)(β=−0.171;p=0.003),thinner subfoveal choroid thickness(β=−0.122;p=0.031),and lower body mass index(BMI)(β=−0.090;p=0.047).Lower VD was associated with more severe DR(β=−0.842;p=0.001),shorter AL(β=0.107;p=0.039),and poorer visual acuity(VA)(β=−0.133;p=0.021).Lower FD was associated with more severe DR(β=−0.891;p<0.001)and with older age(β=−0.142;p=0.004).Conclusions:Quantitative artifact-free DCP metrics are associated with VA,DR severity,AL,subfoveal choroidal thickness,age,and BMI in diabetic patients.The effects of ocular and systemic factors should be considered for meaningful interpretations of DCP changes in DM patients. | Fang Yao Tang Erica O.Chan Zihan Sun Raymond Wong Jerry Lok Simon Szeto Jason C.Chan Alexander Lam Clement C.Tham Danny S.Ng Carol Y.Cheung | 2020 | Eye and Vision2020,7,1: | 7 |
| 3 | Architectural proteins for the formation and maintenance of the 3D genome显示文摘Eukaryotic genomes are densely packaged into hierarchical three-dimensional(3D) structures that contain information about gene regulation and many other biological processes. With the development of imaging and sequencing-based technologies, 3D genome studies have revealed that the high-order chromatin structure is composed of hierarchical levels, including chromosome territories, A/B compartments, topologically associated domains, and chromatin loops. However, how this chromatin architecture is formed and maintained is not completely clear. | 7 | KRAS mutation-induced EndMT of brain arteriovenous malformation is mediated through the TGF-β/BMP-SMAD4 pathway显示文摘Objective Somatic KRAS mutations have been identified in the majority of brain arteriovenous malformations(bAVMs),and subsequent in vivo experiments have confirmed that KRAS mutation in endothelial cells(ECs)causes AVMs in mouse and zebrafish models.Our previous study demonstrated that the KRASG12D mutant independently induced the endothelial-mesenchymal transition(EndMT),which was reversed by treatment with the lipid-lowering drug lovastatin.However,the underlying mechanisms of action were unclear.Methods We used human umbilical vein ECs(HUVECs)overexpressing the KRASG12D mutant for Western blotting,quantitative real-time PCR,and immunofluorescence and wound healing assays to evaluate the EndMT and determine the activation of downstream pathways.Knockdown of SMAD4 by RNA interference was performed to explore the role of SMAD4 in regulating the EndMT.BAVM ECs expressing the KRASG12D mutant were obtained to verify the SMAD4 function.Finally,we performed a coimmunoprecipitation assay to probe the mechanism by which lovastatin affects SMAD4.Results HUVECs infected with KRASG12D adenovirus underwent the EndMT.Transforming growth factor beta(TGF-β)and bone morphogenetic protein(BMP)signalling pathways were activated in the KRASG12D-mutant HUVECs and ECs in bAVM tissue.Knocking down SMAD4 expression in both KRASG12D-mutant HUVECs and ECs in bAVM tissues inhibited the EndMT.Lovastatin attenuated the EndMT by downregulating p-SMAD2/3,p-SMAD1/5 and acetylated SMAD4 expression in KRASG12D-mutant HUVECs.Conclusions Our findings suggest that the KRASG12D mutant induces the EndMT by activating the ERK-TGF-β/BMP-SMAD4 signalling pathway and that lovastatin inhibits the EndMT by suppressing TGF-β/BMP pathway activation and SMAD4 acetylation. | Hongyuan Xu Ran Huo Hao Li Yuming Jiao Jiancong Weng Jie Wang Zihan Yan Junze Zhang Shaozhi Zhao Qiheng He Yingfan Sun Shuo Wang Yong Cao | 2023 | Stroke & Vascular Neurology2023,8,3: | 1 |

| 10 | A likely autotetraploidization event shaped the Chinese mahogany(Toona sinensis)genome显示文摘Chinese mahogany(Toona sinensis) is of considerable medical and economic importance, and its genome has been deciphered. However, the process underlying its polyploidy is unclear, and the chromosomal evolutionary trajectory is poorly understood. Here, by reanalysing the T.sinensis genome, we found evidence of a tetraploidization event(T. sinensis special tetraploidization, TST) that occurred approximately 15-17 million years ago(MYA) after the core eudicot-common hexaploidization(ECH or gamma) event. We characterized the synonymous nucleotide substitution rates(Ks values) of collinear genes and found that T. sinensis genes affected by the TST evolve at a slower rate than Acer yangbiense genes. Furthermore, we identified homologous genes related to polyploidization and speciation and constructed multiple alignments with different reference genomes. Notably, the significant balance of gene retention and loss characterized in the two TST-derived subgenomes suggests an autopolyploid nature of the TST. Moreover, we deduced the chromosomal karyotypes of the two subgenomes and identified 7chromosomal fusions that have shaped the T. sinensis genome;more information is available on a newly constructed karyotype platform(http://gffzze84c5b42976d4cd7hwcv0fubkbu9n6n5x.ffgz.tsg.suse.edu.cn/Toona_sinensis/index.html). The T. sinensis genome preserves the ancestral chromosome structure of dicotyledons well and could serve as a good reference for understanding genomic changes in other Meliaceae and Sapindales plants. In addition, we verified that tandem duplication and the ECH have promoted the expansion of terpene synthase(TPS) genes;conversely, the TST seems to have inhibited expansion of these genes. | 11 | Two-dimensional ferromagnetic superlattices显示文摘Mechanically exfoliated two-dimensional ferromagnetic materials(2D FMs)possess long-range ferromagnetic order and topologically nontrivial skyrmions in few layers.However,because of the dimensionality effect,such few-layer systems usually exhibitmuch lower Curie temperature(TC)compared to their bulk counterparts.It is therefore of great interest to explore effective approaches to enhance their TC,particularly in wafer-scale for practical applications.Here,we report an interfacial proximity-induced high-TC 2D FM Fe3GeTe2(FGT)via A-type antiferromagnetic material CrSb(CS)which strongly couples to FGT.A superlattice structure of(FGT/CS)n,where n stands for the period of FGT/CS heterostructure,has been successfully produced with sharp interfaces by molecular-beam epitaxy on 2-inch wafers.By performing elemental specific X-ray magnetic circular dichroism(XMCD)measurements,we have unequivocally discovered that TC of 4-layer Fe3GeTe2 can be significantly enhanced from 140 K to 230 K because of the interfacial ferromagnetic coupling.Meanwhile,an inverse proximity effect occurs in the FGT/CS interface,driving the interfacial antiferromagnetic CrSb into a ferrimagnetic state as evidenced by double-switching behavior in hysteresis loops and the XMCD spectra.Density functional theory calculations show that the Fe-Te/Cr-Sb interface is strongly FMcoupled and doping of the spin-polarized electrons by the interfacial Cr layer gives rise to the TC enhancement of the Fe3GeTe2 films,in accordance with our XMCDmeasurements.Strikingly,by introducing rich Fe in a 4-layer FGT/CS superlattice,TC can be further enhanced to near room temperature.Our results provide a feasible approach for enhancing the magnetic order of few-layer 2D FMs in wafer-scale and render opportunities for realizing realistic ultra-thin spintronic devices. | Shanshan Liu Ke Yang Wenqing Liu Enze Zhang Zihan Li Xiaoqian Zhang Zhiming Liao Wen Zhang Jiabao Sun Yunkun Yang Han Gao Ce Huang Linfeng Ai Ping Kwan Johnny Wong Andrew Thye Shen Wee Alpha TN’Diaye Simon AMorton Xufeng Kou Jin Zou Yongbing Xu Hua Wu Faxian Xiu | 2020 | National Science Review2020,7,4: | 0 |

| 19 | Chemical Strain of Graphite-Based Anode during Lithiation and Delithiation at Various Temperatures显示文摘Electrochemical lithiation/delithiation of electrodes induces chemical strain cycling that causes fatigue and other harmful influences on lithium-ion batteries.In this work,a homemade in situ measurement device was used to characterize simultaneously chemical strain and nominal state of charge,especially residual chemical strain and residual nominal state of charge,in graphite-based electrodes at various temperatures.The measurements indicate that raising the testing temperature from 20℃ to 60℃ decreases the chemical strain at the same nominal state of charge during cycling,while residual chemical strain and residual nominal state of charge increase with the increase of temperature.Furthermore,a novel electrochemicalmechanical model is developed to evaluate quantitatively the chemical strain caused by a solid electrolyte interface(SEI)and the partial molar volume of Li in the SEI at different temperatures.The present study will definitely stimulate future investigations on the electro-chemo-mechanics coupling behaviors in lithium-ion batteries. | Zeyu Xu Xiuling Shi Xiaoqiang Zhuang Zihan Wang Sheng Sun Kaikai Li Tong-Yi Zhang | 2022 | Research2022,,1: | 0 |
| 20 | 2T1C DRAM based on semiconducting MoS_(2) and semimetallic graphene for in-memory computing显示文摘In-memory computing is an alternative method to effectively accelerate the massive data-computing tasks of artificial intelligence(AI)and break the memory wall.In this work,we propose a 2T1C DRAM structure for in-memory computing.It integrates a monolayer graphene transistor,a monolayer MoS_(2)transistor,and a capacitor in a two-transistor-onecapacitor(2T1C)configuration.In this structure,the storage node is in a similar position to that of one-transistor-one-capacitor(1T1C)dynamic random-access memory(DRAM),while an additional graphene transistor is used to accomplish the nondestructive readout of the stored information.Furthermore,the ultralow leakage current of the MoS_(2)transistor enables the storage of multi-level voltages on the capacitor with a long retention time.The stored charges can effectually tune the channel conductance of the graphene transistor due to its excellent linearity so that linear analog multiplication can be realized.Because of the almost unlimited cycling endurance of DRAM,our 2T1C DRAM has great potential for in situ training and recognition,which can significantly improve the recognition accuracy of neural networks. | Saifei Gou Yin Wang Xiangqi Dong Zihan Xu Xinyu Wang Qicheng Sun Yufeng Xie Peng Zhou Wenzhong Bao | 2023 | National Science Open2023,2,4: | 0 |
